A Comparative Study on Spermatozoan Maturation in the Hibernating Animals (동면동물의 정자성숙과정에 대한 비교 연구)

  • Acrosome reaction usually occures just before fertilization in most mammals, and it has been known that $Ca^{2+}$ plays an important role in the acrosome reaction and albumin also known as a critical factor for spermatozoan activities. The present study has been designed in order to observe maturing processes of the spermatozoa occurred in the ductus epididymidis and to clarify the relationships of $Ca^{2+}$ concentrations with those processes, and to compare the enzymatic activities of ATPase and the lactate dehydrogenase of the spermatozoa in accordance with time before and after the spermatozoan maturation. From the results, we can confirm that most of the bat spermatozoa come to maturity within the epididymal cauda and may pass through capacitation outside the cauda. However it is expected to be studied that the fluctuation of spermatogenic activity depending on temperature changes and their relationships with the ductus epididymidis and their mutual influences.

Anticancer Potency of Terminalia arjuna Bark on N-nitrosodiethylamine-Induced Hepatocellular Carcinoma in Rats

  • The anticancer potency of the ethanolic extract of Terminalia arjuna on N-nitrosodiethylamine (DEN) induced hepatocellular carcinoma in Wistar albino rats was studied. Single intraperitoneal injection of DEN was administrated to induce liver cancer. After two weeks, phenobarbital (PB) was given orally for fourteen weeks to promote the cancer. The cancer bearing animals treated with ethanolic extract of T.arjuna (400 mg/kg body weight) for 28 days. Nucleic acids such as deoxyribonucleic acid (DNA) and ribonucleic acid (RNA) were estimated in liver and kidney of control and experimental animals. Certain marker enzymes viz, alanine aminotransaminase (ALT), aspartate aminotransaminase (AST), acid phosphatase (ACP), alkaline phosphatase (ALP), 5'-nucleotidase (5'ND) and lactate dehydrogenase (LDH) were assayed in serum, liver and kidney of control and experimental animals. The levels of DNA and RNA were significantly increased in cancer bearing animals. The activities of ALT, AST, ACP, ALP, 5'ND, and LDH were significantly (P<0.001) increased in serum of cancer bearing animals. On the other hand, the levels of ALT, AST were decreased (P<0.001) and ACP, ALP, 5'ND, and LDH were significantly increased (P<0.001) in liver and kidney. These changes were reversed to near normal in drug treated animals. These observations suggest that the ethanolic extract of T.arjuna possess anticancer activity.

Antidiabetic Activity of Polysaccharide Extract from Tetragonia tetragonoides in Streptozotocin-induced Diabetic Mice (스트렙토조토신으로 유도한 당뇨 마우스에서 번행초 다당 추출물의 항당뇨 효과)

  • Tetragonia tetragonioides seems to be a promising antiulcer medicinal plant due to the presence of water-soluble polysaccharide and cerebroside as its major constituents. There have been no previous studies using T. tetragonioides polysaccharide extract (TPE) to assess its antidiabetic effect in streptozotocin (STZ)-induced diabetes in mice. This study was designed to evaluate the antidiabetic effect of TPE in diabetic mice, which was established by one-week intraperitoneal injection of 65 mg/kg STZ. After three weeks of TPE treatment at a dose of 100 mg/kg, a maintenance of body weight, a decrement in plasma glucose, and low levels of triglyceride, lactate dehydrogenase, alkaline phosphatase, and glutamic pyruvic transaminase were observed in diabetic mice. Furthermore, the ingestion of TPE lowered the blood glucose levels during the oral glucose tolerance test (OGTT) and restored most of the tested parameters to their normal levels. Therefore, the antidiabetic potential of T. tetragonioides has been demonstrated for the first time by our research.

Effect of Kaempferol on the Cytotoxicity Induced Oxygen Free Radicals in Skin Fibroblast Derived from Human In Vitro

  • In order to evaluate on the effect of kaempferol on the cytotoxicity of oxygen tree radicals, XTT assay was performed to determine the cell viability after skin fibroblasts derived from human (Detroit 51) that were treated with various concentrations of hydrogen peroxide $(H_2O_2)$. And also, the effect of kaempferol on the cytotoxicity induced by H202 that was examined by cell viability, lactate dehydrogenase (LDH) activity and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity in these cultures. $H_2O_2$ decreased cell viability in dose-dependent manner in these cultures and the $XTT_{90}\;and\;XTT_{50}$ values were determined at concentration of $35{\mu}M\;and\;90{\mu}M$ of $H_2O_2$ after skin fibroblasts derived from human were treated with $15{\sim}90{\mu}M$ of $H_2O_2$ for 6 hours, respectively. $H_2O_2$ was highly toxic on cultured skin fibroblasts derived from human by toxic criteria of Brenfreund and Puerner (1984). In the protective effect of kaempferol on $H_2O_2$-induced cytotoxicity, kaempferol increased DPPH radical scavenging activity and significantly decreased LDH activity. From these results, it is suggested that oxygen tree radical, $H_2O_2$, was highly toxic on cultured skin fibroblasts derived from human, and also kaempferol of flavonoid showed the protection on $H_2O_2$-induced cytotoxicity.

Effects of Insulin on Acetyl-CoA Carboxylase in Bovine Mammary Secretory Cells

  • Objectives of this study were to determine effects of insulin on acetyl-CoA carboxylase (ACC) activity and correlate this activity with relative amounts of ACC in MAC-T cells. MAC-T cells were grown in Medium 199 supplemented with fetal bovine serum (5%), cortisol ($1{\mu}g/ml$), and insulin ($1{\mu}g/ml$). At confuluence, the cells were transferred to $100mm^2$ culture dishes coated with the extracelluar matrix. After 10 h of incubation, the media were replaced with media without fetal bovine serum and the concentration of insulin was lowered to 5 ng/ml. After 24 h, the media were changed to contain the varying concentrations of insulin and incubations continued for 48 h. The addition of insulin resulted in increases in the specific activity of ACC. The maximal effects of insulin on the ACC activity occurred at concentrations of insulin, 1,000 ng/ml. In contrast, the relative change in lactate dehydrogenase (LDH) activity in response to increasing insulin concentration was minimal as compared to the effects of insulin on ACC. Transblot and enhanced chemiluminescence (ECL) analysis indicated that the increase in ACC activity in MAC-T cells caused by insulin were due to actual increases in amounts of enzyme.

The Effects of Microcurrent Stimulation on Erector Spinae Fatigue Index (미세전류 자극이 척주세움근의 피로지수에 미치는 영향)

  • This study aimed to compare the effects of microcurrent stimulation and rest on the fatigue index by applying both to subjects who had accumulated fatigue after performing short-duration physical activity. The experiment was performed at S University from December 2012 to February 2013, on 22 healthy men in their 20s, who were right-handed, and without peripheral or musculoskeletal diseases. All subjects lifted a 10-kg box lift and lower 100 times in 15 minutes. Immediately after that, muscle fatigue index (Median Frequency: MF) and blood fatigue index (Creatine Kinase Lactate: CK, Lactate Dehydrogenase: LDH) were measured by using surface electromyography. The subjects were divided into two groups, and microcurrent stimulation and rest were mediated to the experimental and control groups, respectively, for 20 minutes. After intervention, muscle fatigue index and blood fatigue index were measured and the changes in the accumulated fatigue index were compared. Both groups manifested significantly decreased cumulative fatigue after applying the mediation compared to the pre-intervention level (p < .05), the only exception being the fatigue index of the left erector spinae in the rest group. Compared to the rest group, the microcurrent stimulation group showed a significant decrease in muscle fatigue index (p < .05) but no significant differences were found in the blood fatigue index. Given the results of this study, microcurrent stimulation is considered to be helpful in muscle fatigue recovery for workers who have to perform repetitive movements lifting weights, and additional studies on the correlations of the fatigue recovery with respect to the long-term follow-up of fatigue index and mediation time are necessary.

Hepatoprotective Effect of Stamen Extracts of Mesua ferrea L. against Oxidative Stress induced by $CCl_4$ in Liver Slice Culture Model

  • Stamens of Mesua ferrea L. are a well-known herbal drug used in Indian System of Traditional Medicine to treat various diseases. The claimed activity of this plant part is necessitated to investigate antioxidant and hepatoprotective activity. Authenticated plant sample was extracted with hexane, ethanol (EtOH) and water (aq.) using ASE 100 accelerated solvent extractor. Antioxidant activity was evaluated by means of different in vitro assays. Hepatoprotective effect was investigated on carbon tetrachloride induced oxidative stress in liver slice culture model. Cytotoxic marker lactate dehydrogenase (LDH) released in culture medium and the activity of lipid peroxidation along with antioxidant enzymes (AOEs) namely superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) were estimated. Hexane and EtOH extracts were significantly inhibited DPPH, NO, SOD and $ABTS^+$ radical in dose dependent manner. The trade of phenol content was: aq. extract < hexane extract < EtOH extract. A significant correlation was shown by total phenol content and free radical scavenging activity of extracts. The culture system treated with hexane extract, EtOH extract or ascorbic acid exhibited significant depletion in LDH, lipid peroxidation, antioxidative enzymes SOD, CAT and GR. Hexane extract and EtOH extracts of stamen of M. ferrea protected liver slice culture cells by alleviating oxidative stress induced damage to liver cells.

Evaluation of the Antioxidant and Antiproliferative Properties of a Hot-water Extract from Gulfweed, Sargassum fulvellum

  • Sargassum fulvellum (gulfweed) is a widespread seaweed in the coastal areas of northeast Asia. In the present study, we identified the phenolic compounds present in aqueous and ethanolic extracts of S. fulvellum and evaluated their antioxidative properties and their abilities to block cell proliferation using in vitro assays: antioxidant activity was assessed by using a DPPH assay and superoxide anion scavenging activity, anti-tyrosinase activity, and anti-proliferative activity were assessed using MTT and lactate dehydrogenase [LDH] assays in vascular smooth muscle cells. The hot-water ($65^{\circ}C$) extract had a higher phenol content than the ethanolic extract. The hot-water extract showed a statistically significant increase in free radical scavenging activity and a greater ability to reduce proliferation of vascular smooth muscle cells stimulated with platelet-derived growth factor-BB. Taken together, hot-water extracts of S. fulvellum may be an important source of antioxidative and antiproliferative agents.

Biochemical Properties of Lactate Dehydrogenase Eye-Specific C4 Isozyme: Lepomis macrochirus and Micropterus salmoides (젖산탈수소효소 eye-specific C4 동위효소의 생화학적 특성: 파랑볼우럭(Lepomis macrochirus)과 큰입우럭(Micropterus salmoides))

  • The properties of lactate dehydrogenase (LDH, EC 1.1.1.27) eye-specific $C_4$ isozyme were studied by polyacrylamide gel electrophoresis, Western blotting, immunoprecipitation, and enzyme kinetics. Furthermore, we proposed the optimal conditions for measuring the activity of LDH eye-specific $C_4$ isozyme. The isozymes were detected in the cytosol of eye tissues from Lepomis macrochirus and Micropterus salmoides and were more similar to the $A_4$ than the $B_4$ isozyme. LDH/CS in the eye tissue of L. macrochirus was increased in September, so the ratio of anaerobic metabolism was high. The electrophoretic patterns of mitochondrial LDH were similar to those of cytosolic LDH in the eye tissues of L. macrochirus and Micropterus salmoides. LDH eye-specific $C_4$ isozyme from eye tissue was purified by preparative native-PAGE. The activities of LDH eye-specific $C_4$ isozymes in L. macrochirus and M. salmoides were reduced at concentrations greater than 0.2 mM and 0.1 mM of pyruvate, respectively. These concentrations remained at 5.2% and 15.8% as a result of the inhibition by 10 mM of pyruvate, so the degree of inhibition was very high. The LDH activities of eye tissues were reduced at concentrations greater than 22 mM and 24 mM of lactate, respectively, in L. macrochirus and M. salmoides. The ${K_m}^{PYR}$ of eye-specific $C_4$ was 0.088 mM in L. macrochirus and it was 0.033 mM in M. salmoides. The activities of cytosolic and mitochondrial eye-specific $C_4$ isozymes were high in ${\alpha}$-ketobutyric acid. Furthermore, the activities of eye tissue and eye-specific $C_4$ isozyme had to be measured with 0.5 mM of pyruvate and a buffer solution of pH 7.5. As a conclusion, the eye-specific $C_4$ isozyme in M. salmoides has a high affinity for pyruvate and exhibits maximum activity at a lower concentration of pyruvate and at higher concentration of lactate than that in L. macrochirus. Therefore, it seems that the energy produced by the LDH eye-specific $C_4$ isozyme in M. salmoides was used at the first stage of predatory behavior.

The Therapeutic Effects of Korean Red Ginseng on Carbon Tetrachloride- and Galactosamine-induced Hepatotoxicity in Rats (홍삼의 사염화탄소 및 갈락토사민 유발 간독성에 대한 치료효과)

  • In this study, we investgated the effect of Red Ginseng (KRG) on liver damage induced by carbon tetrachloride (CTC) and galactosamine (GalN) in rats using indicator enzymes such as serum alanine/aspartate aminotransferases, sorbital dehydrogenase, lactate dehydrogenase, and ${\gamma}$-glutamyltransferase. Treatment of KRG restored these enzyme activities to near normal level compared to CTC or GalN treatment alone. Treatment of KRG also enhanced hepatic microsomal enzyme system, malondialdehyde formation, and depletion of reduced glutathione content, which were reduced by CTC or GalN. We also found that the decreased activities of glutathione S-transferase and glutathine reductase but not ${\gamma}$-glutamycysteine synthetase after KRG treatment restored to normal level. These results indicate that KRG has potent therapeutic activity against CTC- and GalN-induced hepatotoxicity in rat.
