Expression of Recombinant Hybrid Peptide Gaegurin4 and LL37 using Fusion Protein in E. coli (Glutathione S-Transferase에 융합한 재조합 Hybrid Peptide Gaegurin-LL37의 대장균에서의 발현)

  • Antimicrobial peptides (AMPs) are important components of living organisms acting against Gram-negative and Gram-positive bacterial and fungal pathogens. Cathelicidin human peptides have a variety of biological activities that can be used in clinical applications. AMPs are not produced naturally in large quantities, and chemical synthesis is also economically impractical, especially for long peptides. Therefore, as an alternative, heterologous expression of AMPs by recombinant techniques has been studied as a means to reduce production costs. E. coli is an excellent host for the expression of AMPs, as well as other recombinant proteins, because of the low cost involved and its easy manipulation. However, overexpression of AMPs in E. coli has been shown to cause difficulties resulting from the toxicity of the subsequently produced AMPs. Therefore, fusion expression was theorized to be a solution to this problem. In this study, AMPs were expressed as fused proteins with the glutathione S-transferase (GST) binding protein to protect against the toxicity of AMPs when expressed in E. coli. The LL37, and hybrid gaegurin and LL37 (GGN4(1-16)-LL37(17-32), which we designated as GL32, peptides were expressed as GST-fusion proteins in E. coli and the fusion proteins were then purified by affinity columns. The purified peptides were obtained by removal of GST and were confirmed by western blot analysis. The purified antimicrobial peptides then demonstrated antimicrobial activities against Gram-negative and Gram-positive bacterial strains.

  • Pseudomonas fluorescens strain CL0145A was discovered at the New York State Museum Field Research Laboratory as an effective agent against the environmentally destructive zebra mussel, which has contaminated US waters. Dried cells of the microbe are being commercialized as an environmentally friendly solution to the problem. We found that antibiotic activity against the Gram-positive bacterium Bacillus subtilis is produced and excreted by this strain. We have carried out studies to optimize production of the antibiotic. Studies were begun in a complex corn meal medium. Activity was found in both cells and culture supernates and was maximal after one day of fermentation. Static fermentation conditions were found to be superior to shaken culture. Production of extracellular antibiotic in complex medium was found to be dependent on the content of sucrose and enzyme-hydrolyzed casein. Indeed, production was greater in sucrose plus enzyme-hydrolyzed casein than in the complex medium. Of a large number of carbon sources studied as improvements over sucrose, the best was glycerol. An examination of nitrogen sources showed that production was improved by replacement of enzyme-hydrolyzed casein with soy hydrolysates. Production in the simple glycerol-Hy-Soy medium was not improved by addition of an inorganic salt mixture or by complex nitrogen sources, with the exception of malt extract. In an attempt to keep the medium more defined, we studied the effect of amino acids and vitamins as replacements for malt extract. Of 21 amino acids and 7 vitamins, we found tryptophan, glutamine, biotin, and riboflavin to be stimulatory. The final medium contained glycerol, Hy-Soy, tryptophan, glutamine, biotin, and riboflavin.

  • The 23 representative bacteria were studied for the glucosamine contents which represent the glycolipid content of the cell wall. The distribution of glycolipid in various bacteria was examined and the relationship between the glycolipid contents and the Gram stain was elucidated. The results were as follows: 1. The contents of glucosamine in the glycolipid of Gram negative and variable bacterial cell wall were large ranging from the least 0.04 ${\mu}g$ of Proteus vulgaris to the largest 2.48 ${\mu}g$ of Aerobacter aerogenes. The Gram positive bacteria and only those Gram positive among Bacilli contained less than 0.02 ${\mu}g$ of glucosamine contents. The least glucosamine containing Gram positive bacteria were Corynebacterium sepedonicum and Staphylococcus aureus. It could generally be said that the Gram negative and variable bacteria contain the higher content of glucosamine in the cell wall than the positives. 2. The bacteria were better stained by the Gram solution after the extraction of glycolipid from the cell wall than those without extraction. 3. The four infrared spectra of glycolipids obtained from Aerobacter aerogenes, Bacillus circulans, Pseudomonas fluorescens, and Salmonella typhirurium showed all the similar characteristics. All showed the existence of groups; OH, C-O, C-O-C, $CH_2+CH_3$, amide band, fatty acid ester band and ester carbonyl bond.

Study on the Improving Thermal and Mechanical Properties of Eco-friendly Materials used for Training Ammunition (연습용 탄약 친환경 재료의 내열성 및 기계적 특성 향상에 관한 연구)

  • Unlike live ammunition which has killing power due to the use of high explosives, training ammunition has only the limited explosive effect needed for training purposes, so the risk of accidents is lowered. Because training ammunition is used in large quantities during military drills, the problem of environmental pollution occurs. As most of the waste is left out in the training field, using bio-degradable polymers such as Polylactic Acid (PLA) can provide a solution to these environmental issues. However, bio-degradable polymers such as PLA usually have poor thermal and mechanical properties compared with other general purpose polymers, so they need to be improved before they can be used for military purposes. In this study, Talc is added to the PLA used for the parts of Training Grenades to improve some of their properties and the changes of their thermal and mechanical properties were verified. In the case of the 1 wt.% ~ 5 wt.% PLA/Talc blends, the thermal properties were improved in proportion to the content of Talc, but the best mechanical properties were observed for the 1 wt.% and 3 wt.% PLA/Talc blends.

Legal Approach to the Passage Issues of the Cheju Strait (제주해협 통항문제에 관한 법적 고찰)

  • Considering possible legal and policy problems with regard to the Cheju Strait, a central issue is whether the Cheju Strait should be treated as Korean territorial sea or an international strait The claim that the strait is territorial sea has been based on the use of a straight baseline method of dermarcation With the use of straight baseline, Korea claims that the breadth of the Cheju Strait is only 20.7 miles at its narrowest point and therefore the strait becomes the territorial sea of Korea. The consideration cf marine pollution has weighed heavily in claiming the Cheju Strait as territorial sea. Pollution resulting from the accidents cf tankers caused by fire, collision, or stranding in the Cheju Strait and the Korea Strait would be enormous, affecting the entire coastal waters of the south coasts cf Korea's mainland and Japan's Tsushima Islands areas. Catastrophic pollution in the Cheju Strait could also come from the accidents cf large-size oil tankers passing through the Korea Strait from the Malacca Strait Although the Korean government considers the geographic and socioeconomic conditions sufficient to justify Korea's claim of the Cheju Strait as territorial sea, it believes that declaring it so would raise considerable legal conflicts with maritime states. In view of the legal difficulties and the need to meet the problems arising from the growing vessel traffic in the Cheju Strait, the sea lanes and traffic separation schemes may be considered an alternative to the internationalization of the Cheju Strait Even if the Korean government dose not do so, the regime of innocent passage should be applied to vessels passing through the Strait and should not suspend innocent passage through the Strait. Therefore, the Korean government needs to have a more legal, pragmatic, functional and managerial approach than a purely sovereign and selfish approach to the solution of legal matters of the Cheju strait For this purpose, the UN Convention on the Law of the Sea would serve as a guide and also self-restraint and cooperative approaches would become norms governing the resolution of the law of the sea issues in the Cheju Strait.

Model on the Capillary Action-Induced Dynamics of Contact Lens (모세관 작용에 의한 콘택트 렌즈의 운동 모델)

  • A mathematical model was proposed to analyze the damped motion of contact lens which is initially displaced from the equilibrium position. The model incorporates the differential equations and their numerical solution program, based on the formulations of restoring force arising from the capillary action in the tear-film layer between the lens and cornea. The model predicts the capillary action induced surface tension, time dependence of displacement of lens when it is released from the equilibrium position. It seems that the motion of lens is similar to the typical over-damped oscillation caused by the large viscous friction in the liquid layer between the cornea and lens. The effect of variables such as base curves, lens diameters and thickness of tear film layer were illustrated by the computer simulation of the derived program. The time required for the lens to return to the original position increases as the liquid layer thickness increases and it decreases as the diameter of lens increases. With the certain value of base curve the time interval is found to be minimum. The free vibrations of lenses were also simulated varying the parameters such as base curve, diameter, layer thickness. The resonant frequencies are inversely proportional to the liquid layer thickness and it increases as the lens diameter increases. The resonant frequency of lens has a maximum when the diameter is of certain value. If the external impulse or force of the same frequency as the natural frequency of contact lens acted on the cornea in vivo it may cause an excessive movement and thus it might cause the distortion 10 the lens or be pulled off the eye.

Study on Behaviour of Flood Wave-front Varied with Levee Breach Speed in Flat Inundation Area (평탄지형 제내지에서의 제방붕괴속도에 따른 범람홍수파 선단 거동에 관한 연구)

  • An experimental study was carried out to investigate the characteristics of the propagation distance of a flood wave considering the levee failure speed in a flat inundation area. The Ritter solution for one dimensional flow was considered to formulate the experimental results and a representative form with coefficients of k and m, which consider the three dimensional flow characteristics, was applied. The experiments showed that the propagation velocity of the wave front in the inundation area was influenced by the levee breach speed as well as the initial water level, which is a significant variable representing the flood wave behavior. In addition, coefficients k and m are not constants, but variables that vary with levee breach speed. An empirical formula was also suggested using the experimental results in the form of the relationships between k and m. In this study, a large-scale experiment for flood inundation was carried out to examine the behavior of flooding in the inundated area and the relationships between the levee breach speed and wave-front propagation velocity were suggested based on the experimental results. These research results are expected to be used as the baseline data to draw a flow inundation map, establish an emergency action plan, and verify the two-dimensional numerical model.

Effect of Organic Solvent-Modification on the Electrical Characteristics of the PCBM Thin-Film Transistors on Plastic substrate (플라스틱 기판상에 제작된 PCBM 박막 트랜지스터의 전기적 특성에 대한 유기 용매 최적화의 효과에 대한 연구)

  • Organic thin-film transistors (OTFTs) have received considerable attention because their potential applications for nano-scale thin-film structures have been widely researched for large-scale integration industries, such as semiconductors and displays. However, research in developing n-type materials and devices has been relatively shortage than developing p-type materials. Therefore, we report on the fabrication of top-contact [6,6]-phenyl-C61-butyricacidmethylester (PCBM) TFTs by using three different solvent, o-dichlorobenzene, toluene and chloroform. An appropriate choice of solvent shows that the electrical characteristics of PCBM TFTs can be improved. Moreover, our PCBM TFTs with the cross-linked Poly(4-vinylphenol) dielectric layer exhibits the most pronounced improvements in terms of the field-effect mobility (${\sim}0.034cm^2/Vs$) and the on/off current ratio (${\sim}1.3{\times}10^5$) for our results. From these results, it can be concluded that solvent-modification of an organic semiconductor in PCBM TFTs is useful and can be extended to further investigations on the PCBM TFTs having polymeric gate dielectrics. It is expected that process optimizations using solution-processing of organic semiconductor materials will allow the development of the n-type organic TFTs for low-cost electronics and various electronic applications.

A Study of Political Use of Naval Power in Solving International Conflicts (설득이론을 통한 해군력의 정치적 사용에 관한 고찰)

  • Morgenthau claims that in international relations, there are the economic, political, and military powers that enable a nation to achieve its political and diplomatic goals. This paper explores the possibility of resolving international conflicts with naval power. First, the theoretical and historical perspective, naval power was used as a final resort to force a nation's political or diplomatic objective on an enemy nation when negotiations failed, and this was done through the physical and psychological destruction of the enemy by naval power. But as the use of military power has decreased because of the invention of the nuclear weapon, the existence of a large and capable navy deterrent has become one of the most useful military options among a nation's diplomatic measures. In other words, he focused on the political usefulness of naval power as a deterrent and coercive diplomatic tool for persuading other nations to acquiesce, rather than using naval power and actual military action as a final resort. The reason for this is that compared to army and air force, navy's flexibility, continuity, and the ability to deter are greater. The navy provides excellent accessibility through its wide mobility on the sea, and it has been shown through research that the navy possesses a political usefulness that facilitates the solution of conflicts through presence, naval intervention, and naval blockade. On the other hand, among the factors that could improve the influence of the navy are alliance relations, a reliable and powerful navy, carrots and sticks that it would have to deal with in the case of successful or unsuccessful negotiations, and support from international opinion. On this paper I introduce E.N.Luttwak's naval suasion theory. By the his theory, there are two mode of naval suasion. One is latent naval suasion the other is active suasion. Latent suasion there are deterrent mode and supportive mode. Active naval suasion there are coercive mode and supportive mode. Coercive mode has positive and negative. The limitations of naval suasion have been identified as follows. First, because the objective of the use of naval power is persuading enemy nations, the results are unpredictable. Second, the leaders of all countries possess limited understanding on the complexities of naval power and therefore lack understanding of the usefulness of naval power when choosing options. Third, in case of failure through naval suasion, prestige and reputation of a nation can be damaged. Finally, the following are additional possible research topic. First, a research on the decision making process of choosing naval power as a measure to resolve conflicts is needed. Lastly, research on the size of the navy and types of ships required for efficient naval suasion is needed. Today's world requires cooperative security regime so that middle class navy also requires political use of naval power in solving international conflicts. Therefore, additional research on this topic is needed.

Characterization of Intermediate Conductance $K^+$ Channels in Submandibular Gland Acinar Cells

  • There are some evidences that $K^+$ efflux evoked by muscarinic stimulation is not mainly mediated by large conductance $K^+$ (BK) channels in salivary gland. In this experiment, we therefore characterised non BK channels in rat submandibular gland acinar cells and examined the possibility of agonist effect on this channel using a patch clamp technique. Two types of $K^+$ channels were observed in these cells. BK channels were observed in 3 cells from total 6 cells and its average conductance was $152{\pm}7$ pS (n=3). The conductance of the another types of $K^+$ channel was estimated as $71{\pm}7$ pS (n=6). On the basis of the conductance of this channel, we defined this channel as intermediate conductance $K^+$ (IK) channels, which were observed from all 6 cells we studied. When we increased $Ca^{2+}$ concentration of the bath solution in inside-out mode, the IK channel activity was greatly increased, suggesting this channel is $Ca^{2+}$ sensitive. We next examined the effect of carbachol (CCh) and isoproterenol on the activity of the IK channels. $10^{-5}$ M isoproterenol significantly increased the open probability (Po) from $0.08{\pm}0.02$ to $0.21{\pm}0.03$ (n=4, P<0.05). Application of $10^{-5}$ M CCh also increased Po from $0.048{\pm}0.03$ to $0.55{\pm}0.33$ (n=5, P<0.05) at the maximum channel activity. The degree of BK channel activation induced by the same concentration of CCh was lower than that of IK channels; Po value was $0.011{\pm}0.003$ and $0.027{\pm}0.005$ in control and during CCh stimulation (n=3), respectively. The result suggests that IK channels exist in salivary acinar cells and its channel activity is regulated by muscaricinic and ${\beta}-adrenergic$ agonist. We conclude that IK channels also play a putative role in secretion as well as the BK channels in rat submandibular gland acinar cells.
