Consideration on supplementary matters when preparing radioactive waste self-disposal (방사성폐기물 자체처분 작성시 보완사항에 관한 고찰)

  • Purpose Recently, in the process of examining the self-disposal of radioactive waste by the Korea Institute of Nuclear Safety, it is difficult to reach the final approval process for self-disposal. In connection with this, we intend to increase the processing efficiency of self-disposal and strengthen safety by analyzing cases of recent supplementary matters. Materials and Methods From 2018 to 2021, we compare and review a supplementary requests that preparing the procedures and plans for the self-disposal of radioactive waste by 20 institutions. In this regard, based on the provisions of the Atomic Energy Safety Act, we derive a detailed proposals for the self-disposal of radioactive waste by arranging the review processing period calculation and supplementary requests that occurred during the review process. Results The representative supplementary requests of the Korea Institute of Nuclear Safety are the calculation of the storage period by type and nuclide of radioactive waste, the contents of the packaging container, the RASIS reporting method, the planned storage method for self-disposal, confirmation of the final disposal company, and the storage period of the waste filter Calculation, radioactive labeling, etc. And it is emphasized as important. Conclusion The expected effects of the guidelines reflecting the latest supplements include reduction of the time required for document preparation and increase of work processing efficiency, improvement of storage efficiency in the radioactive waste storage room, and economic cost reduction. If the radioactive waste self-disposal guideline presented in this study is applied to the field, it is thought that it will be helpful in improving the work efficiency of those who are experiencing difficulties.

Safety Assessment of Liquid-Type Herbal Preparations (한약제제(액제)의 안전성 평가)

  • The safety of liquid-type herbal preparations (47 samples) and beverages containing herbal medicines (18 samples), which are distributed in Gyeonggi province was studied by analyzing heavy metals, benzo(a)pyrene, preservatives and sugar content and to explain the differences between medicine and food based on the labeling standards. The herbal preparation is regarded as medicine and can be differentiated from the beverages containing herbal medicine, which is food, based on the fact that the herbal preparation should be labeled with usage, dosage, efficiency and effect. The mean concentrations of Pb, As, Cd, Hg were found to be 0.014 mg/kg, 0.113 mg/kg, 0.004 mg/kg and 0.003 mg/kg in herbal preparation and 0.009 mg/kg, 0.122 mg/kg, 0.003 mg/kg and 0.002 mg/kg in beverages containing herbal medicines, respectively. In 10 cases of herbal preparations and 6 cases of beverages containing Rehmanniae Radix Preparata (Sukjihwang), benzo(a)pyrene was not found. According to preservative test on herbal medicine, dehydroacetic acid was found less than the indicated content in 6 cases out of 37 cases, which labeled with preservatives, and benzoic acid, dehydroacetic acid and methylparaben were contained with 164.0~198.0 mg/kg, 149.8~272.5 mg/kg and 88.4 mg/kg, respectively, in 3 cases out of 10 cases, which was not labeled with preservatives. Among 16 cases of beverages containing herbal medicines, 3 cases of preservative-labeled beverages had the preservatives, and the rest of all, which was not labeled with preservatives, did not show that they contain any preservative, therefore, it was suitable for the labeling standards. As a result of sugar content test, preservative-labeled products were not appreciably different from the others.

  • The methylotrophic yeast Hansenula polymorpha has been extensively studied as a model organism for methanol metabolism and peroxisome biogenesis. Recently, this yeast has also attracted attention as a promising host organism for recombinant protein production. Here, we describe the fabrication and evaluation of a DNA chip spotted with 382 open reading frames (ORFs) of H. polymorpha. Each ORF was PCR-amplified using gene-specific primer sets, of which the forward primers had 5'-aminolink. The PCR products were printed in duplicate onto the aldehyde-coated slide glasses to link only the coding strands to the surface of the slide via covalent coupling between amine and aldehyde groups. With the partial genome DNA chip, we compared efficiency of direct and indirect cDNA target labeling methods, and found that the indirect method, using fluorescent-labeled dendrimers, generated a higher hybridization signal-to-noise ratio than the direct method, using cDNA targets labeled by incorporation of fluorescence-labeled nucIeotides during reverse transcription. In addition, to assess the quality of this DNA chip, we analyzed the expression profiles of H. polymorpha cells grown on different carbon sources, such as glucose and methanol, and also those of cells treated with the superoxide­generating drug, menadione. The profiles obtained showed a high-level induction of a set of ORFs involved in methanol metabolism and oxidative stress response in the presence of methanol and menadione, respectively. The results demonstrate the sensitivity and reliability of our arrays to analyze global gene expression changes of H. polymorpha under defined environmental conditions.

Labeling Efficiency of $^{99m}Tc$-Labeled RBC Due to Labeling Time Change (표지시간 변화에 의한 $^{99m}Tc$과 적혈구 표지효율)

  • For the preparation of $^{99m}Tc$-labeled RBC, $10{\sim}20\;{\mu}g/kg$ of Stannous(II) chloride and $10{\sim}40\;min$ of preparation was used. For finding out the effect of contrast agent, the blood samples were collected in three days, seven days, and 1 months after the diagnostic procedure. In the normal volunteer, the concentration of reducing agent and preparation time did not effect on the radiochemical yield. But in the patients, 10 mg of Stannous(II) chloride and 60 min incubation times was shown high radiochemical yield. Contrast agent has a significant effect on the radiochemical yield. Although the blood samples which were collected after seven days of diagnostic procedure did not effect on the radiochemical yield of $^{99m}Tc$-labeled RBC, but the radiochemical yield of $^{99m}Tc$-labeled RBC which was prepared with a sample of high concentration of contrast agent in blood led to low radiochemical yield. For these samples, the modified method showed high radiochemical yield than previous in vivo preparation method. The recommended method is followed. Blood collecting was performed at 30 minutes after injection of reducing agent, and it is centrifuged for removal of plasma. After addition of $^{99m}TcO^-_4$, sample reservoir was rotated. After addition of normal saline, and it is centrifuged for separation of saline. Then $^{99m}Tc$-labeled RBC was obtained after removal of saline.

Text Region Extraction from Videos using the Harris Corner Detector (해리스 코너 검출기를 이용한 비디오 자막 영역 추출)

  • In recent years, the use of text inserted into TV contents has grown to provide viewers with better visual understanding. In this paper, video text is defined as superimposed text region located of the bottom of video. Video text extraction is the first step for video information retrieval and video indexing. Most of video text detection and extraction methods in the previous work are based on text color, contrast between text and background, edge, character filter, and so on. However, the video text extraction has big problems due to low resolution of video and complex background. To solve these problems, we propose a method to extract text from videos using the Harris corner detector. The proposed algorithm consists of four steps: corer map generation using the Harris corner detector, extraction of text candidates considering density of comers, text region determination using labeling, and post-processing. The proposed algorithm is language independent and can be applied to texts with various colors. Text region update between frames is also exploited to reduce the processing time. Experiments are performed on diverse videos to confirm the efficiency of the proposed method.

Development of a Synthetic Method for [68Ga]Ga-FAPI-04 Using a Cassette-based Synthesizer (카세트 기반 자동합성장치를 사용한 [68Ga]Ga-FAPI-04의 합성방법 연구)

  • [68Ga]Ga-FAPI-04 is a promising radiopharmaceutical that binds specifically to fibroblast activation protein, which is overexpressed in more than 90% of malignant epithelial tumors but not in normal healthy tissue. This study aimed to develop an efficient method for producing 68Ga-labelled FAPI-04 using a cassette-based automated synthesizer. [68Ga]GaCl3 was eluted from an Eckert & Ziegler Medical germanium-68/gallium-68 generator using 2.5 mL of 0.1 M HCl. The synthesis of the [68Ga]Ga-FAPI-04 was performed using different concentrations of HEPES (1~2.5 M; 4-(2-hydroxyethyl) piperazine-1-ethanesulfonic acid) in 3~10 minutes; amounts of FAPI-04 precursor (5~50 ㎍) and reaction temperature (25℃~100℃) were optimized on the BIKBox® synthesizer. The labeling efficiency of [68Ga]Ga-FAPI-04 was greater than 96% (decay corrected) using 25 ㎍ FAPI-04 synthesized in 10 minutes at 100℃ in 2 M HEPES (pH 3.85), and its stability was greater than 99% at 6 hours. The total synthesis time of [68Ga]Ga-FAPI-04 was 32.4 minutes, and the product met all quality control criteria. In this study, we developed and optimized a labeling method using [68Ga]Ga-FAPI-04 using a cassette-based synthesizer. The devised method is expected to be useful for supplying [68Ga]Ga-FAPI-04 for diagnosis in clinical practice.

  • An iodized oil such as Ethiodol or Lipiodol was selectively retained in the tumor vessels of the large hepatomas as well as in the small daughter hepatomas for long periods following the intra-arterial hepatic injection of such contrast material. The specific aim of the study is to deliver a high internal radiation dose to hepatocellular carcinoma (HCC) in an attempt to control the disease. We were able to replace a small fraction of the stable iodine (I-127) of the 37% iodine in Lipiodol by the $I^{-131}$ with 100% exchange efficiency. $I^{-131}$ labeled Lipiodol was injected through the super-selected tumor feeding artery under superselection or into the proper hepatic arterial level of patients who have malignant hepatomas confirmed by aspiration cytology serum AFP and various imaging modalities. Clinical traial was performed on 43 cases during recent 6 months and follow-up observation was carried out. No severe complications or other adverse reactions were encountered until nowdays. $I^{-131}-Lipiodol$ was stable in vivo and no significant activity was noted in the thyroid, stomach, blood and urine after the injection. Only small fraction of radioisotope activity was noticed in the both side of lungs. Tumor to normal liver radio was very high. Therefore, $I^{-131}-Lipiodol$ (or P-32-Lipiodol) will be effective delivering high internal radiation dose to the tumor while delivering small radiation doses to normal tissues. Labeling, tumor dose calculation and preliminary findings will be presented.

A Change Detection Technique Supporting Nested Blank Nodes of RDF Documents (내포된 공노드를 포함하는 RDF 문서의 변경 탐지 기법)

  • It is an important issue to find out the difference between RDF documents, because RDF documents are changed frequently. When RDF documents contain blank nodes, we need a matching technique for blank nodes in the change detection. Blank nodes have a nested form and they are used in most RDF documents. A RDF document can be modeled as a graph and it will contain many subtrees. We can consider a change detection problem as a minimum cost tree matching problem. In this paper, we propose a change detection technique for RDF documents using the labeling scheme for blank nodes. We also propose a method for improving the efficiency of general triple matching, which used predicate grouping and partitioning. In experiments, we showed that our approach was more accurate and faster than the previous approaches.

  • This paper presents a reliable fully automatic labeling system which fits well with languages having well-developed syllables such as in Korean. The ASL System utilize DAC (Divide and Conquer), a control mechanism, based segmentation algorithm to use phonetic and acoustic information with greater efficiency. The segmentation algorithm is to devide speech signals into speechlets which is localized speech signal pieces and to segment each speechlet for speech boundaries. While HMM method has uniform and definite efficiencies, the suggested method gives framework to steadily develope and improve specified acoustic knowledges as a component. Without using statistical method such as HMM, this new method use only phonetic-acoustic information. Therefore, this method has high speed performance, is consistent extending the specific acoustic knowledge component, and can be applied in efficient way. we show experiment result to verify suggested method at the end.

A double-labeling marker-based method for estimating inbreeding and parental genomic components in a population under conservation

  • Objective: The objective of a conservation program is to maintain maximum genetic diversity and preserve the viability of a breed. However, the efficiency of a program is influenced by the ability to accurately measure and predict genetic diversity. Methods: To examine this question, we conducted a simulation in which common measures (i.e. heterozygosity) and novel measures (identity-by-descent probabilities and parental genomic components) were used to estimate genetic diversity within a conserved population using double-labeled single nucleotide polymorphism markers. Results: The results showed that the accuracy and sensitivity of identity-by-state probabilities and heterozygosity were close to identity by descent (IBD) probabilities, which reflect the true genetic diversity. Expected heterozygosity most closely aligned with IBD. All common measures suggested that practices used in the current Chinese pig conservation program result in a ~5% loss in genetic diversity every 10 generations. Parental genomic components were also analyzed to monitor real-time changes in genomic components for each male and female ancestor. The analysis showed that ~7.5% of male families and ~30% of female families were lost every 5 generations. After 50 generations of simulated conservation, 4 male families lost ~50% of their initial genomic components, and the genomic components for 24.8% of the female families were lost entirely. Conclusion: In summary, compared with the true genetic diversity value obtained using double-labeled markers, expected heterozygosity appears to be the optimal indicator. Parental genomic components analysis provides a more detailed picture of genetic diversity and can be used to guide conservation management practices.
