Chungpaesagan-tang which is used for treating patients of brain in cerebrovascular disease frequently from clinical doctor has not reported about the effect of neuronal aptosis caused of brain ischemia. The aim of this study is to investigate effect of Chungpaesagan-tang protecting neuronal cells from being damaged by brain ischemia through using organotypic hippocampal slice cultures. We caused ischemic damage to organotypic hippocampal slice cultures by oxygen and glucose deprivation. And added Chungpaesagan-tang extract to cultures. thereafter we measured area percentage of propidium iodide (PI)-stained neuronal cell, lactate dehydrogenase (LDH) levels in culture media and Terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL)-positive cells. Area percentage of PI-stained neuronal cells and count of TUNEL-positive cells in CA1 and DG area of organotypic hippocampal slice culture were significantly decreased in pertinent density level of Chungpaesagan-tang extract. LDH levels in culture media of organotypic hippocampal slice culture were significantly decreased in pertinent density level of Chungpaesagan-tang extract. Within pertinent density level, Chungpaesagan-tang has cell protection effect that prevents brain ischemia damaging neuronal cells and apoptosis increasing.

This study was performed to investigate detoxifying enzyme activities of carboxylesterase(CE), glutathione S-transferase(GST) and lactate dehydrogenase(LDH) at variable toxicity levels in fresh water fish, carp(Cyprinus carpio L.). The carp was exposed to single and combined pesticides of IBP, isoprothiolane and cartap for 48 hr at sublethal doses, $LC_{10}$ and $LC_{26}$. The detoxifying enzyme activities were assayed for the liver, head and gut of the carp. The enzyme activities we discovered were as follows: Both activities of CE and GST were increased at the sublethal doses but were declined by increasing doses. In the gut, we found that the CE activity had high levels in the treatment groups of isoprothiolane+IBP and isoprothiolane+cartap. In the head, the CE activity had high levels in the treatment groups of cartap, IBP and isoprothiolane. However, the GST activities were inconsistent in the head and gut of the fish. Also, the GST activity was declined by increasing protein contents. The highest LDH activity was shown in the isoprothiolane treated fish, while the lowest activity was observed in the isoprothiolane+cartap treatment.
본 연구의 목적은 비외과적 치주치료시에 부가적으로 사용하기 위해 개발한 테트라싸이클린 젤의 효과를 임상지수, 치은 열구액양 및 치은열구액내 효소활성도 측정을 통해 평가함에 있다. 치주낭 깊이가 4mm 이상 6mm이하인 구치가 편악당 적어도 2개 이상 존재하는 중등도 치주염에 이환된 환자 23명을 선택하여, 한 편악에서는 치근활택술 만을 시행하였고, 다른 한 편악에는 치근활택술 후 테트라싸이클린 젤을 치은연하로 주입한 뒤 치근면 처리하였다. 사용된 임상검사 지수는 치태지수, 치은지수, 치주낭 깊이, 치은부착 상실도 및 탐침시 출혈로, 각각 치료시작 전과 치료 후 4, 12주에 검사하였으며, 치은열구액내 효소활성도 검사를 위해 치은열구액을 치료시작 전과 치료 후 1, 2, 4, 12주에 각각 채취하고 치은열구액 양을 조사한 뒤 Lactate dehydrogenase(LDH)와 Beta-glucuronidase(BG)의 활성도를 측정하였고, 효소활성도는 LDH는 340nm에서, BG는 550nm에서 각각 흡광도를 조사 분석하여 다음과 같은 결과를 얻었다. 1. 임상지수를 측정 비교한 결과 치근활택술 만으로 치료한 군과 치근활택술 후 테트라싸이클린 젤을 병용한 군 모두에서 임상적으로 치은건강의 현저한 향상을 관찰할 수 있었으나, 통계상 두 군간의 유의성있는 차이는 발견되지 않았다. 2. 치은열구액 양은 치근활택술 후 테트라싸이클린 젤을 병용한 군이 치근활택술만 시행한 군보다 치료 후 2주와 4주에 유의성 있게 감소하였다. 3. LDH와 BG 효소 활성도는 두 군 모두 현저히 감소하였고, 비록 통계학적인 유의성은 관찰할 수 없었으나 치근활택술 후 테트라싸이클린 젤을 병용한 군에서 감소량이 더 크게 나타났다.
Background: Upper limb ischemia is less common than lower limb ischemia, and relatively few cases have been reported. This paper reviews the epidemiology, etiology, and clinical characteristics of upper limb ischemia and analyzes the factors affecting functional sequelae after treatment. Methods: The records of 35 patients with acute and chronic upper limb ischemia who underwent treatment from January 2007 to December 2012 were retrospectively reviewed. Results: The median age was 55.03 years, and the number of male patients was 24 (68.6%). The most common etiology was embolism of cardiac origin, followed by thrombosis with secondary trauma, and the brachial artery was the most common location for a lesion causing obstruction. Computed tomography angiography was the first-line diagnostic tool in our center. Twenty-eight operations were performed, and conservative therapy was implemented in seven cases. Five deaths (14.3%) occurred during follow-up. Twenty patients (57.1%) complained of functional sequelae after treatment. Functional sequelae were found to be more likely in patients with a longer duration of symptoms (odds ratio, 1.251; p=0.046) and higher lactate dehydrogenase (LDH) levels (odds ratio, 1.001; p=0.031). Conclusion: An increased duration of symptoms and higher initial serum LDH levels were associated with the more frequent occurrence of functional sequelae. The prognosis of upper limb ischemia is associated with prompt and proper treatment and can also be predicted by initial serum LDH levels.
Mastitis is a highly morbid disease that requires detection at the subclinical stage. Tropical countries like India mainly depend on milch buffaloes for milk. The present study was conducted to investigate whether the trace minerals viz. copper (Cu), iron (Fe), zinc (Zn), cobalt (Co) and manganese (Mn) and enzyme activity of lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and aspartate aminotransferase (AST) in riverine buffalo milk can be used as an indicator of subclinical mastitis (SCM) with the aim of developing suitable diagnostic kit for SCM. Trace elements and enzyme activity in milk were estimated with Atomic absorption Spectrophotometer, GBC 932 plus and biochemical methods, respectively. Somatic cell count (SCC) was done microscopically. The cultural examination revealed Gram positive bacteria as the most prevalent etiological agent. A statistically significant (p<0.01) increase in SCC, Fe, Zn, Co and LDH occurred in SCM milk containing gram positive bacterial agents only. ALP was found to be elevated in milk infected by both gram positive and negative bacteria. The percent sensitivity, specificity and accuracy, predictive values and likelihood ratios were calculated taking bacterial culture examination and $SCC\geq2{\times}10^5$ cells/ml of milk as the benchmark. Only ALP and Zn, the former being superior, were found to be suitable for diagnosis of SCM irrespective of etiological agents. LDH, Co and Fe can be introduced in the screening programs where Gram positive bacteria are omnipresent. It is recommended that both ALP and Zn be measured together in milk to diagnose buffalo SCM, irrespective of etiology.
Objectives : We can find out the experimental reports of Yanggyuksanhwa-tang, which has the function of regulating blood pressure related with cerebral disease, and increasing local cerebral blood stream volume, also has the recoveries for the damage of vessel endothelium, and endothelium hypertrophy caused by angiospasm after subarachnoid hemorrhage, and reduces the contraction of smooth muscle, so simultaneously improves necrosis. The anticancer potency of the ethanolic extract of Terminalia arjuna on N-nitrosodiethylamine (DEN) induced hepatocellular carcinoma in Wistar albino rats was studied. Single intraperitoneal injection of DEN was administrated to induce liver cancer. After two weeks, phenobarbital (PB) was given orally for fourteen weeks to promote the cancer. The cancer bearing animals treated with ethanolic extract of T.arjuna (400 mg/kg body weight) for 28 days. Nucleic acids such as deoxyribonucleic acid (DNA) and ribonucleic acid (RNA) were estimated in liver and kidney of control and experimental animals. Certain marker enzymes viz, alanine aminotransaminase (ALT), aspartate aminotransaminase (AST), acid phosphatase (ACP), alkaline phosphatase (ALP), 5'-nucleotidase (5'ND) and lactate dehydrogenase (LDH) were assayed in serum, liver and kidney of control and experimental animals. The levels of DNA and RNA were significantly increased in cancer bearing animals. The activities of ALT, AST, ACP, ALP, 5'ND, and LDH were significantly (P<0.001) increased in serum of cancer bearing animals. On the other hand, the levels of ALT, AST were decreased (P<0.001) and ACP, ALP, 5'ND, and LDH were significantly increased (P<0.001) in liver and kidney. These changes were reversed to near normal in drug treated animals. These observations suggest that the ethanolic extract of T.arjuna possess anticancer activity.
To evaluate the renal toxicity of the antitumor agent, 5-(piperidonomethylphenyl)-2,3-dihydroimidazo[2,1-a]isoquinoline (SDZ-62-434), rats were treated with SDZ-62-434 of 50 mg/Kg, i.p., once and 10 mg/Kg, i.p., daily for 7 days. The kidney weights and urine volume after and during the treatment were observed. The concentrations of urinary creatinine, protein, and the activities of N-acetyl-$\beta $D-glucosaminidase (NAG), alanine aminopeptidase (AAP), $\gamma$-glutamyl transpeptidase (GGT) and lactate dehydrogenase (LDH) in 24 hr urine were also determined. The kidney weights after acute and subacute administration was not affected. The urine excretions were increased 5 days after the acute administration and increased after the daily 3rd day-administration. The excretion of creatinine was similar as that of urine excretion. The excretion of creatinine was increased 5 days after the acute and subacute administration. However, the protein excretion didn't changed in both treatment. Those indicate that SDZ-62-434 might induce the diuresis and also suggest that diuresis might be due to the some metabolites rather than the compound itself. The urinary activities of NAG and LDH were not affected after the acute treatment. However, the urinary activities of AAP and GGT were slightly increased 3 days after the acute administration but, returned to the control value. In subacute treatment, the activities of GGT was not changed. And the activities of NAG were declined after the 7th day-administration. However, the activities of AAP were significantly increased after the 5th day-administration. Furthermore, the urinary activities of LDH were continuously increased during the subacute administration. These results indicate that the high and subacute administration might induce a weak damage on the kidney cells. Furtherrnore, the present results suggest that SDZ-62-434 might have relatively slow-emerging and mild toxicity to the kidney.
Objectives: Cultivated wild ginseng (cWG), called SanYangSanSam, has been used clinically in patients with chronic fatigue in Korea. Little is known about effects of the ginseng distilled (volatile) components produced during evaporizaiton. Recently, we first identified one major component from cWG distilled extract, panaxydol, by using mass spectrometry. However, functional properties of cWG distilled extract and panaxydol remains elusive. Therefore, the present study evaluated the effect of cWG distilled extract or panaxydol on exercise-induced fatigue in rats. Methods: Fatigue was induced by forced swimming and the immobility time was analyzed in male Sprague-Dawley rats. The animals received intraperitoneally either vehicle, cWG distilled extract, or panaxydol 10 min prior to beginning of the forced swimming test (FST) once daily for 5 days. After the FST on day 5, we also analyzed fatigue-related biochemical levels including blood urea nitrogen (BUN), lactate acid (LAC), and lactate dehydrogenase (LDH) in serum and levels of glycogen in liver and soleus muscle. Results: The forced swimming time in cWG distilled extract (0.6 mL/kg)-treated group was significantly longer than that of control group on day 4 and 5. Panaxydol (0.1 and 0.25 mg/kg)-treated groups showed significantly enhanced performance in the forced swimming, compared to control. In addition, a significant decrease in serum LDH level was found in panaxydol-treated group, while there were no alternations in levels of serum BUN and LAC and glycogen in liver or soleus muscle. Conclusion: The present study demonstrated cWG distilled extract and its active component panaxydol have a function of anti-fatigue.
Objectives : This study was carried out to determine if Dendrobii Herba have protective effect against cell injury induced by various toxic agents in rat kidney slices. Water(DWe) and methanol(DMe) extracts were prepared for this experiment. Methods : Cell injury was estimated by measuring lactate dehydrogenase(LDH). Lipid peroxidation was examined by measuring malondialdehyde, a product of lipid peroxidation. Results : DMe prevented the LDH release by $CCl_4$, menadione, tert-butyl hydroperoxide and mercury treatment in vitro in kidney slices, but DWe prevented the LDH release by $CCl_4$ and mercury. DMe also prevented reduction in GSH and lipid peroxidation induced by $CCl_4$ and mercury. Conclusion : Thus, DMe may have more powerful efficacy on anti-oxidative effects when compared with DWe. And further studies have to be followed concerned with extraction of Dendrobii Herba and its change of effects.
