We have examined the feasibility of using the specific interaction between mistletoe lectin I (ML I) and ${\beta}$-Dgalactose instead of the anti-ML I antibody in developing a homogeneous type competitive binding assay for ML I. We also have examined the feasibility of adapting the biotin/avidin mediated homogeneous assay for this system. Alkaline phosphatase (AKP) was employed as a single substrate enzyme label. The dose-response curve shows a detection range of 1-25 ${\mu}$g/mL and a linear response with a correlation coefficient of 0.99. To demonstrate the analytical utility of this method, 10 ${\mu}$g/mL of ML I was spiked into distilled water. The results show that the mean recovery was 10.03 ${\mu}$g/mL with an SD of 0.18. The difference between the spiked value and the mean recovery was 0.03 ${\mu}$g/mL, with a relative error of 0.3 and 1.6 % of RSD.
Histochemical patterns of lectin binding during development of the rat vomeronasal organ (VNO) were studied to determine whether glycoconjugates are differently expressed after birth. Three types of lectins, Dolichos biflorus agglutinin (DBA), wheat germ agglutinin (WGA), and Ulex europaeus agglutinin I (UEA-I), were studied histochemically in the rat VNO at various stages post-birth: postnatal days 1 and 7, the preweaning period (4 weeks after birth), and at sexual maturity (8 weeks after birth). The free border of the vomeronasal sensory epithelium was positive for both WGA and UEA-I in rats of all ages; whereas, VNO receptor cells and supporting cells were positive only for both WGA and UEA-I from 4 weeks after birth. DBA reactivity was detected in the free border but less so in receptor cells and supporting cells. WGA and UEA-I, but not DBA, showed similar patterns in various ages. In the Jacobson's gland, WGA, UEA-I and DBA were detected in some acini from 4 weeks after birth but not at postnatal days 1 or 7. Collectively, reactivity for three lectins, WGA, UEA-I and DBA, increased in receptor cells and gland acini during postnatal development, possibly contributing to the enhanced chemoreception in rats.

A cDNA of novel immulectin homologue (BmIML), a C-type lectin, was cloned from the silkworm, Bombyx mori. The immulectin cDNA is an open reading frame of 921 bp encoding 307 amino acid residues. The deduced amino acid sequence from the BmIML cDNA contains two C-type carbohydrate recognition domains (CRDs). The BmIML was most similar (61 % protein sequence identity) to the M. sexta immulectin-1, whereas BmIML showed relatively lower identity to the B. mori lipopolysaccharide-binding protein (25% protein sequence identity). These features of BmIML indicate that BmIML is a novel member of C-type lectin superfamily. Northern blot analysis revealed that the BmIML is specifically expressed in the fat body of B. moli larvae.
Previous studies have demonstrated that expression of galectin-3, a member of family of beta-galactoside-binding animal lectin, is associated with pathological conditions including cancer, atherosclerosis, and viral infection. An increase of this lectin has been observed after infection by Kirsten murine sarcoma, human T lymphotropic virus-l (HTLV-l), and human immunodeficiency virus-l (HIV-l). Viral transactivation protein Tax of HTLV-l mediates the increase in the lectin. In case of HIV-1, there are evidences that Tat would be related with increase in galectin-3. We investigated whether Tat directly induced galectin-3 expression in cells. We found that HIV-l tat gene activated galectin-3 promoter in RAW264.7 cells. To demonstrate direct induction of galectin-3 by HIV-l tat, we transfected the tat into a rabbit smooth muscle cell line (Rb1) and obtained RblTatCl-2, a clone of cell stably transfected with tat gene. The Rb1TatCl-2 cells exhibited activation of LTR promoter and up-regulation of galectin-3 transcript as well as protein. Our results indicate that HIV-l tat alone is sufficient to induce the expression of galectin-3. The Rb1TatCl-2 cells could be valuable for study of the effect of HIV-1 tat on expression of cellular genes.
진해만에서 분리한 무독성 Alexandrium tamarense와 이미 보고된 독성을 가진 A. tamarense종 간의 접속공, 제1상판, 후속공의 형태를 상호 비교하였다. 형태적으로 독성종과 거의 일치되지만, HPLC나 생체실험에서 진해만에서 새롭게 분리한 종은 독성이 전혀 나타나지 않았다. lectin반응결과 무독종 A. tamarense은 PNA와 강한 반응을 보여, 세포표면에 lactose나 galactose와 같은 물질이 많이 분포하고 있음을 보였다. 또한, 단백질 전기영동시 무독종은 유독종과 달리 21 kDa 정도 부위에서 종 특이적 밴드를 보였다. 따라서 PNA lectin이나 면역학적 방법을 이용하면 무독종 A. famarense을 신속하게 동정하는데 이용될 수 있다.
목 적 : 본 연구는 어린 연령의 소아에서 선천성 면역력의 중요한 인자인 Mannose Binding Lectin(MBL2) 유전자의 다형성 및 일배체형의 분포를 심한 respiratory syncytial virus(RSV) 감염증 환자군과 정상대조군에서 분석함으로써, MBL2 유전자의 다양성이 소아 RSV 감염증의 발생과 예후에 기여하는지를 밝히고자 하였다. 방 법 : 1993년부터 2000년까지 7년 동안 서울대학교 어린이병원 소아과에서 RSV 감염증으로 치료받은 심한 RSV 감염증 환아 99명을 환자군, 건강한 성인 224명을 대조군으로 선정하였다. 유전형 분석은 선천성 면역의 중추적 역할을 하는 MBL2 유전자의 promoter -221 X/Y와 exon 1의 구조 변이의 분포를 환자대조군에서 비교하여 분석하였다. 결 과 : 환자군 99명의 중앙 연령은 9.5개월로, 6개월 미만 48명(49%), 6~24개월 사이 39명(39%), 그리고 24개월 이상이 12명(12%)이었다. 남아는 58명(59%)이었다. 환자군에서 MBL2 대립유전자 B다형성의 빈도는 유전형 AA 60(61%), 이형접합 AB 34(34%), 그리고 유전형 BB 5(5%)로 나타났다. 환자군의 promoter -221 X/Y 유전형의 분포는 유전형 YY 84(85%), 이형접합 XY 15(15%)이었으며, 유전형 XX는 발견되지 않았다. Exon 1의 구조 변이와 promoter 변이 모두 환자군과 대조군간에 차이가 없었다. MBL2 일배체형의 분포에도 두 군간에 차이가 없었다. 결 론 : 본 연구 결과, 한국인 소아에서 MBL2 유전자의 다형성과 일배체형이 심한 RSV 감염증에 기여하는 예후 인자임을 밝히지 못하였다. 향후 전향적으로 수집한 더 많은 수의 환자군과 적절한 대조군을 통한 연구가 시행되어야 할 것으로 생각한다.
인간의 대장내 점막에 대하여 우수한 점착특성을 갖는 probiotic 유산균주를 선별할 목적으로, colonic mucin-binding assay를 고안하고 최적의 분석 조건을 검토한 결과, microtiter plate의 well에 대한 colonic mucin의 부착은 pH 4.8, biotinylated SLP의 농도는 $5.0\;{\mu}g/mL$, 시판 HRP-conjugated streptoavidin은 24,000배 희석용액, TMB의 발색시간은 10분의 조건에서 측정시 최적의 결과를 나타냈다. 동 조건에서 본 assay system을 이용할 경우, 장내 점착능 측정 및 우수 유산균주의 선별에 있어 신속하고 재현성 있는 결과를 제공할 수 있으며, 인간의 대장에 대한 유산균의 점착특성을 정량적으로 분석할 수 있음을 보여주었다. 공시균주 32종 및 유아 분변 유래의 분리균주 18종을 포함한 총 50종의 유산균주에 대하여, colonic mucin-binding assay를 이용하여 대장 mucin에 대한 결합능을 비교한 결과, L. species FSB-1이 가장 높은 결합능을 보여주었다. 따라서 L. species FSB-1을 대상으로 형태학적 특성, 생리 및 생화학적 특성과 16S rDNA에 대한 부분 염기서열 분석을 포함한 동정실험을 수행한 결과, 장내 점착능 우수균주로 선별된 L. species FSB-1은 Lactobacillus brevis로 최종 동정되었다.
Targeted glycoproteomics is an effective way to discover disease-associated glycoproteins in proteomics and serial affinity chromatography (SAC) using lectin and glycan-targeting antibodies shows glycan diversity on the captured glycoproteins. This study suggests a way to determine glycan heterogeneity and structural analysis on the post-translationally modified proteins through serial affinity column set (SACS) using four Lycopersicon esculentum lectin (LEL) columns. The great advantage of this method is that it differentiates between glycoproteins on the basis of their binding affinity. Through this study, some proteins were identified to have glycoforms with different affinity on a single glycoprotein. It will be particularly useful in determining biomarkers in which the disease-specific feature is a unique glycan, or a group of glycans.
A library of unlimited number of novel lectins with diverse specificities has been previously generated by randomly mutating the carbohydrate-recognition domain of Maackia amurensis hemagglutinin (MAH). To establish the experimental environment capable of selecting high affinity mutant lectins in E. coli, phage display system was adapted. Carbohydrate binding capacity of two phagemid vectors, pComb3 and pComb8 displaying wild-type MAH lectin was assessed. Specific bindings of pComb3 and pComb8 phages expressing w.t. MAH to affinity-purified polyclonal anti-MAH antibody and to glycophorin was demonstrated. Both phages also showed strong hemagglutinating activity to intact but not sialidase-treated human erythrocytes, which is consistent to the specificity of native MAH. Taken together, two different phage display vectors successfully allowed the expression of active MAH as a fusion protein on the surface of bacteriophage, which will lead to preparation of unique plant lectins with high affinity toward a variety of carbohydrate chains.
섬유아세포 표면의 미세구조적 특성과 세포표면에 존재하는 당 단백질 말단 GlcNAc(N-acetylglucosamine)와 NeuNAc(N-acetylneuraminic acid)는 섬유세포의 이동과 세포의 인식에 중요한 역할을 하는 것으로 알려졌다. 따라서 섬유세포의 미세구조적 특성을 전자현미경을 사용하여 관찰하였으며, 당 단백질 말단 GlcNAc와 NeuNAc의 분포를 확인하기 위하여 WGA 황금입자 복합체를 반응시켜 전자현미경으로 관찰하였다. 그 결과 배양섬유세포의 표면에 미세구조적 특성은 세포의 분화 정도와 세포의 부위에 따라 다양한 형태를 형성하며, 세포의 배양시간에 따라 정도의 차이는 있으나 일반적인 미세융모의 분포와 세포질 돌기의 분화는 세포의 주위 환경에 따라 다양한 형태로 분화하는 특성이 있는 것으로 확인되었다. 섬유세포의 세포표면에 분포하는 당 단백질 말단의 일종으로 섬유세포의 이동과 세포인식에 관여하는 lectin WGA 수용체 인 sialic acid (GlcNAc; N-acetylgalactosamine, NeuNAc; N-acetyl neuraminic acid)는 세포질의 조면소포체에서 생성되어 액포상태로 이동되어 섬유세포의 외로 분비되고 분비된 sialic acid는 세포의 표면과 돌기의 표면에 당 단백질 말단으로 분화하여 섬유세포의 이동과 세포인식 등의 섬유세포 기능에 관여하는 것으로 규명되었다.
