젖산 생산을 위한 배지 최적화 (Ootimization of Mekium Components for Lactic Acid Production)

  • Medium components for lactic acid production were optimized with a strain of Lactobacillus sp., isolated by our Lab. Nitrogen source was the key component and manganese ion was also important for lactic acid production in this strain. Optimal concentration of manganese ion was 0.03 g/l as MnSO$_{4}$ 4 - 5 H$_{2}$O base. Other mineral elements, however, had little effect on it. Among the nitrogen sources we examined, yeast extract showed the highest productivity. Yeast extract, the exellent but very expensive medium component, could be partially replaced by soytone until 60% dry base with higher productivity, or by tryptone enforced with vitamines and nucleic acids. In order to replace yeast extract completely, we examined several inexpensive nitrogen sources and their enzymatic hydrolyzates. The hydrolyzate of vital wheat gluten was the best of them.

김치와 김치재료의 콜레스테롤 저하 및 항암효과 (Cholesterol-Lowering Effect and Anticancer Activity of Kimchi and Kimchi Ingredients)

  • The purpose of the paper is to explore the current knowledge on the nutritional evaluation, cholesterol-lowering effect and antitumor activity of kimchi and its ingredients(Korean cabbage, garlic, red pepper powder, ginger and onion). Kimchi contains high contents of nutrients such as vitamins(ascorbic acid, $\beta$-carotene and vitamin B complex), minerals(calcium, potassium, iron and phosphorous), essential amino acids and dietary fiber. Kimch also contains high levels of lactic acid bacteria, allicin, capsaicin, organic acid, phenol compounds, flavonoid and sulfur compounds. The dietary fiber and lactic acid bacteria isolated from kimchi are effective in improving intestinal microflora of human. Isoluble dietary fiber shows anticancer activity, but soluble dietary fiber shows hypocholesterolemic effect. Lactic acid bacteria isolated from kimchi acts as a hypocholesterolemic or anticancer agent. A major ingredient of kimchi is mainly cruciferous and allium family vegetables, which were also reported to prevent cancer and atherosclerosis. It is suggested that kimchi is important not only as one of the traditional fermented Korean food but also as therapeutic agent for carcinogenesis and hypercholesterolemic state.

  • Synthetic biodegradable polymers are of great interest for biomedical applications such as surgical sutures and drug delivery systems. The copolymers of ${alpha}-amino$ acids and ${alpha}-hydroxy$ matrices having the required permeability for drugs. Poly (glycine.co-lactic acid) and poly (glycine-co-glycolic acid) have been synthesized by ring-opening polymerization. Morpholine-2, 5-diane, lactide, and glycolid have been used as starting materials for polydepsipeptides. The synthesized monomers and copoylmers have been identified by NMR and FT-lR spectrophotometer. The thermal properties and glass transition temperatures ($T_g$) of the copolymers have been measured by differential scanning calorimetry. The $T_g$ values of poly (glycine-co-lactic acid) and poly (glycine co.glycolic acid) are increased with increasing mole fraction of morpholine-2, 5-dione in the copolymers.

김치 Starter 용으로 분리한 효모의 동정

  • The eleven strains, which could be used lactic and acetic acids as carbon sources, were isolated from kimchi and identified; the strains were facultative microorganisms which could be grown at low temperature (10$\circ$C) and around pH 3.2. As results of morphological, biochemical and physiological tests, 5 species of 3 genera were identified as Debaryomyces coudertii, Pichia edia, Pichia chambardii, Pichia haplophilia and Saccharomyces fermentati. Each strain was grown in basal media. In acidic resistance and acid utilization test, Saccharomyces sp. YK- 17 and Saccharomyces fermentati YK-19 were grown well in basal and YM media containing 0.3% lactic acid. And two strains were grown in basal and YM media containing O.3% lactic acid and 0.6% acetic acid. Since strain YK-19 was grown better at 10$\circ$C than that in 25$\circ$C, strain YK-19 was known to be a psychrophilic strain.

화학침전을 이용한 발효액의 젖산 회수 및 유기물 특성분석 (Recovery and Characterization of Lactic Acid from Fermentation Broth Using Chemical Precipitation)

  • 유기성 폐기물 소화공정의 발효액에서 젖산(lactic acid)을 회수하기 위해 화학침전법을 평가하였다. 젖산(lactic acid)의 회수율을 높이기 위하여 화학침전제 종류와 교반속도 및 침전시간 등 반응조건이 회수율 향상에 미치는 영향을 살펴보았다. 화학침전제의 종류에 관계없이 주입양이 증가할수록 젖산(lactic acid) 회수율이 증가하는 경향을 보였으며, CaO가 $Ca(OH)_2$$CaCO_3$에 비하여 높은 회수율을 나타냈다. CaO를 사용한 반응조건 최적화 실험결과, 교반속도 180 rpm, 침전시간 24 h, ethanol 주입량 25%(v/v)에서 회수율이 48%로 가장 높게 나타났다. 본 연구에서는 발효액 내 젖산(lactic acid)의 농도만 고려하여 주입할 침전제의 양을 계산하였기 때문에 실제 적용을 위해서는 발효액의 유기산 종류 및 농도를 고려하여 침전제의 투입량을 결정해야할 것이다. 유기물의 정성적 분석(FEEM, SEC) 결과로 볼 때 침전공정은 유기산의 특성에 큰 영향을 미치지 않았다.

오징어 먹즙 첨가에 따른 저염 오징어 젓갈의 비휘발성 유기산 변화 (The Changes of Non-Volatile Organic Acids in Low Salt Fermented Squid Affected by Adding to Squid Ink)

  • Squid ink was added to the low salt fermented squid by 4% of concentration and ripened at 10$^{\cric}C$ for 6 weeks and at 20$^{\cric}C$ for 28 days. The effect of the squid ink on the non-volatile organic acids of low salt fermented squid were investigated. The results are as follows; The non-volatile organic acid in the salt fermented squid without addition of the squid ink was examined and the result showed that lactic and acetic acids were the major organic acids even if very small amount of citric and oxalic acids were detected. In the squid ink added to the low salt fermented squid, total quantity of non-volatile organic acid in the latter part of the ripening was lower than no treatment groups.

동치미의 맛 성분에 관한 연구 (A Study on the Flavor Compounds of Dongchimi)

  • To study the most optimum condition of Dongchimi, the various Dongchimi distinct from the amount of salt, the temperature of fermentation and the ratio of radish to water were examed by sensory evaluation. Also, the content of volatile organic acids and nonvolatile organic acids were investigated by GC, the content of sulfur compounds by GC/MS. Volatile organic acids were identified with those butyl esters and nonvolatile organic acids were done with those TMS derivatives. Sulfur compounds and those decomposed products were extracted by steam distillation In results, the most optimum conditions ware salt 2.4% fermented temperature $4^{\circ}C$, ratio 1:1.5. Volatile organic acids detected were formic, acetic acid, and the amounts were effected by saltness. Nonvolatile organic acids detected were lactio, malio, fumario, tartario acid. Lactic acid was thought to effect overall eating quality. Sulfur compounds were almost the isothiocyanate groups in raw radish, which little in fermented Dongchimi.

In Vitro Antagonistic Activity Evaluation of Lactic Acid Bacteria (LAB) Combined with Cellulase Enzyme Against Campylobacter jejuni Growth in Co-Culture

  • The antibacterial effects of nine lactic acid bacteria (LAB) against Campylobacter jejuni were investigated by using agar gel diffusion and co-culture assays. Some differences were recorded between the inhibition effects measured with these two methods. Only two LAB, Lb. pentosus CWBI B78 and E. faecium THT, exhibited a clear anti- Campylobacter activity in co-culture assay with dehydrated poultry excreta mixed with ground straw (DPE/GS) as the only growth substrate source. It was observed that the supplementation of such medium with a cellulase A complex (Beldem S.A.) enhanced the antimicrobial effect of both LAB strains. The co-culture medium acidification and the C. jejuni were positively correlated with the cellulase A concentration. The antibacterial effect was characterized by the lactic acid production from the homofermentative E. faecium THT and the lactic and acetic acids production from the heterofermentative Lb. pentosus CWBI B78. The antagonistic properties of LAB strains and enzyme combination could be used in strategies aiming at the reduction of Campylobacter prevalence in the poultry production chain and consequently the risk of human infection.

탄소원으로서 입국을 이용한 유산균 발효 (Lactic Acid Fermentation with Rice Koji as a Carbon Source)

  • Recently, several health benefits of rice wine, makgeolli, were known due to the interest on the traditional Korean liquor and the researches on the rice wine are increasing. Organic acids produced during the process of rice wine fermentation play important roles in the taste and flavor. In this study, we have examined the optimal conditions for lactic acid production in rice koji as a carbon source. Skim milk was also used as a supplementary ingredient for the optimization of lactic acid fermentation. Bacterial growth of Lactobacillus sakei was monitored under this condition. The pH, acidity of the culture and the ethanol tolerance of this bacterium were also tested. Through these experiments, we were able to optimize the growth condition of lactic acid bacteria by the addition of skim milk. This was also able to affect the change of pH, acidity, sugar concentration and alcohol tolerance, which might contribute to the improvement of the quality of rice wine. The optimal condition for the growth was 2 days with 10% (w/v) of skim milk concentration. With these results, it was confirmed that rice koji was an effective carbon source for the growth of lactic acid bacteria.

전분질원료(澱粉質原料)를 달리한 고추장의 유기산정량(有機酸定量) (Determination of Organic acids of Kochuzang prepared from various Starch Sources)

  • 전분질원(澱粉質原料)의 종류를 달리하여 양조(釀造)한 3개월 숙성의 찹쌀, 보리쌀, 밀가루, 고구마의 각고추장을 시료로하여 gas chramatography에 의하여 유기산을 분석한 결과 고추장중의 유기산으로 lactic acid, glycolic acid, pyruvic acid, succinic and fumaric acid, malic acid, pyroglutamic acid, citric acid가 모든 시험고추장에서 분리되었고 oxalic acid는 고구마 고추장에서만 검출 되었다. 시험고추장 모두 pyroglutamic acid, pyruvic acid, citric acid가 양적으로 가장 많았으나 밀가루 고추장에서는 이들 함량이 타 고추장에 비하여 다소 적게 나타났다. succinic과 fumaric acid의 함량은 고구마 고추장에서 가장 많았고 보리쌀 고추장이 가장 적었다.
