  • 일반적으로 사용되는 젖산균 선택 배지와 변화시킨 배지 중에 김치 젖산균 분리를 위한 선택배지는 Enterococcus속과 Pediococcus속 선별에 KF Streptococcus 배지가, Lactococcus속 선별에 pH 지시약을 첨가한 M-17배지가, Lnctobacillus속 선별에 m-LBS 배지가, Leuconostoc선별에 PES-3 배지가 우수하였다.

Differential Transformation of Ginsenosides from Panax ginseng by Lactic Acid Bacteria

  • Ginsenosides have been regarded as the principal components responsible for the pharmacological and biological activities of ginseng. The transformation of ginsenosides with live lactic acid bacteria transformed ginsenosides Rb2 and Rc into Rd, but the reactions were slow. When the crude enzymes obtained from several lactic acid bacteria were used for transformation, those from Bifidobacterium sp. Int57 exhibited the most potent transforming activity of ginsenosides to compound K. In comparison, a relatively higher level of Rh2 was produced by the enzymes from Lactobacillus delbrueckii and Leuconostoc mesenteroides. These results suggest that it is feasible to develop a specific bioconversion process to obtain specific ginsenosides using the appropriate combination of ginsenoside substrates and specific microbial enzymes.

Trehalose와 당류가 냉동요구르트의 저장 중 유산균 생존율과 품질특성에 미치는 영향 (Effect of Trehalose and Sugar Alcohol on the Viability of Lactic Acid Bacteria and Quality Characteristics during Frozen Storage of Yoghurt)

  • 본 연구에서는 냉동요구르트 제조 시 trehalose와 sorbitol 첨가가 유산균의 동결변성방지 및 향기성분의 변화에 미치는 영향을 조사하고자 실시하였다. 요구르트를 L. bulgaricus와 S. thermophilus 단독균주 및 혼합균주로 발효시켰을 때 처리구 모두 trehalose와 sorbitol 첨가에 의한 발효과정 중 유산균의 증식효과는 나타나지 않았다. 요구르트를 6주간 냉동저장하는 동안 trehalose와 sorbitol 첨가에 의한 모든 처리구에서 유산균수의 차이가 없었으며, MRS 액상배지에 trehalose와 sorbitol 을 각각 2%, 5% 첨가하여 6주간 냉동저장하였을 때는 5% trehalose 처리구의 동결변성방지 효과가 가장 우수한 것으로 나타났다. 냉동저장 기간 동안 고형분 함량이 12%인 처리구와 20%인 처리구의 생균수는 큰 차이가 나타나지 않아 고형분 함량 차이에 의한 trehalose와 sorbitol 효과에는 차이가 없는 결과를 나타내었다. 요구르트의 동결과 해동을 반복하였을 경우 trehalose와 sorbitol 모두 2% 첨가구보다 5% 첨가구가 우수한 동결변성방지 효과를 나타내었다. 전반적으로 Trehalose 첨가구가 sorbitol 첨가구보다 2%, 5% 모두 동결변성방지 효과가 좋은 결과를 나타내었다. Trehalose와 sorbitol을 첨가하여 요구르트를 제조하였을때 향미성분의 함량은 대조구와 유사하였으며 일반적인 요구르트의 향미를 나타내었다. 냉동저장 중 요구르트 향미성분은 손실되거나, 함량비율의 변화가 발생하지 않았다. 이상의 결과로부터 요구르트의 풍미에는 영향을 미치지 않으면서 냉동저장에 의한 유산균의 동결변성방지제로서의 trehalose첨가가 냉동요구르트의 유산균 안정화에 효과가 있다고 하겠다.

  • The objective of this study is to investigate the effect of Codonopsis lanceolata Bentk et Hook on the growth of lactic acid bacteria and some harmful bacteria. Extract of C. lanceolata was obtained by hydrothermal extraction method and used intactly without concentration. To investigate the inhibitory effect on the growth of harmful bacteria, we used agar dilution method. On agar plate containing more than 25% extract, they showed decreased number of colonies.   • Bacteriocins from lactic acid bacteria have attracted much attention in recent years because of their useful worth in increasing safety and extending shelf life of foods. These substances show an inhibitory effect against some food spoilage bacteria and food-borne pathogens. The inhibitory effect fo the bacteriocin produces by lactic acid bacteria against Listeria monocytogenes(L. monocytogenes) was examined in this study. The culture supernatants of 5 kinds of bacteria among the 10 kinds of testes lactic acid bacteria had the inhibitory activity against Listeria sp., various Gram positive and Gram negative bacteria. Bacteriocin produced by Lactobacillus plantarum(Lact. plantarum) LMG 7945 was the most active toward L. monocytogenes. Bacteriocin production of the Lact. plantarum LMG 7945 cultured on MRS broth was increased late logarithmic phase over early stationary phase. This bacteriocin was stable at heat treatment and acidic pH relatively; The activity was retained after heating at 121$^{\circ}C$ for 15min and was active in the pH range of 2~4 but was lost above pH 5.

  • Bacteriocins in Gram-positive bacteria have attracted much attention because many have a strong antimicrobial activity also against bacteria outside the genera of the producers. Lantibiotics and the pediocin-like bactericins have attracted most attention since they kill a broad spectrum of Gram-positive bacteria including important pathogens. But many other promising Gram-positive bacteriocins have been thoroughly characterized. Recent studies have shown that bacteriocins may playa role in the intestinal flora to protect us against the food-borne pathogens. Bacterial genome sequencing has demonstrated that there may be an arsenal of such compounds and we are only seeing the top of the iceberg. The present review gives a short outlook of the field of bacteriocins with focus on lactic acid bacteria and includes recent findings.

유용미생물을 이용한 육상수조식 양식장 배출물의 재활용 (Recycling Marine Fish Farm Effluent by Microorganisms)

  • 본 연구에서는 광합성세균 또는 젖산균 등과 같은 유용 미생물에 의한 육상수조식 양식장 배출물의 재활용이 가능한지 평가하였다. 제주에 있는 육상수조식 양식장의 배출물을 조사 분석한 결과 배출물은 회분, 수분과 더불어 단백질, 지방으로 구성되어 있었다. 따라서 배출물이 광합성세균 또는 젖산균의 생장에 대한 기질로서 이용 가능한지를 평가하였다. 배출물은 광합성세균 생장의 기질로서 이용할 수 있었으며, 젖산균의 경우에는 당분을 첨가하면 기질로서 이용할 수 있었다. 본 결과는 육상수조식 양식장에서 배출되는 배출물이 유용미생물에 의하여 효과적으로 재활용될 수 있는 가능성을 제시하였다.

  • Lactic acid bacteria in Kimchi fermentation were tested for inhibitory activity against Gram positive bacteria and Gram negative bacteria. The Lactobacillus brevis (KCCM 35464) was found to produce a antimicrobial substance. It showed relatively wide range of inhibition spectrum against gram positive and gram negative bacteria and maintained the inhibitory activity between pH 4.0 and pH 9.0. The antimicrobial substance was obtained in the stationary growth phase and was purified by gel chromatography. The inhibitory effect of the antimicrobial substance on sensitive bacterial strains was determined by filter paper test. The activity of antimicrobial substa- nce was stable at 75$\circ$C. On the basis of its electrophoretic pattern is SDS-PAGE, antimicrobial substance appeared as a single band of 59 KDalton.

김치 발효중 겨자의 첨가효과 (The Additive Effects of Mustard Seed(Brassica juncea) during Fermentation of Kimchi)

  • To investigate the food preservative effects of mustard seed(Brassica juncea), mustard seed were added to Kimchi. Titratable acidity of Kimchi treated with mustard seed was higher than that of non-treated control at initial stage, but it was lower than control after 2 days of fermantation. The number of bacteria and lactic acid bacteria increased rapidly at the initial stage of fermentation and reached plateau by 2 days of fermentation. The number of bacteria and lactic acid bacteria of Kimchi treated with mustard seed was lower than that of control, and the more mustard seed added the less bacteria and lactic acid bacteria were observed.
