Control of Stretching of Tomato (Lycopersicon esculentum Mill.) on Cylindrical Paper Pot Seedling Using High-Salinity Potassium Fertilizers (고농도 칼륨처리를 활용한 원통형 종이포트 토마토묘의 도장억제)

  • This study was conducted to examine the potential of inducing salinity stress on cylindrical paper pot tomato seedlings to inhibit overgrowth. Potassium fertilizers, sulfate of potash (K2SO4), muriate of potash (KCl), and monopotassium phosphate (KH2PO4), were prepared as two solutions of (5 and 10) dS·m-1 salinity level, respectively, to investigate the influence on tomato (Lycopersicon esculentum Mill.) seedling growth. We also investigated the adaptability and survivability of treated tomato seedlings with high-salinity potassium (10 dS·m-1 KCl) to harsh environmental conditions (water deficit, low temperature, and storage conditions). Repeated addition of high-salinity level KCl, K2SO4, or KH2PO4 markedly decreased the dry matter of shoot and root, leaf area, and net assimilate rates (NAR) but increased the stem diameter of seedlings. Among the three sources, the relative growth rate of plant height (RGRH) was most sensitive to KCl addition; increasing salinity levels of KCl solution decreased the RGRH of seedlings. The compactness, which directly reflects the stocky growth index, increased in KCl or KH2PO4 treatments. After a week's water deficit, severely wilted seedlings were observed in control seedlings (untreated with KCl), but no wilted seedlings were observed in the KCl treated seedlings, and the relative water content (RWC) of the untreated seedlings significantly decreased by 23 %, while that of the pretreated seedlings only decreased by 8 %. The increase in ion leakage of KCl treated seedlings at low temperatures was less than that of untreated seedlings. Furthermore, there was far lower damage proportion on pretreated seedlings at (9, 12, and 15)℃ storage temperatures after 20 days, compared with on unpretreated seedlings. Our results suggest that high-salinity potassium fertilizer, especially KCl, is effective in preventing tomato seedling overgrowth, while it also improves tolerance.

Control Efficacy of Bacillus thuringiensis against Herpetogramma luctuosalis on 'Campbell Early' Organic Vineyard (비티제를 이용한 포도 '캠벨얼리' 유기과수원 포도들명나방 방제효과)

  • The experiment of grape leafroller (Herpetogramma luctuosalis) was carried out at Okcheon area from 2007 to 2009 and 2015. The grape leafroller had been occurred at a campbell early' organic vineyard in Okcheon. It's larva was rolling the leaf of grape and ate the leaf. So the leaf of grape decreased. In organic vineyards, adult grape leafroller' generation rate per year showed the first peak in mid-June, the second peak in early -August and the third in mid-September. The larva showed the first peak in early July and the second peak in late August-early September. The grape leaf roller had three generations per year. And it took $60.9{\pm}1.09days$ from egg to adult in growth chamber (VS-91G09M-1300) which the relative humidity conditions was $60{\pm}10%$, temperature $25{\pm}2^{\circ}C$ and photoperiod 16L:8D (The egg : $12{\pm}0days$, larvae : $22.2{\pm}0.22days$, pupa : $10.6{\pm}0.75days$ and adult : $16.1{\pm}0.45days$). It was conducted to find out the effect of microbial pesticide treatments to control H. luctuosalis. The 4 microbial pesticides (Bacillus thurigiensis) were treated twice on the grape leaves in June 11 and 21 at an organic vineyard in Okcheon. On 10 days after last treatment, the control value of all microbial pesticides were more than 95%. When the dates of spraying to the grape leaves were on May 22, June 12 and July 2 each, the effects of microbial pesticide were 73.9%, 93.5% and 43.6% respectively. As a result, it was effective that Bt was sprayed to grape leaves on mid and late June for controling the H. luctuosalis in organic vineyard. And microbial pesticide Bt was thought to be useful to control the grape leafroller in organic vineyard.

Photosynthetic characteristics and growth analysis of Angelica gigas according to different hydroponics methods (당귀의 광합성 특성과 수경재배 방식에 따른 생장 분석)

  • The aim of this study was to investigate which hydroponic system is the optimum for growth and photosynthetic characteristics of Angelica gigas during experiment. Angelica gigas 'Manchu' were sowed and managed under a growth room chamber. The environmental conditions (temperature $22^{\circ}C/18^{\circ}C$ (day/night), relative humidity 50-70%, photosynthetic photon flux density (PPFD) $120{\pm}6{\mu}mol\;m^{-2}s^{-1}$) were maintained for 3 weeks. Forty eight seedlings with 4-5 leaves were transplanted in deep flow technique (DFT), substrate, and spray culture systems [culture bed: 800 (L) ${\times}$ 800 (W) ${\times}$ 400 mm(H)] under $150{\pm}5{\mu}mol\;m^{-2}s^{-1}$ PPFD provided with fluorescence lamps and cultivated for 11 weeks. At the end of the experiment, fresh and dry weights, leaf lenghth and width, SPAD, root fresh, and dry weights, and root volume of Anglica gigas were measured. Photosynthetic rate of Anglica gigas were measured with portable photosynthesis systems to investigate optimum PPFD, $CO_2$ concentration, and air temperature conditions. Fresh and dry weights of Anglica gigas grown in substrate were significantly greater than DFT-treated, but there were not significant with spray treatment. Leaf photosynthesis of Anglica gigas showed the tendency to sharply increase as PPFD was increased from 50 to $200{\mu}mol\;m^{-2}s^{-1}$. Though $CO_2$ saturation point was around $1000-1200{\mu}mol\;mol^{-1}$, increase in air temperature from 16 to $26^{\circ}C$ did not quite affect photosynthesis of Anglica gigas. In conclusion, Anglica gigas may be optimally cultivated with a spray culture system as air temperature, PPFD, and $CO_2$ concentration for environment are controlled at $20{\pm}3^{\circ}C$, $150{\mu}mol\;m^{-2}s^{-1}$, and around $1000{\mu}mol\;mol^{-1}$ for mass production.

A Study on the Development of a Simulation Model for Predicting Soil Moisture Content and Scheduling Irrigation (토양수분함량 예측 및 계획관개 모의 모형 개발에 관한 연구(I))

  • Two types of model were established in order to product the soil moisture content by which information on irrigation could be obtained. Model-I was to represent the soil moisture depletion and was established based on the concept of water balance in a given soil profile. Model-II was a mathematical model derived from the analysis of soil moisture variation curves which were drawn from the observed data. In establishing the Model-I, the method and procedure to estimate parameters for the determination of the variables such as evapotranspirations, effective rainfalls, and drainage amounts were discussed. Empirical equations representing soil moisture variation curves were derived from the observed data as the Model-II. The procedure for forecasting timing and amounts of irrigation under the given soil moisture content was discussed. The established models were checked by comparing the observed data with those predicted by the model. Obtained results are summarized as follows: 1. As a water balance model of a given soil profile, the soil moisture depletion D, could be represented as the equation(2). 2. Among the various empirical formulae for potential evapotranspiration (Etp), Penman's formula was best fit to the data observed with the evaporation pans and tanks in Suweon area. High degree of positive correlation between Penman's predicted data and observed data with a large evaporation pan was confirmed. and the regression enquation was Y=0.7436X+17.2918, where Y represents evaporation rate from large evaporation pan, in mm/10days, and X represents potential evapotranspiration rate estimated by use of Penman's formula. 3. Evapotranspiration, Et, could be estimated from the potential evapotranspiration, Etp, by introducing the consumptive use coefficient, Kc, which was repre sensed by the following relationship: Kc=Kco$.$Ka+Ks‥‥‥(Eq. 6) where Kco : crop coefficient Ka : coefficient depending on the soil moisture content Ks : correction coefficient a. Crop coefficient. Kco. Crop coefficients of barley, bean, and wheat for each growth stage were found to be dependent on the crop. b. Coefficient depending on the soil moisture content, Ka. The values of Ka for clay loam, sandy loam, and loamy sand revealed a similar tendency to those of Pierce type. c. Correction coefficent, Ks. Following relationships were established to estimate Ks values: Ks=Kc-Kco$.$Ka, where Ks=0 if Kc,=Kco$.$K0$\geq$1.0, otherwise Ks=1-Kco$.$Ka 4. Effective rainfall, Re, was estimated by using following relationships : Re=D, if R-D$\geq$0, otherwise, Re=R 5. The difference between rainfall, R, and the soil moisture depletion D, was taken as drainage amount, Wd. {{{{D= SUM from { {i }=1} to n (Et-Re-I+Wd)}}}} if Wd=0, otherwise, {{{{D= SUM from { {i }=tf} to n (Et-Re-I+Wd)}}}} where tf=2∼3 days. 6. The curves and their corresponding empirical equations for the variation of soil moisture depending on the soil types, soil depths are shown on Fig. 8 (a,b.c,d). The general mathematical model on soil moisture variation depending on seasons, weather, and soil types were as follow: {{{{SMC= SUM ( { C}_{i }Exp( { - lambda }_{i } { t}_{i } )+ { Re}_{i } - { Excess}_{i } )}}}} where SMC : soil moisture content C : constant depending on an initial soil moisture content $\lambda$ : constant depending on season t : time Re : effective rainfall Excess : drainage and excess soil moisture other than drainage. The values of $\lambda$ are shown on Table 1. 7. The timing and amount of irrigation could be predicted by the equation (9-a) and (9-b,c), respectively. 8. Under the given conditions, the model for scheduling irrigation was completed. Fig. 9 show computer flow charts of the model. a. To estimate a potential evapotranspiration, Penman's equation was used if a complete observed meteorological data were available, and Jensen-Haise's equation was used if a forecasted meteorological data were available, However none of the observed or forecasted data were available, the equation (15) was used. b. As an input time data, a crop carlender was used, which was made based on the time when the growth stage of the crop shows it's maximum effective leaf coverage. 9. For the purpose of validation of the models, observed data of soil moiture content under various conditions from May, 1975 to July, 1975 were compared to the data predicted by Model-I and Model-II. Model-I shows the relative error of 4.6 to 14.3 percent which is an acceptable range of error in view of engineering purpose. Model-II shows 3 to 16.7 percent of relative error which is a little larger than the one from the Model-I. 10. Comparing two models, the followings are concluded: Model-I established on the theoretical background can predict with a satisfiable reliability far practical use provided that forecasted meteorological data are available. On the other hand, Model-II was superior to Model-I in it's simplicity, but it needs long period and wide scope of observed data to predict acceptable soil moisture content. Further studies are needed on the Model-II to make it acceptable in practical use.

Effects of Drought Stress and Nitrogen Fertilization on Growth and Physiological Characteristics of Pinus densiflora Seedlings Under Elevated Temperature and CO2 Concentration (대기 중 온도 및 CO2 농도 조절에 따른 건조 스트레스와 질소 시비가 소나무의 생장 및 생리적 특성에 미치는 영향)

  • Pinus densiflora is the most widely distributed tree species in South Korea. Its ecological and socio-cultural attributes makes it one of the most important tree species in S. Korea. In recent times however, the distribution of P. densiflora has been affected by dieback. This phenomenon has largely been attributed to climate change. This study was conducted to investigate the responses of growth and physiology of P. densiflora to drought and nitrogen fertiliz ation according to the RCP 8.5 scenario. A Temperature Gradient Chamber (TGC) and CO2. Temperature Gradient Chamber (CTGC) were used to simulate climate change conditions. The treatments were established with temperature (control versus +3 and +5℃; aCeT) and CO2 (control: aCaT versus x1.6 and x2.2; eCeT), watering(control versus drought), fertilization(control versus fertilized). Net photosynthesis (Pn), stomatal conductance (gs), biomass and relative soil volumetric water content (VWC) were measured to examine physiological responses and growth. Relative soil VWC in aCeT significantly decreased after the onset of drought. Pn and gs in both aCeT and eCeT with fertiliz ation were high before drought but decreased rapidly after 7 days under drought because nitrogen fertilization effect did not last long. The fastest mortality was 46 days in aCeT and the longest survival was 56 days in eCeT after the onset of drought. Total and partial biomass (leaf, stem and root) in both aCeT and eCeT with fertiliz ation were significantly high, but significantly low in aCeT. The results of the study are helpful in addressing P. densiflora vulnerability to climate change by highlighting physiological responses related to carbon allocation under differing simulated environmental stressors.

The Effect of Cultivation Period on Growth Characteristics , Palatability and Forage Yield of Soybean Cultivars (대두 품종별 재배기간이 생육특성 , 기호성 및 수량에 미치는 영향)

  • This experiment was canied out to select the soybean of high productivity suited for soiling. and growth characterisitcs, palatability, dry matter and crude protein yield of 10 cultivars of soybean have been evaluated at the experimental forage field. College of Natural Science of Kon-Kuk University in Chungju over three years (1 989-1 09 1 ). The results obtained are summarized as follows: JangBaek showed the tallest plant length from the 70th day to 80th day during cultivation period. and BaekUn showed the tallest at 90th day. PalDal among all cultivars showed the fastest growth rate and the strongest resistant lodging during cultivation period. The stem diameter of 70th day showed highly in order of PalDal and HwangKeum > BaekUn > KwangKyo and JangYeob, the 80th day showed highly in the order of TogYu > BaekCheon > JangBaek and 90th day showed highly in order of BangSa > BaekCheon > JangYeob. Stem hardness was highly increased with increasing of cultivation period, but no significant difference were found among the soybean cultivan. Especially, BaekUn and TogYu of 90th day showed very high as 17.9 and 17.6 kg/cm2, respectively. In the number of branch, PalDal was the lowest during cultivation period(70, 80 and 90th day), and 90th day showed lower two and three times than other soybeans(P<0.05). In the leaf rate, BaekCheon showed the highest according to cultivation period and the lowest in JangBaek. The fresh yield was tended to increase by increasing cultivation period at all cultivars except KwangKyo, but no significant differences were found among cultivars. Dry matter yield of JangYeob was the highest as 5, 861 kg/ ha at 70th day, and that of KwangKyo and TogYu was the highest as 7, 471 and 10, 603 kgha at 80th and 90th day, respectively. Relative palatability of 70th days showed very high at all soybean cultivars(81-100%), and that of 80th and 90th day showed the highest at TogYu, while showed the lowest at PalDal. In crude protein content of the 70th, 80th and 90th day, DanYeob, JangYeob and HwangKeum were the highest as 20.1, 17.9 and 20.4 percentage, respectively. Crude protein yield during cultivation period showed the highest at JangYeob(7Oth : 1, 068, 80th : 1, 211, 90th : 1, 882 kglha), but showed the lowest at BangSa(70th : 61 1, 80th : 814, 90th : 1, 078 kg/ha).

Selection of Lecanicillium Strains for Aphid (Myzus persicae) Control (복숭아혹진딧물 방제를 위한 Lecanicillium 균주 선발)

  • To select efficient entomopathogenic fungal strains of Lecanicillium for the biocontrol of aphid, Myzus persicae, conidial suspension ($1{\times}10\;conidia/ml$) was sprayed onto a detached Chinese cabbage leaf in a petri dish with a dampened filter paper that had 20 nymphs of aphid. Lecancillium strain 4078 and 6543 were the best strains for the biocontrol of aphid at high temperature of $30^{\circ}C$ and low relative humidity (RH) of 85%, respectively. The cumulative mortality of strain 4078 at $30^{\circ}C$ after 3 days was 100% and that of strain 6543 was 90% at 85% RH after 5 days. Strain 4078 also exhibited almost 100% germination ratio of conidia and high rate of mycelial growth at the broad temperature-range of $15{\sim}25^{\circ}C$. The strain 4078 and 6543 were all identified as Lecanicillium species based on the DNA sequences (accession no.: EF026004 and EF026005, respectively) of the ITS regions of the fungi. Excellent production of aerial conidia of strain 6543 was accomplished by using steamed polished rice as the solid culture medium.

Grain Yield and Physiological Responses of Water Stress at Reproductive Stage in Barley (보리 생식생장기의 수분부족이 수량 및 몇 가지 생리적 반응에 미치는 영향)

  • To cope with increasing importance of water stress in food crop production, some physiological characteristics, their cultivar-differences and grain yield of winter barley cultivars in response to water stress during reproductive stages were studied employing three covered-barley cultivars, Milyang 12, Durubori, and Olbori, one naked-barley cultivar, Baegdong, and one two-row malting-barley cultivar, Hyangmaeg. The barley grown in pot-soil was conditioned for 10 days under water stress, varying the time of water stress : 20 days before heading, 10 days before heading and the time of heading. The decrease in growth due to water stress varied greatly with the cultivars and time of water stress. The greatest injury occurred when water stress was imposed for 10 days from 10 days before heading : the culm length of water-stressed plants have shown reduced by 85∼98% of the non-stressed; the number of spikes per plant by 52∼83%; the number of grains per spike by 71∼86%; 1,000-grain weight by 80∼84%; yield per pot by 60∼94%. The number of spikes per plant as one of yield components was most sensitively affected. As a whole, the drought resistance of cultivars was high in the order of Olbori> Milyang 12 and Durubori> Hyangmaeg>Baegdong. On rewatering the plants after termination of the water stress treatment the recovery rate of free proline content and relative turgidity of flag leaf were higher in 3 covered-barley cultivars, and lower in cultivars Baegdong and Hyangmaeg.

Autotrophic Growth of Dendranthema grandiflorum R. 'Bongwhang' Plantlets In Vitro as Affected by PPF, Air Exchange Rate and $\textrm{CO}_2$ Concentration (봉황국화의 자가영양배양시 광도, 환기횟수 및 $\textrm{CO}_2$농도가 기내생육에 미치는 영향)

  • Growth of Dendranthema grandiflorum R. ‘Bongwhang’plantlets, as affected by three levels of photosynthetic photon flux (PPF), 70, 150 and 220 $\mu$mol. $m^{-2}$ . $s^{-1}$ , three levels of C $O_{2}$ concentration, 400-500 (ambient), 1000 and 2000 $\mu$mol.mo $l^{-1}$ , and two levels of number of air exchanges per hour (NAEH), 0.1 $h^{-1}$ and 2.8 $h^{-l}$, was studied. Explants were obtained from photomixotrophically-micropropagated plantlets. Four explants were planted in each 3.7$\times$10$^{-4}$ $m^{3}$ polycarbonate box containing MS medium supplemented with 1.25 meq. $L^{-1}$ $H_{2}$P $O_{4}$$^{[-10]}$ and no added sugar. Explants were cultured under cool-white fluorescent lamps (16 h. $d^{-1}$ ), at 25$\pm$1$^{\circ}C$ temperature, and 70-80% relative humidity. In treatments of 2.8 $h^{-1}$ NAEH, a 10 mm round hole made on the vessel cap was sealed with a microporous filter For higher C $O_{2}$ concentrations in the culture room, C $O_{2}$ gas was provided from a tank of liquefied C $O_{2}$. Fresh and dry weights, height, length of the longest roots, number of leaves, and leaf area significantly increased with increasing PPF and especially, with increasing C $O_{2}$ concentration. Growth was enhanced with increased number of air exchanges per hour (2.8 $h^{-1}$ ). Overall, treatment of 220$\mu$mol. $m^{-2}$ . $s^{-1}$ PPF combined with 2000$\mu$mol.mo $l^{-1}$ C $O_{2}$ and 2.8 $h^{-1}$ NAEH gave the most vigorous growth of Dendranthema grandiflorum R. ‘Bongwhang’ plantlets in vitro.o.

Study on hydroxy fatty acid contents changes and physiological responses under abiotic stresses in transgenic Camelina

  • Hydroxy fatty acid (HFA) is an important industrial resource that known to be extracted from seeds of Castor or Lesquerella. However, mass production of HFA from those crops are difficult because of their behavior or life cycle. In this study, we applied HFA synthesis related gene FAH12, RcPDAT1, RcLPCAT, RcDGAT2, and RcPDCT on bioenergy crop Camelina sativa. Furthermore, we determined NaCl or cold stress tolerance changes of transgenic Camelina. RcFAH12, RcPDAT1, RcLPCAT, RcDGAT2, and RcPDCT genes were cloned into multigene expression vector which is engineered with seed specific promoter of FAE1 or Napin. Combination of HFA genes multi-expression vector constructs were divided into Set3 (RcFAH12, RcPDAT1-2, RcLPCAT), Set4 (RcFAH12, RcDGAT2, RCPDAT1-2, RcLPCAT), and Set5 (RcFAH12, RcDGAT2, RCPDAT1-2, RcLPCAT, RcPDCT). Transgenic HFA synthesis Camelina plants were generated using agrobacterium-mediated vacuum infiltration system. Results of fatty acid composition of T1 transgenic Camelina seeds analyzed by GC-MS showed 9.5, 9.0, and 13.6 % of HFA contents in Set3#6, Set4#8, and Set5#10, respectively. Therefore, seeds of T2 generation were harvest from Set5#10 which is shown highest HFA contents, and, 17.7, 8.1 and 10.5 % of HFA contents were determined in Set5#10-5, Set5#10-8, and Set#10-10, respectively. However, 7.7% of C18:2 and 22.3 % of C18:3 among unsaturated fatty acids were decreased in Set5#10-5 than WT. Meanwhile, we confirmed abiotic stress responses in T2 transgenic Camelina Set5#10-5 and Set5#10-10 under 0, 100, 150, and 200 mM NaCl or 25, 15, and $10^{\circ}C$ temperature for 5 weeks. Both Set5#10-5 and Set5#10-10 showed lower growth in height than WT in control and NaCl condition. Growth of leaf length and width were similar in WT and Set5#10-10 but lower in Set5#10-5 under NaCl stress. Number of opened flowers showed that both transgenic Camelina were lower than WT under normal condition. But, WT and Set5#10-10 showed similar opened flower number in 100 and 200 mM NaCl. In cold stress, 15 and $10^{\circ}C$ treatment for 5 weeks did not showed significant changes in between WT and both transgenic lines even they showed different growth rate in control condition. Taken together, growth and development are delayed by expression of exogenous HFA related genes in transgenic lines but relative abiotic stress sensitivity is similar with WT.
