  • Objectives : We studied Effects of Hagocho(prunella vulgaris L.), Gamgook(chrysanthemum indicum L.) and Galgeun(pueraria Radix) aqua-acupuncture on the hyperlipidemic rat. methods : We investigated lowering lipid effect, oxidative capacity, concentration of TNF-${\alpha}$, IL-6, Leptin and histological consideration in hyperlipidemic rat. Forty male Sprague-Dawley rats weighing about 400g were divided into 5 groups of control, Ⅰ: Hagocho (prunella vulgaris L.)+Gamgook (chrysanthemum indicum L.) and Gokji aqua-acupuncture, Ⅱ: Hagocho(prunella vulgaris L.)+Gamgook (chrysanthemum indicum L.) and Joksamri aqua-acupuncture, Ⅲ: Hagocho(prunella vulgaris L.)+Galgeun(pueraria Radix)and Gokji aqua-acupuncture and Ⅳ: Hagocho (prunella vulgaris L.)+Galgeun(pueraria Radix) and Joksamri aqua-acupuncture. Results : Contents of plasma ${\beta}$-lipoprotein, contents of IL-6, TNF-${\alpha}$ and leptin, Plasma triglyceride and glucose, plasma total cholesterol and LDL-cholesterol, liver total cholesterol, liver triglyceride, plasma and liver TBARS, free fatty acids showed a tendency to decrease in the aqua-acupuncture groups compared to those of control group. The activities of GOT and GPT showed no significantly different in all treatment groups. Values of super oxide dismutase, glutathione peroxidase and catalase activity showed a tendency to increase in the aqua-acupuncture groups. Histological consideration of heart, kidney and liver in the aqua-acupuncture groups showed slight vasodilation and fat accumulation compared to those of normal rat. Conclusions : These results indicated that prunella vulgaris L., chrysanthemum indicum L. and pueraria Radix aqua-acupuncture at gokji(LI11) and Joksamri(ST36) suppressed adipose tissue mass and lipid peroxidation and increased antioxidant system in hyperlipidemic rat.

Effect of Dried Powders or Ethanol Extracts of Garlic Flesh and Peel on Lipid Metabolism and Antithrombogenic Capacity in 16-Month-Old Rats (마늘 육질과 껍질의 건분 및 에탄올추출물이 노령흰쥐의 지방대사와 항혈전능에 미치는 영향)

  • This study was performed to investigate effect of dried powders and ethanol extracts of garlic flesh and peel on lipid metabolism and antithrombogenic capacity in 16-month-old rats. Forty Sprague-Dawley male rats weighing 618.1$\pm$6.5 g were blocked into five groups according to body weight and raised for 3 months with control and experimental diets containing 5% (w/w) of dried powders of garlic flesh or peel, or ethanol extracts from equal amount of each dried powder and control diet. Plasma and liver total lipids, triglyceride and total cholesterol, and plasma HDL-cholesterol, throm-boxane $B_2$ (TX $B_2$), 6-keto-prostaglandin $F_{1a}$ (6-keto-PG $F_{1a}$) concentrations were measured. Total, insoluble and soluble dietary fibers contents were highest in peel powder followed by fresh powder, and those in ethanol extracts of flesh and peel, especially soluble, very low. Plasma and liver total lipids, triglyceride, and total cholesterol concentrations were lower in all the garlic experimental groups compared to Especially, flesh and peel powder lowered plasma total lipids, triglyceride and total cholesterol concentrations markedly, and flesh powder and flesh ethanol extract lowered liver total lipids, triglyceride and total cholesterol concentration remarkably. Plasma TX $B_2$ concentrations in garlic experimental groups were lower than that of control group, and 6-keto-PG $F_{1a}$ concentrations. In garlic experimental groups were higher than that of control group. Flesh ethanol extract group showed the lowest TX $B_2$ and the highest 6-keto-PG $F_{1a}$ concentrations among experimental groups, so TX $B_2$/6-keto-PG $F_{1a}$ ratio in flesh ethanol extract group was significantly lower than that of control group. Moreover, clotting time was significantly increased in flesh ethanol extract group as compared to control group. In conclusion, intakes of dried powders and ethanol extracts of garlic flesh and peel were effective in lowering lipid levels of liver and plasma. And also flesh ethanol extract diet was most effective in antithrombogenic activity among garlic experimental groups as TX $B_2$/6-keto-PG $F_{1a}$ ratio in flesh ethanol extract group was significantly lower and clotting time was significantly increased in this group as compared to control group.ntrol group.

  • The study was conducted to compare the components such as proximate composition, total dietary fiber(TDF) content, acid detergent fiber(ADF) content, lignin, water binding capacity(WBC), amino acid composition and lipid components from brown Job's tears, dehulled Job's tears, brown Yeomjoo and bran of Job's tears. The results from this study are summerized as follows: Total dietary fiber(TDF) content of Job's tears, brown Job's tears, brown Yeomjoo and bran of Job's tears were 2.70%, 3.86%, 4.33% and 13.3% each. Water-binding capacity(WBC) of TDF and ADF were $2.63{\pm}0.02g\;H_2O/g$ TDF and $5.89{\pm}0.15g\;H_2O/g$ ADF each. In amono acids composition of samples, glutamic acid content was the highest and the next was leucine. Chemical score of leucine in dehulled Job's tears was very high(189), in contrast lysine was very low$(22{\sim}23)$ So lysine was a first limitting amino acid in Job's tears and Yeomjoo. Neutral lipid contents were 90.89%-96.55%, glycolipid contents were 2.35%-7.48% and phospholipid contents were very low. The major fatty acids of lipid fractions were palmitic acid. oleic acid and linoleic acid.

Inhibitory Effects of Opuntia humifusa on 7, 12-Dimethyl-benz[a]anthracene and 12-O-tetradecanoylphorbol-13-acetate Induced Two-stage Skin Carcinogenesis

  • Opuntia humifusa, member of the Cactaceae family, was previously demonstrated to have radical scavenging, anti-inflammatory and anti-proliferative effects in in vitro models. It was suggested that O. humifusa could function in the prevention of carcinogenesis. To investigate the in vivo chemopreventive effect of O. humifusa, mice were fed a diet containing either 1% or 3% following 7, 12-dimethylbenz[a] anthracene (DMBA) and 12-O-tetradecanoylphorbol-13-acetate (TPA) induction of skin carcinogenesis. Significant decrease in the numbers of papilloma and epidermal hyperplasia were observed in mice fed with O. humifusa, compared to the control group. O. humifusa also upregulated high total antioxidant capacity and level of phase II detoxifying enzyme such as superoxide dismutase and glutathione S-transferase activity in the skin. Lipid peroxidation activity level was measured in skin cytosol and significantly inhibited in 3% OH fed group compared to the control group. These results suggest that O. humifusa exerts chemopreventive effects on chemical carcinogenesis in mouse skin and that prevention effects are associated with reduction of oxidative stress via the modulation of cutaneous lipid peroxidation, enhancing of total antioxidant capacity especially in phase II detoxifying enzyme system and partial apoptotic influence.

Functional Activities of Ethanol Extracts from Flammulina velutipes (팽이버섯 에탄올 추출물의 기능적 특성)

  • This study was conducted to evaluate the antioxidative effect and antimutagenic capacity of ethanol extracts of Flammulina velutipes by employing biological and biochemical assays. The $IC_{50}$ of MDA with BSA conjugation reaction, lipid peroxidation and scavenging effect on DPPH radical in ethanol extracts of Flammulina velutipes was found to be 28.39 mg/assay, 9.33 mg/assay and 144.61 mg/assay respectively. Therefore, the most effective antioxidative capacity of ethanol extracts of Flammulina velutipes was $Fe^+$-induced linoleate peroxidation, among the method used this study. The indirect and direct antimutagenic effects of the ethanol extracts of Flammulina velutipes were examined by the Ames test using Salmonella typimurium TA98 and TA100. The inhibition rates on indirect mutagenicity mediated by 2-anthramine and on direct mutagenicity mediated by sodium azide in Salmonella typimurium TA100 and mediated by 2-nitrofluorene in Salmonella typimurium TA98 were 0%, respectively. These findings indicate that ethanol extracts of Flammulina velutipes have no effects on indirect and direct mutagenicity. Based on these results, it believed that the ethanol extracts of Flammulina velutipes has antioxidative capacities, and is a the candidate for the prevention and dietetic treatment of chronic diseases and the development of antioxidative functional food.

Effects of taeeumjoweetang Herbal-Acupuncture on plasma and liver lipid composition and antioxidative capacity in rat fed high fat diet (태음조위탕(太陰調胃湯) 약침(藥鍼)이 고지방 급여 흰쥐의 혈장 및 간장의 지질구성과 항산화능에 미치는 영향)

  • Objectives : In order to measure the Effects of taeeumjoweetang Herbal- Acupuncture at joksamri(ST36) and kwanweun(CV4) on liver and plasma lipid composition and antioxidative capacity in rat fed high fat diet. Results : Concentrations of plasma triglyceride, total cholesterol and LDL-cholesterol showed a tendency to increase in the high fat diet group. However Herbal-Acupuncture groups showed a lower values than control groups. HDL-cholesterol showed a tendensy to decrease in high 131 fat diet groups and in high fat diet groups, these values showed no significantly different. Liver total cholesterol values showed no significantly different in all treatment groups. Triglyceride concentration showed a high value in control group and other treatment groups showed no significantly different. plasma GOT and GPT values showed a tendency to increase in high fat diet group. however these values decreased in Herbal-Acupuncture group. The concentration of TBARS in liver and plasma showed a high values in high fat diet group, however these values showed a tendency to decrease in aqua- acupuncture group. Glutathione peroxidase, superoxide dismutase and catalase activity values showed a low values in high fat diet group, however these values showed a tendency to increase in Herbal-Acupuncture group.

Anti-oxidant Effect of Agastache rugosa on Oxidative Damage Induced by $H_2O_2$ in NIH 3T3 Cell

  • The plant Agastache rugosa Kuntze has various physiological and pharmacological activities. Especially, it has been regarded as a valuable source for the treatment of anti-inflammatory and oxidative stress-induced disorders. However, little has been known about the functional role of it on oxidative damage in mammalian cells by ROS. In this study, we investigated the DPPH radical, hydroxyl radical, hydrogen peroxide and intracellular ROS scavenging capacity, and $Fe^{2+}$ chelating activity of the extracts from Agastache rugosa. In addition, we evaluated whether the extract can be capable of reducing $H_2O_2$-induced DNA and cell damage in NIH 3T3 cells. These extracts showed a dose-dependent free radical scavenging capacity and a protective effect on DNA damage and the lipid peroxidation causing the cell damage by $H_2O_2$. Therefore, these results suggest that Agastache rugosa is useful as a herbal medicine for the chemoprevention against oxidative carcinogenesis.

Antioxidant Activities of Bromotopsentin from the Marine Sponge Spongosorites sp. (해면으로부터 분리된 Bromotopsentin의 항산화활성)

  • Bromotopsentin (BSM) is a bisindole alkaloid compound, which is recognized as a metabolite of the marine sponge Spongosorites sp. In this study, the antioxidant activity of BSM was investigated. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assay, the trolox equivalent antioxidant capacity (TEAC) assay, the superoxide radical scavenging (NBT) assay, the lipid peroxidation and hydroxyl radical-induced DNA damage assays were carried out to evaluate the antioxidant activity of BSM. It was found that BSM had stronger scavenging activity on the stable free radical DPPH and superoxide radical than L-ascorbic acid with an $IC_{50}$ value of 62 and 64 ${\mu}M$, respectively. The TEAC value which indicated the total antioxidant capacity of BSM was about 0.8, which was also stronger than L-ascorbic acid. About 1.3 ${\mu}M$ of BSM induced 50% inhibition of lipid peroxidation. 60 nM of BSM exhibited a significant protective activity against DNA strand scission by hydroxyl radical on pBR322 DNA. Taken together, we suggest that BSM possesses strong antioxidant activity, and could be a valuable new addition to the list of anti-aging chemotherapeutic agents.

Effects of Powdered Siho(Bupleuri Radix) on Serum and Liver Lipid Composition and Antioxidative Capacity in Rat Fed High Oxidized Fat (시호(Bupleuri Radix)분말이 과산화지질을 급여한 흰쥐의 혈장 및 간장지질구성과 항산화능에 미치는 영향)

  • Effects of powdered siho(bupleuri radix) on serum and liver lipid composition and antioxidative capacity were investigated in rat fed high oxidized fat. Sixty male Sprague-Dawley rats weighing 161.25$\pm$2.51g were blocked into four groups according to body weight and raised seven weeks with basal diet(normal group), basal diet and 10% oxidized fat(control group), basal diet, 10% oxidized fat and 2% powdered siho(2% powdered siho group) and basal diet, 10% oxidized fat and 3% powdered siho(3% powdered siho group). Feed intake, body weight gain and feed efficiency were not significantly different among oxidized fat diet groups. The values of total cholesterol and triglyceride in plasma and liver showed no significantly different(p>0.05) in the oxidized fat diet groups. Plasma HDL-cholesterol showed atendency to increase in powdered siho diet groups. The values of thiobarbituric acid in plasma and liver were lower in the powdered siho diet groups than control group. Plasma GOT and GPT activity showed a tendence to decrease in powdered siho groups. Liver glutathione peroxidase activity showed a tendence to increase in powdered siho diet groups. Thus, it seems that powdered siho had some antioxidative elements.

Fuctional Relationship between Rate of Fatty Acid Oxidation and Carnitine Palmitoyl Transferase I Activity in Various Rat Tissues

  • Lipids play many structural and metabolic roles, and dietary fat has great impact on metabolism and health. Fatty acid oxidation rate is dependent on tissue types. However there has been no report on the relationship between the rate of fatty acid oxidation and carnitine transport system in outer mitochondrial membrane of many tissues. In this study, the rate of fatty acid oxidation and carnitine palmitoyltransferase (CPT) I activity in the carnitine transport system were measured to understand the metabolic characteristics of fatty acid in various tissues. Palmitic acid oxidation rate and CPT I activity in various tissues were measured. Tissues were obtained from the white and red skeletal muscles, heart, liver, kidney and brain of rats. The highest lipid oxidation rate was demonstrated in the cardiac muscle, and the lowest oxidation rate was in brain. Red gastrocnemius muscle followed to the cardiac muscle. Lipid oxidation rates of kidney, white gastrocnemius muscle and liver were similar, ranging from 101 to 126 DPM/mg/hr. CPT I activity in the cardiac muscle was the highest, red gastrocnemius muscle followed by liver. Brain tissue showed the lowest CPT I activity as well as lipid oxidation rate, although the values were not significantly different from those of kidney and white gastrocnemius muscle. Therefore, lipid oxidation rate was highly (p<0.001) related to CPT I activity. Lipid oxidation rate is variable, depending on tissue types, and is highly (p<0.001) related to CPT I activity. CPT I activity may be a good marker to indicate lipid oxidation capacity in various tissues.
