In this study Carotenoprotein from Styela clava were extracted, and purified by ammonium sulfate fraction, Sephadex G- 200 gel chromatography and DEAE-cellulose ion exchange chromarography. The purified carotenoprotein from styela clava had absorption maxima of 487nm, 463nm and 280nm, and the carotenoid liberated from carotenoprotein had 478nm and 452nm with inflexion. One miligram of caroteno-protein contained 0.35 $\mu$g of carotenoid. The carotenoprotein had an approximate molecular weight of 398 kDa (gel filtration). SDS-PAGE showed only a single polypeptide chain with a molecular weight of 62.4 kDa. The amino acid composition of the carotenoprotein were mainly glutamic acid(11.48%), valine(10.75%), leucine (10.45%), aspartic acid(9.94%), while cysteine and tryptophan were not found. The carotenoprotein contained lipid as structure units. In the carotenoprotein, the major fatty acids were oleic acid, plamitoleic acid and palmiunsaturated fatty acids 33.6%, saturated fatty acids 18.6%. In addition, the levels of higher unsaturated fatty acids were high as much as 30.8% of the total fatty acids. Carotenoid was extracted from the carotenoprotein by acetone. Thin layer chromatography showed only carotenoid to be present. Its chemical reactivities and spectroscopic properties were studied and elucidated as astaxanthin.
Oryza sativa cv. Heugjinjubyeo, an anthocyanin-pigmented rice variety, is well known to contain high levels of bioactive phytochemicals, anthocyanin, quinolone alkaloids and phenolic acids. Here, we studied the inhibitory effect of Oryza sativa cv. Heugjinjubyeo bran on the absorption of dietary fat in streptozotocin-induced diabetic Sprague-Dawley male rats. For these experiments, experimental animals were divided into four groups: normal, diabetic-control and two experimental groups that were fed 1.0 g or 2.0 g/kg body weight/day of Oryza sativa cv. Heugjinjubyeo bran supplement for 14 days. As a result, liver glycogen levels increased significantly by 65% and 32% in groups receiving 1.0 g and 2.0 g/kg body weight/day, respectively, compared to diabetic-control. Liver cholesterol levels were significantly lower by 8.3% and 14.5% in the groups fed 1.0 g and 2.0 g of anthocyanin- pigmented rice extracts, respectively.

In this study, we investigated the quality characteristics of noodles supplemented with lyophilized leg-meat powder from the red snow crab, Chionoecetes japonicus (CP). The noodles were prepared by adding 0, 2, 4, 6, 8, 10% of CP on a wheat-flour basis. The ash, crude protein and crude lipid contents of noodles increased with increasing CP content. The cooked weight, volume and water absorption values of noodles decreased, whereas the turbidity increased, with increasing CP content. The L values of the noodles decreased with increasing CP content, whereas the a and b color values increased. The hardness, gumminess and chewiness values increased with increasing CP content increased, while springiness and cohesiveness showed no significant differences among the groups. In the sensory evaluation, cooked noodles with 4-6% CP had greater overall acceptability than the noodles with other CP levels.
The main objective of this study was to investigate possible application of citron (Citrus junos Sieb. ex TANAKA) seeds, which are massively produced as by-products during citron tea process, into functional food materials. First of all, citron seeds were germinated and produced citron shoots were examined for their functional properties. When contents of flavonoids in citron seeds and their germinated shoots were compared, naringenin, neohesperitin, and hesperitin were remarkably increased in shoots after germination while naringin and didymin were decreased. Concentrations of limonin and nomilin were decreased by germination otherwise their unidentified derivatives were newly formed. A methanol extract of citron shoot had lower $IC_{50}$ values [0.13 and 0.07 mg/mL for 2,2-diphenyl-1-picryl-hydrazyl (DPPH) and ABTS, respectively] than citron seed extract in radical scavenging activities. Addition of 500 mg/mL of citron shoot extract suppressed fat accumulation in 3T3-L1 adipocytes by 36.9%. Oral administration of olive oil along with citron shoot extract (33 mg/kg body weight) to Sprague Dawley rats effectively inhibited absorption of lipid into a body by decreasing blood triglyceride levels from 105.1 to 74.9 mg/dL 2 hr after olive oil administration. According to these results, citron shoot extract as a rich source of flavonoids can be utilized for functional food ingredients with effective antioxidant and anti-adipogenic properties.
Fresh grass carp was used to produce surimi and 50 g/kg, 100 g/kg, or 150 g/kg pork back fat was added. The water distribution, thiobarbituric acid reactive substances (TBARS), myoglobin oxidation, color parameter (L*, a*, and b*), heme and non-heme iron content of samples were determined to analyze the effects of different fat content on the oxidation of myoglobin and lipids during multiple freeze-thaw cycles of grass carp surimi. Both multiple freeze-thaw cycles and increased fat content lead to an increase in TBARS, a blue shift in the absorption peak of myoglobin porphyrin, a decrease in heme iron content, and an increase of non-heme iron content. Repeated freeze-thaw caused a decrease in immobilised water content and L*, and caused an increase in a* and b*. Increased fat content caused an increase in immobilised water content, L* and a*, and caused a decrease in b*.
This study compared the physicochemical and gelatinization properties of naked barley, tetrastichum barley, and waxy barley. Compared to tetrastichum barley and waxy barley, naked barley had shorter and rounder grains with a 1.43 length/width ratio. Tetrastichum barley had lower crude protein, crude lipid, and crude ash content and higher amylose content compared to naked barley and waxy barley. The L, a, b color values of waxy barley were significantly higher than those of naked barley and tetrastichum barley. The water absorption index (WAI) and the water soluble index (WSI) were highest in waxy barley. The X-ray diffraction pattern was type A in all samples, and the peak intensity was highest in waxy barley. The maximum viscosity, cooling viscosity, breakdown, and setback of amylogram properties were the highest in tetrastichum barley. The thermal properties through the differential scanning calorimeter showed that the waxy barley had higher values of the onset, peak, conclusion temperature and enthalpy (?H). In conclusion, the variety of barley influenced the physicochemical and gelatinization properties, which could be important factors in the manufacture of processed foods. These results would thus useful inputs for the manufacturing of these foods using barley.
Cyanobacteria are the dominant micro flora in rice-fields, contributing significantly to fertility as a natural biofertilizer. Recent studies show a continuous depletion of the stratospheric ozone layer, and the consequent increase in solar UV-B (280-315 nm) radiation reaching the Earth's surface. UV-B radiation causes reduction in growth, survival, protein content, heterocyst frequency and fixation of carbon and nitrogen in many cyanobacteria. UV -B induced bleaching of pigments, disassembly of phycobilisomal complexes, thymine dimer formation and alterations in membrane permeability have also been encounterd in a number of cyanobacteria. However, certain cyanobacteria produce photoprotective compounds such as water soluble colorless mycosporine-like amino acids (MAAs) and the lipid soluble yellow-brown colored sheath pigment, scytonemin, to counteract the damaging effects of UV-B. Cyanobacteria, such as Anabaena sp., Nostoc commune, Scytonema sp. and Lyngbya sp. were isolated from rice fields and other habitats in India and screened for the presence of photoprotective compounds. A circadian induction of the synthesis of MAAs by UV -B was noted in a number of cyanobacteria. Polychromatic action spectra for the induction of MAAs in Anabaena sp. and Nostoc commune also show the induction to be UV-B dependent peaking at 290 nm. Another photoprotective compound, scytonemin, with an absorption maximum at 386 nm (also absorbs at 300, 278, 252 and 212 nm), was detected in many cyanobacteria. In conclusion, a particular cyanobacterium having photoprotective compounds may be a potent candidate as biofertilizer for crop plants.
Previously, we have shown that green tea extract lowers the intestinal absorption of cholesterol, fat, and other fat-soluble compounds. We conducted this study to determine whether green tea extract affects the rate of $^{14}C$-oleic acid esterification into various lipids in the intestinal mucosa of rats. Male Sprague-Dawley ruts were had free access to a nutritionally adequate AIN-93G diet and deionized water. Initially, the rat's mucosal content of total lipids was measured following 1 mL olive oil administration with (green tea group) or without (control group) 100 mg green tea extract powder. At 1 h and 5 h, intestinal segments were extracted for total lipid analysis. Secondly, to measure mucosal esterification rates of lipids, an abdominal incision was made along the midline, and a 10-cm long jejunal segment of the small intestine was ligated in situ. Then, micellar solutions with or without green tea extract were injected into the ligated jejunal segments and incubated for 10 mill. The micellar solution contained $200.0\;{\mu}$ Ci $^{14}C$-oleic acid, $200.1\;{\mu}mol$ unlabelled oleic acid, $66.7\;{\mu}mol$ 2-monooleoylglycerol, $66.7\;{\mu}mol$ palmitoyl-sn-glycero-3-phosphocholine, 2.2 mmol glucose, $50.0\;{\mu}mol$ albumin, and 16.5 mmol Na-taurocholate per L of phosphate buffered saline (pH, 6.3) with or without 8.87 g green tea extract powder. At 10 min, each rat was sacrificed by cervical dislocation under anesthesia and the segment was removed for lipid analysis. Significant differences were observed in mucosal triglyceride content at 1 h and 5 h in ruts given green tea extract. Significant differences in the rate of $^{14}C$-oleic acid esterification into triglycerides and phospholipids fractions were observed between control and green tea groups. However, There were no significant differences in other lipid fractions. These results indicate that the lowered esterification rates of $^{14}C$-oleic acid into triglycerides and phospholipids fractions is attributable to presence of green tea extract. This may be associated with an inhibitory effect of green tea catechin on the mucosal processes of lipids, leading to the inhibition of intestinal absorption of lipids.
In this study, we aimed to investigate the effects of dietary supplementation with fruiting body of Agaricus brasiliensis (AB) mushroom on the lipid profiles of serum and histological patterns of liver of high cholesterol-fed rats. Five-week-old, female Sprague-Dawley albino rats were divided into three groups of 8 rats each, including a normal control-diet (NC) group, a high-cholesterol diet (HC) group, and a group fed high-cholesterol diet supplemented with 5 % powder of Agaricus brasiliensis fruiting bodies (HC+AB). Total serum cholesterol, low density lipoprotein (LDL), and triglyceride (TG) concentrations in the HC+AB group were significantly reduced when compared with those in the HC group. Body weight in the HC+AB group was significantly lower than that in the HC group, whereas no adverse effects were observed on the levels of plasma albumin, creatinine, blood urea nitrogen, uric acid, glucose, and total protein. In the HC+AB group, liver enzyme activities related to liver function, such as GOT and GPT, presented values lower than those in the HC group and were very similar to the ones in the NC group. Excretion of total lipid and cholesterol in feces in the HC+AB group was significantly higher than that in the NC and HC groups, indicating that mushroom feeding inhibits the absorption of lipid cholesterol in the intestine. Liver histopathological analyses revealed that rats fed with HC diet developed fat liver disease, whereas only small amounts of fat were deposited in the livers of the HC+AB group. In conclusion, the results suggest that fruiting body powder of A. brasiliensis provides health benefits to high-cholesterol-fed rats by lowering body weight and the risk of atherogenic lipid profile.
Oxidative changes in the rice lipids under accelerated condition were studied by measuring the changes in weight gain, total carbonyl compound, malonaldehyde, and fatty acid composition. Rice lipids were prepared by extraction with either n-hexane or ethanol from polished rice grain and purified by Folch's method. The lipid preparations were either incubated in dark at $40^{\circ}C$ or irradiated with ultra-violet light for a period of 55 days. Weight gain by oxygen absorbed sharply increased within 3 days in the rice lipids under UV light irradiation. However, with the rice lipids at $40^{\circ}C$ incubation a moderate increase in weight was observed only after 45 days storage. Their induction periods were one day (hexane extracted, under UV light), 2 days (ethanol extracted, under UV light), 30 days (hexane extracted, at $40^{\circ}C$), and 40 days (ethanol extracted, at $40^{\circ}C$) respectively. Oxidative rancid odor appeared at the end of the induction period. Total carbonyl compound and malonaldehyde markedly increased within 7 days, and decreased in the rice lipids under ultra violet light irradiation, while at $40^{\circ}C$ incubation they were continued to increase slowly through out the storage. The hexane extracted lipid were less stable than ethanol extracted lipid on the basis of oxygen absorption, malonaldehyde and other carbonyl compound formation. With the hexane extracted lipid during 55 days incubation at $40^{\circ}C$, the contents of linoleic and linolenic acids decreased, while the oleic, stearic, and palmitic acids increased.
