해송의 생육을 저해하는 Ca/Al 및 Mg/Al의 한계 비율 (Critical Ratios of Ca/Al and Mg/Al in Nutrent Solution Limiting Growth of Pinus thunbergii)

  • Acid deposition in forest adjacent to the industrial complexes causes soil acidification resulting in the leaching of cations, decreases of buffering capacity and increases of toxic metal concentrations such as Al, Fe, Mn and Cu in soil solution. Changes of nutrient availability equilibria by acid deposition have been known to retard the growth of pine trees. Objective of this research was to assess the critical ratios of Ca/Al and Mg/Al limiting the growth of Pinus thunbergii in the hydroponic culture. The Ca concentration and Ca/Al ratio in stalks of pine tree were increased as increasing Ca/Al molar ratio in the nutrient solution, but were not changed when the Ca/Al molar ratio was adjusted to greater than 1. Growth of Pinus thunbergii was inhibited at the Ca/Al molar ratio lower than l due to the Ca deficiency. The molar ratios of Ca/Al in the needles of Pinus thunbergii showed the similar tendency with the stalks. This indicated that Ca/Al molar ratio of 1 in the growth media was the critical level limiting the growth of Pinus thunbergii. Concentration of Mg and Mg/Al molar ratios in the stalks of pine tree were increased as increasing Mg/Al molar ratio in nutrient solution. Molar ratios of Mg/Al in the needles were increased as increasing Mg/Al ratios in nutrient solution up to 0.83, which was the critical level limiting the growth of Pinus thunbergii.

점도변화에 의한 식물유의 전이 에스테르화 추산 (The Transesterification Prediction of Vegetable Oils Based on Viscosity Changes)

  • The transesterification of vegetable oils into Biodiesel at $60^{\circ}C$ was performed on the rotary viscometer. The overall yield(%) of fatty acid methyl ester from canola oil at optimum conditions was 95%. The viscosities of fatty acid methyl esters were predicted by Orrick and Erbarr's model. The overall yield increased as the viscosities of fatty acid methyl esters decreased. The limiting molar ratio of methanol to oil appeared to be 1:5. The content of sodium hydroxide as the optimum catalyst appeared to be 0.5wt%.

  • Bio-diesel as fatty acid methyl ester was derived from such oils as soybean, peanut and canola oil by lipase catalyzed continuous trans-esterification. So the activation of lipase(Novozym - 435) was kept to be up to 4:1, the limiting molar ratio of methanol to oil under one-step addition of methanol due to the miscibility of oil and methanol through the static mixer for 4hrs and the elimination of glycerol on the surface of lipase by 7wt% silica gel. Therefore the overall yield of fatty acid methyl ester from soybean oil appeared to be 98% at 50$^{\cdot}C$ of reaction temperature under two-steps addition of methanol with 2${\times}$2:1 of methanol to oil molar ratio at an interval of 5.5hrs, 7wt% of lipase, 24 number of mixer elements, 0.2ml/min of flow rate and 7wt% of silica gel.

  • Available nutrients in soil solution play key roles on the growth of plants, but the equilibria in soil solution can be disturbed by acid precipitation. In this study, we investigated the relationships between element concentrations in the needles of Pinus thunbergii and chemical properties of forest soil solutions in the industrial complexes as an effort to find the possible limiting factor(s) causing the forest decline. The Ca/Al molar ratios in needles of Pinus thunbergii collected from the control sites were 18. However, at Onsan and Ulsan industrial complexes, those were decreased to the ranges from 10 to 11 for the one-year old needles and from 9 to 10 for the two-year old needles. The Mg/Al molar ratios showed similar tendencies with the Ca/Al molar ratios of the needles of Pinus thunbergii. In the A horizon, there existed a significant correlation between Mg concentrations in the needles of Pinus thunbergii and Ca/Al molar ratio of forest soil solution. Calcium concentrations in the needles of Pinus thunbergii in the B horizon were also significantly correlated with Ca/Al molar ratios of forest soil solutions. The uptakes of Ca and Mg by Pinus thunbergii were mainly limited by Al in the soil solutions of the A horizon and by Mn and Al in the soil solutions of the B horizon.

공단 인접지역 산림토양용액의 특성 (Characteristics of Soil Solution Extracted from Forest Soils Adjacent to Heavy Industrial Complexes)

  • Forest soils were collected from various sites adjacent to the heavy industrial complexes and soil solutions were extracted to characterize the chemical properties and to find factors affecting forest decline by acid deposition. Concentrations of $NO_3{^-}$, $SO{_4}^{2-}$, $F^-$ and Al in the soil solutions collected from industrial complexes were 2-33 times higher than those from the non-industrial areas. pH and Al concentrations were significantly correlated with $NO_3{^-}$ and $SO{_4}^{2-}$ concentrations. Forest soils from Onsan and Ulsan regions had very low Ca and Mg concentrations in the soil solutions. In these sites, the molar ratios of Ca/Al and Mg/Al were also lower than 2 and 1, respectively. Aluminum concentrations in the A and B horizons were 547 and $683mg\;kg^{-1}$, respectively, which were considered to be high enough to inhibit tree growth. Magnesium deficiency in A horizon and high concentrations of Al and Mn in B horizon were considered as the major limiting factors for tree growth by inhibiting the uptake of Ca and Mg and causing the imbalance of nutrients in both soil solution and trees.

  • By utilizillg whole cell enzyme of the Xantho-monas citri IFO 3835, cephalexin is synthesized directly from 7-amino-deacetoxy cephalosporanic acid (7-ADCA) and phenyl glycine methyl ester (PGM). To date, cephalexin has been manufactu-red by chemical process involving fairly large number of steps to protect the amino group of phenly glycine and carboxyl group of 7-ADCA. However, the enzymatic process involves only a single step with 85% conversion in 90 minutes. The fermentation variables studied indicate that oxygen transfer is limiting step in the enzyme production. Optimum conditions for enzymatic reaction were 37 C, pH 6.0, and the optimum substrate molar ratio of PGM to 7-ADCA was 2. Other variables that are related to the biochemical properties of whole cell enzyme temperature stability, pH stability, kinetic constants, reusing effect, enzyme loading effect were also evaluated.

염화에텐류 화합물 및 전자공여체가 VC 탈염소화 속도에 미치는 영향 (The Effect of Chlorinated Ethenes and Electron Donor on VC Dehalogenation Rate)

  • Anaerobic reductive dehalogenation of perchloroethene (PCE) was studied with lactate as the electron donor in a continuously stirred tank reactor (CSTR) inoculated with a mixed culture previously shown to dehalogenate vinyl chloride (VC). cis-1,2- dichloroethene (cDCE) was the dominant intermediate at relatively long cell retention times (>56 days) and the electron acceptor to electron donor molar ratio (PCE:lactate) of 1:2. cDCE was transformed to VC completely at the PCE to lactate molar ratio of 1:4, and the final products of PCE dehalogenation were VC (80%) and ethene (20%). VC dehalogenation was inhibited by cDCE dehalogenation. Propionate produced from the fermentation of lactate might be used as electron donor for the dehalogenation. Batch experiments were performed to evaluate the effects of increased hydrogen, VC, and trichloroethene (TCE) on VC dehalogenation which is the rate-limiting step in PCE dehalogenation The addition of TCE increased the VC dehalogenaiton rate more than an increase in the $H_2$ concentration, which suggests that the introduction of TCE induces the production of an enzyme that can comtabolize VC.

동물성 유지를 이용한 바이오디젤 생산의 반응조건 (Reaction Condition for Biodiesel Production from Animal Fats)

  • 동물성 유지로부터 알칼리 촉매를 사용한 전이에스테르화 반응에 있어 적정 반응조건을 탐색한 결과는 다음과 같다. 1 L 규모의 회분식 반응기에서 적정 반응조건은 반응온도 65$^{\circ}C$, KOH 1.0% (w/w), 그리고 유지와 메탄올의 몰 비 (1:15)의 조건에서 20분 동안 반응 시에 98.9%의 높은 지방산 메틸에스테르 함유량을 얻을 수 있었다. 또한, 생산한 지방산 메틸에스테르의 물성을 분석해 본 결과, 점도는 4.2 (40$^{\circ}C$, cp)로 감소하였으며, 산가 0.08 (mg KOH/g), 밀도 865 ($15^{\circ}C,\;kg/m^3$)로 국내 바이오 디젤 품질규격을 만족시키는 결과를 얻었다. 알칼리 촉매를 이용한 동물성 유지의 전이에스테르화에 관한 연구를 통하여 식물성 유지와 더불어 바이오디젤의 원료 유지로써 동물성유지의 사용 가능성을 확인할 수 있었다.

Visualization of Gene Transfer into Live Cells Using Fluorescent Semiconductor Nanocrystals

  • We have developed the method for the conjugation of biotinylated DNA to streptavidin-coated QDs. QD-DNA conjugates and a high-sensitive fluorescence imaging technique are adopted to visualize gene transport across the membrane of the live cell in real time. Endocytotic cellular uptake of oligonucleotide and electrically-mediated plasmid DNA transfer into the live cell are monitored by a quantitative microscopic imaging system. Long-term kinetic study enables us to reveal the unknown mechanisms and rate-limiting steps of extracellular and intracellular transport of biomolecules. We designed experimental protocols to conjugate the oligonucleotide or the plasmid DNA to commercially available streptavidin-coated QDs. Gel electrophoresis is used to verify the effect of incubation time and the molar ratio of QDs and DNA on the conjugation efficiency. It is possible to fractionate the QD-DNA conjugates according to the DNA concentration and obtain the purified conjugates by a gel extraction technique.

  • DMSO와 $H_2O$용매 중에서 결합구조가 밝혀진 동공의 크기를 달리한 토륨(IV) 착물들의 전기전도도를 측정하여 해리현상과 전해질의 행동을 알아 보고 DMSO용매중에서 polarography와 cyclic voltammography적 거동을 조사하여 합성착물의 산화환원반응 메카니즘, 가역성을 알아 보고 환원과정에 관여하는 전자수 및 착물의 확산계수를 계산하였다. 그 결과 반양성자성 용매에서 모든 착물들은 1:1전해질로, 물에서는 1:4전해질로 각각 행동하고, DMSO용매중에서 각 착물의 환원반응은 1전자 1단계의 환원반응으로 가역적이며 확산계수는 착물종에 따라 $5.831{\times}10^{-6}{\sim}6.900{\times}10^{-6}$이었다. 그리고 모든 착물의 물분자는 -1.8V(대조전극, SCE)이상에서 분해를 일으켜 수소기체를 발생한다.
