A Comparison between Diploid and Tetraploid Cultivars of Lolium multiflorum Lam, italicum (이탈리안 라이그라스의 2배체와 4배체 품종간 비교)

  • The influence of ploidy level on agronomic characteristics in italian ryegrass, Lolium multiflorum Lam, italicum, was studied using diploids (2x) and tetraploids (4x) on the upland of Livestock Experiment Station, Suweon. The results are summarized as follow: 1. Tetraploid cultivars are in general dark green in leaf color and strong glittered. 2. Diploid cultivars are more resistant to cold than tetraploids but degree of resistance to cold depends on cultivars. The di-and tetraploids did not differ in summer survival and coldand heat resistance of di-Itetraploid cultivars were not related. 3. Tetraploid cultivars have larger leaf-blades, more rapid growth and higher leaf ratio to stem by heading than diploids. 4. Tetraploids possess significantly lower dry matter content than diploids. 5 . Fresh- and dry weight of tetraploids are similar with that of diploids but tetraploids are more leafy than diploids. 6. Seasonal yield depends more on the type of the cultivar than on its tetraploid nature.

RT-PCR Detection of Three Non-reported Fruit Tree Viruses Useful for Quarantine Purpose in Korea

  • A simple and reliable procedure for RT-PCR detection of Apple stem pitting virus (ASPV), Cherry rasp leaf virus (CRLV), and Cherry necrotic rusty mottle virus (CNRMV) was developed. Two virus specific primer sets for each virus were found to specifically detect each virus among fourteen sets of designed oligonucleotide primers. Total RNAs extracted from healthy and from ASPV-,CRLV- and CNRMV-infected plant tissues were used to synthesize cDNA using oligo dT primer and then amplified by virus-specific primers for each virus. Each primer specifically amplified DNA fragments of 578 bp and 306 bp products for ASPV (prAS CP-C and prAS CP-N primers, respectively); 697 bp and 429 bp products for CRLV (prCR4 and prCR5-JQ3D3 primers, respectively); and 370 bp and 257 bp products for CNRMV (prCN4 and prCN6-NEG 1 primers, respec-tively) by RT-PCR. DNA sequencing of amplified DNA fragments confirmed the nature of each amplified DNA. Altogether, these results suggest that these virus specific primer sets can specifically amplify viral sequences in infected tissues and thus indicate that they can be used for specific detection of each virus.

Multiplex RT-PCR Assay for the Detection of Apple stem grooving virus and Apple chlorotic leaf spot virus in Infected Korean Apple Cultivars

  • To develop the diagnostic method for the viral infection in apple, the partial genes corresponding to the N-terminal region of RNA polymerase of Apple stem grooving virus (ASGV) and coat protein of Apple chlorotic leaf spot virus (ACLSV) were characterized from the infected apple cultivars in Korea. Based on the nucleotide sequences of the characterized partial genes, the virus gene-specific primers were designed for the detection of ASGV and ACLSV infected in species of Malus. The RT-PCR using the primers for the genes of ASGV and ACLSV successfully gave rise to 404 and 566 bp DNA fragments, respectively. Using those viral gene-specific primers, the multiplex RT-PCR assays were also established to diagnose the mixed infection by ASGV and ACLSV simultaneously. Furthermore, the control primers, which have to be included for the RT-PCR as an internal control, were designed using the nucleotide sequence of the gene encoding elongation factor $1{\alpha}(EF1{\alpha})$. This multiplex RT-PCR including the control primers provides more reliable, rapid and sensitive assay for the detection of ASGV and ACLSV infected in Korean apple cultivars.

Management Methods on the Growth Activation of Rhododendron mucronulatum Habitat in Mt. Biseul (비슬산 진달래군락지의 생육활성화 방안)

  • This study was carried out to find growth increasing methods of Rhododendron mucronulatum in Mt. Biseul. 4 treatments(heavy pruning, light pruning, non-pruning+fertilization, non-pruning(control)) were used in the study area with different pruning methods. Growth characteristics of new shoot, number of creation of flower buds and leaf buds in new shoot were analysed. Significant differences were observed among the treatments and stem diameter degree. Diameter, length of new shoots and the number of flower buds, leaf buds per new shoots increased in the pruning treatments. But heavy pruning showed more effective growth increasing way than light pruning. Non-pruning and fertilization treatments were not effective to increase growth of Rhododendron mucronulatum. And pruning at the end of May was the most effective way for growth of new shoot.

Anatomy of Bupleurum latissimum Nakai (Apiaceae), an Endemic Species of Korea (한국특산식물 섬시호의 해부학적 연구)

  • Anatomical characters of the Bupleurum latissimum Nakai, an endemic species of Korea, were investigated to confirm its phylogenetic relationships. Compare to other species with anatomical characters, B. latissimum is very similar with B, euphorbioides and B, longeradiatum in point of lacking of pith in the stem, shape of involucres, number of vascular bundles in radical leaf and cauline leaf, and lacking stomata in adaxial leaf surface. The other hand, protruded pollen aperture character appears in B. latissimum and B. euphorbioides. On the based of anatomical characters, therefore, B. latissimum has closest relationships with B. euphorbioides and B. longeradiatum. It also needs molecular study including Asian species in order to confirm phylogenetic position and speciation process apparently.

Insecticidal activity of native plant extracts against Culix pipiens pallens and Musca domestica (자생식물 추출물의 모기 및 집파리에 대한 살충활성)

  • Methanol extracts of 9 kinds of native medicinal plants(Taraxacum platycarpum, leaf; Pinus densiflora, leaf; Artemisia prinseps, leaf; Allium tuberosum, leaf; Cassia obtussifolia, whole; Sophora angestifolia, root; Stemonae sessilifolia, root; Lonicera japonica stem, leaf, flower; and Clivia miniata) were investigated for insecticidal effect. Methanol extracts of Pinus densiflora leaves and Sophora angestifolia radix showed relatively good insecticidal activity against Culex pipiens pallens larvae. Strong larvicidal activity against the Musca domestica larvae was produced from methanol extracts of Taraxacum platycarpum leaves and Allium tuberosom leaves. while Stemonae radix showed moderate activity. All materials tested revealed little or weak insecticidal activity against M. domestica adults.

Characteristics of Absorption and Accumulation of Inorganic Germanium in Panax ginseng C. A. Meyer

  • The characteristics of absorption and accumulation of inorganic germanium in Panax ginseng C. A. Meyer were examined. In 4-year-old P. ginseng, the germanium content of the field soil increased with increased amounts and frequencies of inorganic germanium application, while chemical components of the soil, such as available phosphate and exchangeable calcium, potassium, and magnesium, decreased with the increased inorganic germanium application. In the 4-year-old P. ginseng, the germanium content was highest in the rhizome and increased in the order of stem, leaf, lateral root, and main root, suggesting that inorganic germanium was absorbed from the root and translocated to the stem and leaf via the rhizome. As for changes in ginsenosides in 4-year-old P. ginseng rhizomes, the contents of ginsenosides $Rb_1$, $Rb_2$, Re, and Rf decreased as the germanium content in soil increased. Ginsenosides $Rb_1$, $Rb_2$, Rc, Re, and Rf in the main root also decreased with increasing germanium content in the main root. The results suggest that inorganic germanium treatment may increase organic germanium in harvested P. ginseng, thus enhancing the medicinal effi cacy of ginseng products.

Factors Affecting In vitro True Digestibility of Napiergrass

  • Changes of in vitro true digestibility (IVTD) of Napiergrass (Pennisetum purpureum) were determined by a filter bag system, and their relationships to chemical composition, leaf to stem ratio, plant height, geographic location, climatic factors and harvest interval were studied and used to develop prediction models for the crude protein (CP), acid-detergent fiber (ADF), and neutral-detergent fiber (NDF) contents and IVTD. Partitioning the total variance of IVTD of Napiergrass showed that 80% was attributable to the effect of harvest interval. Days of growth, plant height, leaf/stem ratio, CP, ADF and NDF of Napiergrass had highly significant relationships (p<0.01) with IVTD. The highest coefficient of correlation between the ADF, NDF, and IVTD of Napiergrass and growth degree days was obtained when the base temperature was set at $0^{\circ}C$. Growth degree days could predict ADF, NDF, and IVTD of Napiergrass more accurately than plant height, and plant height is not suitable to predict IVTD.

Growth Characteristics by Shading Rates in Panax ginseng C. A. Meyer (해가림 투광 정도에 다른 인삼의 생육 특성)

  • To compare the growth pattern of ginseng plant under between conventional shading(light transmittance rate 3%) and polyethylene net shading(light transmittance rate 10%), the distribution of leaf area, specific leaf weight (S. L. W), leaf and stem dry weight and changes in light intensity were investigated in 2, 4 and 6 year old ginseng plant populations. Light transmittance rate(L.T.R.) was 3% at front line, 2% at middle line and 1.5% at rear line under conventional shading but it was 12, 10 and 8% under polyethylene net(P.E) shading, respectively. In 2 year old population, there was a little difference in the growth characteristics investigated between conventional and P.E. shading. In 4 year old field, the leaf area, stem and leaf dry weight decreased in large amount in the order of middle, and rear line on ridge under conventional shading, but with a little difference under P.E. shading. And these trends enlarged in 6 year old field with appearance of a large part of shoot over furrow from ridge planted ginseng. Root yield index was much lowered at the rear 3rd, 4th and 5th line of the conventional shading bed, but there was no yield difference among lines except 5th lines under the P.E. shading with higher yield by 28% than conventional shading.
