  • This study was performed to evaluate the effect of the herbal extract mixture of Cnidium officinale, Petasites japonicus and Coptis chinensis (CPC) on the lipids metabolism in mice with tyloxapol. ICR mice weighing between 30-40 g were divided into four groups: normal group, 600mg/kg tyloxapol injected group, $50\;{\mu}g/g$ CPC treated group 6h after tyloxapol injection (SAM1), and tyloxapol and CPC treated group (SAM2), respectively. Tyloxapol or CPC was injected intraperitoneally. Tyloxapol caused an elevation of total cholesterol (TC), triglycerides(TG), and LDL-cholesterol and a decrease of HDL-cholesterol. In addition, tyloxapol induced accumulation of lipid including cholesterol in both the liver and kidney. Serum levels of TC, TG and LDL-cholesterol were decreased whereas HDL-cholesterol was increased by CPC. CPC increased in HDL-cholesterol /total cholesterol ratio and lowered atherogenic index. The levels of TC, TG and LDL-cholesterol by CPC were rather lower in SAM2 than SAM1. CPC also inhibited lowering HDL-cholesterol by tyloxapol. CPC reduced lipid blots and cholesterol particles in both the deposition and size in the liver and kidney with tyloxapol. These results suggest that CPC might be expected to be beneficial for protection and treatment of hyperlipidemia by the disturbance of lipid metabolism.

  • The Galkun-Whanglyeon-Whanggum-Tang, an officially standardized mixture of traditional herbal medicines used in Korea and China, consists of Pueraria lobata, Scullellaria baicalensis, Coptis chinensis and Glycyrrhiza uralensis at a ratio of 6:9:3:2.4. In this study, we evaluated the effect of lowering lipid accumulation in blood by treatment of Galkun-Whanglyeon-Whanggum-Tang in Apo E(-/-) atherosclerotic animal model. ApoE/mice fed with 1.25% cholesterol, 7.5% cocoa butter and 0.5% sodium cholate diet were orally given vehicle or Galkun-Whanglyeon-Whanggum-Tang(10, 100 and 300 mg/kg/day) for 12 weeks. Serum levels of triglyceride(TG), total cholestrerol(TC), low-density lipoprotein(LDL) and high-density lipoprotein(HDL) were analyzed, and PPAR-${\alpha}$ and PPAR-${\gamma}$ were examined by Western blotting analysis. Galkun-Whanglyeon- Whanggum-Tang decreased serum levels of TG, TC and LDL, but not HDL in ApoE/mice. In parallel, Galkun-Whanglyeon-Whanggum-Tang treatment showed the increased activity of PPAR-${\alpha}$ and PPAR-${\gamma}$ in hepatocytes. In summary, Galkun-Whanglyeon-Whanggum-Tang can reduce lipid accumulation in blood, and this effect might be accompanied by the upregulation of PPAR-${\alpha}$ and PPAR-${\gamma}$ in Apo E(-/-) atherosclerotic mouse model.

  • To investigate the anti-obesity and cholesterol-lowering effects of germinated brown rice (GBR), male Sprague-Dawley rats were randomly divided into the following 4 groups and fed with high fat and cholesterol diets for 5 weeks; control group fed with experimental diet, rice group fed with diet containing 50% rice, brown rice group fed with diet containing 50% brown rice and GBR group fed with diet containing 50% GBR. All groups showed no significant difference in body weight, but the GBR group showed the lowest value in body weight, $471.2{\pm}17.8\;g$. Body weight gain and FER of GBR group, $310.6{\pm}14.7\;g$ and $41.8{\pm}2.0%$, respectively, were also lower than those of other groups. Blood triglyceride and cholesterol levels of the GBR group were $54.0{\pm}23.4\;mg/dL$ and $64.8{\pm}14.7\;mg/dL$, respectively, which were significantly lower than those of rice groups and were the lowest values among the experimental groups. The weights of epididymal and kidney fat of GBR group also showed the lowest values compared to other groups. The liver total lipid and total cholesterol of the GBR group, $216.3{\pm}35.7\;mg/g$ liver and $16.5{\pm}0.7\;mg/g$ liver, respectively, were lower than those of other groups, and total lipid, total cholesterol and triglyceride of GBR group showed the same results. These results suggest that GBR diet reduces body weight and fat gain, and has cholesterol-lowering effect.

  • This study aims to investigate the changes in the lipid composition and angiotensin-converting enzyme activity in serum of rats exposed to microwave radiation, and to examine improving effects of green tea catechin to the lipid composition. The microwave-exposed rats received the normal and one of 3 diets: catechin free (MW-0C), 0.25% catechin (MW-0.25C) or 0.5% catechin (MW-0.5C). Rats were sacrificed 6th day after microwave radiation (2.45 ㎓, 15 min). The concentration of serum triglyceridein MW-0C group was increased by 85%, compared with the normal group, but that of MW-0.25C group with 0.25% catechin supplementation was to 17% lower, compared with the MW-0C group. There was no significant difference between normal group and MW-0.5C group. Total-cholesterol and LDL-cholesterol concentrations were increased by 21.4% and 38.6%, respectively, by microwave irradiation. The concentration of HDL-cholesterol in MW-0C group was lower to 29.3%, but it was maintained at the normal level by catechin supplementation. There was no significant difference among four groups in HLD-cholesterol/LDL-cholesterol. Atherogenic index in MW-0C group was increased by 56%, compared with the normal group. Angiotensin-converting enzyme (ACE) activity in lung tissue of MW-0C group was increased by 59 %, while that of MW -0.25C and MW -0.5C group were maintained at the normal level. ACE activity of MW-0C group in serum was increased by 122.8%, compared with the normal group. Catechin supplementation group was significantly reduced, compared with the MW-0C group ACE activity. In conclusion, microwave irradiation increased the serum triglyceride and cholesterol concentrations and ACE activity which are considered as the blood pressure increasing agents. However, catechin supplementation decreased the level of triglyceride, total cholesterol, LDL cholesterol, and ACE activity, which maybe consider catechin as being agent of lowering effect for blood lipid profile for athero-genesis.

  • This study was performed to evaluate the effect of mushrooms powder (Lentinus edodes, Ganoderma lucidum, Pleurotus ostreatus; 5:3:2, w/w/w) of on the lipid concentration in female Sprague-Dawley rats for 4 weeks. Experimental groups were divided into two dietary groups, the cholesterol diet (Cholesterol group) and the cholesterol diet supplemented mixed mushrooms powder (Mushroom group). The concentration of total cholesterol in serum was significantly decreased by 57.4% in mushroom group compared to cholesterol group. The concentration of HDL-cholesterol was significantly increased by 230% in mushroom group compared to cholesterol group. At the same time, atherogenic index was also significantly decreased by 68.4% in mushroom group compared to cholesterol group. The concentration of triglyceride in liver was significantly increased by 50% in mushroom group compared to cholesterol group. However, the concentrations of triglyceride and phospholipid in serum and cholesterol and phospholipid in liver had no significant difference both groups. This study suggested that mixed mushroom powders exert a cholesterole-lowering effect in hyperlipidemic female rats.

  • We evaluated the effects of newly developed functional food containing Bacillus polyfermenticus SCD as the main material on the lipid and antioxidant metabolisms of hypercholesterolemic rats. Thirty male SD rats were divided into three groups after a 1-week adaptation period and were fed with a high fat-cholesterol diet (control), or with a high fat-cholesterol diet supplemented with low or high doses ($3.1\;{\times}\;10^6\;cfu/day$ or $3.1\;{\times}\;10^8\;cfu/day$) of B. polyfermenticus SCD and other physiological active materials for 6 weeks. Both doses of B. polyfermenticus SCD significantly reduced hepatic total cholesterol and triglycerides, while increasing the fecal excretion rates of total lipid, total cholesterol and triglycerides. B. polyfermenticus SCD increased the total radical trapping antioxidant potential (TRAP). The erythrocytic glutathione peroxidase activity in the B. polyfermenticus groups was significantly lower than that in the control group. Plasma TRAP levels exhibited a highly significant negative correlation with hepatic total cholesterol while a significant positive correlation was detected between fecal total cholesterol and plasma TRAP. This hypolipidemic and antioxidative effect of B. polyfermenticus SCD seemed to be unrelated to its dosage. These results suggest that functional food containing B. polyfermenticus SCD can improve oxidative stress and hepatic lipid profiles by enhancing the excretion of cholesterol and triglycerides in feces of rats fed with high fat-high cholesterol diet.

  • We previously showed that a methanol extract of Paeoniae radix decreased total cholesterol level in rats with hyperlipidemia. In order to isolate the active ingredient(s), the methanol extract of Paeoniae radix was fractionated with chloroform/methanol(4:1) solution and isolate into soluble part and insoluble part of the the methanol extract. Above two parts were tested on the experimentally induced hypercholesterolemia in rats for the lowering effect of serum lipoprotein contents. Hyperlipidemia was induced on male Wistar rats by feeding high choleserol diet for 7 days. After oral administration of above samples for 4 weeks, serum lipid profile was verified on these rats by measuring total cholesterol, triglyceride, high density lipoprotein cholesterol and low density lipoprotein cholesterol. The chloroform/methanol(4:1) soluble part and insolule part showed lowering activity of total cholesterol level and triglyceride level at 4 week point significantly(p<0.01 and p<0.05) compare with the control group and the soluble part was more effective.

  • This study was performed to investigate the effects of dried powder and juice of Aster scaber on lipid metabolism and antioxidative capacity in rats. Twenty one male Sprague-Dawley rats weighing 170.9$\pm$15.1g were blocked into 3 groups according to body weight and raised for 4 weeks with diets containing 5%(w/w) dried powder and juice from the same amount of Aster scaber. Food intake, body weight gain, food efficiency ratio and organ weights were not different among all the experimental groups. On lipid metabolism, both dried powder and juice supplementation decreased plasma total lipid, triglyceride(TG), total cholesterol and liver cholesterol levels by increasing fecal lipid excretions. Plasma thiobarbituric acid reactive substance(TBARS) concentrations were significantly decreased in dried powder and juice groups, whereas liver TBARS concentrations were not affected by experimental diets. Red blood cell(RBC) superoxide dismutase(SOD) activities were significantly increased by dried powder and juice supplementation, while activities of other antioxidative enzymes in RBC and liver were decreased or showed no changes. In conclusion, both Aster scaber dried powder and juice had the effects on lowering lipid levels by increasing fecal lipid excretions, and inhibiting of plasma lipid peroxidation in animals and the effects of juice were higher than that of dried powder. • This study was performed to investigate the effects of different fat diets on the serum lipid composition and on the morphology of liver tissue of the spontaneously hypertensive rats(SHR), Rats were fed for 17 weeks one of four fat diets, control(corn oil : beef tallow=1:1), beef tallow, grapeseed oil and soybean oil. Blood was withdrawn by heart puncture from each SHR, and used to determine the serum levels of total cholesterol, HDL-cholesterol, trigly ceride and phospholipid. No significant difference in body weight was observed in all diet groups. the total serum cholesterol was significantly lower in SHR fed grapeseed oil than that of other groups. The cholesterol lowering effect of grapeseed oil seemed to results from its high contents of phytosterol and linoleic acid. Also, serum HDL-cholesterol, triglyceride and phospholipid contents were low in all the groups. photochemical microscopic observations of the liver tissue revealed the congestion of sinosoid which is regarded as the characteristics of SHR, but no significant difference was observed among the groups.
