A sampling and estimation method for monitoring poultry red mite (Dermanyssus gallinae) infestation on caged-layer poultry farms

  • Background: The poultry red mite, Dermanyssus gallinae, is a serious problem in the laying hen industry worldwide. Currently, the foremost control method for D. gallinae is the implementation of integrated pest management, the effective application of which necessitates a precise monitoring method. Objectives: The aim of the study was to propose an accurate monitoring method with a reliable protocol for caged-layer poultry farms, and to suggest an objective classification for assessing D. gallinae infestation on caged-layer poultry farms according to the number of mites collected using the developed monitoring method. Methods: We compared the numbers of mites collected from corrugated cardboard traps, regarding with length of sampling periods, sampling sites on cage, and sampling positions in farm buildings. The study also compared the mean numbers of mites collected by the developed method with the infestation levels using by the conventional monitoring methods in 37 caged-layer farm buildings. Results: The statistical validation provided the suitable monitoring method that the traps were installed for 2 days on feed boxes at 27 sampling points which included three vertical levels across nine equally divided zones of farms. Using this monitoring method, the D. gallinae infestation level can be assessed objectively on caged-layer poultry farms. Moreover, the method is more sensitive than the conventional method in detecting very small populations of mites. Conclusions: This method can be used to identify the initial stages of D. gallinae infestation in the caged-layer poultry farms, and therefore, will contribute to establishment of effective control strategies for this mite.

Anticancer Activity of Periplanetasin-5, an Antimicrobial Peptide from the Cockroach Periplaneta americana

  • Cockroaches live in places where various pathogens exist and thus are more likely to use antimicrobial compounds to defend against pathogen intrusions. We previously performed an in silico analysis of the Periplaneta americana transcriptome and detected periplanetasin-5 using an in silico antimicrobial peptide prediction method. In this study, we investigated whether periplanetasin-5 has anticancer activity against the human leukemia cell line K562. Cell growth and survival of K562 cells treated with periplanetasin-5 were decreased in a dose-dependent manner. By using flow cytometric analysis, acridine orange/ethidium bromide (AO/EB) staining and DNA fragmentation, we found that periplanetasin-5 induced apoptotic and necrotic cell death in leukemia cells. In addition, these events were associated with increased levels of the pro-apoptotic proteins Fas and cytochrome c and reduced levels of the anti-apoptotic protein Bcl-2. Periplanetasin-5 induces the cleavage of pro-caspase-9, pro-caspase-8, pro-caspase-3, and poly (ADP-ribose) polymerase (PARP). The above data suggest that periplanetasin-5 induces apoptosis via both the intrinsic and extrinsic pathways. Moreover, caspase-related apoptosis was further confirmed by using the caspase inhibitor carbobenzoxy-valyl-alanyl-aspartyl-[O-methyl]-fluoromethylketone (Z-VAD-FMK), which reversed the periplanetasin-5-induced reduction in cell viability. In conclusion, periplanetasin-5 caused apoptosis in leukemia cells, suggesting its potential utility as an anticancer therapeutic agent.

Improvement Effect of the Water Extract from the Root of Cirsium japonicum var. ussuriense on Type II Collagen-induced Rheumatoid Arthritis Animal Models (엉겅퀴 뿌리 물 추출물의 류마티스 관절염 동물 모델에 대한 개선 효과)

  • Background: The roots of Cirsium japonicum var. ussuriense (RCJ) have been used as traditional medicine in Korea for hematuria and hematemesis. These extracts exert anti-oxidative and anti-inflammatory effects by scavenging for free radical and regulating the inflammatory response. However, the effect of RCJ on rheumatoid arthritis (RA) has not been elucidated. Thus, we evaluated the water extract of RCJ (WRCJ) using type II collagen-induced RA models. Methods and Results: RA was induced by immunization with type II collagen. All experimental materials were orally administered daily for three weeks. The positive control group was administered with 0.2 mg/kg methotrexate (n = 7), while the experimental group was administered with WRCJ (100 or 500 mg/kg, n = 7). Serum levels of TNF-alpha, Interleukin 6 (IL-6), and type II collagen IgG (CII) were measured using ELISA. Administration of 500 mg/kg WRCJ decreased the levels of TNF-alpha, IL-6, and CII. Moreover, WRCJ treatment diminished swelling of hind legs and infiltration of inflammatory cells in RA models' synovial membrane. Conclusions: These results indicate that WRCJ could improve RA, reduce inflammatory indicators and synovial inflammation. However, further experiments are required to determine how WRCJ can influence the signal transduction pathway in RA.

Antioxidant Effect of Edaravone on the Development of Preimplantation Porcine Embryos against Hydrogen Peroxide-Induced Oxidative Stress

  • Edaravone (Eda) is a potent scavenger of inhibiting free radicals including hydroxyl radicals ($H_2O_2$). Reactive oxygen species (ROS) such as $H_2O_2$ can alter most kinds of cellular molecules such as lipids, proteins and nucleic acids, cellular apoptosis. In addition, oxidative stress from over-production of ROS is involved in the defective embryo development of porcine. Previous study reported that Eda has protective effects against oxidative stress-like cellular damage. However, the effect of Eda on the preimplantation porcine embryos development under oxidative stress is unclear. Therefore, in this study, the effects of Eda on blastocyst development, expression levels of ROS, and apoptotic index were first investigated in preimplantation porcine embryos. After in vitro fertilization, porcine embryos were cultured for 6 days in PZM medium with Eda ($10{\mu}M$), $H_2O_2$ ($200{\mu}M$), and Eda+$H_2O_2$ treated group, respectively. Rate of blastocyst development was significantly increased (P<0.05) in the Eda treated group compared with only $H_2O_2$ treated group. And, we measured intracellular levels of ROS by DCF-DA staining methods and investigated numbers of apoptotic nuclei by TUNEL assay analysis is in porcine blastocyst, respectively. Both intracellular ROS levels and the numbers of apoptotic nucleic were significantly decreased (P<0.05) in porcine blastocysts cultured with Eda ($10{\mu}M$). More over, the total cell number of blastocysts were significantly increased (P<0.05) in the Eda-treated group compared with untreated group and the only $H_2O_2$ treated group. Based on the results, Eda was related to regulate as antioxidant-like function according to the reducing ROS levels during preimplantation periods. Also, Eda is beneficial for developmental competence and preimplantation quality of porcine embryos. Therefore, we concluded that Eda has protective effect to ROS derived apoptotic stress in preimplantation porcine embryos.

The Role and Application of Biomarkers and Surrogate Endpoints for New Drug Development : Focused on Diabetes Mellitus and Osteoporosis (당뇨병 및 골다공증 치료제의 효율적인 신약개발을 위한 생체표지자 및 대리 결과 변수의 역할 및 활용)

  • Recently, the FDA (Food and Drug Administration) of the United States and many advanced countries remark biomarkers and surrogate endpoints as a critical path tool on model based drug development. Economic, technical and social profit on model based drug development like a reduction of the length of research and development have been achieved. Therefore we summarize previous studies about biomarkers and surrogate endpoints and suggest a development direction of therapeutic agents. In diabetes mellitus (DM) and osteoporosis, there are remarkable increases in number of patients and most of patients take medicine during their whole lifetime. For this reason, many patients with DM and osteoporosis have a tolerance on their medicine. We expect that research and development on biomarkers and surrogate endpoints will contribute to new drug development on DM and osteoporosis. Biomarkers for DM are blood levels of glucose, insulin, ${HbA}_{1c}$, CRP, alpha-glucosidase, adiponectin and DPP-4. Among these, validated surrogate endpoints for DM are blood levels of glucose, insulin and ${HbA}_{1c}$ Biomarkers for osteoporosis are BMD, BMC, trabecular volume, ICTP, DPD, osteocalcin, the activity of osteoclast and production of osteoblast. The validated surrogate endpoints for osteoporosis are BMD only. This review summarizes all suggested biomarkers and surrogate endpoints in DM and osteoporosis. The biomarkers are classified by drugs, and the method of validation for surrogate endpoints is suggested. This information would contribute to suggest a direction of DM and osteoporosis therapeutic agent development.

Hair follicle development and related gene and protein expression of skins in Rex rabbits during the first 8 weeks of life

  • Objective: We aimed to observe hair follicle (HF) development in the dorsal skin and elucidate the expression patterns of genes and proteins related to skin and HF development in Rex rabbits from birth to 8 weeks of age. Methods: Whole-skin samples were obtained from the backs of Rex rabbits at 0, 2, 4, 6, and 8 weeks of age, the morphological development of primary and secondary HFs was observed, and the gene transcript levels of insulin-like growth factor-I (IGF-I), epidermal growth factor (EGF), bone morphogenetic protein 2 (BMP2), transforming growth factor ${\beta}-1$, 2, and 3 ($TGF{\beta}-1$, $TGF{\beta}-2$, and $TGF{\beta}-3$) were examined using quantitative real-time polymerase chain reaction (PCR). Additionally, Wnt family member 10b (Wnt10b) and ${\beta}$-Catenin gene and protein expression were examined by quantitative real-time PCR and western blot, respectively. Results: The results showed significant changes in the differentiation of primary and secondary HFs in Rex rabbits during their first 8 weeks of life. The IGF-I, EGF, $TGF{\beta}-2$, and $TGF{\beta}-3$ transcript levels in the rabbits were significantly lower at 2 weeks of age than at birth and gradually increased thereafter, while the BMP2 and $TGF{\beta}-1$ transcript levels at 2 weeks of age were significantly higher than those at birth and gradually decreased thereafter. ${\beta}$-Catenin gene expression was also significantly affected by age, while the Wnt10b transcript level was not. However, the Wnt10b and ${\beta}$-catenin protein expression levels were the lowest at 2 and 4 weeks of age. Conclusion: Our data showed that a series of changes in HFs in dorsal skin occurred during the first 8 weeks. Many genes, such as IGF-I, EGF, BMP2, $TGF{\beta}-1$, $TGF{\beta}-2$, $TGF{\beta}-3$, and ${\beta}$-Catenin, participated in this process, and the related proteins Wnt10b and ${\beta}$-Catenin in skin were also affected by age.

The Effect of Hyeonggaeyeongyotang Extract on the Fertility, Early Embryonic Development in Wistar rats by Oral Gavage Administration (형개련교탕(荊芥連翹湯) 추출물의 경구투여(經口投與)가 rat의 수태능(受胎能) 및 초기(初期) 배발생(胚發生)에 미치는 영향(影響))

  • Objectives : this study was to access the effect of Hyeonggaeyeongyotang water extracts, a polyherbal formula has been used as folk medicine, on the fertility and early embryonic development of male and female Wistar rats when administered by oral gavage. Methods : In male rats, Hyeonggaeyeongyotang extract were dosed 4 weeks before pairing and 2 weeks after mating including the mating periods up to termination after necropsy of the majority of the females. In female rats, they were dosed 2 weeks before pairing, and from Day 0 to Day 7 of gestation. This study was conducted in accordance with the recommendations of the KFDA Guideline [2005-60] for Detection of Toxicity to Reproduction for Medicinal Products. Results: 1. No Hyeonggaeyeongyotang extract treatment-related changes on the clinical signs and mortalities, the Food consumptions, the Body weights and gains were demonstrated in all dosed levels tested in this study except for 500ml/kg-dosing male group in which a significant(p<0.05) increase of body gains was detected during day 0-7 after dosing. 2. No Hyeonggaeyeongyotang extract treatment-related changes on the pre-coital intervals, the estrus cycles, the mating index, conception rate and fertility index were demonstrated in all dosed levels tested in this study. 3. No Hyeonggaeyeongyotang extract treatment-related gross findings on reproductive organs, the weights of reproductive organs, histopathological findings on reproductive organs, the corpora lutea number, implantation site number, live fetus number, number of resorpted embryo and pre-and post-implatation loss were demonstrated in all dosed levels tested in this study. Conclusions : Base on the results, it is considered that the NOAEL (No-Observed-Adverse-Effect Level) for fertility and early embryonic development toxicity of Hyeonggaeyeongyotang extract was under 2000ml/kg/day in Wistar male and female rats because there no treatment-related changes on the fertility and early embryonic developmental index were demonstrated in all dosed levels tested.

Chemical Resistance of Striped Rice Borer, Chilo Suppressalis, and Green Rice Leafhopper, Nephotettix cincticeps (이화명충과 끝동매미충의 약제저항성)

  • The study was planned to detect resistance levels of striped rice borers (Chilo suppressalis Walker) and green rice leafhoppers(Nephotettix cincticeps Uhler) from different localities to organophosphate and carbamate insecticides using topical application. Strains of overwintering striped rice borers were collected from 7 different areas in Gyeonggi Province and they were kept under conditions of about $25^{\circ}C$ for 10 days. Overwintered and reared larvae of striped rice borers were compared for response to insecticides. The strains of green rice leafhopper were collected from Iri, Milyang, Pyongtaek, Echeon and Suweon. Insects were multiplied in the lab. All insecticides tested were organphosphates(MPP, MEP and Diazinon) and carbamate (NAC), which have been used in control of rice insect pests for over past 10 years. The results obtained were as follows; a. With MPP compound, resistant levels of the Joam, Suweon, and Echeon strains of C. suppressalis were from 2 to 4 times as compared with the Yangpyong strain. b. With MEP insecticide, resistant levels of the Kimpo, Pyongtaek, Yongin, Echeon, Suweon, and Joam strains of C. suppressalis were from 2 to 6 times as compared with the Yangpyong strain. c. With Diazinon, resistant levels of the Suweon strain of C. suppressalis were 3 times greater than the Yangpyong strain; but the resistant levels of Kimpo, Pyongtaek, Yongin, and Echeon strains approximated the later. 4. The overwintered larvae of C. suppressalis were more tolerant to the insecticides than the larvae reared in the lab. e. With MEP, resistant levels of the Iri, Milyang, and Pyongtaek strains of N. cincticeps to the Suweon strain showed 13, 8, 7 and4 times, respectively. f. With MPP, resistant levels of the Iri and Pyongtaek strains of N. cincticeps were 7 and 4 times as compared with the Suweon strain, respectively. g. With NAC, resistant levels of the several strains of if. cincticeps showed no difference, and seemed not to have developed resistance yet.

Analyzing the Levels of Vocational key competencies Required by Radiological Technologist Job Description in Blind Hiring process (블라인드 채용 시 방사선사 직무기술서에서 요구하는 직업기초능력 수준 분석)

  • The objective of this study was to prepare the baseline data for vocational key competence of radiologists by analyzing the communication skills, problem-solving skills, self-development capability, interpersonal skills, and vocational ethics, which are required by NCS job descriptions for radiologists, of college students majoring in radiology. This study surveyed 79 sophomore and junior college students majoring in radiology at S University from Nov 19 to Nov 30, 2018, using questionnaires and analyzed the levels of vocational key competencies. The analysis results of vocational key competences levels showed that the level of interpersonal skills was the highest (71.2 points for juniors and 74.9 points for seniors). The levels of problem-solving skills and self-development capability were low. Conflict management capability, which is a sub-domain of interpersonal skills, was 77.5 and 80.5 points for juniors and seniors, respectively, which were the highest, while leadership ability was the lowest. When the sub-domains of self-development capability were analyzed, self-recognition capability was the highest (70.0 points for juniors and 74.8 points for seniors) and the career development capability was the lowest. Colleges are required to develop a customized regular curriculum that is well-linked to the clinic in order to improve the vocational key competencies required by the NCS job description to prepare for the blind hiring process. Colleges also put efforts to educate radiologists who can actively cope with rapidly changing medical environment by running various non-regular programs.

The Bacteriological Quality of Seawater in Kamak Bay, Korea (가막만해역 해수의 세균학적 수질 평가)

  • From 1998 to 2008, the bacteriological quality of seawater in Kamak Bay, Korea, was monitored. The total and fecal coliforms in seawater were monitored as bacterial indicators, and the level of fecal coliform contamination was used to evaluate seawater quality. In the southern shellfish growing area of Kamak Bay, the observed fecal coliform geometric mean (GM) and estimated 90th percentile did not exceed the quality standards for fecal coliforms in seawater in Korea and th USA (GM<14 MPN/100 mL, 90th percentile<43 MPN/100 mL). The GM and estimated 90th percentile of the fecal coliform levels in the adjacent northern area of Kamak Bay were higher than in the southern area. Stations near wastewater discharge sites and urban areas in the north had the lowest quality and did not meet the bacteriological seawater quality criteria. The high fecal coliform levels in the Seonso coastal region near wastewater discharges and northern urban areas declined significantly following dredging and operation of a sewage treatment plant.
