Protective Effects of Ginkgo Biloba Leaf Extract(GBE) against 1,2,4-benzenetriol Induced Toxicity in Vitro (Ginkgo biloga 잎 추출물의 1,2,4-benzenetriol에 대한 항산화 효과에 대한 연구)

  • Ginkgo biliba has been used for bronchitis and asthma in oriental countries and its leaf extract(GBE) contains 24% ginkgoflavone glycoside and 6% terpenoid. Flavonoids and terpenoids are known to have various antioxidant effects such as scavenging of free radicals and chelation of transtional metals. Antioxidant effect of GBE against 1,2,4-benzenetriol(BT), one of toxic metabolites of benzene, was demonstrated throughbsister chromatid exchange(SCE) analysis, single cell gel electrophoresis(SCGE) analysis, DNA cleavage assay and lipid peroxidation production analysis. The means of SCE frequencies at 10, 25 and 50$\mu$M concentration of BT were 7.72, 8.02, 9.22 respectively. In addition of GBE with concentration of 50, 200 and 500$\mu\textrm{g}$/$m\ell$, SCE frequencies were decreased significantly.(p<0.05) According to SCGE analysis, BT induced DNA damage in a dose-dependent manner at concentration of 10 and 50 $\mu$m and the DNA damage induced by BT was significantly protected by GBE(p<0.001). No genotoxicity was observed by GBE treatment alone on DNA cleavage. The effect of BT on lipid peroxidation product, Malondiadehyde(MDA), was increased with concentration of BT(10 and 50 $\mu$M) and reduction in MDA was noted when GBE was added. From above results it is suggested that GBE could protect the cell and DNA from pro-oxidant effect by reactive oxigen species induced by BT.

Effects of Paraquat on Ascorbic Acid and Malondialdehyde Contents, and Superoxide Dismutase Activity in Spinach Chloroplasts under Light and Dark

  • The amounts of ascorbic acid in chloroplasts treated with light and light+paraquat (PQ) were reduced by 81 and 82% of initial level, respectively at 24 hr at incubation. And those treated with dark and dark+PQ were decreased by W and 55% of the original level, respectively. Malondialdehyde (MDA) contents at 24 hr of dark and dark+PQ treatment were increased by 6 and 31% of the initial level, respectively. When chloroplasts were treated with light and light+PQ, MDA contents after 24 hr were increased by 88 and 146% of the initial level, respectively. SOD activities treated with light and light+PQ were increased by 10 and 20% of the initial level, respectively for 3 hr and thereafter reduced by 46 and 49% of the original level, respectively at 24 hr. However, the SOD activities treated with dark and dark+PQ were decreased by 37 and 30% of the initial level, respectively. It is considered that PQ triggers the oxidation of ascorbic acid, the induction of lipid peroxidation and the inactivation of SOD under light so that PQ has inhibitors effect on the pathway of plant metabolism. Effect of Bromocriptine on 6-Hydroxydopamine-induced Lipid Peroxidation and Cytotoxicity in vitro and in vivo

  • The present study was to evaluate the protective effects of bromocriptine, which is known as $D_2$ dopamine receptor agonist and used for the treatment of patients with Parkinson's disease (PD), on 6-hydroxydopamine (6-OHDA)-induced neurotoxicity in vitro and in vivo. Lipid peroxidation product (malondialdehyde; MDA) produced by the administration of 6-OHDA was profoundly reduced following the treatment of bromocriptine in a dose-dependent manner in rabbit brain homogenate. Quinone formation by 6-OHDA autoxidation was also attenuated, and its effect was as potent as other antioxidants. Pretreatment of bromocriptine reduced the cytotoxicity of 6-OHDA on SH-SY5Y neuroblastoma cell lines dose-dependently. The loss of striatal dopamine and its metabolite, DOPAC (dihydroxyphenylacetic acid) as well as increase of MDA production caused by intrastriatal injection of 6-OHDA was significantly recovered following the treatment of bromocriptine. The present study clearly showed that bromocriptine had a protective action against 6-OHDA-induced neurotoxicity. These results suggest that bromocriptine has the antioxidant properties, which could be another advantage for delaying the progress of Parkinson's disease.

Oxidative Stress and Antioxidant Status during Transition Period in Dairy Cows

  • The study was conducted on 20 Holstein X Sahiwal cross bred dairy cows, with an average milk production of $2,752{\pm}113.79$ liters in $284{\pm}5.75$ days during a single lactation, that were divided in to two groups of 10 animals. We investigated the oxidative stress and antioxidant status during the transition period in dairy cows. In this study, plasma level of MDA was considered as an indicator of lipid peroxidation and SOD, catalase, GSH and GSHPx as antioxidants. The lipid peroxidation was significantly (p<0.001) higher in cows during early lactation as compared to the cows in advanced pregnancy. A significant positive correlation (r = +0.831, p<0.01) was determined between MDA and catalase in early lactating cows. In early lactating cows, blood glutathione was significantly lower than in advanced pregnant cows. However, early lactating cows showed non-significant negative correlation for all antioxidant enzymes with lipid peroxidation. In conclusion, dairy cows seemed to have more oxidative stress and low antioxidant defense during early lactation or just after parturition than advanced pregnant cows, and this appears to be the reason for their increased susceptibility to production diseases (e.g. mastitis, metritis, retention of fetal membranes etc.) and other health problems.

Physiological and Biochemical Responses of Fifteen Rice Cultivars to UV-B Radiation

  • This study was conducted to examine the physiological and biochemical responses against UV-B radiation in the seedling of 15 different rice cultivars, having the different physiological sensitivities. Out of 15 rice cultivars tested, moderate and susceptible groups showed significant decreases in biomass and RGR (relative growth rate). Contents of total chlorophyll were reduced remarkedly by irradiation of UV-B. In all rice cultivars tested, the content of chlorophyll a was strongly decreased, while the contents of chlorophyll b were slightly reduced without showing clear different among three groups and 15 cultivars. Carotenoid content was largely reduced by UV-B radiation, whereas polyamine content was moderately increased. The contents of MDA (malondialdehyde) that reflect the level of lipid peroxidation of cell membranes were clearly increased by UV-B stress, showing higher content in susceptible cultivars than moderate and torelant cultivars. The physiological important parameters highly related to visible injury were leaf color, chlorophyll, carotenoid, and lipid peroxidation, whereas biomass and polyamines were not closely correlated. Based on this results, it was concluded that changes of visible injury and the contents of chlorophyll and MDA could be adequately applied and utilized as physiological indicators to UV-B radiation.

Effect of Scutellaria baicalensis Georgi Aquacupuncture on Oxidant-induced Cell Injury in Renal Cortical Slices (황금약침액(黃芩藥鍼液)이 신장조직(腎臟組織)에서 Oxidant에 의한 세포손상(細胞損傷)에 미치는 영향(影響))

  • Objective : This study was undertaken to determine if Scutellaria baicalensis Georgi (SbG) extract exerts protective effect against oxidant-induced cell injury in renal proximal tubular cells. Methods : The cell injury was evaluated by lactate dehydrogenase (LDH) release in rabbit renal cortical slices and lipid peroxidation was estimated by measuring malondialdehyde (MDA). t-Butylhydroperoxide (tBHP) was used as a model of oxidant. Results : tBHP at 1 mM increased LDH release and lipid peroxidation, which were prevented by SbG in a dose dependent manner over concentration range of 0.001-0.1%. SbG provided the protective effect against oxidant-induced reduction in PAH uptake by renal cortical slices and microsomal Na+-K+-ATPase activity. SbG attenuated tBHP-induced depletion of reduced glutathione. 0.2 mM $HgCl_2$ increased LDH release and lipid peroxidation, which were completely prevented by 0.05% SbG. Conclusion : SbG prevents oxidant-induced impairment in membrane transport function.

The Effects of Soybean Peptide on Lipid Peroxidation, Antioxidant Enzyme and Inflammatory Cytokines in Elite Taekwondo Player

  • The purpose of this study was to investigate the effects of soybean peptide on lipid peroxidation, antioxidant enzyme and inflammatory cytokines in male high school taekwondo players. The subjects were divided into 2 groups, which were a soybean peptide intake group (S-peptide, n=13) and a placebo intake group (Placebo, n=10). The s-peptide group took 4 g a day of soybean peptide for 4 weeks. Blood samples were taken from the antecubital vein at before taekwondo performance, after 2 and 4 weeks taekwondo performance. The plasma was analyzed for the antioxidant enzyme activity factor, lipid peroxidation and cytokines. As a result, the s-peptide group decreased lipid peroxidation (MDA) and increased the antioxidant enzyme activity factor (TAS and CAT). The plasma concentration of inflammatory cytokines, IL-6 and TNF-${\alpha}$ were significantly decreased in the s-peptide group after 4 weeks and showed significant differences between the groups. These results indicate that the intake of soybean peptide was positively improved on antioxidant enzyme and inflammatory cytokines in taekwondo player.

Age Related Changes of Microsomal Fatty Acid Composition and Lipid Peroxidation in 2-acetylaminofluorene Treated rats. (2-Acetylaminofluorene의 투여와 나이에 따른 쥐의 세포막 지방산 조성 및 지질과산화물 생성의 변화)

  • For studying the effect of different dietary fats on carcinogenesis, fatty acid composition of membrane and lipid peroxidation were measured. Male Sprague-Dawley rats were fed on diet containing 15%(w/w) beef tallow or soybean oil. A single dose of 50 mg 2-AAF/kg B.W. was injected i.p. in each diet group 10 times. Rats were sacrifled after 1, 5, 10, and 15 weeks from the first injection. By 2-AAF injection, !ipid peroxidation increased slightly compared to control group. The rats fed on different fats had similar MDA production and those fed on soybean oil had slightly higher free radical concentration measured by ESR. In young rats, iipid peroxidation level was high and hydroxy radical production was higher in soybean oil group than in beef tallow group. With age, the lipid peroxidation values were decreased initially then increased. The fatty acid composition in microsomal membrane was reflected by dietary fatty acid composition. In soybean oil group, monoenoic acid was lower and polyunsaturated fatty acid was higher than beef tallow group. Linoleic acid contents showed the most discrepancy among groups. By 2-AAF treatment, iinoleic acid content and unsaturation index increased in soybean oil group. But in beef tallow group, there was no difference in fatty acid contents.

Protective Effect of Antioxidants on the Reoxygenation Injury in Hypoxic Myocardium of Rat (저산소 심장의 산소 재공급에 따른 심근 손상에 있어서 항산화제의 보호 효과)

  • The effect of antioxidants on the myocardial cellular damage which occurs during reoxygenation of hypoxic myocardium was examined in isolated rat hearts. The roles of oxygen free radical and lipid peroxidation in reoxygenation injury of myocardium were also investigated. In Langenorff preparation of isolated rat heart, which was made hypoxic by perfusion with the substrate free, hypoxic cardioplegic solution ($37^{\circ}C$, 90 min), the release of cytosolic enzymes (creatine phosphokinase, lactic dehydrogenase) and a lipid peroxidation product, malondialdehyde into the coronary effluent were abruptly increased by reoxygenation. The release of enzymes was closely parallel to that of MDA. These increases of enzymes and lipid peroxidation product were suppressed to various degrees in the presence of scavengers of superoxide anion (superoxide dismutase, 10,000 U), hydrogen peroxide (catalase, 25,000 U) and hydroxyl radical (dimethyl sulfoxide, 10%). A natural antioxidant, ${\alpha}-tocopherol$(4.5 uM) and a synthetic one, butylated hydroxytoluene (2 uM) suppressed the release of cytosolic enzymes with the concomittent reduction of lipid peroxidation as measured by malondialdehyde release into the coronary effluent. These effects of antioxidants were dose dependent, and were more pronounced when the antioxidants were administered throughout hypoxic and reoxygenation periods than given during reoxygenation period only. These results suggest that cytotoxic oxygen free radicals produced in the myocardium during reoxygenation may be responsible fur the myocardial cellular injury by enhancing the lipid peroxidation of cellular membranes. Furthermore, the antioxidants may exert protective effect against reoxygenation damage of hypoxic myocardium through the inhibition of lipid peroxidation reaction.

  • ln an attempt to define the effects of hyperbaric oxygen treatment on the lipid peroxidation and oxygen See radical reactions in rats exposed to carbon monoxide, we studied malondialdehyde(MDA) level and activities of catalase and superoxide dismutase in the heart of the rats exposed to carbon monoxide. Male Sprague-Dawley albino rats weighing 240 to 260gm were used. Experimental groups consist of Control group (=breathing with air), HBO group(=exposed to hyperbaric oxygen(HBO, 3ATA, 100%) after air breath), CO group(=exposed to CO(3,970 ppm) after air breath), CO-Air group(=exposed to CO after air breath followed by air breath) and CO-HBO group(=exposed to CO after ai. breath followed HBO treatment). The CO group showed significantly higher MDA level, catalase activity and SOD activity as compared to that of control group. The CO-HBO group showed significantly lower MDA level as compared to that of CO group, and did not show significantly lower catalase activity and SOD activity as compared to that of CO group. These results suggest that the excessive oxygen free radicals is an important determinant in pathogenesis of Co-induced cardiotoxicity and HBO inhibits the lipid peroxidation caused by excessive oxygen free radicals in the heart of the rats exposed to carbon monoxide.
