자병취(Saussurea chabyoungsanica Im)의 외부형태학적 연구 (The Morphological Study of Saussurea chabyoungsanica)

  • 자병취 (Saussurea chabyoungsanica Im..)에 대한 실체를 파악하기 위해 전반적인 형태학적 형질을 재검토하여 지역간 변이의 폭을 분석하였다. 태백산맥계에 속하는 만덕봉, 석병산, 석개재, 덕항산의 능선부 등 모두 석회암 지대에서 발견된 자병취와 북한에 자생하는 것으로 보고된 산골취의 외부형태에 대한 PCA분석 결과, 자병취 집단은 산골취 집단과 뚜렷이 구분되었고, 잎, 화관, 포엽의 형질이 주요 요인으로 나타났다. 또한 자병취의 지역별 외부형태의 분석결과, 석병산 지역의 자병취집단이 다른 지역의 자병취 집단과 구분되었으며 이는 환경구배현상에서 오는 차이가 반영된 것으로 생각된다.

한국산 팥꽃나무과의 비교형태학적 연구 (A comparative morphological study of Thymelaeaceae in Korea)

  • 한국산 팥꽃나무과 분류군 5속 10종(Daphne속 5종, Diarthron속 1종, Edgeworthia속 1종, Stellera속 1종 및 Wikstroemia속 2종)에 대해 비교형태학적 연구를 수행하였다. 야외 조사와 국내외 주요 표본관에 소장된 표본을 이용하여 팥꽃나무의 주요 형태형질을 정의하고, 변이의 폭을 파악하여, 각 분류군의 한계를 설정하고 유연관계를 추론하였다. 그 결과 한국산 팥꽃나무과 분류군들은 형태형질에 의해서 뚜렷이 구분되었다. 팥꽃나무속의 경우 한국산 분류군만 고려하면 산닥나무속과 쉽게 구분되었으나, 팥꽃나무(D. genkwa)는 잎의 배열과 털의 분포 및 자방의 형태에 있어서 산닥나무속과 공통적인 특징을 보이고 있었다. Daphne group에 대해 보다 많은 분류군과 다른 종류의 형질분석을 통하여 자세한 연구가 필요한 것으로 사료된다.

Intraspecific genetic variation in Corynandra chelidonii (Angiosperms: Cleomaceae) as revealed by SCoT, ISSR and RAPD analyses

  • The genetic diversity of two subpopulations of Corynandra chelidonii, one of terrestrial and the other of aquatic environments, was measured with molecular markers, such as start codon targeted (SCoT), inter simple sequence repeats (ISSR), and random amplification of polymorphic DNA (RAPD). The traditional morphological traits such as habitat, habit, leaf morphology, the colour of the sepals and petals, number of stamens, and seed morphology formed the base for their realization as two varieties, C. chelidonii var. pallae and C. chelidonii var. chelidonii. The polymorphism between the two variants was 100% with the primers SCoT-2 and OPA-1 and 4, while maximum polymorphism was detected with ISSR-2, SCoT-3, and OPA-3. The study used, for the first time, more than one molecular marker to assess the genetic variation underscoring the morphological variation in Corynandra chelidonii (L.f.) Cochrane & Iltis. The study justifies the recognition of the two subpopulations of Corynandra chelidonii from aquatic and terrestrial environments as two distinct varieties, C. chelidonii var. pallae (Reddy & Raju) V.S.Raju and C. chelidonii var. chelidonii, respectively, based on the traditional taxonomic evidence.

한국산 수거머리말(Zostera caulescens Miki.)의 형태 및 생태적 특성에 대한 연구 (Ecological and Morphological Characteristics of Zostera caulescens Miki (Zosteraceae) in Korea)

  • 한국에 자생하는 거머리말속(Zostera, Zosteraceae)에 포함되는 수거머리말(Z. caulescen Miki)의 형태 및 생태학적 특성을 파악하기 위하여 생육환경, 외부형태학적 형질, 잎과 종자의 미세 구조를 비교 분석하였다. 우리나라 해역에서는 남해연안의 거제 가배리와 진도 죽림리에서만 수거머리말의 실체가 확인되었으며, 생육지의 환경은 파랑의 영향이 적은 내만의 수심 $3.0{\sim}6.5m$의 사니질 또는 니사질의 퇴적환경에서 출현하였다. 생식지의 길이, 잎 너비, 엽맥 수, 잎 표피 세포의 크기와 수, 종피의 늑골 수, 생식지 상에 수관 형성 여부, 잎 가장자리와 중앙의 두께 및 색조의 차이, 종자 표면의 anthocyanin색소 집적에 따른 자주색의 반점,잎 표피세포의 기본모양에서 수거미리 말은 뚜렷하게 구별되었다. 남해안 가배와 죽림의 집단에서 생육지에 따른 외부 형태학적 차이는 나타나지 않았으나, 하계의 생육밀도와 생물량은 가배리$(367.3m^{-2},\;725.7g\;dry\;wt\p;m^{-2})$에서 죽림리$(112.5m^{-2},\;392.0\;g\;dry\;wt\;m^{-2})$보다 높게 나타났다. 지상부의 생물량은 지하부보다 높게 나타났으며, 생육밀도와 생물량은 생식지의 개체수와 개체 당 생물량에 따라 큰 차이를 나타내었다.

A Preliminary Study on Growth and Habitat Characteristics of Zostera marina (Zosteraceae) in Gamak Bay, Yeosu

  • This study was performed to obtain basic information on the ecology of Zostcra marina and to promote efficient conservation of this species which has been decline in Gamak Bay, Yeosu, Korea. Whater column characteristics and eelgrass morphology at Anpori, Jangsuri and Wonpori were investigated every month from December 1999 to November 2000. The water temperature, salinity and pH at the three sites were 10.0-27.0${\circ}C$,29.4-34.7% and 8.1-8.5, respectively. The water temperature at Anpori tended to be slightly lower than that at the other locations; the salinity at Wonpori from July to November was a little lower than that of the other locations. The concentrations of $NO_2$-N, $NO_3$_N, $NH_4$-N, $PO_4$-P and $Si(OH)_4$-Si at the three sites were 0.9-1.3, 2.0-6.2, 7.8-9.0, 3.0-3.6 and 22.2-30.2 uM, respectively. The concentration of $NO_3$-N at Wonpori from June to November was somewhat lower than that at the other locations; that of NH4-N at Jansuri was somewhat lower than the others. The mean shoot height and leaf width of the Anpori, Jangsuri and Wonpori populations were 80.6 cm and 0.9 mm, 90.0 cm and 1.0 mm, and 95.3 cm and 1.0 mm, respectively. The mean total shoot weight of the Anpori, Jangsuri and Wonpori ones was 24.5,31.0 and 29.7 & respectively. The mean leaf and branch numbers of the Anpori, Jangsuri and Wonpori populations were 16.5 and 2.6, 16.1 and 2.4 and 15.4 and 2.6 individuals, respectively. The correlation coefficients between shoot height and water temperature, leaf width and total shoot weight, leaf number and branch number, and $Si(OH)_4$-Si and $NO_3$-N were 0.726, 0.692, 0.862, and 0.693, respectively. The coefficients between shoot height and $NO_3$-N, total shoot weight and NO_3$-N, water temperature and $Si(OH)_4$-Si, water temperature and salinity, and water temperature and $NO_3$-N were -0.716, -0.536, -0.775, -0.685 and -0.685, respectively. The first four principal components explain 71.1% of the total sample variance. For axis 1, shoot height and water temperature tended to correlate with the population of Jansuri, followed by the Wonpori population, and $Si(OH)_4$-Si and $NO_3$-N tended to correlated strongly with the Anpori population. For axis 2, total weight, leaf width, leaf number and branch number showed a tendency to correlate with the Anpori and Jangsuri populations. For axis 3, the Anpori population tended to be influenced by $NO_2$-N and $PO_4$-P. For axis 4, the Wonpori and Jangsuri populations tended to be affected by salinity. The tendency, however, differed according to season.

한반도에 자생하는 한국잔디류(Zoysia spp.)의 분포 및 형태적 특성 (Distribution and Morphology Characteristics of Native Zoysiagrasses (Zoysia spp.) Grown in South Korea)

  • 본 연구는 국내 해안 및 도서지역에 자생하고 있는 한국잔디류를 지역별로 수집하여 형태적 특성 및 지역별 변이 정도를 알아보고자 수행되었으며, 수집된 유전자원 보존 평가할 수 있는 체계를 확립하는 것이 목적이다. 16개시 군 단위에서 159개체 자생 한국잔디를 수집하였으며, 이들의 초장, 엽폭, 엽장, 엽각도, 종자길이, 종자폭, 종자길이와 폭의 비 및 화수당 종자수 등의 형태적 특성을 조사하였다. Z. japonica류, Z. sinica류, Z. macrostachya류, Z. matrella류 등은 수집된 지역에 따라 초장, 엽장, 엽각도, 화수당 종자수 등에서 큰 변이를 보였다. 반면에 엽폭, 종자길이, 종자폭, 종자길이와 폭의 비율 등에서는 큰 변이를 보이지않았다. 중간적 형태를 보이고 있는 hybrid zoysiagrass류는 조사 형질 간에 다양한 변이를 보이는 것을 확인할수 있었다. 상기 형태적 특성 조사 결과 및 수집된 유전 자원들은 앞으로 고품질 잔디육종을 위한 귀중한 자료로 활용가치가 높다고 판단된다.

Beta 함수 기반 기온에 따른 양파의 잎 수 증가 예측 (Estimation of Onion Leaf Appearance by Beta Distribution)

  • 생물계절은 작물의 발달 시기를 결정하며, 생육기 온도에 의해 크게 영향을 받는다. 과정 기반 모델(PBM)에서 엽면적은 생물계절 및 형태 모듈의 결합에 의해 동적으로 시뮬레이션된다. 따라서 잎 발달 속도 또는 최종 잎 수의 예측은 전체 작물 모델의 성능에 영향을 주게 된다. 기온에 따른 잎 축적 속도 결정을 위한 데이터는 SPAR 챔버로부터 수집되었다. 온도의 함수로서 발달 속도를 설명하기 위해 베타 분포 함수(Yan and Hunt(1999)에 의해 제안됨)가 사용되었으며, 최적온도와 임계온도는 각각 26.0℃와 35.3℃로 추정되었다. 모델 추정치는 기온 모델에 생장기의 일 평균 기온을 입력하여 얻은 양파 잎의 일별 증가량을 누적한 결과이며, 모델 평가를 위해 온도구배하우스에서 관찰된 양파 잎의 누적 개수를 모델 추정치와 비교하였다. 본 연구에서 잎 수 추정 모델의 결정계수(R2)와 RMSE 값은 각각 0.95와 0.89였다.

Etiology of Apple Leaf Spot Caused by Colletotrichum spp. in China

  • Glomerella leaf spot pathogens can infect apple leaves, causing extensive necrosis and premature defoliation, as well as necrotic spots on fruit. In recent years, the disease has been reported with increasing frequency in China, and appears to be spreading rapidly in some apple-producing areas. In this study, fungal isolates from diseased apples leaves collected in Henan and Shaanxi provinces were analyzed based on morphology, cultural characters, pathogenicity and molecular phylogenetics. It was found that Glomerella leaf spot of apple was caused by two pathogens, Colletotrichum fructicola and C. aenigma. Pathogenicity tests showed that C. fructicola and C. aenigma could infect apple leaves of cultivar Golden Delicious, as well as Gala, Qinguan, Pink Lady, Pacific Rose, Golden Century and Honeycrisp, all of which include Golden Delicious in their parentage. In wound inoculation experiments, C. fructicola and C. aenigma were pathogenic to fruit of Gala, Qinguan, Golden Delicious, Pacific Rose, Starkrimson and Fuji. With non-wounded fruit, C. fructicola was pathogenic to Gala and Golden Delicious, and C. aenigma was pathogenic to Gala. It is concluded that the two pathogens could be differentiated according to pathogenicity to leaves and fruits of different apple cultivars.

Anti-adipogenic effect of mulberry leaf ethanol extract in 3T3-L1 adipocytes

  • BACKGROUND/OBJECTIVES: Adipogenesis is part of the cell differentiation process in which undifferentiated fibroblasts (pre-adipocytes) become mature adipocytes with the accumulation of lipid droplets and subsequent cell morphological changes. Several transcription factors and food components have been suggested to be involved in adipogenesis. The aim of this study was to determine whether mulberry leaf ethanol extract (MLEE) affects adipogenesis in 3T3-L1 adipocytes. MATERIALS/METHODS: The 3T3-L1 adipocytes were treated with different doses of MLEE for 8 days starting 2 days post-confluence. Cell viability, fat accumulation, and adipogenesis-related factors including CCAAT-enhancer-binding protein alpha ($C/EBP{\alpha}$), peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$), $PPAR{\gamma}$ coactivator 1 alpha (PGC-$1{\alpha}$), fatty acid synthase (FAS), and adiponectin were analyzed. RESULTS: Results showed that MLEE treatments at 10, 25, 50, and $100{\mu}g/ml$ had no effect on cell morphology and viability. Without evident toxicity, all MLEE treated cells had lower fat accumulation compared with control as shown by lower absorbances of Oil Red O stain. MLEE at 50 and $100{\mu}g/ml$ significantly reduced protein levels of $PPAR{\gamma}$, PGC-$1{\alpha}$, FAS, and adiponectin in differentiated adipocytes. Furthermore, protein level of $C/EBP{\alpha}$ was significantly decreased by the treatment of $100{\mu}g/ml$ MLEE. CONCLUSION: These results demonstrate that MLEE treatment has an anti-adipogenic effect in differentiated adipocytes without toxicity, suggesting its potential as an anti-obesity therapeutic.

남해 서부연안의 사구미 만에서 거머리말(Zostera marina)과 애기거머리말(Z. japonica)의 개체생태학 (The autecology of Zostera marina and Z. japonica at Sagumi Bay in the southwestern coast of Korea)

  • The autecology of the Zostera marina and Z. japonica was studied in populations growing in the same locality (Sagumi Bay, southwestern coast of Korea). Environmental factors and plant characteristics were examined monthly from August 2008 to September 2011. Along intertidal zone, Z. japonica (0.1-0.5 m above mean lower low water, MLLW) occurred above Z. marina (0.5-2.5 m MLLW). Tidal exposure at low tide during day was the highest in the spring and the lowest in the summer. Underwater Irradiance showed seasonal fluctuation that was the highest in spring and summer caused by tidal pattern. Strong seasonal patterns in water temperature appeared to control the seasonal variations in morphology, biomass and leaf growth. The seasonal pattern of Z. japonica resembled that of the Z. marina in morphological characteristics, above-and below-ground biomass, whereas it differed in shoot density and leaf elongation. Despite some similarities in seasonal growth patterns, the patterns of Z. japonica were lagged by 2 month of Z. marina. Seasonal variation in the above biomass of Z. marina was caused by changes in density and plant size, whereas that of Z. japonica was mainly caused by changes in shoot density. Zostera marina was more sensitive to high temperatures than Z. japonica, and the increasing water temperature during the summer became the factor that inhibits the growth of the Z. marina. Zostera Japonica, there is no clear change according to the amount of the light. It is because its habitat locates above that of Zostera marina so that the amount of the light that is necessary to growth is enough and in this condition, any preventing factor does not seem to work at all. Although underwater light getting into Zostera marina's habitat is very low level and there is no any hindrance to the survival of them, it prevents them from their productivity a bit.
