  • The present study was conducted to examine metabolic risk factors and total antioxidant capacity (TAC) of Korean females living in Seoul and to investigate the relationship between the metabolic risk factors and serum TAC. A total of 353 females aged between 20 and 64 participated in the study. Obesity indicators, blood pressure, serum lipid profile and fasting blood glucose were measured as metabolic risk factors. Ferric reducing antioxidant power (FRAP) assay was employed to determine serum TAC of subjects. Obesity indicators such as body mass index, waist circumference and waist-hip ratio were significantly higher in the participants aged $\geq$ 50 y (older group) than in the participants aged 20-49 y (younger group) (p < 0.001). Blood pressure, serum total cholesterol (TC), triglyceride (TG) and fasting blood glucose were also significantly higher in the older group than in the younger group (p < 0.001), demonstrating significant positive correlations between age and MS risk factors. The association between FRAP and MS risk factors were also investigated. FRAP values showed significant positive correlations with age (p = 0.001), serum TG (p = 0.002) and TC (p = 0.03). A tendency of positive association between FRAP and waist circumference was observed without any significant difference (p = 0.06). Increased serum FRAP with central obesity and serum lipids may be interpreted as results of activation of antioxidant defense system against oxidative stress induced by metabolic syndrome (MS) constituent factors. However, to verify the function of FRAP as a potential biomarker of susceptibility to MS various contributors to the plasma antioxidant capacity and their biological relevance related to MS should be elucidated further.

폐경기 여성의 식이 지방산 및 혈청 인지질의 지방산 조성과 혈청 지질 농도의 관계 (Correlation of Dietary and Serum Phospholipid Fatty Acid Composition and Serum Lipid Levels in Postmenopausal Women)

  • The purpose of this study was to investigate the correlation of dietary and serum phospholipid fatty acids composition and serum lipid levels in postmenopausal women. Data about anthropometry, dietary intake, physical activities, serum lipid profile and serum phospholipid fatty acid composition were collected from eighty-five postmenopausal women. The subjects were classified as normocholesterolemia (NC), moderate hypercholesterolemia(MC) and phyperchollesterolemia(HC) according to their serum total cholesterol (TC) levels based on The Guideline for Korean Hyperlipidemia set/published by The Committee for Hyperlipidemia in Korea. The results were as follows. Total energy intake and the ratio of energy intake to energy expenditure were positively related, while dietary fiber intake was negatively related, with serum TC level. Dietary fat intake was positively related with serum LDL-C level. Dietary cholesterol, saturated fatty acid (SFA), monounsaturated fatty acid (MUFA) intake and the ratio of ingested saturated fat and cholesterol to calories (RISCC) were positively related, while the P/S ratio of dietary fat was negatively related, with serum TC and LDL-C levels. Serum phospholipid fatty acids composition was not significantly different among the three groups. SFA of serum phospholipid fatty acids was positively related, while P/S and M/S ratios of serum phospholipid fatiy acids were negatively correlated, with serum TC and LDL-C levels. We recommend that the dietary P/S ratio of postmenopausal women is increased by reducing their dietary SFA intake. With these changes in the diet, serum phospholipid fatty acid composition could be also changed, and serum lipids levels could be improved.

성인의 연령별 설탕 섭취상태와 혈중 지질과의 관련성 (Relation between Sugar Intake and Serum Lipids in Korean Adults according to Age)

  • The purpose of this study was to estimate sugar intake and relation between it and blood lipid profiles in Korean adult according to age. The 346 subjects were measured anthropometric assessment, dietary intake using 24-hour recall method, and serum lipid profiles. The average age of the subjects of 20-49, 50-64, over 65 years were 40.8, 57.0, 70.2 years, respectively. The mean height and weight were significantly decreased with increment of age(p<0.001, p<0.01). The mean energy, food, and sugar intakes of the age groups were decreased with increment of age(p<0.05, p<0.001, p<0.001), that is, 1649.8 ㎉/1253.6 g/36.1 g for 20~49 years, 1555.4 ㎉/1157.0 g/31.4 g for 50~64 years, 1404.4 ㎉/893.5 g/17.1 g for over 65 years. The major foods consumed sugar of the age groups were watermelon, sugar, ice cream for 20-49 years, watermelon, sugar, peach for 50-64 years, watermelon, sugar, kimchi for over 65 years. Carbohydrate intake of the subject adjusted age provided significantly negative correlation with serum HDL-cholesterol(p<0.05). However, there was not significant correlation between sugar intake and serum lipid profile. Based on these results, further studies on effects on blood lipid of sugar and carbohydrate intakes were needed for proper carbohydrate intake.

국내 침엽수재 추출성분의 혈액지질 저하 효과 연구 (Hypolipidemic Effects of Korean Softwood Components)

  • 본 연구는 한국에 널리 분포되어 있는 대표적인 침엽수중에 혈액지질에 영향을 미치는 성분이 있는지를 검색하는 것을 목적으로 시도되었다. 1차적으로 미국측백과 뚝향나무의 유기 용매와 수용성 추출물을 흰쥐에게 경구 투여하여 체중 증가, 장기 무게, 혈액지질성분 및 혈당의 변화를 관찰하였다. 추출물의 섭취는 체중증가와 간, 신장, 비장과 같은 장기무게에 유의적인 차이가 없어 일반적인 성장에는 영향을 주지 않는 것으로 보인다. 그러나 체내 저장지방은 뚝향나무의 유기용매층 섭취군에서 유의적으로 낮았다. 그리고 혈청내 총 지방량은 고지방 대조군이 저지방 대조군에 비해 유의적으로 높았으며, 뚝향나무의 유기용매층이 유의적으로 낮아 뚝 향나무의 유기용매층이 혈액 지질저하 효과가 있는 것으로 선정되었다. 2차적으로 뚝향나무의 유기용매층을 3분획하여 횐쥐를 대상으로 혈액지질성분과 혈당, 빈혈지표등을 관찰하였다. 결과를 보면 JCDM2가 체내 지방축적량을 유의적으로 낮추었으며, 혈청내 총 콜레스테롤과 중성지방량이 유의적이지는 않으나 낮은 경향을 보였다. 그리고 JCDM2의 50%이상을 차지하고 있는 물질로 communic acid가 순수 분리되었다. 본 연구는 아직 시도되지 않았던 수목의 혈액내 지질 저하효과를 관찰하는 기초연구로 실험 결과, 뚝향나무의 유기용매층에서 효과가 있었으며 그 주요 물질은 communic acid로 관찰되었다.

뽕잎 분말 첨가 두부 섭취가 충남 일부 지역에 거주하는 흡연 남자 성인의 손목 골밀도, 생화학 골대사 지표 및 혈청 지질 성상에 미친 영향 (Effects of Mulberry-Leaf Powder Tofu Consumption on Carpal Bone Mineral Density, Biochemical Bone Turnover Markers and Serum Lipid Profiles in Smoking Male Adults Living in Choongnam)

  • The effects of mulberry-leaf powder Tofu(MPT) on anthropometric measurements, including bone mineral density(BMD) in the right carpus, biochemical bone turnover markers, serum levels of lipids and macrominerals, were investigated in 30 smoking male adults who lived in Choongnam were given MPT(100 g/day) for 4 weeks. The average ages, number of smoked cigarettes and packyear were 22.38 years, 15.12/day and 3.54 years, respectively. The nutrient contents per 100 g MPT were 86.10 kcal energy, 8.98 g protein, 0.53 mg fiber, 211.33 mg Ca and 1.59 g fat. Anthropometric measurements, including dietary intake using the 24-hours recall method, carpal BMD using DEXA, serum levels of protein, albumin and glucose, lipid profiles (cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol) with Al(atherosclerosis index), HTR, CRF, LHBt, some biomarkers of BMD(serum alkaline phosphatase activity, osteocalcin, urinary DPD), and serum macrominerals(Ca, Ca/P ratio, Mg) and Pb were analyzed before and after consumption of MPT. After MPT consumption, dietary intakes of plant protein, total Ca and plant Ca increased significantly, but there were no significant differences in anthropometric measurements, BMD with bone metabolism markers, serum levels of protein, albumin or glucose, lipid profiles with AI, HTR, LHR and CRF.

Characterization of gender-specific bovine serum

  • Animal cell cultures generally require a nutrient-rich medium supplemented with animal serum. Adult bovine serum contains a variety of nutrients including inorganic minerals, vitamins, salts, proteins and lipids as well as growth factors that promote animal cell growth. To evaluate the potential use of gender-specific bovine serum (GSBS) for cell culture, the biochemical properties of male serum (MS), female serum (FS) and castrated-male serum (CMS) were investigated. Overall, the chemical profile of GSBS was similar to that of bovine references except for glucose, creatine kinase, lactate dehydrogenase and potassium. FS showed elevated total protein and sodium concentrations compared to MS and CMS. Proteins present in MS, FS and CMS but absent in fetal bovine serum (FBS) were selected by two-dimensional gel electrophoresis and identified by peptide mass fingerprinting. Some of the identified proteins are known to be involved in immune responses and the others have unknown physiological roles. Moreover, it was found that some proteins such as alpha-2-macroglobulin appeared to be gender-specific with higher contents in FS. Insulin and testosterone was significantly higher in MS, and $17{\beta}$-estradiol and estrone were higher in FS, as compared to the other sera. Taken together, the results indicate that each GSBS has a different ratio of components. Differences in serum constituents may affect cell cultures in a different manner and could be beneficial, depending on the specific aim of cell cultures.

Differential Expression Profiling of Salivary Exosomal microRNAs in a Single Case of Periodontitis - A Pilot Study

  • Exosomes are Nano-sized lipid vesicles secreted from mammalian cells containing diverse cellular materials such as proteins, lipids, and nucleotides. Multiple lines of evidence indicate that in saliva, exosomes and their contents such as microRNAs (miRNAs) mediate numerous cellular responses upon delivery to recipient cells. The objective of this study was to characterize the different expression profile of exosomal miRNAs in saliva samples, periodically isolated from a single periodontitis patient. Unstimulated saliva was collected from a single patient over time periods for managing periodontitis. MicroRNAs extracted from each phase were investigated for the expression of exosomal miRNAs. Salivary exosomal miRNAs were analyzed using Affymetrix miRNA arrays and prediction of target genes and pathways for its different expression performed using DIANA-mirPath, a web-based, computational tool. Following the delivery of miRNA mimics (hsa-miR-4487, -4532, and -7108-5p) into human gingival fibroblasts, the expression of pro-inflammatory cytokines and activation of the MAPK pathway were evaluated through RT-PCR and western blotting. In each phase, 13 and 43 miRNAs were found to be differently expressed $({\mid}FC{\mid}{\geq}2)$. Among these, hsa-miR-4487 $({\mid}FC{\mid}=9.292005)$ and has-miR-4532 $({\mid}FC{\mid}=18.322697)$ were highly up-regulated in the clinically severe phase, whereas hsa-miR-7108-5p $({\mid}FC{\mid}=12.20601)$ was strongly up-regulated in the clinically mild phase. In addition, the overexpression of miRNA mimics in human gingival fibroblasts resulted in a significant induction of IL-6 mRNA expression and p38 phosphorylation. The findings of this study established alterations in salivary exosomal miRNAs which are dependent on the severity of periodontitis and may act as potential candidates for the treatment of oral inflammatory diseases.

Determination of niacin profiles in some animal and plant based foods by high performance liquid chromatography: association with healthy nutrition

  • Vitamin $B_3$ (niacin) is essential for all living cells and plays a central role in energy metabolism and oxidative phosphorylation. Vitamin $B_3$, a water-soluble vitamin, is present in the form of nicotinic acid and nicotinamide, a monocarboxylic acid derivative of pyridine. While nicotinic acid is commonly effective in lowering cholesterol levels, unlike nicotinic acid, nicotinamide is ineffective on lipids. Presence rates of nicotinic acid and nicotinamide, which are the available forms of vitamin $B_3$, are different for each food. However, the studies in the literature are generally based on the analysis of total amount of vitamin $B_3$ in foods and the studies determining the profile of vitamin $B_3$ in foods are limited. The aim of the study was to determine the vitamin $B_3$ profiles of 10 kinds of animal based food and 10 different plant based food samples. In this study, 10 kinds of animal based food samples consisting of veal (veal steak fillet), chicken (breast), turkey meat (thigh), goat meat (leg, belly), lamb (leg, back, arm), mutton (belly), bovine meat (loin) and 10 different plant based food samples namely; barley, rye, wheat (bread), wheat (durum), oat, rice, dried pea, green lentil, red lentil and chickpea were studied by high performance liquid chromatography using post-column derivatization system. The presence rates of nicotinic acid and nicotinamide were determined in the meat samples as 30% and 70% and as 87% and 13% in the cereal and legume samples, respectively. Nicotinic acid levels were found in low amounts in the meat samples. The amounts of nicotinic acid in the cereal and legume samples were significantly higher than the meat samples. Consequently, the plant based foods such as cereals and legumes, with a ratio of 87% nicotinic acid presence, standout as the best source of nicotinic acid and encouraging regular intake of those cereals and legumes containing rich nicotinic acid would remove nicotinic acid deficiency in human.

Comparison of Lipid Profiles in Head and Brain Samples of Drosophila Melanogaster Using Electrospray Ionization Mass Spectrometry (ESI-MS)

  • Drosophila melanogaster (fruits fly) is a representative model system widely used in biological studies because its brain function and basic cellular processes are similar to human beings. The whole head of the fly is often used to obtain the key function in brain-related diseases like degenerative brain diseases; however the biomolecular distribution of the head may be slightly different from that of a brain. Herein, lipid profiles of the head and dissected brain samples of Drosophila were studied using electrospray ionization-mass spectrometry (ESI-MS). According to the sample types, the detection of phospholipid ions was suppressed by triacylglycerol (TAG), or the specific phospholipid signals that are absent in the mass spectrum were measured. The lipid distribution was found to be different in the wild-type and the microRNA-14 deficiency model ($miR-14{\Delta}^1$) with abnormal lipid metabolism. A few phospholipids were also profiled by comparison of the head and the brain in two fly model systems. The mass spectra showed that the phospholipid distributions in the $miR-14{\Delta}^1$ model and the wild-type were different, and principal component analysis revealed a correlation between some phospholipids (phosphatidylethanolamine (PE), phosphatidylinositol (PI), and phosphatidylserine (PS)) in $miR-14{\Delta}^1$. The overall results suggested that brain-related lipids should be profiled using fly samples after dissection for more accurate analysis.

Screening Moringa species focused on development of locally available sustainable nutritional supplements

  • BACKGROUND/OBJECTIVES: The leaves of Moringa oleifera (MO) and Moringa stenopetala (MS) commonly grown in Ethiopia possess potential nutritional and medicinal value. The aim of this study was to evaluate the nutritional and functional characteristics of the dried leaf powder from two Moringa species to develop sustainable nutritional supplements for Ethiopians from locally grown plant sources. MATERIALS/METHODS: Freshly harvested and air-dried MO and MS leaves were authenticated and the nutritional contents, such as protein, ash, lipids, and selected vitamins and minerals, were analyzed using standard analytical procedures. Amino acid compositions were also determined by an amino acid analyzer. Nine-week-old mice were randomly divided into four groups to investigate the anti-obesity effects of Moringa. The first group was fed a basal diet, the second group a high-fat diet, and the others were fed a high-fat diet containing 0.1% Moringa leaf powder from each species. After seven weeks, serum indices related to lipid profiles from each mouse were analyzed. RESULTS: The present study revealed high protein (28-29%) and ash (7-11%) contents. Glutamic acid, aspartic acid, proline, and leucine were the most abundantly found amino acids in both species. The predominant minerals in the leaf powder were calcium (826-1,530 mg/100 g), potassium (794-904 mg/100 g), and magnesium (286-431 mg/100 g). Pyridoxine (475.06 mg/100 g) and vitamin E (34.2 mg/100 g) were found only in MS. Niacin was found only in MO at 32.21 mg/100 g, whereas ascorbic acid was found in both species (3.89 and 6.19 mg/100 g dry weight for MO and MS, respectively). The results of the animal study showed that mice on a high-fat diet containing 0.1% MO leaf powder alleviated the elevation of cholesterol, triglycerides, and low-density lipoprotein cholesterol induced by the high fat diet. MO was more effective than MS in preventing hypercholesterolemia and fat deposition. CONCLUSION: The findings in this work confirmed that Moringa leaves of both MO and MS possessed high nutritional value but MO was better at preventing the harmful effects of the high-fat diet than MS.
