A Survey on Mushroom Uses (버섯의 이용실태에 관한 조사연구)

  • The main purpose of this research is to survey mushroom uses by housewives in Taegu metropolitan region to provide basic knowledge for the development of mushroom cooking methods. We have done statistical analyses of 472 questionnaires on mushroom uses. We found the following results: many respondents regarded mushrooms as one of natural, or health foods. The most well-known mushrooms turned out to be Pleurotus ostreatus, Dried Lentinus edodes, Ganoderma lucidum, Agaricus bisporus and Tricholoma matsutake in the order. The higher in income and year of education, the more species of mushroom are known to respondents. Anticancer effect of mushrooms are known very well and 56.4% of respondents eat Pleurotus edodes often. Preference between mushrooms and their characteristics have been turned out to be significantly different. Preferred characteristics are taste, texture and aroma: respondents preferred texture of Pleurotus ostreatus, and taste and aroma of Tricholoma matsutake. Main purchasing place for mushrooms turned out to be market. Majorproblems of mushroom uses are price, development of cooking methods, storage and preservation of mushrooms.

  • Mitochondria in the L. edodes was separated and purified by stepped sucrose density gradient centrifugation. The activity of mitochondrial ATP synthase has been investigated during various illumination times at each wavelength within the range of 400 nm to 700 nm. The stimulation of above activity increased by two times compared with nonilluminated control group when the illumination was given for 15 seconds at 470 nm wavelength. The optimal pH and temperature of this light-induced mitochondrial ATP synthase were 7.5 and $54^{\circ}C$, respectively. The activity of this enzyme increased by 26%, 25% and 14%, respectively, when there were 1 mmole $Fe^{3+}$, 0.5 mmole $Fe^{2+}$, and 5 mmole ${SO_4}^{2-}$ ion, and was inhibited by 5 mmole $Co^{2+}$, 5 mmole $Mn^{2+}$, 1 mmole $Ca^{2+}$, 0.1 mmole $Na^+$, 5 mmole $CN^-$, and 0.1 mmole ${CO_3}^{2-}$ ion. But $Na^+$ and $K^+$ ion did not affect the activity of enzyme.

  • Among the Basidiomycetes, Pleurotus ostreatus 301 and Lentinus edodes 3-1 were chosen because of their good enzyme productivity and rapid mycelial growth in rice straw medium. Their cultural conditions adequate for the enzymes production and effects of various materials and inorganic salts added to thd rice straw media were investigated. L. edodes 3-1 was excellent in productivity of cellulase and xylanase, and P. ostreatus 301 in protease. The optimum conditions for enzyme production were at $30^{\circ}C$ in cellulase production and at $25^{\circ}C$ in xylanase and protease production, with 75% moisture content and 5.0-6.0 initial pH. The appropriate cultural periods, for enzyme production were 30 days and 35 days for P. ostreatus 301 and L. edodes 3-1, respectively. Among the various materials added, defatted soybean, defatted rape seed, or defatted sesame were all effective to enzyme production but reduced mycelial growth. Rice bran was also effective, particularly at 30% concentration. The addition of inorganic salts showed effective to enzyme production. Among inorganic salts, optimum concentration of $CaCO_3$ was 5%, and that of $CaSO_4$ was 2%.

  • Lentinus lepideus, known as train wrecker fungus, has been used for nutritional and medicinal purposes. Recently, commercial cultivation technique and a new cultivar of the mushroom were developed. To investigate the genetic diversity and phylogenetic relationship for identifying the mushroom strains and cultivar, one commercial and 13 strains of Lentinus lepideus from different geographical regions of Korea were analyzed by ITS regions of rDNA and RAPD of genomic DNA. Three strains of Lentinus edodes were also used for the analysis. The size of the ITS1 and ITS2 regions of rDNA from the different strains varied from 173 to 179 bp and 203 to 205 bp, respectively. The sequence of ITS1 was more variable than that of ITS2, while the 5.8S sequences were identical with 156 base pairs. A phylogenetic tree based on the ITS region sequences indicated that selected strains could be classified into four clusters, while 3 strains of L. edodes was divided into a new cluster. Ten primers out of 20 arbitrary primers used in the RAPD-PCR efficiently amplified the genomic DNA. The numbers of amplified DNA bands varied with the primers and strains, with polymorphic DNA fragments in the range from 0.2 to 2.6 kb. The results showed that phylogenetic relationship among Korean strains of Lentnus lepideus is high, but genetic diversity is low.

  • Mitochondria in the L. edodes was purified by linear sucrose density gradient centrifugation. The mitochondrial ATPase activity was investigated by various wavelength illumination for 30 min at dark state. The mitochondrial ATPase activity was stimulated 1.6 fold by 680 nm illumination compared with dark control group. The mitochondrial ATPase activity of different light illumination time at 680 nm was stimulated 2.3 fold at 5 minutes compared with dark control group. Its optimum pH and temperature were found to be 7.5 and $59^{\circ}C$ after illumination for 5 minutes at 680 nm. The mitochondrial ATPase activity was activated by 5 mmol $Fe^{3+}$, 0.1 mmol $Fe^{2+}$, 0.1 mmol $Mg^{2+}$, 0.5 mmol $K^{+}$, and 0.1 mmol $Ca^{2+}$ ion. But, the enzyme was inhibited by 5 mmol $Na^{+}$ ion.

  • The equilibrium moisture content(EMC) of Shiitake mushroom is and important factor because it has a close relationship to storage and drying problems. The determination of the EMC for Shiitake mushroom was made in atmospheres of various constant humidities at four different constant temperatures and the fitting of the five selected EMC models were performed with the experimental EMC data. The desorption equilibrium moisture contents for Shiitake mushroom were increased as the temperature was decreased and the relative humidity was increased. The significant difference of the equilibrium moisture content was appeared between the cap and the stipe of Shiitake mushroom. The Henderson-Thompson model was fitter than the others with the experimental data.

  • This study was carried out to obtain the basic data on artificial culture of Hohenbuehelia petaloides. The optimum medium are glucose peptone medium (GP), Hennerberg medium, Phellinus igniarius medium (PIM), Lentinus edodes medium (LEM), Czapek dox medium. The optimum condition for the mycelial growth was $30^{\circ}C$ and pH 6.0. The carbon sources such as dextrine, fructose and lactose were favorable to mycelial growth. The optimal concentrations of carbon sources are 10% dextrin and fructose. As nitrogen sources, tryptone, casamino acid and histidine appeared to be favorable. The optimal concentrations of nitrogen sources are 1% soy tone and 0.3% ammonium nitrate. The optimal concentration of yeast extract is 0.4%. The mineral nutrients of $KH_2PO_4$, $K_2HPO_4\;and\;MgSO_4{\cdot}7H_2O$ were effective and the optimal concentrations were 0.046, 0.1 and 0.05%, respectively.

Water-solubility of β-Glucans in Various Edible Mushrooms - Research Note -

  • The amount of $\beta-glucans$ in 12 edible mushroom species was determined and their water-solubility was assessed. A large variability in $\beta-glucans$ content was observed in the mushroom species, ranging from 4.71 to $46.20\%$ on a dry basis. Gyrophora esculenta, Lentinus edodes, Coriolus versicolor, Ganodenna lucidum, and Flammulina velutipes had high levels of $\beta-glucans$ Soluble $\beta-glucans$ content, which plays a key role in the physiological effects of mushrooms, also varied greatly according to the mushroom species, ranging from 2.12 to $19.66\%$. Water-solubility of $\beta-glucans$ in the edible mushrooms, as a percentage of total $\beta-glucans$ content varied from 42.55 to $73.35\%$.

  • 표고버섯의 재배는 농촌 부업뿐만 아니라 농한기의 노동력 이용면에서 유리하고, 영양학적으로 혈압강하물질, 빈혈치료물질, 제암물질 등이 함유되어 있음이 밝혀져 영양식품, 또는 건강식품으로 그 진가를 더욱 평가받게 되었으며 주로 동양에서 많이 소비되었으나 서양에서도 소비가 증가하여 수요는 날로 증가하고 있어 표고재배 사업의 전망은 매우 밝다(이, 1982). 그러나 아직까지 고품질 버섯의 생산이나 버섯의 상품성을 높이기 위한 연구들은 미흡한 실정이다. 국내의 표고버섯은 생산량의 50% 이상이 건조되어 유통되므로 표고버섯의 건조특성에 관한 연구는 필수적이며 특히 건조 뿐 아니라 가공 및 저장의 가장 기초적인 자료로 이용되는 평형함수율에 관한 명확한 구명이 요구된다. 버섯 배지를 이용한 tyrosinase 저해제 발효

  • Tyrosinase is an enzyme which catalyzes an enzymatic browning of some foods and in vivo synthesis of melanin. In order to produce natural and edible inhibitor of the enzyme which is expected to have whitening effect on melanogenesis, a microorganism was selected from fermented foods. It was named as NU-7, and cultured in mushroom (Lentinus edodes, Shiitake) media. Optimal media to produce tyrosinase inhibitor was formulated by varing nitrogen or carbon content. If glucose content was in a range of 3-20% and ammonium sulfate was in a range of 0-0.25%, production of inhibitor was independent of cell mass. Addition of ammonium sulfate as a nitrogen source had little effect on inhibitor production. Production of inhibitor (Y) was proportionally related to shiitake content (X) with a regression equation of Y= -0.96X$^{2}$ + 13.07X + 14.43 (R = 0.96). These results indicate that shiitake and glucose are necessary for the production of tyrosinase inhibitor. In the analysis of mycotoxin in culture broth, aflatoxin was not detected, suggesting that it would be probably edible.
