Cells in a culture process transform with time and produce many overlapping cells in their vicinity. We are interested in a separation algorithm for images of overlapping cells taken using a fluorescence optical microscope system during a cell culture process. In this study, all cells are assumed to have an ellipse-like shape. For an ellipse fitting-based method, an improved least squares method is used by decomposing the design matrix into quadratic and linear parts for the separation of overlapping cells. Through various experiments, the improved least squares method (numerically stable direct least squares fitting [NSDLSF]) is compared with the conventional least squares method (direct least squares fitting [DLSF]). The results reveal that NSDLSF has a successful separation ratio with an average accuracy of 95% for two overlapping cells, an average accuracy of 91% for three overlapping cells, and about 82% accuracy for four overlapping cells.
Fitting the projected wire-frame model to the detected edge pixels on images by using least-squares approach, called Least-squares Model-image Fitting (LSMIF), is the key of the Model-based Building Extraction (MBBE). It is implemented by iteratively adjusting the model parameters to minimize the squares sum of distances from the extracted edge pixels to the projected wire-frame. This paper describes a series of experiments and studies on various factors affect the fitting results, including the edge detectors, the weighting rules, the initial value of parameters, and the number of overlapped images. The experimental result is not only helpful to clarify the influences of each factor, but is also able to enhance the robustness of the LSMIF algorithm.

The size of microarray gene expression data is very big and its observation process is also very complex. Thus missing values are frequently occurred. In this paper we propose the sequential partial least squares(SPLS) regression fitting method to estimate missing values for time course gene expression data that has correlations among observations over time points. The SPLS method is to combine the sequential technique with the partial least squares(PLS) regression fitting method. The usefulness of method proposed is evaluated through some simulation study for three yeast time course data.
We consider the problem of determining the circle of best fit to a set of data points in the plane. In [1] and [2] several algorithms already have been given for fitting a circle in least squares sense of minimizing the geometric distances to the given data points. In this paper we present another new descent algorithm which computes a parametric represented circle in order to minimize the sum of the squares of the distances to the given points. For any choice of starting values our algorithm has the advantage of ensuring convergence to a local minimum. Numerical examples are given.
A new quadratic response surface modeling method is presented. In this method, the incomplete small composite design (ISCD) is newly proposed to .educe the number of experimental runs than that of the SCD. Unlike the SCD, the proposed ISCD always gives a unique design assessed on the number of factors, although it may induce the rank-deficiency in the normal equation. Thus, the singular value decomposition (SVD) is employed to solve the normal equation. Then, the duality theory is used to newly develop the conservative least squares fitting (CONFIT) method. This can directly control the ever- or the under-estimation behavior of the approximate functions. Finally, the performance of CONFIT is numerically shown by comparing its'conservativeness with that of conventional fitting method. Also, optimizing one practical design problem numerically shows the effectiveness of the sequential approximate optimization (SAO) combined with the proposed ISCD and CONFIT.

This paper presents a new circular curve fitting approach of articulated manipulators, based on least square. The approach aims at gaining the interpolation of circle from n data points, under the condition that the fitted circle should pass both a starting point and an ending point. First a spherical fitting should be performed, using least squares. Then the circular curve fitting can be resulted from the intersection of the fitted sphere and the plane obtained from 3 points, i. e., a starting point, an ending point and the center of a sphere. The proposed algorithms are shown to be efficient by using MATLAB-based simulation.

The modeling and parameter estimation of a damped one-degree-of-freedom mass-spring system is examined. This paper presents a method for estimating the system parameters (damping coefficients and natural frequency) from measured free-vibration motion of a system that is modeled to include both subcritical viscous damping and kinetic Coulomb friction. The method applies a commercially available least-squares curve-fitting software function to fit the known solution of the equations of motion to the measured response. The method was tested through numerical simulation, and it was applied to experimental data collected from a laboratory mass-spring apparatus. The mass of this apparatus translates on linear bearings, which are the primary source of light inherent damping. Results indicate that the curve-fitting method is effective and accurate for both perfect and noisy measurements from a lightly damped mass-spring system.

In this paper, we describe a method for fuzzy polynomial regression analysis for fuzzy input--output data using shape preserving operations for least-squares fitting. Shape preserving operations simplifies the computation of fuzzy arithmetic operations. We derive the solution using mixed nonlinear program.

The Total Least Squares (TLS) method is introduced in comparison with the conventional Least Squares (LS) method. The principles and mathematical models for both methods are summarized and the comparison results from their applications to a simple geometric example, fitting a straight line to a set of 2D points are presented. As conceptually reasoned, the results clearly indicate that LS is more susceptible of producing wrong parameters with worse precision rather than TLS. For many applications in surveying, can adjustment computation and parameter estimation based on TLS provide better results.

Classical principal component analysis (PCA) can be formulated as finding the linear subspace that best accommodates multidimensional data points in the sense that the sum of squared residual distances is minimized. As alternatives to such LS (least squares) fitting approach, we produce LMS (least median of squares) and LTS (least trimmed squares)-type PCA by minimizing the median of squared residual distances and the trimmed sum of squares, in a similar fashion to Rousseeuw (1984)'s alternative approaches to LS linear regression. Proposed methods adopt the data-driven optimization algorithm of Croux and Ruiz-Gazen (1996, 2005) that is conceptually simple and computationally practical. Numerical examples are given.
