A Study on Identifying Factors Affecting Improvement for Academic Libraries Multimedia Services Using Six Sigma Methodology (6시그마 도구를 활용한 대학도서관 멀티미디어 서비스 개선 변수 도출에 관한 연구)

  • This study analyzed the process of multimedia services and indexed the core quality characteristics through the tools employed in 'Define' and 'Measure' steps of the 6 Sigma DMAIC methodology. To achieve the goal, as a case study of 'S' University Central Library, first, brainstorming technique was used to tap the librarians, second, interview with the half-time student workers, third, user's opinion on homepage was analyzed and lastly, the interview with the users. This study also drew some potential causal factors which would likely to affect the multimedia service using Process Map and C&E Diagram. Those factors were prioritized using X-Y matrix and Pareto Chart. This study revealed 14 factors affecting multimedia services: 'presence/absence of the persons who are in charge of selection', 'cycle of selection materials', 'presence/absence of seperated multimedia room', 'materials arrangement method', 'notification of popular materials', 'notification of documents arrivals', 'methods of noticing new arrivals', 'methods of ordering materials', 'cycle of checking order status', 'types of persons requesting materials', 'methods of requesting materials', 'checking of the status of the returned materials', 'methods of noticing materials in housed', and 'methods of ways to return overdue materials'.

Identification of a Gene for Aerobic Growth with a SoxS Binding Sequence in Escherichia coli by Operon Fusion Techniques

  • Eight Escherichia coli cells with aerobic growth deflects were isolated by the insertion of ${\lambda}placMu53$, a hybrid bacteriophage of ${\lambda}$ and Mu, which created transcriptional fusion to lacZY. Two of these mutants, CLIO and CLl2, were irradiated with UV to obtain specialized transducing phages. The phages that took out the neighboring chromosomal DNA of the related gene responsible for deflective aerobic growth were identified. The in vivo cloned chromosomal sequence revealed that the mutated gene of CLIO was located at min 34.5 on the Escherichia coli linkage map and 1,599,515 on the physical map. The physical map indicated that there were 7 cistrons in the operon. We named this operon oxg10. The promoter sequence of oxg10 exhibited a possible binding site far SoxS, a transcriptional regulator that activates the transcription of various SoxRS regulon genes. Transferring the oxg10:: ${\lambda}placMu53$ mutation into the wild-type strain, RZ4500, resulted in the inhibition of normal aerobic growth, while the salute mutation in strain MO inhibited aerobic cell growth completely. The full operon sequences of oxg10 were cloned from the Excherichia coli genomic library. The mutated gene of CLl2 was identified to be a sucA gene encoding the ${\alpha}$-ketoglutarate dehydrogenase El component in the TCA cycle.

Molecular Cloning of nifHD from Rhizobium sp. SNU003 (Rhizobium sp. SNU003의 nifHD 클로닝)

  • Genes for dinitrogenase reductase (nifH) and dinitogenase a subunit (nifD) were found to be located on 7.9 kb of EcoRI, 6.5 kb of Sail, 7.3 kb of HindlII and 4.4 kb of Pstl fragments of the genomic blot of Rhizobium sp. SNU003. a symbiotic strain from root nodule of Canavalia lineata. Nine recombinant phage nif-clones were selected from the genomic library constructed by using EMBL-3 BamHI arms of bacteriophage lambda. Among them. Rnif-6 had insert DNA of 15.3 kb. in which 7.6 kb of BamHI!SacI fragment contained nifHD region. Therefore, the 7.6 kb fragment was subcloned into pUC19 and partial restriction map was constructed. As the results, nifH and nifD were found to be located continuously on 4.5 kb of BamHI/BglIl in the genome of Rhizobium sp. SNU003 strain.

  • Molccular cloning of nod genes from Bradvrhizobium sp. SNU001, a nitrogen-fixing symbiont isolated from thc root nodules of soybean (Clycine trim) . was carried out. nod genes were found to be located on thc genome of the symbiont by gcnomic hybridization with 4.5 kb EcoRI/HndIII fragment (nod DABC) of Rhizohium meliloti as probe. Genomic library of this symbiont was constructed using h phage EMBL3-BanlHI vector. from which five nod positive clones were sclectcd by primary and secondary screening methods. The partial restriction map of inserted genomic DNA of h CNS-l(c1one 2) was constructed. and 3.9 kh Bun7HI fragment. which showed strong hybridization signal to the probe, was subcloned into pBS KS(+) plasmid vector. Partial restriction inap ot' a selected subclone (pBjCNS-I) was constructed and nod DABC was found to be located on the 1.8 kb KpnI/Sacl fragment of this subclone.

  • This study was conducted to grasp senior education programs with the purpose of providing senior education services and researching program development of public libraries for elderly population of an aging society. Senior education programs provided by 33 Senior Welfare Centers and 41 public libraries in Seoul were compared in terms of, 6 subjects such as health, literacy/education, information education, death preparation education, hobbies and employment/volunteer. The Senior Welfare Center provided much more senior programs, whereas the public libraries appeared that the number of institutions participating in senior services was very few, and the program provision for elderly population appeared to be extremely insufficient. A specific course schedule of the program of 'making of a happiness map' of death preparation education, which is one theme among them, was developed.

  • Cataloging principles, cataloging rules, and encoding formats should considered LRM acceptance because LRM replaced FRBR as the conceptual model. This study identifies considerations for BIBFRAME acceptance of expression and representative expression attributes in LRM by using literature reviews and expert interviews. Primarily, work in BIBFRAME without expression as entity could map to work and expression of LRM and sustain expression by linking 2 works (work and expression). Second, BIBFRAME must consider association between representative expression attributes and specific expressions whose values can be transferred to the representative expression attributes. Third, representative expression attributes are different according to work types in LRM, and language, media, intended audience, and scale, that can be used as representative expression attributes in BIBFRAME, should be changed in class. Fourth, relation properties should be articulated for expanding networks between expressions originated from work in BIBFRAME. This study analyzes LRM and BIBFRAME by focusing on expression entity and representative expression attributes. More LRM study is needed on cataloging principles and cataloging rules.

Korea Brassica Genome Project: Current Status and Prospective (배추 유전체열구의 현황과 전망)

  • Brassica rape is an important species used as a vegetable, oil, and fodder worldwide. It is related phylogenically to Arabidopsis thaliana, which has already been fully sequenced as a model plant. The 'Multinational Brassica Genome Project (MBGP)'was launched by the international Brassica community with the aim of sequencing the whole genome of B. rapa in 2003 on account of its value and the fact that it has the smallest genome among the diploid Brassica. The genome study was carried out not only to know the structure of genome but also to understand the function and the evolution of the genes comprehensively. There are two mapping populations, over 1,000 molecular markers and a genetic map, 2 BAC libraries, physical map, a 22 cDHA libraries as suitable genomic materials for examining the genome of B. rapa ssp. pekinensis Chinese cabbage. As the first step for whole genome analysis, 220,000 BAC-end sequences of the KBrH and KBrB BAC library are achieved by cooperation of six countries. The results of BAC-end sequence analysis will provide a clue in understanding the structure of the genome of Brassica rapa by analyzing the gene sequence, annotation and abundant repetitive DHA. The second stage involves sequencing of the genetically mapped seed BACs and identifying the overlapping BACs for complete genome sequencing. Currently, the second stage is comprises of process genetic anchoring using communal populations and maps to identify more than 1,000 seed BACs based on a BAC-to-BAC strategy. For the initial sequencing, 629 seed BACs corresponding to the minimum tiling path onto Arabidopsis genome were selected and fully sequenced. These BACs are now anchoring to the genetic map using the development of SSR markers. This information will be useful for identifying near BAC clones with the seed BAC on a genome map. From the BAC sequences, it is revealed that the Brassica rapa genome has extensive triplication of the DNA segment coupled with variable gene losses and rearrangements within the segments. This article introduces the current status and prospective of Korea Brassica Genome Project and the bioinformatics tools possessed in each national team. In the near future, data of the genome will contribute to improving Brassicas for their economic use as well as in understanding the evolutional process.

  • Recently, the technology of autonomous driving, one of the core of the fourth industrial revolution, is developing, but sensor-based autonomous driving is showing limitations, such as accidents in unexpected situations, To compensate for this, HD-map is being used as a core infrastructure for autonomous driving, and interest in the public and private sectors is increasing, and various studies and technology developments are being conducted to secure the latest and accuracy of HD-map. Currently, NGII will be newly built in urban areas and major roads across the country, including the metropolitan area, where self-driving cars are expected to run, and is working to minimize data error rates through quality verification. Therefore, this study analyzes the spatial relationship of reference objects in the attribute structuring process for rapid and accurate renewal and production of HD-map under construction by NGII, By applying the attribute input automation methodology of the reference object in which spatial relations are established using the library of open source-based PyQGIS, target sites were selected for each road type, such as high-speed national highways, general national highways, and C-ITS demonstration sections. Using the attribute automation tool developed in this study, it took about 2 to 5 minutes for each target location to automatically input the attributes of the spatial relationship reference object, As a result of automation of attribute input for reference objects, attribute input accuracy of 86.4% for high-speed national highways, 79.7% for general national highways, 82.4% for C-ITS, and 82.8% on average were secured.

A Study on Ontology and Topic Modeling-based Multi-dimensional Knowledge Map Services (온톨로지와 토픽모델링 기반 다차원 연계 지식맵 서비스 연구)

  • Knowledge map is widely used to represent knowledge in many domains. This paper presents a method of integrating the national R&D data and assists of users to navigate the integrated data via using a knowledge map service. The knowledge map service is built by using a lightweight ontology and a topic modeling method. The national R&D data is integrated with the research project as its center, i.e., the other R&D data such as research papers, patents, and reports are connected with the research project as its outputs. The lightweight ontology is used to represent the simple relationships between the integrated data such as project-outputs relationships, document-author relationships, and document-topic relationships. Knowledge map enables us to infer further relationships such as co-author and co-topic relationships. To extract the relationships between the integrated data, a Relational Data-to-Triples transformer is implemented. Also, a topic modeling approach is introduced to extract the document-topic relationships. A triple store is used to manage and process the ontology data while preserving the network characteristics of knowledge map service. Knowledge map can be divided into two types: one is a knowledge map used in the area of knowledge management to store, manage and process the organizations' data as knowledge, the other is a knowledge map for analyzing and representing knowledge extracted from the science & technology documents. This research focuses on the latter one. In this research, a knowledge map service is introduced for integrating the national R&D data obtained from National Digital Science Library (NDSL) and National Science & Technology Information Service (NTIS), which are two major repository and service of national R&D data servicing in Korea. A lightweight ontology is used to design and build a knowledge map. Using the lightweight ontology enables us to represent and process knowledge as a simple network and it fits in with the knowledge navigation and visualization characteristics of the knowledge map. The lightweight ontology is used to represent the entities and their relationships in the knowledge maps, and an ontology repository is created to store and process the ontology. In the ontologies, researchers are implicitly connected by the national R&D data as the author relationships and the performer relationships. A knowledge map for displaying researchers' network is created, and the researchers' network is created by the co-authoring relationships of the national R&D documents and the co-participation relationships of the national R&D projects. To sum up, a knowledge map-service system based on topic modeling and ontology is introduced for processing knowledge about the national R&D data such as research projects, papers, patent, project reports, and Global Trends Briefing (GTB) data. The system has goals 1) to integrate the national R&D data obtained from NDSL and NTIS, 2) to provide a semantic & topic based information search on the integrated data, and 3) to provide a knowledge map services based on the semantic analysis and knowledge processing. The S&T information such as research papers, research reports, patents and GTB are daily updated from NDSL, and the R&D projects information including their participants and output information are updated from the NTIS. The S&T information and the national R&D information are obtained and integrated to the integrated database. Knowledge base is constructed by transforming the relational data into triples referencing R&D ontology. In addition, a topic modeling method is employed to extract the relationships between the S&T documents and topic keyword/s representing the documents. The topic modeling approach enables us to extract the relationships and topic keyword/s based on the semantics, not based on the simple keyword/s. Lastly, we show an experiment on the construction of the integrated knowledge base using the lightweight ontology and topic modeling, and the knowledge map services created based on the knowledge base are also introduced.

  • To clone the gene coding for steroid $\Delta^1$-dehydrogenase of Arthrobacter simplex, its genomic library was constructed with a , $\lambda$gt11 expression vector and immunoscreened with antiserum against the enzyme. One positive clone was found to carry a 1.6-kb EcoR I restriction endonuclease fragment of A. simplex DNA. The restriction map of the 1.6-kb EcoR I fragment was determined after cloning of the DNA into pBS vector.
