복분자(Rubus coreanum Miquel) 즙을 첨가한 호상 요구르트의 품질 특성 (Quality Characteristics of Curd Yogurt with Rubus coreanum Miquel Juice)

  • 복분자 착즙액을 첨가한 호상 요구르트를 제조하기 위하여 skim milk에 S. thermophilus, L. acidophilus와 L. casei를 혼합한 균주를 접종하여 복분자 착즙액이 요구르트의 품질 특성에 미치는 영향을 조사하였다. pH는 전반적으로 발효기간중 감소하는 경향을, 산도는 증가하는 경향을 나타냈다. 생균수 변화에 있어서 복분자 착즙액의 첨가는 농도에 따라 생육을 촉진하기도 하고 저해하기도 하는 것으로 나타났으나, 4% 첨가농도까지는 안정된 발효를 유도하는 결과를 보였다. 성분 분석 결과를 살펴보면, 대조구와 복분자 착즙액 첨가구별 단백질, 유당, 유리지방산 함량에서는 시료간 비슷한 값을 보였고, 복분자 착즙액의 첨가량에 따라 총고형분함량은 증가하였다. 기호도 검사에서는 3% 복분자즙 첨가구가 색, 향, 맛, 후미 및 종합적인 기호도에서 가장 높은 선호도를 나타내었다.

Relation of Calcium Activity in Milk and Milk Production of Holstein Cows in Hot Season

  • The content of Ca in milk exceeds the typical saturation level of Ca salts, which is necessary for neonate growth. This calcium is distributed between the casein micelles in the colloidal and aqueous phases. Information on the properties of calcium activity in the aqueous phase is limited compared with that on the properties of bound or sequestrated calcium. The objectives of this study were to evaluate the changes in calcium activity in fresh milk using an ion-selective electrode and to assess the relationship between calcium activity and milk production in hot season. Milk samples collected from 10 cows at the National Agricultural Research Center for Kyushu Okinawa Region in June to October (Min-Max: 7.2-$35.2^{\circ}C$, 24.3-100% RH) were analyzed on total calcium concentrations and calcium activity. We observed that the rectal temperature of the cows increased according to elevation of ambient temperature but that the pH of the collected milk ($6.61{\pm}0.01$ (Mean${\pm}$SEM)) was not significantly influenced by rectal and ambient temperature. Total calcium concentrations and calcium activity in fresh milk decreased in July (Min-Max: 21.1-$33.5^{\circ}C$, 48.9-100.0% RH) compared with the values after August (Min-Max: 18.1-$35.0^{\circ}C$, 26.5-96.2% RH) (p<0.05); however, there was no significant correlation between the two parameters. The ratio of calcium activity to total calcium concentration decreased after August compared with the values in June and July (p<0.05). The calcium activity in fresh milk was positively correlated with milk yield (r = 0.45, p<0.01) and negatively correlated with milk lactose content (r = -0.53, p<0.01). These results suggest that the calcium activity in milk could be affected by ambient temperature and might be associated with milking production in hot season.

Holstein 젖소 초유의 성분 변화에 관한 연구 (Changes in Compositions of Holstein Colostrum during Lactation Period)

  • 초유의 이용성을 향상시키고, 그 품질을 보존하기 위한 방안의 일환으로 분만 후 12시간 간격으로 착유한 초유를 이용하여 시간의 경과에 따른 초유의 성분 변화를 조사하였다. 그 결과, 초유의 단백질, 지방, SNF, 그리고 총 고형분은 분만 후 12시간부터 48시간까지 급속도로 감소하였다. 그러나, 유당은 분만 12시간 후의 함량이 가장 낮았으며, 회분 함량은 거의 변화하지 않았다. 분만 후 12시간 및 24시간의 IgG함량은 그 이후에 착유한 것보다 높았으며(p<0.05), 12시간 후의 IgG 함량은 큰 차이가 없는 것으로 나타났다. 분만 후 시간의 경과에 따른 초유 중의 지방산 조성은 거의 변화하지 않았으나, lauric acid, myristic acid, 그리고 총 포화 지방산 함량은 경산우에서 감소하는 경향을 나타내었다. 미경산우의 capric acid, lauric acid, myristic aid, 그리고 palmitoleic acid는 경우산의 초유보다 높은 함량을 나타내었다.

Preparation and Characterization of a Polar Milk Lipid-enriched Component from Whey Powder

  • Milk fat globule membrane (MFGM) is a lipid carrier in mammals including humans that consists mainly of polar lipids, like phospholipids and glycolipids. In this study, a process to enrich polar lipids in commercial butter and whey powder, including polar lipids of MFGM, was developed. WPC (whey protein concentrate) 60 was selected as the most suitable raw material based on the yield, phospholipid, protein, and lactose content of the polar lipid fraction obtained by ethanol extraction of two WPC (WPC60 and WPC70) and two buttermilk (A and B). After fractionation under optimum conditions, the polar-lipid enriched fraction from WPC60 contained 38.56% phospholipids. The content of glycolipids, cerebroside, lactosylceramide, ganglioside GM3, ganglioside GD3, was 0.97%, 0.55%, 0.09%, and 0.14%, respectively. Rancimat results showed that the oxidation stability of fish oil increased with an increase in the polar-lipid fraction by more than 30 times. In addition, the secretion of IL-6 and TNF-α decreased in a concentration-dependent manner after treatment of RAW 264.7 cells with 0.1 to 100 ppm of the polar lipid fraction. In this study, polar lipid concentrates with antioxidant and anti-inflammatory activity, were prepared from milk processing by-products. The MFGM polar lipid concentrates made from by-products are not only additives for infants, but are also likely to be used as antioxidants in cooking oils and as active ingredients for functional foods.

원료유(原料乳)의 이상유무(異常有無)와 전기전도도(電氣傳導度) (Measurement of Electrical Conductivity for Detection of Abnormality of Raw Milk)

  • 목장(牧場)으로 부터 납유(納乳)된 혼유상태(混乳狀態)의 수송관우유(輸送罐牛乳)의 이상유무판정(異常有無判定)을 위한 한 방법(方法)으로서 전기전도도측정법(電氣傳導度測定法)의 적용가능성(適用可能性) 여부(如否)를 유당함량(乳糖含量), 염소농도(鹽素濃度) 및 koestler number와 관련시켜 검토(檢討)하였다. 시료유(試料乳)의 전기전도도(電氣傳導度)와 koestler number와의 상관계수(相關係數)는 0.76이었다. 43개 시료유중(試料乳中) koestler number 3.5를 기준(基準)으로 한 이상유(異常乳)의 비율(比率)은 14.0%, 염소농도(鹽素濃度) 0.14%를 기준(基準)으로 하면 15.3%, 그리고 전기전도도(電氣傳導度) $6.260({\mu}mho/cm){\times}10^3$을 기준(基準)으로 한 비율(比率)은 14.0%였다. 가수(加水), 가염(加鹽)된 시료유(試料乳)의 이상유무판정(異常有無判定)에는 전기전도도측정(電氣傳導度測定)외에 보조적(補助的)으로 비중측정(比重測定)이 필요(必要)하였다.

WPI의 이화학적 특성과 항산화성에 관한 연구 (A Study on the Physicochemical Properties and Antioxidative Activity of Whey Protein Isolate)

  • In this study, physicochemical properties and the antioxidative activity of whey protein isolate(WPI) for com germ oil were measured. The pH of WPI was 6.26, and the titrable acidity was 0.18%. The WPI’s moisture content was 5.2% and each of the other element content such as lactose, crude protein, crude ash and crude fat was found to be 0.8%, 90.7%, 2.7% and 0.6%, respectively. The amounts of active SH group in WPI 9 ${\mu}$ M-g and total colony counts of bacteria was 5.9 ${\times}$ 10$^3$ CFU-g. ${\alpha}$-Lactalbumin, ${\beta}$-lactoglobulin and bovine serum albumin(BSA) were shown in WPI as major protein by electrophoresis. The antioxidative effect of WPI and other antioxidants on com germ oil used as substrate was determined by peroxide value(POV) and conjuqated dienoic acid value(CDV). By these results, the order of antioxidative effects could be defined as BHT 0.02%>ascorbic acid 0.1%>WPI 0.1%>WPI 0.02%>ascorbic acid 0.02%>control>tocopherol 0.02%>tocopherol 0.1%, respectively. Also the induction period of com germ oil added with WPI was longer by 1.6 times than that of control(none added any antioxidant). Therefore the fact suggested that WPI could be utilized as a good antioxidative agents.

Comparison of Oven-drying Methods for Determination of Moisture Content in Feed Ingredients

  • An accurate assessment of moisture content in feed ingredients is important because moisture influences the nutritional evaluation of feedstuffs. The objective of this study was to evaluate various methods for moisture content determination. In Exp. 1, the weight loss on drying (LOD) of corn, soybean meal (SBM), distillers dried grains with solubles (DDGS), whey permeate, whey powder, spray-dried porcine plasma (SDPP), fish meal, and a mixed diet of these 7 ingredients were measured by oven drying at $135^{\circ}C$ for 2 h. Additionally, the samples were dried at $105^{\circ}C$ for 3, 6, 9, 12, or 15 h. The LOD contents of the DDGS, whey permeate, and whey powder measured by drying at $135^{\circ}C$ for 2 h were greater than the values measured by drying at $105^{\circ}C$ for 3 h (p<0.05). All samples except SDPP (p = 0.70) dried at $105^{\circ}C$ for 6, 9, 12, or 15 h caused more LOD compared with the samples dried for at $105^{\circ}C$ for 3 h (p<0.05). The LOD contents of the individual ingredients were additive when dried at $105^{\circ}C$ regardless of drying time. In Exp. 2, moisture contents of corn, SBM, wheat, whey permeate, whey powder, lactose, and 2 sources of DDGS (DDGS1 and DDGS2) were measured by the Karl Fischer method, oven drying at $135^{\circ}C$ for 2 h, and oven drying at $125^{\circ}C$, $115^{\circ}C$, $105^{\circ}C$, or $95^{\circ}C$ for increasing drying time from 1 to 24 h. Drying samples at $135^{\circ}C$ for 2 h resulted in higher moisture content in whey permeate (7.5% vs 3.0%), whey powder (7.7% vs 3.8%), DDGS1 (11.4% vs 7.5%), and DDGS2 (13.1% vs 8.8%) compared with the Karl Fischer method (p<0.05). Whey permeate and whey powder were considerably darkened as the drying time increased. In conclusion, drying samples at $135^{\circ}C$ for 2 h is not appropriate for determining the moisture content in whey permeate, whey powder, or DDGS as well as the mixed diet containing these ingredients. The oven-drying method at $105^{\circ}C$ for 5 to 6 h appears to be appropriate for whey permeate and whey powder, and at $105^{\circ}C$ for 2 to 3 h for DDGS.

Effect of Feeding Strategies on Milk Production of Holstein Dairy Cows Managed by Small-Farmers Alpine Grassland in Korea

  • The main objective of this experiment was to investigate effects of two different feeding systems on body weight, milk yield, milk composition, and mineral and fatty acids content of Holstein dairy cows' milk. Sixteen of 25 months-old Holstein dairy cows were assigned to two groups (n=8) to study effects of the feeding system for 150 days. Two feeding systems were compared for five months; Group 1 was housed indoors and mainly fed a concentrate diet, Group 2 was maintained outdoors for five-seven hours/day on various kinds grass in a pasture. The experiment was conducted June-October 2017. Results revealed the indoor-fed cows had higher body weight, that was significant compared with the outdoor-based feeding system of Holstein dairy cows (p<0.05). Indoor-raised milking cows had higher milk yield (32.45 kg) as compared with pasture-raised milk yield (26.44 kg). Cows fed indoors significantly increased milk yield, total protein content, lactose, citric acid level, and lowered level of total solid and free fatty acids relative to the pasture-fed milking cows (p>0.05). There were higher levels of mineral content and fatty acid content in the milk of indoor-fed dairy cows than the pasture-raised dairy cows (p>0.05). Our study results demonstrated the potential benefits of the indoor feeding system for increased body weight, milk yield, mineral and fatty acids content summer through autumn when low pasture growth rates and quality may otherwise limit production.

뽕잎추출액을 첨가한 발효유의 이화학적 특성 (Physicochemical Characteristics of Fermented Milk Containing Mulberry Leaf Extract)

  • 뽕잎추출액을 첨가한 발효유의 이화학적 특성을 분석한 결과, pH는 배양시간이 경과함에 따라 뽕잎추출액 첨가군의 pH가 낮아졌으며, 산도는 발효유의 뽕잎추출액 첨가 농도가 증가함에 따라 증가하였다. 생균수는 뽕잎추출액 0.5% 첨가군의 생균수가 가장 많았고, 뽕잎추출액의 첨가량에 따라 무지유고형분, 조단백질, 조지방 및 조회분 함량은 증가하였으며, 수분과 lactose 함량은 유의하게 감소하였다(p<0.05). 무기질함량에서는 칼슘, 마그네슘, 칼륨 함량이 뽕잎추출액 첨가군에서 유의하게(p<0.05) 증가하였고, 색도는 발효유의 뽕잎추출액 함량이 높을수록 L값과 a값은 낮았으며 b값은 증가되었다. 관능검사에서 색은 뽕잎추출액 1.0% 첨가군에서 가장 높았으며, 향미는 뽕잎추출액 0.5% 첨가군이 가장 높았고, 맛과 질감은 뽕잎추출액 1.0% 첨가군이 가장 높아 전체적인 기호도에서 뽕잎추출액 1.0% 첨가군이 높게 나타났다. 따라서 뽕잎추출액을 첨가한 발효유의 이화학적 특성과 관능적 특성을 분석한 결과 뽕잎추출액 1%를 첨가한 발효유가 가장 적합하였다.

유청분말을 이용한 생고분자 필름의 제조 (Preparation of Whey Powder-Based Biopolymer Films)

  • 본 연구는 산업 부산물인 유청분말과 필름 matrix를 보조하는 sodium caseinate를 혼합 이용하여 생고분자 필름을 제조하기 위한 필름용액의 최적 pH를 결정하였으며, 각기 다른 혼합비의 유청분말-sodium caseinate 혼합물로 제조한 생고분자필름의 기계적 성질에 미치는 가소제의 영향을 조사하고, 유청분말의 혼합비율을 높게 하였을 때 나타나는 낮은 필름의 기계적 성질을 가교제를 첨가하여 개선함으로써 최대한 유청분말을 이용할 수 있는 혼합비율과 제조조건을 결정하였다. 필름제조 결과, 유청분말과 sodium caseinate의 혼합비율이 70 : 30까지 필름으로써의 기계적 기능을 부여했으며, 그 이상의 유청분말 함량에서는 유청분말 중의 높은 lactose의 함량에 기인한 끈적거리는 현상이 나타나 기계적 성질에 대한 실험을 할 수 없었다. pH에 따른 필름의 성상은 필름의 pH가 5.0이하이었을 때에는 필름용액이 겔화되어 유리판에 부을 수가 없었으며, pH 6.0과 7.0일 경우에는 필름용액 중 원료 단백질이 완전히 용해되지 않아 입자가 존재하였다. 필름용액의 pH를 10.0으로 조절하였을 때 최대 인장강도(TS)와 최대 신장률(E)이 각각 15.2 MPa과 191.8%를 보여 가장 필름형성에 적합한 pH로 나타났다. 가소제로서 glycerol의 사용은 부적합한 것으로 나타났으며, 가소제로서 sorbitol을 필름원료 혼합물의 30%와 40%로 첨가하였을 때 제조된 필름의 최대 인장강도는 각각 10.8 MPa자 6.6 MPa로, 또한 최대 신장률은 각각 96.5%와 149.7%로 나타나 가장 필름형성에 적합한 것으로 나타났다. 필름의 기계적 성질에 미치는 가교제의 영향은 가소제로써 sorbitol 30%를 사용하였을 때에는 sodium citrate가, 가소제로써 sorbitol 40%를 사용하였을 때는 sodium chloride가 가장 많은 영향을 주는 것으로 나타났다. 결론적으로 본 연구에서는 필름용액의 pH를 10.0으로 조절하고 가소제와 가교제를 첨가함으로써 혼합원료 중 유청분말의 비율을 70%까지 사용하여 양호한 기계적 성질을 지닌 생고분자 필름을 제조할 수 있었다.
