본 연구에서는 임신진단등의 간편 현장진단(point of care test, POCT)에 주로 사용되고 있는 멤브레인 측면흐름 분석기법을 사용하여 인유두종 바이러스(Human papillomavirus, HPV)의 특정 서열을 검출할 수 있는 DNA array를 개발하였다. HPV type 6, 11, 16, 18, 31, 45에 특이적인 DNA 탐침들을 측면흐름 분석용 멤브레인 표면에 고정하고, biotin이 label된 MY09/11 primer를 사용하여 얻어진 HPV PCR 반응 결과물과 탐침 사이에 hybridization 반응을 유도하였다. 이후 streptavidin이 label된 colloidal gold가 교잡물의 biotin과 반응함으로써 DNA hybridization 결과를 육안으로 확인할 수 있었다. 본 연구를 통해 개발된 HPV DNA lateral flow membrane array는 기존의 HPV DNA chip 기법과 비교하여 경제적이고 편리하게 주요 HPV type을 확인할 수 있음을 보여주었다.
A low-cost, simple strip reader system using a linear movement mechanism of CD-ROM deck has been developed to characterize a lateral flow membrane-based immunochromatographic assay. The test strip reader was assembled by a CD-ROM deck and home-made optical head especially designed for immunoassays. The optical head for detecting reflected light from the test strip surface consists of green light-emitting diode, large area silicon photodiode, and anodized aluminum mounting block providing a slit structure for cutting light from the LED. The stepping motor of the deck was operated in the full step mode, whose distance of each reading point is about 0.15mm. The performance of the strip reader was tested by analysis of HBV(hepatitis B virus) antigen test kit. This strip reader can be useful for inexpensive, disposable, and membrane-based assays that provide visual evidence of the presence of an analyte in a liquid sample.

Purpose: This study was performed to develop a lateral flow strip sensor for the detection of pathogenic Escherichia coli O157:H7 in various samples. Also, feasibility of using an image analysis method to improve the interpretation of the strip sensor was evaluated. Methods: The lateral flow strip sensor has been fabricated based on nitrocellulose lateral-flow membrane. Colloidal gold and E. coli O157:H7 antibodies were used as a tag and a receptor, respectively. Manually spotted E. coli O157:H7 antibody and anti-mouse antibody on nitrocellulose membrane were used as test and control dots, respectively. Feasibility of the lateral flow strip sensor to detect E. coli O157:H7 were evaluated with serially diluted E. coli O157:H7 cells in PBS or food samples. Test results of the lateral flow strip sensor were measured with an image analysis method. Results: The intensity of the test dot started to increase with higher concentration of the cells were introduced. The sensitivities of the sensor were both $10^4$ CFU/mL Escherichia coli O157:H7 spiked in PBS and in chicken meat extract, respectively. Conclusions: The lateral flow strip sensor and image analysis method could detect E. coli O157:H7 in 20 min, which is significantly quicker than conventional plate counting method.
The inertial migration of a two-dimensional elastic capsule in a channel flow was studied over the Reynolds number range $1{\leq}Re{\leq}100$. The lateral migration velocity, slip velocity, and the deformation and inclination angle of the capsule were investigated by varying the lateral position, Reynolds number, capsule-to-channel size ratio(${\lambda}$), membrane stretching coefficient(${\Phi}$), and membrane bending coefficient(${\gamma}$). During the initial transient motion, the lateral migration velocity increased with increasing Re and ${\lambda}$ but decreased with increases in ${\Phi}$, ${\gamma}$ and the lateral distance from the wall. The initial behavior of the capsule was influenced by variation in the initial lateral position ($y_0$), but the equilibrium position of the capsule was not affected by such variation. The balance between the wall effect and the shear gradient effect determined the equilibrium position. As Re increased, the equilibrium position initially shifted closer to the wall and then moved towards the channel center. A peak in the equilibrium position was observed near Re=30 for ${\gamma}=0.1$, and the peak shifted to higher Re as ${\gamma}$ increased. Depending on the lateral migration velocity, the equilibrium position moved toward the centerline for larger ${\gamma}$ but moved toward the wall for larger ${\Phi}$ and ${\gamma}$.
Among semi-quantitative or fully quantitative lateral flow assay readers, an image sensor-based instrument has been widely used because of its simple setup, cheap sensor price, and compact equipment size. For all previous approaches, monochrome CCD or CMOS cameras were used for lateral flow assay imaging in which the overall intensities of all colors were taken into consideration to estimate the analyte content, although the analyte related color information is only limited to a narrow wavelength range. In the present work, we introduced a color CCD camera as a sensor and a color decomposition method to improve the sensitivity of the quantitative biosensor system which utilizes the lateral flow assay successfully. The proposed setup and image processing method were applied to achieve the quantification of imitatively dispensed particles on the surface of a porous membrane first, and the measurement result was then compared with that using a monochrome CCD. The compensation method was proposed in different illumination conditions. Eventually, the color decomposition method was introduced to the commercially available lateral flow immunochromatographic assay for the diagnosis of myocardial infarction. The measurement sensitivity utilizing the color image sensor is significantly improved since the slopes of the linear curve fit are enhanced from 0.0026 to 0.0040 and from 0.0802 to 0.1141 for myoglobin and creatine kinase (CK)-MB detection, respectively.
Staphylococcal enterotoxin is a very common cause of food poisoning. Conventional detection methods for the toxin including enzyme-linked immunosorbent assays (ELISAs), chemiluminescence (ECL), and polymerase chain reaction (PCR) assays require a lot of time, efforts, and expert technicians. Lateral flow strip kits have shown great potential for the rapid detection of foodborne pathogens. The lateral flow strip kit is widely used in clinical settings because it is easy to use, fast, and cost effective. A typical lateral flow strip kit uses colloidal gold to generate a visual signal. However, the lateral flow strip kit based on colloidal gold has limited sensitivity to fulfill food safety regulation requirements. This study was performed to develop a rapid test kit for pathogenic staphylococcal enterotoxin B (SEB) in food samples. The rapid detection kit was fabricated based on a nitrocellulose lateral-flow strip. Cellulose nano beads and SEB antibodies were used as the tag and receptor, respectively, to improve the detection performance. Manually spotted SEB antibody and anti-rabbit antibody on the surface of the nitrocellulose membrane were used as test and control spots, respectively. The feasibility of the rapid test kit to detect SEB in samples was evaluated. The sensitivity of the kit was 10 ng/mL SEB spiked in PBS. Additionally, the rapid test kit could detect 1 ng/mL of SEB in chicken meat extract.
We explored the dynamic motions and the lateral equilibrium positions of an elastic capsule in channel flow at moderate Reynolds number varying Re, aspect ratio, size ratio, membrane stretching and bending coefficient. The transition of tank-treading/swinging to tumbling motion was observed in the simulations and the transition of dynamic motions for capsules resulted in different trend of the variation in the lateral equilibrium positions. Though other conditions were similar, the capsule with tumbling motion migrated closer to the wall than that with tank-treading motion.
We report a case of non communicating hydrocephalus due to membranous obstruction of Magendie's foramen. A 37-year-old woman presented with intracranial hypertension symptoms caused by the occlusion of Magendie's foramen by a membrane probably due to arachnoiditis. As far as the patient's past medical history is concerned, an Epstein-Barr virus infectious mononucleosis was described. Fundoscopic examination revealed bilateral papilledema. Brain magnetic resonance imaging demonstrated a significant ventricular dilatation of all ventricles and turbulent flow of cerebelospinal fluid (CSF) in the fourth ventricle as well as back flow of CSF through the Monro's foramen to the lateral ventricles. The patient underwent a suboccipital craniotomy with C1 laminectomy. An occlusion of Magendie's foramen by a thickened membrane was recognized and it was incised and removed. We confirm the existence of hydrocephalus caused by fourth ventricle outflow obstruction by a membrane. The nature of this rare entity is difficult to demonstrate because of the complex morphology of the fourth ventricle. Treatment with surgical exploration and incision of the thickened membrane proved to be a reliable method of treatment without the necessity of endoscopic third ventriculostomy or catheter placement.
본 논문에서는 수용성의 CdSe/ZnS 양자점을 합성하고 이에 항체기능성을 도입하여 lateral flow immunoassay (LFIA) 플랫폼에 융합하여 폐암 질병진단에 활용 가능한 단백질 바이오마커[예: 인간 혈청 아밀로이드 A-1 (hSAA1)]의 농도 분석에 적용하고자 한다. 면역분석법 센서 스트립은 니트로셀룰로오즈 막에 테스트라인과 대조라인으로 각각 항hSAA1 단일클론항체(10G1)(anti-hSAA1)와 항chicken IgY (anti-chicken IgY)를 스프레이하여 제작하였다. 이와 함께, 유기상에서 합성된 CdSe/ZnS 양자점은 카르복실기로 변형된 알케인티올기를 이용한 리간드 교환방법으로 수용성으로 전환하였으며, 이에 타겟 단백질인 hSAA1에 특이적으로 결합 가능한 항체인 항hSAA1 단일클론항체(14F8)로 컨쥬게이션하여 형광검출용 입자[QDs-anti hSAA1 (14F8)]로 사용하였다. 제작된 LFIA 스트립 위에 순차적으로 다른 농도의 hSAA1과 QDs-anti hSAA1 (14F8)의 복합체를 흘려주면, 테스트라인에 anti hSAA1 (10G1)/hSAA1/QDs-anti hSAA1 (14F8) 샌드위치 복합체가 형성되어 양자점에 의한 발광신호가 검출됨을 측정하였다. 최적화된 측방흐름이 가능한 완충용액 조건에서 100 nM 농도의 hSAA1 단백질의 유무를 5 min 안에 눈으로 확인 가능하였다.
Slabs in buildings and bridge decks, which are restrained against lateral displacements at the edges, have ultimate strengths far in excess of those predicted by analytical methods based on yield line theory. The increase in strength has been attributed to membrane action, which is due to the in-plane forces developed at the supports. The benefits of compressive membrane action are usually not taken into account in currently available design methods developed based on plastic flow theories assuming concrete to be a rigid-plastic material. By extending the existing knowledge of compressive membrane action, it is possible to design slabs in building and bridge structures economically with less than normal reinforcement. Recent research on building and bridge structures reflects the importance of membrane action in design. This paper describes the finite element modelling of membrane action in reinforced concrete slabs through optimisation of a simple concrete model. Through a series of parametric studies using the simple concrete model in the finite element simulation of eight fully clamped concrete slabs with significant membrane action, a set of fixed numerical model parameter values is identified and computational conditions established, which would guarantee reliable strength prediction of arbitrary slabs. The reliability of the identified values to simulate membrane action (for prediction purposes) is further verified by the direct simulation of 42 other slabs, which gave an average value of 0.9698 for the ratio of experimental to predicted strengths and a standard deviation of 0.117. A 'deflection factor' is also established for the slabs, relating the predicted peak deflection to experimental values, which, (for the same level of fixity at the supports), can be used for accurate displacement determination. The proposed optimised concrete model and finite element procedure can be used as a tool to simulate membrane action in slabs in building and bridge structures having variable support and loading conditions including fire. Other practical applications of the developed finite element procedure and design process are also discussed.
