Effects of Ukgan-san on the L-thyroxine-induced Rat Hyperthyroidism (억간산이 갑상선기능항진 유발 백서에 미치는 영향)

  • This study was to elucidate the effects of Ukgan-san on the hyperthyroidism induced by sodium levothyroxine. Hyperthyroidisms were induced by continuous subcutaneous treatment of LT4, 0.3 mg/kg, once a day for 27days, and 1,500, 1,000 and 500 mg/kg of Ulkansan extracts were orally administered, once a day for 15 days from 12th LT4 treatment, and the changes on the body, thyroid gland, liver and epididymal fat pad weights, serum triiodothyronine(T3), thyroxine(T4), thyroid-stimulating hormone(TSH), asparte aminotransferase (AST) and alanine aminotransferase (ALT) concentrations, hepatic lipid peroxidation(LPO), glutathione(GSH), superoxide dismutase(SOD) and catalase(CAT) activities were investigated with throid gland, liver and epididymal fat pad histopathological changes. The effects of Ukan-san extracts were compared with that of propyl thiouracil(PTU), a standard antithyroidic drug 10 mg/kg(intraperitoneally). 1,500 and 1,000 mg/kg of Ukan-san extracts reversed all LT4-induced hyperthyroidisms and these effects indicating their potential in the regulation of hyperthyroidism. Further, the Ukan-san extract normalized LT4-induced liver oxidative stresses, and also reduced liver damages and epididymal fat pad reducements suggesting its antioxidative and relative organ protective nature. However, nor favorable effects on LT4-induced hyperthyroidisms were detected in Ulkansan 500 mg/kg treated rats as compared with LT4 control rats in the present study. These effects of Ukan-san may help improvement of hyperthyroidisms and accompanied various organ damages.

The Effect of Cimicifuga heracleifolia on Carben Tetrachloride-induced Hepatotoxicity in Rats (승마추출물이 흰쥐의 사염화탄소 유발 간독성에 미치는 효과)

  • The effect of Cimicifuga heracleifolia on CC1$_4$-induced hepatotoxicity was investigated. Cimicifuga heracleifolia has been used to diaphoresis, antipyretics and detoxification in oriental remedy. We examined the effect of Cimicifuga heracleifolia methanol extract by blood chemical analysis and histopathologic examination. ALT and AST were decreased by 38% and 67% in pretreatment group of Cimicifuga heracleifolia compared to CC1$_4$control group respectively. There were significant changes neither in total protein, albumin, triglyceride and cholesterol nor in BUN and creatinine. In histopathologic examination, there were severe necrosis and hemorrhage with infiltration of inflammatory cells around the central vein, zone 3 in the liver of CC1$_4$ treated rat. Ballooning degeneration of hepatocytes were frequently noted in the periphery of the hemorrhagic necrosis. In Cimicifuga heracleifolia pretreatment group, we observed mild degree of ballooning degeneration and inflammatory cell infiltration. No gross necrosis was observed. We measured malondialdehyde (MDA) formation by TBA method. It showed that the formation of MDA in Cimicifuga heracleifolia pretreatment group was decreased compared to the $CCl_4$control group. We got the result of the effect of Cimicifuga heracleifolia on CC1$_4$-induced hepatotoxicity by decreasing serum ALT and AST. It seems that the decrease of lipid peroxidation is related to the recovery effect.

Protective Effects of Chlorogenic Acid against Experimental Reflux Esophagitis in Rats

  • Esophageal reflux of gastric contents causes esophageal mucosal damage and inflammation. Recent studies show that oxygen-derived free radicals mediate mucosal damage in reflux esophagitis (RE). Chlorogenic acid (CGA), an ester of caffeic acid and quinic acid, is one of the most abundant polyphenols in the human diet and possesses anti-inflammatory, antibacterial and anti-oxidant activities. In this context, we investigated the effects of CGA against experimental RE in rats. RE was produced by ligating the transitional region between the forestomach and the glandular portion and covering the duodenum near the pylorus ring with a small piece of catheter. CGA (10, 30 and 100 mg/kg) and omeprazole (positive control, 10 mg/kg) were administered orally 48 h after the RE operation for 12 days. CGA reduced the severity of esophageal lesions, and this beneficial effect was confirmed by histopathological observations. CGA reduced esophageal lipid peroxidation and increased the reduced glutathione/oxidized glutathione ratio. CGA attenuated increases in the serum level of tumor necrosis factor-${\alpha}$, and expressions of inducible nitric oxide synthase and cyclooxygenase-2 protein. CGA alleviates RE-induced mucosal injury, and this protection is associated with reduced oxidative stress and the anti-inflammatory properties of CGA.

  • Metabolic syndrome such as, hypertension, diabetes, obesity, dyslipidemia etc are well known lifestyle diseases which threaten a health state. The purpose of this study is to evaluate antioxidant status and reactive oxygen species (ROS) in various stressful conditions. A group of persons (n=28) were enrolled in this study and stress status was evaluated using questionnaire of psychological well-being index (PWI-SF) and hematologic analysis of free oxygen radical defense (FORD), free oxygen radical test (FORT), lipid profiles and serum cortisol level. Analysis was done according to stress index, BMI status and overtime working. Blood samples from diabetics were used for control. Results are as in followings: Higher PWI-SF group showed higher FORT. Obese persons with BMI > $25kg/m^2$ showed statistically lower FORD (p<0.05) and higher FORT level (p<0.01). When compare with diabetics, an overweight group had similar levels in FORD and FORT while a normal body weight group showed quite different with diabetics in FORD (p<0.001). Higher ROS group has higher triglyceride, CRP and insulin levels but lower in FORD and HDL-cholesterol levels. Overtime working at night showed no meaningful result against our expectations. These results suggested that obesity showed most unfavorable correlations in an antioxidant status in various situations.

Effects of Dietary Nutrient Content, Feeding Period, and Feed Allowance on Juvenile Olive Flounder Paralichthys olivaceus at Different Feeding Period and Ration

  • We examined the effects of dietary nutrient content, feeding period, and feed allowance on compensatory growth, food use, chemical composition, and serum chemistry of juvenile olive flounder Paralichthys olivaceus. We placed 720 juvenile fish into 24 400-L flow-through round tanks (30 fish per tank).A $2{\time}2{\time}2$ factorial design (diet: control (C) and high protein and lipid (HPL) ${\time}$ feeding period: 8 and 6 weeks ${\time}$ feed allowance: 100% and 90% of satiation) was applied. Fish were hand-fed twice daily, based on the designated feeding schedule. Weight gain and food consumption were affected by both the feeding period and feed allowance, but not by diet. The food efficiency ratio was not affected by diet, feeding period, or feed allowance, but the protein efficiency ratio and protein retention were affected by diet and feeding period, respectively. We found that the full compensatory growth of fish was not achieved at a restricted feeding allowance.

Effect of InJinORyungSan on the nephrotoxicity in rat (인진오령산이 흰쥐의 신독성(腎毒性)에 미치는 영향(影響))

  • This study investigated the effect of InJinORyungSan on the nephrotoxicity in rat treated with cyclosporin A. Control group were injected with cyclosporin A alone. whereas test group were injected with cyclosporin A and InJinORyungSan extract. In the control group, blood urea nitrogen(BUN), serum creatinine(S-Cr) and renal lipid peroxidation(LPO) level were significantly increased, but renal superoxide dismutase(SOD) activity was significantly decreased. In the kidney of control group, the destruction of distal convoluted tubules(DCT) and proximal convoluted tubules(PCT) were observed in renal cortex, lymphocytes and fibroblast were appeared in the portion of DCT destruction. However, in the test group, BUN, S-Cr and renal LPO level were significantly decreased as compared with control group, on the other hand, renal SOD activity was significantly increased. In the kidney of test group, the destruction of DCT and PCT were repaired as compared with control group. These results demonstrated that InJinORyungSan. can be attributed to recovery from nephrotoxicity, We consider that activated SOD by InJinORyungSan suppress renal LPO or production of free radicals induced by cyclosporin A.

Effect of Acron Extracts on the Antioxidative Enzyme System (도토리추출물이 흰쥐의 체내 항산화효소계에 미치는 영향)

  • This study was conducted to investigate the effect of acorn extracts on the antioxidative enzyme system. Male Sprague-Dawley rats(110$\pm$10g) were fed on containing normal and high fat diets. They were orally administrated (0.02g/100g B.W) of acorn ethylacetate-extract and water-extract at the same time once a day, respectively. Net weight gain and feed efficiency ratio were increased in high fat diet groups and decreased by acorn extracts administration. The effect of acorn extracts on hepatic glutathione peroxidase(GSH-Px), glutathione S-transferase(GST) and catalase activities did not show significancy in normal fat diet groups. GST and catalase activities and lipid peroxidation content(LPO) were significantly increased in high fat diet groups and this increment were decreased by acorn extracts administration. However GSH-Px activity and GSH content were decreased in high fat diet groups but increased by acorn extracts administration. The activities of lactate dehydrogenase(LDH), alkaline phosphatase(ALP) and aminotransferase in serum were significantly increased in high fat diet group but these increment reduced in acorn extracts administration groups. These results indicate that acorn extracts could improve the liver function and prevent the metabolic diseases by hyperlipidemia.

Effects of a Cardiac Rehabilitation Program on Health Behavior and Physiologic Parameters for Myocardial Infarction Patients (심장재활 프로그램이 심근경색증 환자의 건강행위와 생리적 지수에 미치는 효과)

  • Purpose: The purpose of this study was to investigate the effects of a cardiac rehabilitation program for patients with myocardial infarction. Method: The subjects were 45 myocardial infarction patients(22 for experimental group and 23 for the control group). Data were collected through questionnaire survey, blood test, and the treadmill test, between October 1, 1999 and December 31, 2000. As for data analyses, paired and unpaired t-test and $\chi^2$ test were adopted using the SAS program. Result: 1. The post-test revealed that increase in compliance score of health behavior was significantly higher in the experimental group than in the control group. 2. The increase in HDL cholesterol was significantly higher in the experimental group than in the control group, in the post-test. 3. The experimental group showed significantly higher duration of exercise time and maximal METs than the control group after the program. Conclusion: The above findings indicate that a cardiac rehabilitation program was effective in increasing compliance of health behaviors, serum HDL cholesterol level, duration of exercise time, and maximal METs in patients with myocardial infarction. Accordingly, we can adopt the individualized cardiac rehabilitation programs as a nursing intervention.

Effects of Pine Needle Extracts on Enzyme Activities of Serum and Liver, and Liver Morphology in Rats Fed High Fat Diet (솔잎추출물이 고지방식이를 급여한 흰쥐의 혈청, 간장의 효소 및 간조직구조에 미치는 영향)

  • The effects of pine needle extracts on lipid contents and antioxidative enzyme activities in liver of rat were evaluated. Thirty male Sprague-Dawley rats were divided into five groups and fed high fat diets for four weeks. Each group was administered with pine needle extract in the following doses: control, water ; WE-3, hot water extract(3% PN) ; WE-6, hot water extract(6% PN) ; AE-3, acetone extract(3% PN) ; AE-6, acetone extract(6% PN). The results obtained from the experiment were as follows: GOT activities were not significantly different among experimental groups but GPT activities were significantly low in the experimental groups compared to the control group. Liver superoxide dismutase(SOD) activity of pine needle extract administered groups was higher than that of control group. Catalase activities of liver had a similar tendency to SOD activities, but were not significantly different among the groups. Liver TBARS of WE-3 WE and AE-6 groups were slightly lower than those of other groups. Microscopic observation of liver tissue revealed that pine needle extracts increased cellular swelling, which was markedly increased in WE-6 group compared with control group.

Effect of Circadian Rhythms on the Bromobenzene Metabolism in Rats (랫드에 있어서 주야 시차가 Bromobenzene 대사에 미치는 영향)

  • To investigate the circadian variation in the bromobenzene metabolism, bromobenzene(400 mg/kg body weight) was intraperitoneally administered to the rats every other day for 6 days both in the night; 24:00 and the day; 12:00. Each group of animals was sacrificed at 8hr after last injection of bromobenzene. The contents of hepatic CYP were more increased in control rats of night phase than those of day phase but in case of bromobenzene treatment there were no differences in hepatic CYP between rats of the night phase and those of day phase and the injection of prednisolon inhibited the hepatic CYP content in rats. Furthermore, the decreasing rate of hepatic glutathione contents to the control was higher in rats of day phase than those of night phase by the bromobenzene treatment. And the hepatic glutathione S-transferase activities were increased both in control and bromobenzene treated rats of the night phase than those of day phase. On the other hand, liver weight per body weight(%), hepatic lipid peroxide content, serum levels of alanine aminotransferase were more increased both in bromobenzene-treated and control rats of the night phase than those in the day phase. These results indicate that the rats of night phase may induce more accelerated formation of bromobenzene 3,4-oxide from bromobezene than those of day phase in rats.
