Triglyceride solid lipid with medium chain fatty acid, tricaprin (TC), was used as a core matrix of lipid nanoparticles (LN) to solubilize water-insoluble paclitaxel and enhance the stability of nanoparticles by immobilization of incorporated drug in the solid core during storage at low temperature. In the present study, TC-LN containing paclitaxel was prepared by hot melt homogenization method using TC as a core lipid and phospholipids as stabilizers. The particle size of TC-LN containing paclitaxel was less than 200 nm and its zeta potential was around -40 mV. Calorimetric analysis showed TC core could be solidified by freezing and thawing in the manufacturing process in which the hot dispersion should be prepared at elevated temperature and subsequently cooled to obtain solid lipid nanoparticles. The melting transition of TC core was observed at $27.5^{\circ}C$, which was lower than melting point of TC bulk. The particle size of TC-LN remained unchanged when kept at $4^{\circ}C$. Paclitaxel containing TC-LN showed comparable anticancer activity to the Cremophore ELbased paclitaxel formulation against human ovarian (OVCAR-3) and breast (MCF-7) cancer cell lines. Thus, lipid nanoparticles with medium chain solid lipid may have a potential as alternative delivery system for parenteral administration of paclitaxel.

Apolipoprotein A-I and A-IV are protein constituents of high-density lipoproteins although their functional difference in lipoprotein metabolism is still unclear. To compare anti-atherogenic properties between apoA-I and apoA-4, we characterized both proteins in lipid-free and lipidbound state. In lipid-free state, apoA4 showed two distinct bands, around 78 and $67{\AA}$ on native gel electrophoresis, while apoA-I showed scattered band pattern less than $71{\AA}$. In reconstituted HDL (rHDL) state, apoA-4 showed three major bands around $101{\AA}$ and $113{\AA}$, while apoA-I-rHDL showed almost single band around $98{\AA}$ size. Lipid-free apoA-I showed 2.9-fold higher phospholipid binding ability than apoA-4. In lipid-free state, $BS_3$-crosslinking revealed that apoA-4 showed less multimerization tendency upto dimer, while apoA-I showed pentamerization. In rHDL state (95:1), apoA-4 was existed as dimer as like as apoA-I. With higher phospholipid content (255:1), five apoA-I and three apoA-4 were required to the bigger rHDL formation. Regardless of particle size, apoA-I-rHDL showed superior LCAT activation ability than apoA-4-rHDL. Uptake of acetylated LDL was inhibited by apoA-I in both lipid-free and lipid-bound state, while apoA-4 inhibited it only lipid-free state. ApoA-4 showed less anti-atherogenic activity with more sensitivity to glycation. In conclusion, apoA-4 showed inferior physiological functions in lipid-bound state, compared with those of apoA-I, to induce more pro-atherosclerotic properties.
A 10-week feeding trial with four dietary protein levels (22%, 32%, 42% and 52%) and two dietary lipid levels (8% and 17%) was conducted to investigate the optimum dietary protein and lipid levels for growth of long-nosed barbel fingerlings. Survival rate of fish was not affected by either the dietary protein or the dietary lipid level. Weight gain and feed efficiency were affected by the dietary protein level (P<0.01), but not by the lipid level, and increased with the dietary protein level at the both lipid levels. Weight gain and feed efficiency of fish fed the 52% protein diets with 8-17% dietary lipids were not significantly different from those of fish fed the 42% protein diets with 8-17% dietary lipids and 32% protein diet with 17% dietary lipid. Daily feed intake of fish was not affected by either dietary protein or dietary lipid level. Protein efficiency ratio and protein retention rate of fish fed the 32% protein diet with 17% dietary lipid were significantly higher than those of fish fed the 52% protein diets with 8-17% dietary lipids. Moisture content of fish fed the diets containing 8% lipid were higher than those of fish fed the diets containing 17% dietary lipid at each protein level. Crude lipid content of fish fed the diets containing 17% dietary lipid were higher than that of fish the fed the diet containing 8% dietary lipid at each protein level. The results of this study indicated that 32% protein and 17% lipid could be the optimum dietary level for growth of juvenile long-nosed barbel.

A feeding trial was conducted to determine the optimum dietary protein and lipid levels for the growth of juvenile muddy loach Misgurnus anguillicaudatus. Eight experimental diets (designated as P20L7, P20L14, P30L7, P30L14, P40L7, P40L14, P50L7 and P50L14) were formulated to contain 20%, 30%, 40% and 50% protein with 7% and 14% lipid. Three replicate groups of muddy loach (1.5 g/fish) were fed one of the experimental diets ad libitum for 8 weeks. Survival of fish fed the P20L14 diet was lower than in other groups (P<0.05). Specific growth rate (SGR) and feed efficiency (FE) of fish were affected by dietary protein (P<0.05) and lipid (P<0.005) levels. SGR and FE of fish tended to decrease with increasing dietary lipid level at the same protein level. There were no significant differences in SGR and FE among fish fed diets containing different protein levels ranging from 30% to 50% with the same lipid level. Whole body moisture and lipid contents were affected by dietary protein and lipid levels. Crude lipid content of fish fed the 14% lipid diets was higher than that of fish fed 7% lipid diets at each protein level. The results of this study indicate that a diet containing 30% protein with 7% lipid would be suitable for the optimum growth and effective feed utilization of juvenile muddy loach.
This study was designed to find out the effects on the lipid metabolism of two herbs used in folk treatment in Korea The two herbs were A: Equisetum arvense L. (Seddugi), B: Eunymus alatus Siebold (Hwasalnamoo). The experiment was performed with the male rats of Sprague-Dawley which fed on high calorie and animal fat supplemented diets from which 30% of the total calories can from lard. Experimental groups were divided into standard (ST) and two other(A, B) groups for the each herb. Each of the two experimental groups consisted of three subgroups (I, II, III) according to the three different doses of the folk remedy used, which were five times and ten times the dose commonly used in a folk remedy. During the experimental period, total food intake and body weight gains were measured, and FER, CER, PER were calculated at the same times. Serum levels of triglyceride, total lipid, ttal cholesterol, HDL-cholesterol, and total lipid as well as cholesterol contents of the liver were measured at the time of sacrifice. Epididymal fat pads were weighted and an indes was calculated. The results of the effects on the lipid metabolism were as follows : 1.Total food intake and body weight gains increased in group A & B. 2. The serum levels of the total lipid and triglyceride decreased in AIII, BI and BIII.3. The serum level of cholesterol decreased in group B, while level of HDL-Cholesterol were increased in all groups and most effectively in group B. 4. The contents of total liver lipid increased in groups A&B. cholesterol decreased in groups A&B. In conclusion, effects on the lipid metabolism associated with the kinds of herbs a diet were the increase of body weighta and the decrease of total lipid and cholesterol levels in serum and liver.
In the presence of a photosensitizer, rose bengal, phooxidatlon in serum total lipids has been studied and the effects of ginseng water extract and saponins on it have reviewed. In the presence of rose bengal, serum total lipids undergo photooxidation and produce lipid hydroperoxides. On the other hand, ginseng water extract and dial saponins largely inhibit photooxidation and decrease the amount of lipid hydroperoxides in serum total lipids.
In order to elucidate the mechanism of the leaf-burning disease of ginseng (Panax ginseng C.A. Meyer), the relationships between thylakoid membrane peroxidation and chlorophyll bleaching were investigated in comparison with the ones of soybean (Glycine max L). When I measured the rate of lipid peroxidation in the thylakoids of ginseng and soybean by irradiation of light(60 w.m-2), it was identified that, the remarkably lower rate of lipid peroxidation was found in the ginseng thylakoid than the case of soybean. When lipid peroxidation of ginseng thylakoid was induced in the dark, chlorophyll contents of thylakoid was not changed. The results suggest that lipid peroxidation does not affect the chlorophyll bleaching in ginseng thylakoid. Thylakoid membrane peroxidation as well as chlorophyll bleaching was closely related with photosynthetic electron transport. But, according to the quenching experiment active oxygen species induced lipid peroxidation may be different species in the case of chlorophyll bleaching.
Synaptic vesicles (SVs) are key structures for synaptic transmission in neurons. Numerous membrane-associated proteins are sorted from the Golgi complex to the axon and the presynaptic terminal. Protein-protein and protein-lipid interactions are involved with SV targeting in neurons. Interestingly, many SV proteins have lipid binding capability, primarily with either cholesterol or phosphoinositides (PIs). As examples, the major SV protein synaptophysin can bind to cholesterol, a major lipid component in SVs, while several other SV proteins, including synaptotagmin, can bind to PIs. Thus, lipid-protein binding plays a key role for the SV targeting of synaptic proteins. In addition, numerous SV proteins can be palmitoylated. Palmitoylation is thought to be another synaptic targeting signal. Here, we briefly describe the relationship between lipid binding and SV targeting.
Lipoprotein lipase (LPL) I san enzyme that catalyzed the hydrolysis of triacylglycerols of chylomicrons and VLDL to produce 20acylglycerols and fatty acids. The enzyme, LPL, is localized on the surface of the capillary endothelium and is widely distributed in extrahepatic tissues including heart, skeletal muscle and adipose tissue. LPL has been isolated from boving milk by affinity chromatography on heparin-separose in 2 M NaCL, 5mM barbital buffer, pH 7.4. To elucidate the lipid-binding regin, LPL was digested with trypsin and then separated by gel filtration. Lipid binding region of LPL has been investigated by recombining LPL peptides with DMPC vesicles. Proteolytic LPL fragments with DMPC were reassembled and stabilized by cholate. Lipid-binding region of LPL was identified by a PTH-automated protein sequencer, as AQQHYPVSAGYTK. The analysis of the secondary structure of the lipid-binding peptides revealed a higher probability of $\alpha$-helix structure compared to the whole LPL protein. The prediction of hydrophobicity of lipid -binding region was highly hydrophobic (-1.1) compared to LPL polypetide(-0.4).
The experimentally cultivated potatoes of 4 varieties, Irish Cobbler, Warba, Shimabara, and Saco were dried in a frozen state, powdered subsequently and subjected to the extraction of free and bound lipids. Constituents of the prepared lipids were fractionated, quantified, and compared by the methods of column and gas-liquid chromatographies. The results were summarized as follows : 1. The total crude lipid content in potato on a dry weight basis was 0.57 % of which 0.2 % was free lipid and 0.37 % was bound lipid. 2. The neutral lipid content in the free lipid was 14.9 %, approximately 3 times as much as the 4.5 % contained in the bound lipid, whereas the glycolipid content in the free lipid was 15.1 %, slightly less than 22.2 % contained in the bound lipid. However, the phospholipid content was 33.9 % in the bound lipid, approximately 4.5 times as much as the 7.2 % contained in the free lipid. This fact revealed that the bound lipid consisted mainly of polar lipid, while the free lipid consisted of neutral lipids, glycolipids and phospholipids in about the same proportion. 3. The main fatty acids constituting more than 90 % in the free and bound lipids were linoleic, palmitic and linolenic acids. The content of the saturated fatty acid was slightly less in the free lipid than in the bound lipid, whereas the unsaturated fatty acids were more abundant in the free lipid.
