The dietary fiber alginic acid has no nutritional value; however, it decreases the utilization of nutrients by the body and can prevent obesity, hyperlipidemia, arteriosclerosis, constipation, and colon cancer. The low-molecular-weight alginic acid polymannuronate improves serum and liver lipid metabolism by decreasing cholesterol levels in high-cholesterol groups and may be used to control high blood pressure. Previously, we showed that polymannuronate, a physiologically active agent from seaweed, has a lipid-lowering effect and preventative role in colon cancer. In addition, the differentiation of polymannuronate-treated adipocytes was inhibited, triggering decreased leptin expression. This study examined ways to increase dietary satisfaction with and improve the nutritional quality of polymannuronate using hamburger buns supplemented with polymannuronate. Twenty male Sprague-Dawley rats were divided into two groups: the control group was fed hamburger buns containing 6% gluten, while the experimental group was fed hamburger buns containing 6% gluten and 10% polymannuronate. The serum triglyceride, phospholipid, and total and free cholesterol levels of the rats in the experimental group were decreased compared with those of the controls. The serum aspartate aminotransferase activity levels did not differ between the two groups. This study demonstrates that polymannuronate has beneficial effects on lipid metabolism and may be commercially useful.
In this study, we demonstrated the in vitro and in vivo formation of carcinogen-lipid adduct and its correlation with DNA or protein adducts. The lipids from serum or hepatocyte membranes of Spragu-Dawley rats. human serum, and standard major lipids were in vitro reacted with benzo[a]pyrene(BP) and BP metabolites. 7,8-Dihydroxy-9,10-epoxy-7,8,9,10-tetrahydrobenzo[a]-pyrene(BPDE-I), an ultimate carcinogenic form of BP, was covalently bound to triglyceride(TG). BPDE-I-TG adducts isolated by thin-layer chromatography (TLC) were further detected by high performance liquid chromatography(HPLC). TGs, including triolein, tripalmitin and tristearin, showed positive reactions with BPDE-I. However, cholesterol, phospholipids(Phosphatidylcholine, phosphatidyl-ethanolamine, phosphatidyl-inositol and sphingomyelin) and nonesterified fatty acids(palmitic acid, oleic acid, linoleic acid and stearic acid) did not react with BPDE-I. In addition, other BP metabolites (BP-phenols and -diols) did not react with TG, which TG appeared to be the most reactive lipid yet studied with respect to its ability to form an adduct with BPDE-I. There was a clear-cut dose-respect to its ability to form an adduct with BPDE-I-lipid adduct in vitro between TG and [1,3-3H]BPDE-I. In an animal study, BPDE-I-TG was also formed in the serum of rats orally treated with BP(25 mg/rat). Also, obvious correlations between [3H]BP related-biomolecule adducts (DNA, protein) or lipid damage and the BPDE-I-TG adduct were obtained in various tissues of mice i.p. treated with [3H]BP. These data suggest that TG can form an adduct with BPDE-I, as do other macromolecules (DNA, RNA, and protein). Therefore, a carcinogen-lipid adduct would be a useful biomarker for chemical carcinogenesis research and cancer risk assessment.
The purpose of this study is to verify the relation between vegetarian diet and the risk factors of cardiovascular disease. To do the present study the vegetarians were selected;245 Buddhist nuns(age: 23-79yrs). For control subjects, 235 healthy female adults(age: 23-79yrs) were selected. They were the teachers, the nurses and the housekeepers living in Chinju Gyeongsang Namdoo Province. Study period was from October 1996 to February 1997. The contents are consist of survey, anthropometric measurement, and clinical examination. The average ages of the subjects were 44.20yrs for vegetarians and 40.52yrs for non-vegetarians respectively. Average body mass indice(BMI) of vegetarians and non-vegetarians were 22.47 and 21.08, WHR was 0.85 and 0.84, percentage of body fat was 28.79 and 26.55 and the average duration of vegetarian diet of the vegetarians was 13.16 years. On the nutrient related lipid, fat, the energy ratio of fat, saturated fatty acid, total fatty acid, monounsaturated fatty acid and total-cholesterol were significantly lower(p<0.01) but the ratio of p/s was significantly higher(p<0.01) in vegetarians than non-vegetarians. The vegetarians were significantly higher than the non-vegetarians in intake of fiber, vitamin C. In vegetarians, variables that significant negative correlation with fiber were total-cholesterol, AI, blood sugar. In non-vegetarians, variables that significant negative correlation with fiber were TG, total-cholesterol, blood sugar, systolic blood pressure. In both subjects, the serum lipid concentration and the blood pressure were getting lower with fiber intakes. Consequently, vegetable diet can be considerably effective in making the level of the risk factors causing in cardiovascular disease lower. (Korean J Nutrition 34(3) : 313∼321, 2001)
Hyperlipidemia has been treated as one of the most important etiological cause factor in 21th century. The cortex and root of Acanthopanax sessiliflorus (AR), a herbal medicine, have been used for several diseases including cardiovascular diseases in Oriental countries. Recently, we reported that AR has anti-hyperlipidemic action. BS-01 was made using extract from AR. For these reasons, we investigated the effects of BS-01 as anti-hyperlipidemic drug through measurement of body weights, cholesterol levels, total lipid, phospholipid in serum. In our results, mice induced by high fat diet elevated body weight level compared to naive group. And total lipid in serum was also elevated by induction of hyperlipidemia. In BS-01 group, body weight of mice was lowered significantly compared to that of control group. Oral administration of BS-01 decreased total cholesterol and triglyceride level back to that of naive mice. HDL and LDL cholesterol levels were not affected by BS-01. In addition, total lipid level, which elevated by induction of hyperlipidemia was also lowered by oral administration of BS-01. Finally, free fatty acid level was lowered in BS-01 group. These results demonstrate that BS-01 lowered body weight and titers involved in hyperlipdemia such as total cholesterol, triglyceride, free fatty acid and total lipid. In these results, we demonstrate that BS-01 has anti-hyperlipidemic action.
The effect of dietary zinc deficiency and age on lipid peroxide level was investigaed in rats. Zinc level in serum and liver were also measured. Fifty Sprague-Dawly male rats aging 8 months(older rats) and 2 months(younger rats) were used as experimental animal. Zinc deficient diet(1.1ppm) and normal zinc diet(36.5ppm) were used as experimental diets. Rats in each age group were divided into zinc deficient(ZnDF), zinc pair-fed(ZnPF) and zinc ad-libitum(ZnAL) to remove the variances of food intake. After 4 weeks of experimetal period, rats were sacrificed. Thiobarbituric acid reactive substance(TBARS) levels in plasma and liver, lipofuscin and conjugated diene levels in liver were measured as lipid peroxide index. Food intakes of all groups were not different because zinc deficiency did not reduce food intake in ZnDF group. Younger rats gained weight continuously, while older rats lost weight in the begining of experiment and regained afterwards. In older rats, serum zinc level was decreaed while plasma TBARS. level was increased in ZnDF group. In younger rats, plasma TBARS concentration was increased in dietary zinc deficient rats although serum zinc concentration was not reduced. Liver zinc concentration was significantly higher in older rats comparing to younger rats. However, there was no difference among the three dietary groups. Liver TBARS level was not different by age or dietary zinc level. However it was tended to be higher in older rats. However there was no difference by the dietary zinc level. In both age groups, ZnDF group significantly increased plasma TBARS levels, which suggested dietary zinc deficiency could increase lipid peroxidation in part. Significantly higher levels of lipofuscin and conjugated diene in older rats suggested lipid peroxidation was accelerated by aging.
■ Objectives This study was designed to investigate the effects of Ephedra Herba hexane fraction on lipid levels in serum and lipid accumulation in liver tissue in Hyperlipidemic mice. ■ Methods Hyperlipidemia was induced by providing high fat diet for 4 weeks. Normal group was provided with normal diet. CTL groupwas provided with high fat diet. Ephedra Herba hexane fraction group was provieded with high fat diet and administered orally in the concentration of 1.5mg/kg body weight/day for 2 weeks. In this experiment, effects on total cholesterol, HDL-cholesterol, triglyceride, AST, ALT, fasting blood glucose in serum were measured. In addition histopathological changes in liver tissue were also observed. ■ Results Ephedra Herba hexane fraction did not affects weight gain, serum AST and ALT in hyperlipidemic mice. Oral administration of Ephedra Herba hexane fraction lowered levels of total cholesterol and triglyceride, which were elevated by induction of hyperlipidemia. In addition, Ephedra Herba hexane fraction group showed downward tendency of lipid accumulation compared with CTL group. Finally, administration of Ephedra Herba hexane fraction lowered fasting blood glucose significantly. And Ephedra Herba hexane fraction also ameliorates anti-oxidative stress systems in internal organs which play key role in disease prevention. ■ Conclusion These results suggest that Ephedra Herba hexane fraction can prevent lipid accumulation in liver tissue through regulation of dyslipidemia and hyperglycaemia.
Objective:The Herbal Remedy for Diabetes Mellitus-01 (HRDM-01) is composed of 11 species of medicinal plants. HRDM-01 is used as anti-hyperglycaemic agent. Anti-hyperglycaemic agents in western medicine often have hepatotoxicities. Therefore, we investigated safety of HRDM-01, especially in liver functions. Methods:We investigated the effects of HRDM-01 on liver function measuring serum AST, ALT and LDH levels and histopathological changes of liver tissue using photomicroscope. In addition, we also investigated the effects on serum lipid levels such as total cholesterol, HDL-cholesterol and triglyceride. Results:In our experiment, single injection of streptozotocin elevated levels of AST and ALT in serum. But LDH level was not affected. In addition, our animal model showed elevated levels of total cholesterol and triglyceride in serum. 30 days treatment with HRDM-01 lowered serum AST level. Serum levels of ALT and LDH did not affected. In addition, HRDM-01 lowered serum triglyceride level significantly. In histopathological observation, we did not find any abnormal changes in all experimental groups. Conclusions:Briefly, HRDM-01 did not elevate serum levels of ALT, LDH, total cholesterol and did not affect histopathological changes in liver tissues. Moreover, HRDM-01 lowered serum AST, triglyceride, which are elevated by induction of diabetes mellitus. These results suggest the safety of HRDM-01 in diabetic treatment.
This study was undertaken to investigate the relationship between the life style and the nutritional status of serum antioxidant vitamins and lipids in university male and female students. 48 male and 49 female students attending Andong university, aged between 18 and 25 years, were selected. Questions about the life styles including dietary intakes, food habits, smoking, drinking alcohol, exercise, stress were answered. And serum levels of antioxidant vitamins and lipids were determined. Average serum levels of total cholesterol, LDL C, HDL C, and triglyceride in male and female subjects were 158.6$\pm$32.7, 177.3$\pm$33.8; 86.4$\pm$26.0, 109.0$\pm$31.2; 46.0$\pm$10.7, 49.9$\pm$12.4; 131.2$\pm$22.5, 91.7$\pm$ 38.6mg/dl respectively. Average serum levels of antioxidant vitamin A, E and C in male and female subjects were 42.6$\pm$12.3, 31.4$\pm$9.8 g/dl, 1.11$\pm$0.38, 1.15$\pm$0.29mg/dl and 164.66 $\pm$65.01, 220.06$\pm$80.11 g/dl respectively. There was no significant difference between smoking habits and either serum lipids or antioxidant vitamins level. The serum vitamin C level of drinkers was significantly lower(p=0.038), but serum lipids(total cholesterol, LDL C, and triglyceride) were higher than non alcoholic subjects. The subjects with severe stress had lower in HDL C and higher in atherogenic index than others. This result indicates that oxidative stress may be increased in stressful environment from undesirable life styles and influence the status of serum lipid and antioxidant vitamins.
Objectives : This study was designed to investigate protective effects of Sayeoksanhap-Pyeongweisan-Gamiban(SPG) on hematological changes in rats damaged by CCl4 Methods : We investigated the effects of SPG on hematological changes in terms of AST/ALT levels, Total cholesterol, HDL-cholesterol and triglyceride in serum and lipid peroxidation in rat liver. Results : Administration of SPG decreased AST level in serum effectively. In addition, lipid peroxidation in liver tissue, which was elevated by injection of CCl4, was decreased by administration of SPG. In contrast, ALT level, Total cholesterol, HDL-cholesterol and triglyceride in serum were not affected by administration of SPG. Conclusion : These results suggest that oral administration of SPG was useful to protect liver against CCl4 by its antioxidant action in liver tissues.
The purpose of this study was to investigate the effect of dietary Morus alba L.(Bong-ip, B), Glycyrrhizae glabra(Gam-chei, C), Pinus densiflora(Sol-lp, S) and Angelica gigas(Dang-gi, D)powder on serum composition in rats(Sprague-Dawley male rats, 100-110g). Serum TG(triglyceride, p<0.01), total cholesterol, glucose, total protein, albumin, GGT$({\gamma}-glutamyl$ transferase, p<0.05) were significantly increased D group than that of nomal and other groups, but UA(uric acid, p<0.05) was significantly decreased, and C group(p<0.05) was significantly increased. but C group of urine(p<0.05) was significantly decreased. Also, B and S groups(p<0.05) of BUN(blood urea nitrogen), S group(p<0.05) of ALP(alkaline phosphokinase, Band C(p<0.05) of CPK(creatinine phosphokinsae, p<0.05) were significantly increased. B, S and C groups were better than D group for lipid metabolism, and pretection to liver. Also, B and C groups of glucose were same as normal diet, so Morus alba L. was good food for lipid metabolism and hypoglycemic effect.
