Quantitative Analysis of Icariside E5 and Vanilloyl Icariside E5 from the Seed of Capsicum annuum. L (고추씨로부터 Icariside E5와 Vanilloyl Icariside E5의 함량분석)

  • Capsicum annuum L. (CA) is reported to be the most widely cultivated species of the Capsicum throughout the world. Its fruit and the seeds have been used as spice after drying and grinding. It is reported that the fruit and seeds contain capsaicinoid such as capsaicin and dihydrocapsaicin, carotenoid such as capsanthin and ${\beta}$-carotene with its anti-tumor effect, anti-obesity effect, anti-diabetic action. Present study, we isolated two lignans, icariside $E_5$ and vanilloyl icariside $E_5$ from the seed of CA and validation and content determination were confirmed using High-Performance Liquid Chromatography (HPLC). As a result, the contents of the lignans, icariside $E_5$ and vanilloyl icariside $E_5$ were 0.28%, 0.085%, respectively in the extract.

Isolation and Quantitative Analysis of Schisandrin, Gomisin A and Gomisin M2 From Schisandra chinensis (오미자로부터 Schisandrin, Gomisin A, Gomisin M2의 분리 및 함량 분석)

  • The lignan compound of Schisandra chinensis Baill. (Schizandraceae) has been reported to have a variety of biological activities such as liver protection, neuroprotection, anti-oxidant and anti-diabetes. In this study, we investigated a quantitative analytical method for schisandrin, gomisin A and gomisin $M_2$ isolated from S. chinensis by high-performance liquid chromatography-ultraviolet spectroscopy (HPLC-UV). The isolated compounds were identified by the analysis of 1H- and 13C-NMR spectroscopic data. The results showed that calibration curves of three compounds indicated great linearity with a correlation coefficient ($R^2$) of schisandrin 0.9983, gomisin A 0.9982 and gomisin $M_2$ 0.9986. The limits of detection (LOD) of schisandrin, gomisin A and gomisin $M_2$ were 0.14, 0.07 and $0.05{\mu}g/ml$ and the limits of quantification (LOQ) were 0.42, 0.22 and $0.14{\mu}g/ml$. Intra-day and inter-day precisions of schisandrin were 0.40~1.44%, 0.07~1.02% gomsin A were 0.22~0.52%, 0.10~0.63%, gomisin $M_2$ were 0.40~0.99%, 0.81~2.88%. In result, contents of schisandrin, gomisin A and gomisin $M_2$ in ethanol extract of S. chinensis were $25.95{\pm}0.15$, $2.51{\pm}0.02$ and $2.17{\pm}0.07mg/g$.

Selection of Superior Resources through Analysis of Growth Characteristics and Physiological Activity of Schisandra chinensis Collection (오미자 수집종의 생육특성 및 생리활성 분석을 통한 우수자원 선발)

  • Background: Various Schisandra chinensis (SC) varieties grow in diverse regions in Korea. However, there is no valid scientific evidence of these varieties. This study aimed to select the excellent resources in terms of the growth characteristics, antioxidant activities, and analysis of the active compounds of the SC collection. Method and Results: In total, 154 resources of SC were collected from various regions of Korea. The growth characteristics were measured by the number of fruit bunches, fruit number, and weight of 100 fruits. The antioxidant activities were investigated by analyzing the total flavonoid and total polyphenol contents and the radical scavenging activity of DPPH and ABTS. Schizandrin A, schizandrin c, gomisin a, and gomisin N were analyzed by HPLC. Each resource showed different growth characteristics. Among the antioxidative effects, the highest 20 resources showed high antioxidant activities in selected 29 resources. Analysis of the SC lignan index showed that all resources contained more than 1.16% of active compounds. Conclusions: All of the selected 29 SC resources were shown to have excellent growth characteristics, antioxidant activities, and bioactive compound richness. Especially, SC-004, SC-007, and SC-154 showed the best growth characteristics, and SC-22, SC-40, and SC-45 showed the best antioxidant activities and bioactive compound richness.

Screening of Lignan Compounds and Antioxidant Activity of Chungkukjang Fermented with Defatted Sesame Flour (참깨 탈지박을 첨가하여 제조한 청국장의 생리활성 및 리그난 성분 탐색)

  • This study is on the effect of oil seed by-products added to Chungkukjang. For this, we designed three cases: Chungkukjang was added in with defatted sesame flour before fermented (DSFBF), added with defatted sesame flour after fermented (DSFAF) and with no adding (control). In each case, the common ingredients and the active antioxidant ingredients were examined and compared and the effects were analyzed. According to microanalysis result, carbohydrate content Chungkukjang the DSFAF 24.97%, control 23.86%, DSFBF Chungkukjang 20.21% as compared to control and Chungkukjang DSFAF relatively low carbohydrate content. The moisture contents in DSFBF (55.98%) or DSFAF (52.83%) were higher than that in control (48.89%). Chungkukjang crude ashes in DSFBF (1.48%) or DSFAF (2.41%) were much lower than in control (6.45%). The proportions of crude lipid in DSFBF (3.30%) or DSFAF (3.93%) were higher than in control (1.77%) by about 2%. As for crude protein, the percentage in DSFAF (15.86%) was lower than that of DSFBF (19.03%) or of control (19.03%). There was no meaningful difference in biological activity measurement as total phenolic contents were 1.26 mg/mL in DSFBF, 1.14 mg/mL in DSFAF and 1.26 mg/mL in control. But electron donating ability was meaningfully more active in DSFBF (21.30%) than in control (20.24%). The superoxide dismutase (SOD)-like activity in DSFBF (68.48%) was twice higher than in control (34.01%), which may imply that DSFBF contain some ingredients that can scavenge superoxide anion radically. In hydroxyl radical scavenging activity, DSFAF scores 96.87%, which is the highest with 96.40% in DSFBF and 95.73% in control. Relative antioxidative effects in DSFBF was 47.92%, which is comparable to 47.06% in control. As a result of extraction and quantitative HPLC analysis of sesamin and sesamolin extracted from the samples, DSFBF contained 3.04$\pm$0.21 mg/g of sesamin, which is meaningfully higher than 2.41$\pm$0.14 mg/g in DSFAF. Content of sesamolin was higher in DSFBF (1.36$\pm$0.09 mg/g) than DSFAF (1.12$\pm$0.07 mg/g) or in control. We can conclude that biologically active and effective ingredients could be found more in DSFBF than in DSFAF or in control. This study conveys not only the meaning that oil seed by-products can be used as an ingredient for making Chungkukjang functional food, but also the possibility that oil seed by-products themselves could become excellent functional food.

Characterization of compounds and quantitative analysis of oleuropein in commercial olive leaf extracts (상업용 올리브 잎 추출물의 화합물 특성과 이들의 oleuropein 함량 비교분석)

  • Olive (Olea europaea L.) leaves, a raw material for health functional foods and cosmetics have abundant polyphenols including oleuropein (major bioactive compound) with various biological activities: antioxidant, antibacterial, antiviral, anticancer activity, and inhibit platelet activation. Oleuropein has been reported as skin protectant, antioxidant, anti-ageing, anti-cancer, anti-inflammation, anti-atherogenic, anti-viral, and anti-microbial activity. Despite oleuropein is the important compound in olive leaves, there is still no quantitative approach to reveal oleuropein content in commercial products. Therefore, a validated method of analysis has to develop for oleuropein. In this study, the components and oleuropein content in 10 types of products were analyzed using a developed method with ultra-performance liquid chromatography to quadrupole time-of-flight mass spectrometry, charge of aerosol detector, and photodiode array. The total of 18 compounds including iridoids (1, 3, 4, 14, and 16-18), coumarin (2), phenylethanoids (5, 9, and 11), flavonoids (6-8, 10, 12, and 13), lignan (15), were tentatively identified in the leaves extract based high resolution mass spectrometry data, and the content of oleuropein in each product was almost identical between two detection methods. The oleuropein in three commercial product (A, G, H) was contained more over the suggested content, and it of five products (B, E, H, I, J) were analyzed within 5-10% error range. However, the two products (C, D) were found far lower than suggested contents. This study provides that analytical results of oleuropein could be a potential information for the quality control of leaf extract for a manufactured functional food.

Arctiin inhibits adipogenesis in 3T3-L1 cells and decreases adiposity and body weight in mice fed a high-fat diet

  • BACKGROUND/OBJECTIVES: The purpose of this study was to examine the effects and associated mechanisms of arctiin, a lignan compound found in burdock, on adipogenesis in 3T3-L1 cells. Also, the effects of arctiin supplementation in obese mice fed a high-fat diet on adiposity were examined. MATERIALS/METHODS: 3T3-L1 cells were treated with arctiin (12.5 to $100{\mu}M$) during differentiation for 8 days. The accumulation of lipid droplets was determined by Oil Red O staining and intracellular triglyceride contents. The expressions of genes related to adipogenesis were measured by real-time RT-PCR and Western blot analyses. For in vivo study, C57BL/6J mice were first fed either a control diet (CON) or high-fat diet (HF) to induce obesity, and then fed CON, HF, or HF with 500 mg/kg BW arctiin (HF + AC) for four weeks. RESULTS: Arctiin treatment to 3T3-L1 pre-adipocytes markedly decreased adipogenesis in a dose-dependent manner. The arctiin treatment significantly decreased the protein levels of the key adipogenic regulators $PPAR{\gamma}$ and $C/EBP{\alpha}$, and also significantly inhibited the expression of SREBP-1c, fatty acid synthase, fatty acid-binding protein and lipoprotein lipase. Also, arctiin greatly increased the phosphorylation of AMP-activated protein kinase (AMPK) and its downstream target phosphorylated-acetyl CoA carboxylase. Furthermore, administration of arctiin significantly decreased the body weight in obese mice fed with the high-fat diet. The epididymal, perirenal or total visceral adipose tissue weights of mice were all significantly lower in the HF + AC than in the HF. Arctiin administration also decreased the sizes of lipid droplets in the epididymal adipose tissue. CONCLUSIONS: Arctiin inhibited adipogenesis in 3T3-L1 adipocytes through the inhibition of $PPAR{\gamma}$ and $C/EBP{\alpha}$ and the activation of AMPK signaling pathways. These findings suggest that arctiin has a potential benefit in preventing obesity.

Supercritical Fluid Extraction of Sesame Oil with High Content of Sesamol (초임계 유체를 이용한 세사몰 고함유 참기름 추출 연구)

  • Studies for the commercial production of sesame oil using th supercriticl carbon dioxide were made. Characteristics of sesame oil containing one of natural antioxidant 'sesamol', which only exist at sesame seed were also studied during the supercritical fluid extraction. Among the various factors influencing the sesamol contents in the sesame oil, the roasting time and temperature were checked, because sesamol can be converted from sesamol in through pyrolysis. We found that the sesamol content was increased rapidly under the condition of roasting temperature over $200^{\circ}C$ with longer roasting time. The sesamol content was increased as the temperature and pressure increased, which was caused by increase of solubility of sesamol against sesamol oil. And the sesamol content was increased also with lower speed of supercritical fluid, which increased the contact time with the raw material. The sesamol content was also increased using water increase up to $1\%$ as the entrainer. When the extraction performance with the supercritical fluid was compared to the conventional compressed extraction, the sesamol content was increased up to 11.5 times with the entrainer.
