  • The purpose of this study was to an in estigate the antibiotic susceptibiliity of Actinoobacillus actinomycetemcomitans isolated from the subgingival plaque to adult periodontitis. Seven bacterial strains were tested for their susceptibility to 10 antimicrobial agents under disk diffusion method and broth dilution methold, Seven patients with deep pocket(6mm) were selected for this study. They had not taken antibiotics for 6 months and no history of dental treatment for 6 months and no history of dental treatment for 6 months before this study. The results were as follows : 1. For the antibiotic disk diffusion method, seven A. actinimycetemcomitans were tested with 10 antimicrobial agents which comprised penicillin, gentamycin, clindamycin, lincomycin, ampicillin, erythromycin, tetracycline, amikacin, chloramphenicol, and vancomycin. The sensitive antibiotics were tetracycline, vancomycin, and chloramphenicol and the resistant antibiotics were clindamycin, and lincomycin. The other antimicrobial agents were less active. 2. From the study of determination on the minimal inhibitory concentration(MIC) by broth dilution method, the MIC of tetracycline to seven strains of the A. actinomycetemcomitans was $0.5-1.0{\mu}g/ml$, that of clindamycin was $32{\mu}g/ml$. These data suggest that tetracycline may be valuable drugs in the elimination of A. actinimycetemcomitans from the patients with adult periodontitis

  • Lincomycin, an inhibitor of plastid protein synthesis, was found to block the synthesis of apoprotein P700 with a molecular mass of 72 kDa and the assembly of the Chl a-protein of PS I. Synthesis of the polypeptides of 48, 43.5, and 32 kDa of the PS II complex is also suppressed. This process is accompanied by the disappearance of the PS Two reaction center Chl a at 683 nm, and of the PS One reaction center Chl a at 690, 696, and 705 nm on the fourth derivative of the absorption spectra at 77K. Lincomycin does not affect the synthesis of LHC subunits. It increases the content of the two main Chl forms of LHC at 648 nm (Chl b) and 676 nm (Chl a). The low-temperature fluorescence ratio F736/F685 is also increased. However, the effect of cycloheximide (an inhibitor of cytoplasmic protein synthesis) leads to the reduction of polypeptides of the light-harvesting Chl a/b-protein complex in the range of 29.5-22 kDa. Under these conditions, the relative amount of Chl b and the F736/ F685 fluorescence ratio decrease significantly. This is obviously the result of blocking the LHC I and LHC II synthesis. At the same time rifampicin and actinomycin D (inhibitors which block transcription in chloroplast and nuclear genome, respectively) inessentially affect the characteristics of these complexes.

  • These studies were conducted to investigate the effect of dietary supplemental lincomycin at different food waste supplements on performance of broiler chicks. Diets were based corn, soybean meal and contained 3,100kcal/kg ME, 21% CP for the rest two weeks. Three levels of dietary lincomycin(LM; 0,2.2, 4.4mg/kg) and food waste(FW; 0, 7.5, 15%) were respectively inclusion in both experiments. In experiment 1, weight gain of birds fed 4.4mg/kg LM was significantly higher than no LM treatments(P<0.05). It tended to increase in 7.5% food waste treatment compared to that of control, but was not significantly different. FCR of chicks fed 2.2mg/kg LM supplement was significantly improved compared to that of other treatments Chicks fed food waste showed inferior FCR to control as dietary FW level increased(P<0.05). There was no significance in blood components among treatments. In experiment 2, weight gain of chicks fed 15% FW exhibited higher weight gain than that of other treatments. As FW supplement increased, the feed intake increased significantly(P<0.05) There was no significance in blood components except serum calcium. The results of these experiments indicated that hicks fed 4.4mg/kg LM supplements maximized growth rate of all treatments, and improved FCR in 2.2mg/kg LM.

  • During the period from August through October, 1988, 50 isolates of Edwardsiella tarda were isolated from 6 diseased cultured Channa argus in Dunchi island and Myung-ghi, near Pusan in Korea were examined by studying their biochemical and antibiotical reactions. The ill animals moved slowly and irregular-formed swimming at the surface of the corner. The symthoms were necrosis with hemorrhage on the body surface, head, gill region, and mouth. Some fish were observed dropsy of the belly. The bacteria grew slowly on Double Salmonella-Shigella agar, 24h, at $37^{\circ}C$ to form relatively small size (2mm diameter), smoothed and convexed form with transient or black in center of the colonis. They gave negative reactions to Voges-Proskauer, Simmon's citrate, urea, KCN (in growth), gelatin, arginine dehydrolase, phenylalanine deaminase and many sugars. The isolates showed positive reactions to $H_2S$ (in KIA agar), indol, Methyl-Red, motility, lysine and ornithine decarboxylase, and gas from glucose. 8 drugs tested as chloramphenicol, colistin, gentamicin, kanamycin, lincomycin, nalidixic acid spectinomycin, and tetracycline. All cultures were resistant to colistin, lincomycin and spectinomycin respectibly, but sensitive to kanamycin and nalidixic acid. Three strains showed resistance to chloramphenicol and 2 isolates among them were resistant to two drugs(gentamicin and tetracycline), coincidentally.

  • Streptococcus faecalis var. liquefaciens was examined for the presence of plasmid deoxyribonucleic acid. An analysis by agarose gel electrophoresis revealed the presence of at least four plasmids of approxymately 6.8, 5.2, 2.6, and 2.1 Mdal. Two plasmid cured strains were obtained by novobiocin treatment. SKR2, which lost 5.2 mdal plasmid (pSK2) and 2.1 Mdal plasmid(pSK4) was sensitive to lincomycin and erythromycin. However, all cured strains showed identical response as parental strain in sugar fermentation, temperature sensitivity, proteolytic activity, and liquefaction of gelatin. The results imply that pSK2 or pSK4 is associated with antibiotic resistance of Str. faecalis var. liquefaciens.

  • To understand the characteristics of 29 toxigenic strains of Corynebacterium diphtheriae isolated in Seoul area, type classification, biochemical properties and antibiotic susceptibility pattern to 9 kinds of antibiotics were investigated. The results obtained were summerized as follows; I. Among the 29 strains, gravis type was the overwhelming majority(24 strains), followed by intermedius type(3 strains) and mitis type(2 strains). II. Fermentation of glucose, maltose, lactose, trehalose and mannitol, nitrate reduction and urease were tested. All strains fermented glucose, but not sucrose, lactose, mannitol and trehalose. 9 strains fermented maltose and 20 strains did not. Nitrate was reduced by 28 strains but not by one strain. In urease test one strain showed positive, 28 strains negative. III. Antibiotic susceptibility test to penicillin G, chloramphenical, kanamycin, lincomycin, streptomycin, terramycin, erythromycin and gentamycin were carried out. The MIC of erythromycin(0.025 ${\mu}g/ml$ 26 strains and 0.05 ${\mu}g/ml$ 3 strains) was the lowest, followed by ampicillin, lincomycin and penicillin G. Streptomycin showed the highest MIC.

  • We evaluated the antibiotic susceptibilities, hemolytic activities, and technological properties of 36 Staphylococcus xylosus strains and 49 S. pseudoxylosus strains predominantly isolated from fermented soybean foods from Korea. Most of the strains were sensitive to chloramphenicol, erythromycin, gentamycin, kanamycin, lincomycin, oxacillin, tetracycline, and trimethoprim. However, 23 strains exhibited potential phenotypic acquired resistance to erythromycin, lincomycin, and tetracycline. Based on breakpoint values for staphylococci from the Clinical and Laboratory Standards Institute, >30% of the isolates were resistant to ampicillin and penicillin G, but the population distributions in minimum inhibitory concentration tests were clearly different from those expected for acquired resistance. None of the strains exhibited clear α- or β-hemolytic activity. S. xylosus and S. pseudoxylosus exhibited salt tolerance on agar medium containing 20% and 22% (w/v) NaCl, respectively. S. xylosus and S. pseudoxylosus strains possessed protease and lipase activities, which were affected by the NaCl concentration. Protease activity of S. pseudoxylosus was strain-specific, but lipase activity might be a characteristic of both species. This study confirms the potential of both species for use in high-salt soybean fermentation, but the safety and technological properties of strains must be determined to select suitable starter candidates.

  • In this study, it was found that determined the photocatalytic degradation of antibiotics (lincomycin, LM) with various catalyst composite of titanium dioxide ($TiO_2$), hydroxyapatite (HAP) and germanium (Ge) under UV-A irradiation. At first, various type of complex catalysts were investigated to compare the enhanced photocatalytic potential. It was observed that in order to obtain the removal efficiencies were $TiO_2/HAP/Ge$ > $TiO_2/Ge$ > $TiO_2/HAP$. The composition of $TiO_2/HAP/Ge$ using a statistical approach based on mixture analysis design, one of response surface method was investigated. The independent variables of $TiO_2$ ($X_1$), HAP ($X_2$) and Ge ($X_3$) which consisted of 6 condition in each variables was set up to determine the effects on LM ($Y_1$) and TOC ($Y_2$) degradation. Regression analysis on analysis of variance (ANOVA) showed significant p-value (p < 0.05) and high coefficients for determination value ($R^2$ of $Y_1=99.28%$ and $R^2$ of $Y_2=98.91%$). Contour plot and response curve showed that the effects of $TiO_2/HAP/Ge$ composition for LM degradation under UV-A irradiation. And the estimated optimal composition for TOC removal ($Y_2$) were $X_1=0.6913$, $X_2=0.2313$ and $X_3=0.0756$ by coded value. By comparison with actual applications, the experimental results were found to be in good agreement with the model's predictions, with mean results for LM and TOC removal of 99.2% and 49.3%, respectively.
