Lipid Peroxidation Induced by the Reaction of Cytochrome c with Hydrogen Peroxide

  • Lipid peroxidation induced by the reaction of cytochrome c with $H_2O_2$ was investigated. When linoleic acid micelles or phosphatidyl choline liposomes were incubated with cytochrome c and $H_2O_2$, lipid peroxidation was increased in cytochrome c and $H_2O_2$ concentrations-dependent manner. Radical scavengers, azide, formate and ethanol prevented lipid peroxidation induced by the cytochrome c/$H_2O_2$ system. Iron specific chelator, desferoxamine also prevented the cytochrome c/$H_2O_2$ system-mediated lipid peroxidation. These results suggest that lipid peroxidation may be induced by the cytochrome c/$H_2O_2$ system via the generation of free radicals. Carnosine, homocarnosine and anserine are present in the muscle and brain of many animals and human. Previous studies show that these compounds have an antioxidant function. In the present study, carnosine, homocarnosine and anserine significantly prevented the cytochrome c/$H_2O_2$ system-mediated lipid peroxidation. Carnosine and related compounds also inhibited the free radical-generating activity of cytochrome c. The results suggest that carnosine, homocarnosine and anserine may prevent lipid peroxidation induced by the cytochrome c/$H_2O_2$ system through a free radical scavenging.

  • This study was conducted to investigate the effects of chitosan, chitosan oligomer and beef tallow of different levels on lipid metabolism in rats, Seventy male rats of the Sprague-Dawley strain were blocked into 10 groups according to body weight , and were raised for 4 weeks. Dietary fat levels were 20% en and 40% en, and chitosan and chitosan oligomer were given at levels of 05, 3%, and 5%(wt/wt) of diet. The results are summarized as follows, chitosan oligomer supplement decreased serum total lipid, as chitosan 양, and exhibited a tendency to decrease serum total cholesterol. Chitosan oligomer supplement tended to increase the HDL cholesterol ; total cholesterol ration as chitosan did. Liver total lipid and triglyceride concentration were lower in high fat groups than in low fat groups. Liver total lipid concentrations was decreased slightly by chitosan and sigfnificantly by chitosna ligomer. Epididymal fat pad total lipid. total cholesterol and triglyceride concentration were slightly decreased by chitosan and chitosan oligomer supplement, especially in high fat groups. fat absorptivity was decreased by low fat level, chitisan and chitosan oligamer supplement. Fecal excretion of total lipid and triglyceride were increased by high fat level, chitosan and chi샌무 oligomer supplement. However, fecal excretion of total cholesterol was increased by high fat level and chitosan supplement. This indicated that chitosan and chitosan oligomer were effective in interfering with lipid and triglyceride absorption, In conclusion, chitosan oligomer at levels of 3% and 5% has more effective lipid, cholesterol and triglyceride lowering activity than chitosan.

Age-and Sex-Related Differences in Serum Levels of Lipid Peroxide, Retinol an d$\alpha$-Tocopherol in Korean Adults

  • The present study was aimed to investigated whether there are age-and sex-related differences in serum levels of lipid peroxides, retinol , and $\alpha$-tocopherol in Korean adults. The subjects were 441 persons , including 268 men and 173 women. Those of each sex were divided into four age group : 20-29, 30-39, 40-49 and 50-65 years, and their lifestyles and serum levels were compared . Men smoked and drank less as age increased , but 99.9% of women of did not smoke and only 11% drank more than once a month. Lipid peroxides of males increased gradually with age, while those of females showed greater levels in the 50-65 years group thon younger groups. Lipid peroxides, retinol and $\alpha$-tocopherol concentrations, which were adjusted for age, were significantly higher in males than in females. Lipid peroxide levels adjusted for total lipid were positively correlated with age in males than in females. Serum levels of $\alpha$-tocopherol adjusted for total lipid were positively correlated with age both in males and in females , while retinol was neither. The results indicate that serum levels of lipid peroxides, retinol , and $\alpha$-tocopherol are affected by age and that the response could be different between males and females.

쌀눈기름의 급여가 KK 당뇨 마우스의 지질대사에 미치는 영향 (Supplementary effect of the rice germ oil on Lipid Metabolism in Diabetic KK Mice)

  • This study was carried out to investigate the supplementary effects of the rice germ oil compared with soy bean oil on lipid metabolism of non-insulin dependent diabetic mice. Forty diabetic KK mice were fed two kinds of experimental diets with 20% lipid from soy bean oil as a control(CO) and rice germ oil(RG) for 8 weeks, respectively. Diet intake, body weight, organs weights and lipids levels of serum, liver and feces were measured. There was no significant difference in food and water intake, body weight gain and organs weights between experimental groups. But the concentrations of serum total cholesterol and LDL-cholesterol were lower in RG group than in CO group. The hepatic total lipid and total cholesterol levels of RG group were significantly lower than those of CO group. The contents of total lipid, triglyceride and total cholesterol excreted in feces of RG group were higher than those of CO group. These results suggested that rice germ oil can reduce serum total cholesterol and LDL-cholesterol and hepatic total lipid concentration of non-insulin dependent diabetic mice compared with soybean oil due to increasing fecal lipid excretion. But we need to investigate the hypolipidemic effect of this oil by supplementary level and period.

근육식품에서 지방산화에 대한 피틴산, 저장기간 및 온도의 영향 (Effects of Phytic Acid Content, Storage Time and Temperature on Lipid Peroxidation in Muscle Foods)

  • Phytic acid, making up 1~5% of the composition of many plant seeds and cereals, is known to form iron-chelates and inhibit lipid peroxidation. Thiobarbituric acid reactive substances (TBARS), as an indication of lipid peroxidation, were measured in beef round, chicken breast, pork loin, and halibut muscle after the meats were stored for 0, 1, 3, 5, and 7 days at various temperatures [frozen (~2$0^{\circ}C$), refrigerator (4$^{\circ}C$), and room temperature ($25^{\circ}C$)]. Phytic acid effectively inhibited lipid peroxidation in beef round, chicken breast, halibut, and pork loin muscle (p<0.05). The inhibitory effect of phytic acid was dependent on concentration, storage time, and temperature. At frozen temperature, the inhibitory effect of phytic acid was minimal, whereas at room temperature, the inhibitory effect of phytic acid was maximal, probably due to the variation of the control TBARS values. At the concentration of 10 mM, phytic acid completely inhibited lipid peroxidation in all the muscle foods by maintaining TBARS values close to the level of the controls, regardless of storage time or temperature (p<0.05). The rate of lipid peroxidation was the highest in beef round muscle, although they had a close TBARS value at 0 day. Addition of phytic acid to lipid-containing foods such as meats, fish meal pastes, and canned seafoods may prevent lipid peroxidation, resulting in improvement of the sensory quality of many foods and prolonged shelf-life.

  • Lipid oxidation is one of the most important non-microbial causes of meat quality deterioration. However, there have been different/conflicting views concerning the primary catalysts of lipid oxidation in meat. This presentation provides brief overviews of lipid oxidation mechanism in general and catalysis of lipid oxidation in meat, and then focuses on inter-species differences in lipid oxidation potential, using results from our studies on meats (beef, pork and chicken) at retail and the respective meats of uniform postmortem history. The inter-species differences have highlighted the relative roles of meat pigment (myoglobin) content, catalase activity, and the concentration of oxidation substrates (particularly polyunsaturated fatty acids) in determining the lipid oxidation potential of raw meat versus cooked meat.

  • Membrane lipid peroxidation processes yield products that may react with DNA to cause mutations. Lipid hydroperoxides from linoleic acid in the presence of transition metal ions caused strand breaks in plasmid DNA. DNA damage induced by reactive aldehydes known to be produced by decomposition of lipid hydroperoxides, such as 4-hydroxynonenal or rnalondialdehyde, was repaired by endonucleases and exonuclease III which resulted in the increase of single strand breaks in DNA. Lipid hydroperoxides as well as malondialdehyde and 4-hydroxynonenal also caused mutations in the pUC18 lacZ' gene when measured as a loss of ${\alpha}-cornplementation$. In conclusion. the lipid peroxidation could be an important intermediary event in DNA damage and mutation by oxidative stress.

  • The present study was carried out to investigate the effects of magnesium supplement levels and periods on lipid metabolism in male Sprague-Dawley rats given low protein and magnesium deficient diets. The effect of magnesium supplement levels and periods on lipid metabolism in rats given a low protein and magnesium deficient diet for 2 weeks were investigated. Serum total lipid and triglyceride contents were significantly lower in magnesium supplement group compared with magnesium deficient group. Serum HDL-cholesterol/total cholesterol ratio was significantly increased as magnesium supplement level was increased. Liver total lipid, triglyceride, total cholesterol and phospholipid contents were significantly lower in magnesium supplement group than those in magnesium deficient group. Serum ALP, GOT and GPT activities were significantly decreased in magnesium supplement group compared with magnesium deficient group. In summary, the effect of magnesium supplement on lipid metabolism and enzyme activities were significant and we can see that magnesium supplement level propered to be requirement level(400 mg/kg diet)in the other cases except serum lipid contents.

탄닌 투여시 인체의 혈청 지질개선 효과에 미치는 영향 (Effects of Tannin on Lipid metabolism in 6 College Women)

  • In order to observe the effect of tannin on plasma lipid status in human, tannin was supplemented with increasing amounts from 0 to 0.5, 1.0, 2.5g/d early week until 4th week. The results obtained from this research are a follows. Concentration of total lipid in plasma was significantly lower compared to the control group when the level of tannin supplementation was 0.5g/day while the level of tannin 1.0g/day and 2.5g/day did not affect to change the total lipid level. There seems to be no obvious improvement of the plasm lipid status respect to different tannin levels. Despite the result, the excretion of total bile acids in feces was increased and total lipid in plasma was reduced significantly during first time supplementation compared the control.

Combined Effects of Sex Hormones and Dietary Oils on Lipid Peroxidation

  • The effects of orchildectomy with/without testosterone replacement in wale rats, and ovarectomy with estrogen replacement in female rats on lipid peroxidation were studied in male and female rats fed with diets fortified with 20% w/w, soybean oil or palm oil for 4 months. Serum, liver and heart homogenates were assayed for malonaldehyde and conjugated diene levels. Orchidectomy was found to reduce levels of lipid peroxidation products in the serum, liver and heart. Testosterone replacement did not increase the lipid peroxidation products to levels in the non-orchildectomised rats, while estrogen did not influence lipid peroxidation significantly. Palm oil decreased, but soybean oil increased lipid peroxidation in the liver and heart of both the castrated and sex hormone-replaced male and female rats.
