Plasminogen Activator 및 Plasmin-like Protease활성도의 변화와 쥐 피부암의 악성 (Relation of the Activities of Plasminogen Activator and Plasmin-like Protease with Malignant Behavior of Skin Tumor of Rats)

  • 피부암의 형성과 피부 조직에서 나타나는 단백질 분해활성의 변화와의 상과관계를 조사하기 위하여, 쥐의 피부에 7,12-dimethylbenzanthracene과 photbor ester를 차례로 처리하였다. 이 결과, 암이 유발된 피부 조직에서는 대조군에 비하여 plasminogen Activator(PA)의 활성도는 약 3배가 높게 나타났다. 더우기, PA의 활성도는 암 유발전(proteolytic activity)의 조직에서도 약6배가 정도 증가하였다. 반면,casein과 insulin의 분해활성도는 암조직에서 현저히 감소되었고 anti-trypsin의 활성도는 대조군의 것과 거의 비슷하게 나타났다. 따라서, PA 및 PLP활성도의 증가현상은 피부암에서 특징적으로 나타나는 것으로 보이며, 이 암의 전이 및 침투에 밀접한 관계가 있는 것으로 사료된다.

Specific Biological Activity of Equine Chorionic Gonadotropin (eCG) Glycosylation Sites in Cells Expressing Equine Luteinizing Hormone/CG (eLH/CG) Receptor

  • Equine chorionic gonadotropin (eCG), produced by the endometrial cups of the placenta after the first trimester, is a specific glycoprotein that displays dual luteinizing hormone (LH)-like and follicle-stimulating hormone (FSH)-like effects in non-equid species. However, in equidaes, eCG exhibits only LH-like activity. To identify the specific biological functions of glycosylated sites in eCG, we constructed the following site mutants of N- and O-linked glycosylation: eCGβ/αΔ56, substitution of α-subunit56 N-linked glycosylation site; eCGβ-D/α, deletion of the O-linked glycosylation sites at the β-subunit, and eCGβ-D/αΔ56, double mutant. We produced recombinant eCG (rec-eCG) proteins in Chinese hamster ovary suspension (CHO-S) cells. We examined the biological activity of rec-eCG proteins in CHO-K1 cells expressing the eLH/CG receptor and found that signal transduction activities of deglycosylated mutants remarkably decreased. The EC50 levels of eCGβ/αΔ56, eCGβ-D/α, and eCGβ-D/αΔ56 mutants decreased by 2.1-, 5.6-, and 3.4-fold, respectively, compared to that of wild-type eCG. The Rmax values of the mutants were 56%-80% those of wild-type eCG (141.9 nmol/104 cells). Our results indicate that the biological activity of eCG is greatly affected by the removal of N- and O-linked glycosylation sites in cells expressing eLH/CGR. These results provide important information on rec-eCG in the regulation of specific glycosylation sites and improve our understanding of the specific biological activity of rec-eCG glycosylation sites in equidaes.

Reactive Oxygen Species Depletion by Silibinin Stimulates Apoptosis-Like Death in Escherichia coli

  • Silibinin is the major active component of silymarin, extracted from the medicinal plant Silybum marianum. Silibinin has potent antibacterial activity; however, the exact mechanism underlying its activity has not been elucidated. Here, we investigated the novel mechanism of silibinin against Escherichia coli. Time-kill kinetic assay showed that silibinin possess a bactericidal effect at minimal inhibitory concentration (MIC) and higher concentrations (2-and 4-fold MIC). At the membrane, depolarization and increased intracellular $Ca^{2+}$ levels were observed, considered as characteristics of bacterial apoptosis. Additionally, cells treated with MIC and higher concentrations showed apoptotic features like DNA fragmentation, phosphatidylserine exposure, and caspase-like protein expression. Generally, apoptotic death is closely related with ROS generation; however, silibinin did not induce ROS generation but acted as a scavenger of intracellular ROS. These results indicate that silibinin dose-dependently induces bacterial apoptosis-like death, which was affected by ROS depletion, suggesting that silibinin is a potential candidate for controlling bacteria.

드렁허리(Monopterus albus) 혈청내 lgM유사 면역글로불린의 구조적 특성 (Structural Characterization of an IgM-like Immunoglobulin in the Serum of Swamp Eel, Monopterus albus)

  • BSA로 면역시킨 드렁허리의 혈청에서 lgM 유사면역글로불린의 정제한 후 그 구조적 특성을 구명하였다. 면역후 항혈청에서는 항원특이 항체능이 증가되었다. 정제된 lgM 유사면역글로불린은 199kD의 분자량을 갖는 단량체 4개가 결합된 800kD의 단백질이었으며, 단량체는 70kD인 H-사슬 2개와 29.5kD인 L-사슬 2개가 결합된 형태였다. 또한 드렁허리의 lgM 유사면역글로불린은 lgM의 특성과 같이 포유류 적혈구에 대하여 약한 응집력을 나타냈다.

Anxiolytic-like Effects of Saponin and Polysaccharide Fractions Extracted from White and Red Ginsengs in the Elevated Plus-Maze Model

  • Ginseng has been widely used for the management of anxiety and emotional instability, but there is little experimental evidence supporting these clinical applications. The anxiolytic-like effect of ginseng saponin and polysaccharide fractions of white (WG) and red ginsengs (RG) was investigated using the elevated plus-maze test. The saponin (SF) and polysaccharide (PF) fractions were orally administered to male ICR mice for 3 days and behavioral test for the anxiolytic activity were performed. SF significantly increased the time-spent open arms and number into the in the open arm entries. However, PF weakly increased the time-spent in the open arms, but did not increase number into the open ann entries. The WG showed more potent anxiolytic-like effect than that of RG. The anxiolytic-like activities were antagonized by flumazenil, but not by esmolol. These findings suggest the saponin fractions of WG and RG promote the anxiolytic-like activity by antagonizing GABN/benzodiazepine receptors in mice.

Expression and phosphorylation analysis of soluble proteins and membrane-localised receptor-like kinases from Arabidopsis thaliana in Escherichia coli

  • Molecular and functional characterization of proteins and their levels is of great interest in understanding the mechanism of diverse cellular processes. In this study, we report on the convenient Escherichia coli-based protein expression system that allows recombinant of soluble proteins expression and cytosolic domain of membrane-localised kinases, followed by the detection of autophosphorylation activity in protein kinases. This approach is applied to regulatory proteins of Arabidopsis thaliana, including 14-3-3, calmodulin, calcium-dependent protein kinase, TERMINAL FLOWER 1(TFL1), FLOWERING LOCUS T (FT), receptor-like cytoplasmic kinase and cytoplasmic domain of leucine-rich repeat-receptor like kinase proteins. Our Western blot analysis which uses phospho-specific antibodies showed that five putative LRR-RLKs and two putative RLCKs have autophosphorylation activity in vitro on threonine and/or tyrosine residue(s), suggesting their potential role in signal transduction pathways. Our findings were also discussed in the broader context of recombinant expression and biochemical analysis of soluble and membrane-localised receptor kinases in microbial systems.

용매에 따른 양송이 추출물의 생리활성 효과 (Physiological Activities of Agaricus bisporus Extracts as Affected by Solvents)

  • 갈색과 흰색 양송이를 열수, 50% 에탄올 및 100% 에탄올 등의 추출용매를 사용하여 건물 중량의 10배에 해당되는 부피(w/v)일 때, 추출물의 생리활성을 탐색하였다 전자공여 작용의 경우 갈색양송이의 50% 에탄을 추출물에서 66.54%의 높은 전자공여능을 나타내었으며, SOD 유사활성을 측정한 결과 갈색양송이의 열수 추출물이 55.42%로 다른 추출물에 비해 높은 활성을 보여주었다. Tyrosinase저해활성의 경우 두 종류 모두 100% 에탄을 추출물에서 높은 저해활성을 보여주었으며, 갈색양송이가 흰색양송이보다 다소 높은 활성을 나타내었다. 특히 비교물질인 0.1% L-ascorbate와 유사한 활성을 보여주었다. 총 폴리 페놀 함량의 경우 두 종류 모두 열수 추출물보다 50%, 100% 에탄을 추출물에서 높은 함량을 나타내었다. 아질산염 소거작용을 측정한 결과 pH 1.2일 때 갈색양송이와 흰색양송이 모두 소거능이 높게 나타났다.

마이크로웨이브 추출조건에 따른 볶음 새송이버섯의 품질 특성 변화 (Change in Quality Properties of Extracts from Roasted Pleurotus eryngii by Microwave-Assisted Extraction Condition)

  • 볶음처리한 새송이버섯의 마이크로웨이브 추출조건, 즉 에탄올 농도, 추출시간 및 마이크로웨이브 power에 따른 품질특성을 살펴보았다. 총 페놀성 화합물 함량, 전자공여능 및 pH 1.2에서의 아질산염소거능은 50% 에탄올로 추출한 경우 높게 나타났다. SOD 유사활성은 75% 에탄을 농도로 추출하였을때 가장 높은 활성을 나타났다. 가용성 고형분 및 총 페놀성 화합물 함량은 추출시간이 증가할수록 증가하다가 7분일 때 가장 높게 나타났으며, 전자공여능은 추출시간이 증가할수록 감소하는 것으로 나타났으며, SOD유사활성 및 아질산염소거능의 경우추출시간에 따른 유의적인 차이가 없는 것으로 나타났다. 마이크로웨이브 power 의 경우 100 W로 추출하였을 때 가용성 고형분, 총 페놀성화합물 함량 및 전자공여능 가장 높게 나타났다. SOD 유사 활성은 25 W일 때 높은 활성을 나타내었다. 그러나 아질산염 소거능은 마이크로웨이브 power에 영향을 많이 받지 않는 것으로 나타났다.

Phenolic Compounds from the Fruit Body of Phellinus linteus Increase Alkaline Phosphatase (ALP) Activity of Human Osteoblast-like Cells

  • Secondary metabolites from the fruit body of Phellinus linteus were evaluated for their proliferative effect on human osteoblast-like cells. 3-[4,5-Dimethylthiazole-2-y1]-2,5-diphenyl-tetraxolium bromide (MTT) assay and alkaline phosphatase (ALP) activity assay were used to assess the effect those isolates on the human osteoblast-like cell line (Saos-2). Activity-guided fractionation led to the isolation of ALP-activating phenolic compounds through the extraction of P. linteus, solvent partitioning, and repeated silica gel and octadecyl silica gel (ODS) column chromatographic separations. From the result of spectroscopic data including nuclear magnetic resonance (NMR), mass spectrometry (MS), and infrared spectroscopy (IR), the chemical structures of the compounds were determined as 4-(4-hydroxyphenyl)-3-buten-2-one(1), 2-(3',4'-dihydroxyphenyl)-1,3-benzodioxole-5-aldehyde (2), 4-(3,4-dihydroxyphenyl)-3-buten-2-one (3), 3,4-dihydroxybenzaldehyde (4), and protocatechuic acid methyl ester (5), respectively. This study reports the first isolation of compounds 1-3 and 5 from P. linteus. In addition, all phenolic compounds stimulated proliferation of the osteoblast-like cells and increased their ALP activity in a dose-dependent manner ($10^{-8}$ to $10^{-1}\;mg/mL$). The present data demonstrate that phenolic compounds in P. linteus stimulated mineralization in bone formation caused by osteoporosis. The bone-formation effect of P. linteus seems to be mediated, at least partly, by the stimulating effect of the phenolic compounds on the growth of osteoblasts.

콩깍지 추출물의 항산화·항염·항비만 및 에스트로겐 유사활성 평가 (Antioxidant, Anti-inflammatory, Anti-obesity and Estrogen-like Activities of Soybean Pod Extracts)

  • In this study, soybean pods of 45 soybean landraces (or varieties) were classified as yellow (19 samples), black (23 samples), or black in green (3 samples) based on soybean seed coat color. Total polyphenol and flavonoid contents were measured, and antioxidant, anti-inflammatory, anti-obesity, and estrogen-like activities were assessed. Total polyphenol and flavonoid content ranges were 24.13-108.03 mg GAE/g and 3.31-72.02 mg CE/g, respectively, and were highest in the black group followed by the yellow group and were least in the black in green group, while ABTS and DPPH activities followed the order black in green > black > yellow. Estrogen-like and estrogen receptor-α activity ranges were 29.06-35.58 pg/mL and 7.05-10.13 pg/mL and were followed the order yellow > black > black in green and black in green > yellow > black, respectively. Nitric oxide (NO) inhibitory and UCP-1 activities followed the same order as estrogen receptor-α activities. Our findings suggest that soybean pods are excellent sources of antioxidants and high-quality functional materials.
