Development of an Efficient In Vitro Screening Method for Selection of Resistant Lily Cultivars Against Fusarium oxysporum f. sp. lilii (백합 구근부패병 저항성 검정을 위한 기내 간편 검정법 개발)

  • The soilborne fungus Fusarium oxysporum f. sp. lilii (Fol) is a serious threat to all lily cultivars, especially infecting bulbs and flowers. It has become increasingly important to develop varieties resistant against the bulb rot disease. Genetic diversity of cultivars and reliable screening methods are required for this purpose. Here, an efficient in vitro screening system for evaluating resistance to Fol in 38 in vitro-grown lily plants was investigated. Various factors including culture conditions of Fol, inoculum density, appropriate plant materials, inoculation method and duration, and incubation period of plant materials after inoculation were combined to optimize the screening method. As a result, we suggest optimal conditions for an in vitro screening system for the selection of Fol-resistant lily cultivars as follows. Fol was grown on potato dextrose agar (PDA) medium for 6 days at $25^{\circ}C$ in darkness and used as working inoculation. Spore suspensions were prepared (inoculum density: $1.0{\times}10^4$ $spores{\cdot}mL^{-1}$), and then leaf segments $1.5{\times}2.0cm^2$ were inoculated by dipping for 22 hours at $25^{\circ}C$ in dark. Later, leaves were cultured on 0.6% agar plates at $25^{\circ}C$ and 50% humidity with a photoperiod of 16 hours light/8 hours dark (fluence rate of $40{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$) to examine the progress of bulb rot. After 7 days, disease levels were classified into indices 1 (no symptom) to 6 (serious bulb rot). Soil inoculation of Fol carried out with resistant or susceptible lily cultivars that had been selected through in vitro screening confirmed the reproducibility of results. Therefore, the in vitro screening method established in this study is efficient and reliable for selection of lily cultivars resistant against bulb rot disease.

'Asian Flame' an Asiatic Hybrid Lily with Unspotted Red Petals (무반점 적색 아시아틱 나리 '아시안플레임' 육성)

  • An Asiatic lily cultivar 'Asian Flame' was bred in 2007 at National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA) of Korea. The crossing was conducted between female parent A94-34 ('Montreux' ${\times}$ 'Anabelle') and male parent 'Fire' by stigmatic pollination at Suwon in 1996. First selection was done and tentatively named as A98-98 in 1998. After in vitro multiplication and bulb production of A98-98 line, growth and flowering characteristic tests has been conducted from 2005 to 2007. The evaluation of characteristics and preference were surveyed at a lily flower show of NIHHS in 2007 in Taean. The new cultivar 'Asian Flame' flowers in middle of June and grows to an average of 122.7 cm in height. Flowers bloom upward-facing, red (RHS 45B) petals. Year-round flowering is possible by storage of the bulb under $-1.5^{\circ}C$ conditions. For forcing culture, it is necessary to add calcium to the fertilizer or remove side scales of large bulb to prevent leaf scorch.

  • An Asiatic lily cultivar 'Yesol' was developed in 2005 at National Institute of Horticultural and Herbal Science (NIHHS), Rural Development Administration (RDA), Korea. The cross was made in 1992 between Asiatic lily 'White Bird', a white colored cultivar, and 'Connecticut King', yellow colored cultivar. Preliminarily selection was done as 'A94-11' in 1994. Multiplication and bulb formation, and characteristic tests were conducted from 1996 to 2004. The evaluation of this line was performed and named as 'Wongyo C1-27' in 2005 that was registered as 'Yesol' to the registration office of Korea Seed & Variety Service. 'Yesol' flowers at the end of June and grows average 32.4 cm. It flowers upward-facing, yellow (RHS, Y7D) with the size of flower 14.7 cm. The average petal length and width are 8.2 cm and 2.6 cm, respectively. Leaves are 4.8 cm long and 2 cm wide. The weight and size of bulb are 18.9 g and 11.8 cm, respectively. Year-round flowering can be obtained by storing the bulb under -1.5$^{\circ}C$ conditions.

  • Cryopreservation is one of the ideal and suitable methods for long-term storage of plant germplasm. The plant contaminated with diseases and pathogens are decreased the multiplication rate, survival rate and high quality of plants after cryopreservation. The aim of this work was to improve a micropropagation method for lily in Korea, which are cultivars and indigenous plant. In the last process of rinsing scales after surface-sterilization, we tried to control the diseases and pathogens lived within the tissue by rinsing in 0.03% sodium hypochlorite (NaClO) instead of sterile distilled water. Bulb scales of Lilium were cultured in vitro on MS medium supplemented with Plant Preservative Mixture (PPM). The newly small bulb formed from bulb-scales was transferred to MS medium. We checked the non-contamination and survival rate after 2 weeks in culture. Non-contamination was shown to be 70 to 90% in formed small bulbs. This study will help to mitigate microbial contamination in Lilium species micropropagation for cryopreservation.

Effects of Bulb Circumference and Cultivar on the Cut Flower Quality in Lily. (백합의 품종 및 구근크기가 절화품질에 미치는 영향)

  • This study was carried out to investigate cut flower quality bulb circumference and cultivars affecting 7 different cultivars to produce high quality cut flower lilies. To determine the effects of bulb circumference on growth and cut flower quality of 7 different cultivars, Lilium oriental hybrid 'Chilli', 'Ismael', 'Medusa', 'Mother's Choice', 'Siberia', 'Sorbonne', and 'Yelloween' were tested. After cut flower quality experiment, the highest 1st flower bud length and cut flower height were 'Medusa' and 'Mother's Choice' respectively. Most plants had over $30^{\circ}$ stem bending angle except for 'Mother's Choice', and 'Ismael'. The study shows that the bigger bulb circumference has the better 1st peduncle diameter, inflorescence length, stem diameter, number of flowers, and fresh weight. Bulb circumference affects the 1st flower bud length, cut flower height and number of flowers. Stem bending angle, however, is affected by neither bulb circumference nor cultivar. In conclusion, growth, cut flower quality and vase life were affected by the bulb circumference rather than cultivar.

Characteristics of Self-compatible Variety from Native Lilium tigrinum Thunberg (우리나라 자생 참나리에서 선발된 자가결실성 2배체 품종의 특성(特性))

  • This study was carried out to breed a self-compatible variety of Tiger Lily from the seedlings originated from Chinju city, Gyeongsangnam-do. The morphological characteristics, ploidy, and the resistance of seedlings to virus infection were investigated. A progeny test was also conducted to examine whether the propagated progenies had the same characteristics as the mother plant. The self-compatible diploid lily variety developed in this study showed a tall type like native triploid lily, Lilium tigrinum, and bulbils were formed on both lily varieties. The morphological characteristics of the flowers and leaves were not much different between the varieties, while the size of the bulbils and bulbs showed significant differences. The percentage of seed generation by self-pollination was 72.6% for the self-compatible variety, while there was no seed generated for native triploid lily. The number of chromosome was 2n = 26 (x = 13, diploid) for the self-compatible variety, while 2n = 39 (x = 13, triploid) in native lily variety. The progenies of the self-compatible diploid lily variety showed the same characteristics as those of its mother plant in morphology, seed germination, and polyploidy. The mother plant of the self-compatible diploid lily variety showed 58% pollen germination and the 2-year-old and 3-year-old progenies showed similar germination percentages. The pollen grains of Korean native triploid lily, however, never germinated.

Cloning of Superoxide Dismutase (SOD) Gene of Lily 'Marcopolo' and Expression in Transgenic Potatoes

  • Differential display reverse transcription PCR (DDRT-PCR) analysis was performed on lily 'Marcopolo' bulb scale for isolation of expressed genes during bulblet formation. Cu/Zn lily-superoxide dismutase (LSOD) of 872 bp gene, with ability to scavenge reactive oxygen in stress environment, was isolated. Northern blot analysis showed expression levels of LSOD maximized 12 days after bulblet formation. Ti plasmid vectors were constructed with sense and antisense expressions of LSOD gene and transformed into potato. Southern blot analysis of transgenic potatoes revealed different copies of T-DNA were incorporated into potato genome. In transgenic potatoes, lily SOD gene was overexpressed in sense lines and not in antisense lines. In native polyacrylamide gel electrophoresis analysis, additional engineered LSOD was detected in sense overexpressed transgenic line only. Transgenic potatoes were subjected to oxidative stress, such as herbicide methyl viologen (MV). Transgenic potato lines with sense orientation exhibited increased tolerance to MV, whereas in antisense lines exhibited decreased tolerance. In vitro tuberization of transgenic potato with sense orientation was promoted, but was inhibited in transgenic potato with antisense orientation.

Inhibition of Penicillium Bulb Rot by Fungicides and Culture Broth of Antagonistic Bacteria during Storage of Lilium Oriental Hybrids (살균제 및 길항세균 배양액 처리에 의한 백합 저장구근의 부패 발생억제)

  • This study was conducted to find out the proper chemicals and doses for controlling the bulb disease of Lilium Oriental hybrids 'Capablanca' and 'Marco Polo' cultivars during storage, and to ascertain any disadvantage of the treatment on shoot growth and flower development. Bulbs of 'Casablanca' and 'Marco Polo' cultivars were dipped in some fungicide solutions, including azoxystrobin, benomyl+thiram, and prochloraz, and culture broth of antagonistic bacteria for one hour. The rot was apparently inhibited by azoxystrobin and prochloraz solution dippings and it was not by benomyl+thiram in bulbs produced domestically. To improve physiological integrity, calcium chloride was treated. However, this treatment had no positive effects on bulb rot of lily. When bulbs were dipped in one-fifth of diluted broth. But the bulb rot was inhibited effectively as compared with untreated during storage. Benomyl+thiram and that with calcium inhibited the sprouting in 'Casablanca' and 'Marco Polo' cultivars during cultivation. Calcium and benomyl+thiram treatments decreased growth of leaf and flower but with no statistically significant difference. Other fungicidal materials treated to bulbs showed no effect on vegetative growth and flower quality.

The first selection was done and was tentatively named as 'A95-68' in 1995. After in vitro multiplication and bulbing production, growth and flowering characteristic tests were conducted from 1996 to 2003. The evaluation of characteristics was performed and named as 'Wongyo C1-19' in 2005 that was registered as 'Yeri' to the registration office of Korea Seed & Variety Service. 'Yeri' flowered at the first of July and grew average 34.6 cm stem in length. Flowers bloomed facing upward, unspotted in petals and deep purple (RHS, RP58A). The size of flower was 13.3 cm. Mean petal length and width was 7.7 cm and 2.7 cm, respectively. Leaves were 5.1 cm long and 0.5 cm wide, respectively. The weight and size of bulb were 9.6 g and 11.7 cm, respectively. Year-round flowering can be done by storing the bulb under $-1.5^{\circ}C$ conditions. For forced cultivation, it was necessary to add calcium to the fertilizer or remove side scales to prevent leaf scorch. It was needed to control Botrytis disease in wet season.

  • An Asiatic lily cultivar 'Apricot King' was bred in 2007 at National Horticultural Research Institute (NHRI), Rural Development Administration (RDA), Suwon, Korea. The cross was made in 2002 between Asiatic lily 'Orlando', a light orange colored cultivar, and 'Corrida', yellow and pink colored cultivar. Preliminarily selection was done as 'A04-72' in 2004. Multiplication and bulb formation, and characteristic tests were conducted from 2005 to 2006. The evaluation of this line was performed and named as 'Wongyo C1-84' in 2007 that was registered as 'Apricot King' to the registration office of Korea Seed & Variety Service. 'Apricot King' flowers at the end of June and grows average 121.6 cm. It flowers upward-facing, dark green throat and yellow orange (RHS 20A) with the size of flower 17.9 cm. Mean petal length and width is 12.8 cm and 3.7 cm, respectively. Leaves are 16.1 cm long and 2.1 cm wide. It shows light orange stigma and pollen. The weight and size of bulb is 64.2 g and 17.6 cm, respectively. For long-term storage, bulbs can be stored under -1 to $-2^{\circ}C$ for year-round forcing.
