The objective of this study was to identify lactic acid bacteria (LAB) isolated from kimchi and to evaluate its characteristics and functional properties for application in fermented dairy products as a probiotic or commercial starter culture. Eight stains isolated from kimchi were selected through an investigation of phenotypic characteristics. Two strains (DK211 and DK303) were identified as Lactobacillus plantarum, another two (DK207 and DK215) as Lactobacillus paracasei, and one (DK301) as Lactobacillus sakei. The remaining three strains were identified as species of Weissella. All selected Lactobacillus strains had acid and bile tolerance, even though there was wide variation in the ability of each strain. DK303 showed a remarkably higher proteolytic activity. There were no significant differences in β-galactosidase activity among the tested strains, except that DK301 showed no activity. Auto-aggregation varied between 82.1 and 90.0%, and hydrophobicity values ranged from 0.5 to 51.6%.The strongest auto-aggregation and hydrophobicity were observed in DK211. All selected strains showed better 1,1-diphenyl-2-picrylhydrzyl (DPPH) scavenging activity than commercial strains. DK211, DK215, DK301, and DK303 had effective inhibitory activity against all pathogens tested except E. coli. When selected strains were used for yogurt preparation as a single starter culture, the time required to reach target titratable acidity (0.9) was 11-12 h. The yogurt fermented with DK211 had favorable panelists ratings for most sensory attributes, which were comparable with yogurt fermented with a commercial strain. The results suggest that strains isolated from kimchi could be potential probiotic and starter cultures for use in yogurt manufacturing.
The aim of this study was to investigate the efficacy of Lactobacillus spp. W44 and J124 strain isolated from Kimchi on the growth rate, average daily gain (ADG), feed conversion and change of intestine microflora in the weaning piglets. In the experiment 1, growth rate was significantly increased to $28.5\;{\pm}\;4.3\;kg$ and $27.6\;{\pm}\;3.3\;kg$ after oral administration of W44 and J124 strain as feed additives, respectively (p<0.05). ADG and feed conversion were also significantly improved after administration during 44 experiment days (p<0.05). In the analysis of intestinal microflora, the number of Lactobacillus spp. in the experiment groups was significantly increased 100 to 1,000 times compared to those of control group. In the experiment 2, the efficacy of W44 and J124 strains on the growth rate, ADG, feed conversion and change of intestine microflora were reconfirmed significantly (p<0.05). However, there were no significant differences on feed conversion in the W44 bead and J124 bead groups. In summary, our results suggest that W-44 and J-124 stains from Kimchi have a significant effect on the weight gain and feed conversion, and it may be useful probiotic strains for the weaning piglets as feed additives.
산란계의 분변으로부터 생균제 특성을 가진 유산균을 분리하기 위하여, 무작위 선발법과 agar well diffusion assay법을 사용하여 다수의 유산균을 1차 선발하였으며 이 중에서 대장균 억제능이 가장 우수한 CPM-7 균주를 분리하였다. 최종 선발된 CPM-7 균주는 형태학적 특징, 당 이용성 및 16S rRNA 서별 분석을 통하여 Lactobacillus salivarius CPM-7으로 동정되었다. L. salivarius CPM-7은 내산성 및 내담즙성이 우수한 것으로 나타났는데, pH 2에서 30분 동안 및 pH 3에서 6시간 동안 생균수가 거의 유지되었으며, bile salt 0.2%가 첨가된 MRS 배지에서 증식할 수 있었다. L. salivarius CPM-7은 a-galactosidase를 포함한 다수의 효소를 생산하는 것으로 확인되었으며, 돼지의 장 상피세포에 흡착할 수 있었다. L. salivarius CPM-7의 배양액 및 중화액은 자돈 설사를 유발하는 것으로 알려진 E. coli K88에 대한 강력한 억제능을 나타내었다.
본 연구에서는 원유와 사일리지, 전통발효식품 등으로부터 유방염 원인균에 대한 항균성을 가지는 유산균을 분리하였고, 이들 중 원유에서는 79, 91-3 균주와 전통발효식품에서는 SA131, NK24, NK34, 32, 44-1 균주, 사일리지에서는 253, 261, 262, 263, 265 균주, 닭분변에서 B32, C23 균주에서 유방염 원인균에 대한 비교적 높은 항균력을 확인할 수 있었다. 이를 통해 새로운 항생제 대체물질의 가능성을 확인할 수 있었다.
Objective: The aim of the present study was to investigate the effect of yeast with bacteriocin and Lactobacillus cultures (mixture of Lactobacillus agilis BCRC 10436 and Lactobacillus reuteri BCRC 17476) supplements, alone or in combination, on broiler chicken performance. Methods: A total of 300, 1-d-old healthy broiler chickens were randomly divided into five treatment groups: i) basal diet (control), ii) basal diet+0.25% yeast (Saccharomyces cerevisiae) (YC), iii) basal diet+0.25% yeast with bacteriocin (BA), iv) basal diet+Lactobacillus cultures (LAB), and v) basal diet +0.25% yeast with bacteriocin+Lactobacillus cultures (BA+LAB). Growth performance, cecal microbiota, cecal fermentation products, and blood biochemistry parameters were determined when chickens were 21 and 35 d old. Results: The supplementation of YC, BA, and BA+LAB resulted in a significantly better feed conversion rate (FCR) than that of the control group during 1 to 21 d (p<0.05). The LAB supplementation had a significant effect on the presence of Lactobacillus in the ceca at 35 d. None of the supplements had an effect on relative numbers of L. agilis and L. reuter at 21 d, but the BA supplementation resulted in the decrease of both Lactobacillus strains at 35 d. The BA+LAB supplementation resulted in higher short chain fatty acid (SCFA) in the ceca, but LAB supplementation significantly decreased the SCFA at 35 d (p<0.05). All treatments tended to decrease ammonia concentration in the ceca at 21 d, especially in the LAB treatment group. The BA supplementation alone decreased the triacylglycerol (TG) concentration significantly at 21 d (p<0.05), but the synergistic effect of BA and LAB supplementation was required to reduce the TG concentration at 35 d. The YC supplementation tended to increase the plasma cholesterol at 21 d and 35 d. However, the BA supplementation significantly decreased the cholesterol and low density lipoprotein cholesterol level at 35 d. In conclusion, the BA+LAB supplementation was beneficial to body weight gain and FCR of broiler chickens. Conclusion: The effect of BA and LAB supplementation may be a result of the growth of lactic acid bacteria enhancement and physiological characterization of bacteriocin, and it suggests that the BA and LAB supplementation level or Lactobacillus strain selection should be integrated in future supplementation designs.
The beneficial effects of lactic acid bacteria (LAB) have been intensively investigated in recent decades with special focus on modulation of the host intestinal microbiota. Numerous discoveries of effective probiotics are driven by a significantly increasing demand for dietary supplements. Consequently, technological advances in the large-scale production and lyophilization are needed by probiotic-related industries for producing probiotic LAB for commercial use. Our study had a dual objective, to determine the optimum growth medium composition and to investigate appropriate cryoprotective additives (CPAs) for Lactobacillus salivarius, and compare its responses with other Lactobacillus species. The one-factor-at-a-time method and central composite design were applied to determine the optimal medium composition for L. salivarius cultivation. The following composition of the medium was established (per liter): 21.64 g maltose, 85 g yeast extract, 1.21 ml Tween 80, 6 g sodium acetate, $0.2g\;MgSO_4{\cdot}7H_2O$, $0.02g\;MnSO_4{\cdot}H_2O$, $1g\;K_2HPO_4$, $1.5g\;KH_2PO_4$, $0.01g\;FeSO_4{\cdot}7H_2O$, and 1 g sodium citrate. A cryoprotective additive combination comprising 10% (w/v) skim milk and 10% (w/v) sucrose supplemented with 2.5% (w/v) sodium glutamate was selected for L. salivarius, and its effectiveness was confirmed using culture-independent methods in the freeze-dried cells of the Lactobacillus strains. In conclusion, the optimized medium enhanced the species-specific cultivation of L. salivarius. On the other hand, the cryoprotective effects of the selected CPA mixture may also be dependent on the bacterial strain. This study highlights the necessity for precise and advanced processing techniques for large-scale production of probiotics in the food and feed industries.
김치로부터 강력한 H. pylori 생육 저해활성을 보이는 균주를 분리, 동정하여 Lb. plantarum NO1으로 명명하였다. Lb. plantarum NO1은 H. pylori 뿐만 아니라 그람 양성균 및 그람 음성균들에 넓은 범위의 저해활성을 나타내었다. Lb. plantarum NO1의 배양 상징액을 H. pylori배양액에 첨가한 후 H. pylori의 urease 활성을 측정한 결과 Lb. plantarum NO1의 강한 urease 억제활성($40{\sim}60%$ 저하)을 확인할 수 있었다. AGS위암 세포주에 H. pylori를 부착시킨 후 Lb. plantarum NO1의 배양액을 첨가하여 AGS세포에서 H. pylori 탈착능을 측정한 결과 Lb. plantarum NO1은 유산균 배양액 무첨가보다 33% 이상 높은 H. pylori 탈착능을 나타내었으며, 비교구로 사용된 Lb. rhamnosus GG, Lb. sakei SI3에 비해 더 우수한 H. pylori 탈착능을 나타내었다. 분리균주의 장내 생존성 여부 확인을 위하여 내산성, 인공위액에서 2시간동안 처리한 결과 Lb. plantarum NO1이 초기균수$(10^9CFU/ml)$를 유지하면서 높은 저항성을 나타내었다. Oxgall 농도 0.3%와 0.5%의 인공담즙에서 24시간 처리한 후에도 초기균수$(10^9CFU/ml)$를 유지하였다. 뿐만 아니라 인공위액에서 생존한 균주를 연속적으로 인공담즙으로 처리하였을 때에도 높은 생존율$(10^8{\sim}10^9CFU/ml)$를 유지하였다. Lb. plantarum NO1의 용혈성 반응 유무 결과 용혈반응이 일어나지 않았으므로 인체에 안전하다는 것을 간접적으로 확인할 수 있었다. 본 연구에서 김치로부터 분리한 H. pylori억제 유산균 Lb. plantarum NO1은 장내에 생존 가능성도 높으며, 동시에 위에서 효과적으로 H. pylori를 억제 할 수 있을 것으로 기대되어진다.
흑마늘 농축액을 첨가한 요구르트의 품질특성 평가를 위하여 탈지분유에 0.5~1.5% (w/v) 흑마늘 농축액을 혼합하고 Streptococcus thermophilus와 Lactobacillus delbrueckii subsp. bulgaricus의 혼합균주를 접종하여 $42^{\circ}C$에서 24시간 배양하였다. 흑마늘 농축액 첨가량에 따른 유산균의 생육정도, 요구르트의 이화학적 및 관능적 특성을 검토하였다. 흑마늘 농축액이 첨가된 요구르트의 pH는 발효 6시간 이후부터 pH 6.0 이하로 대조군에 비해 더 빨리 산성화 되었으며, 발효 24시간에 적정산도는 흑마늘 농축액의 첨가량이 높을수록 증가율이 더 높아 흑마늘 농축액 1.5% 첨가군의 경우 발효 전에 비해 약 5.7배 증가하여 0.74%였다. 흑마늘 농축액을 첨가한 요구르트의 생균수는 발효 3시간까지는 흑마늘 첨가량이 증가할수록 생육정도가 감소하였으나 발효시간이 경과함에 따라 대조군과 유의적인 차이가 없었다. 흑마늘 농축액의 첨가량이 증가할수록 요구르트의 점도는 감소하는 경향이었으나 통계적인 유의차는 없었고, 색도는 흑마늘 농축액의 첨가량이 증가할수록 명도(L값)는 감소하였고 적색도(a값)와 황색도(b값)는 증가하였다. 흑마늘 농축액의 첨가량이 증가할수록 전체적인 기호도가 감소하였는데 이상의 모든 분석 결과를 종합하여 볼 때 요구르트 제조시 흑마늘 농축액의 적정 첨가 농도는 1% 미만이 적합한 것으로 판단된다.
막걸리내 유산균의 특성을 연구하기 위해 알코올 내성이 있는 P. acidilactici K3를 첨가하여 막걸리를 담갔을 때와 시중에 판매되고 있는 생막걸리에 P. acidilactici K3를 첨가하여 한 달간 저장했을 때의 이화학적 변화와 젖산균수의 변화를 조사하였다. 그 결과, P. acidilactici K3를 첨가하여 발효하였을 때의 젖산균 생균수는 이를 첨가하지 않은 막걸리보다 3배 이상 많았으나 알코올 함량은 큰 차이를 보이지 않았으며 당도, 환원당, 총당, 탁도, pH, 산도, 아미노태 질소함량은 큰 차이를 보이지 않았다. 또한 시판되는 생막걸리에 P. acidilactici K3를 첨가하여 저장기간 동안 이화학적 변화를 관찰한 결과, 젖산균 생균수는 P. acidilactici K3를 첨가한 생막걸리는 $10^7CFU/ml$을 유지하였으며, 알코올함량, 탁도, 당도, 환원당, 총당, pH, 총산, 아미노태 질소함량은 큰 차이가 없었다. 본 실험결과, 알코올 내성 젖산균 P. acidilactici K3가 막걸리의 이화학적 성질에 큰 영향을 미치지 않으며, 막걸리 발효균주 및 첨가제로의 응용이 가능함을 확인하였다.
마늘 분말을 첨가한 요구르트의 품질특성 평가를 위하여 탈지분유에 0.1~0.5%(w/v) 마늘 분말을 혼합하고 Streptococcus thermophilus와 Lactobacillus delbrueckii subsp. bulgaricus의 혼합균주를 접종하여 $42^{\circ}C$ 에서 24시간 배양하였다. 마늘 분말 첨가량에 따른 유산균의 생육정도, 요구르트의 이화학적 및 관능적 특성 을 검토하였다. 마늘 분말이 첨가된 요구르트의 적정산도는 0.78~0.81%로 마늘 분말이 첨가되지 않은 대조군(0.73%)보다 높았다. 마늘 분말을 첨가한 요구르트의 생균수는 0.3% 첨가군에서만 상대적으로 낮았고 여타 실험군은 대조군과 유의적인 차이가 없었다. 마늘분말의 첨가량이 증가함에 따라 요구르트의 점도도 상승하여 0.5% 마늘 분말이 첨가된 요구르트의 점도가 가장 높았으며, 색도는 대조군과 마늘 분말 첨가 요구르트 사이에 유의적인 차이가 없었다. 관능적 특성을 조사한 결과 마늘 분말의 첨가에 따라 색은 더 높은 선호도를 보였고, 마늘 맛이나 향은 마늘 분말의 첨가량에 비례적으로 증가하였으며, 전반적인 기호도는 0.1%와 0.3% 첨가시는 대조군과 유의차가 없었다. 이상의 결과를 종합하여 볼 때 요구르트 고유의 품질특성이나 유산균의 생육 및 관능적 특성에 영향을 미치지 않는 범위에서 첨가 가능한 마늘 분말의 적정 수준은 0.3% 미만으로 판단된다.
