Objective: The study was conducted to investigate the dephosphorylation of Pseudomonas aeruginosa flagellin (PA FLA) by sweet potato purple acid phosphatase (PAP) and the effect of the enzyme on the flagellin-mediated inflammatory response in the A549 lung epithelial cell line. Methods: The activity of sweet potato PAP on PA FLA was assayed at different pH (4, 5.5, 7, and 7.5) and temperature (25℃, 37℃, and 55℃) conditions. The release of interleukin-8 (IL-8) and the activation of nuclear factor kappa- light-chain-enhancer of activated B cells (NF-κB) in A549 cells exposed to PA FLA treated with or without sweet potato PAP was measured using IL-8 and NF-κB ELISA kits, respectively. The activation of toll-like receptor 5 (TLR5) in TLR5-overexpressing HEK-293 cells exposed to PA FLA treated with or without sweet potato PAP was determined by the secreted alkaline phosphatase-based assay. Results: The dephosphorylation of PA FLA by sweet potato PAP was favorable at pH 4 and 5.5 and highest at 55℃. PA-FLA treated with the enzyme decreased IL-8 release from A549 cells to about 3.5-fold compared to intact PA FLA at 1,000 ng/mL of substrate. Moreover, PA-FLA dephosphorylated by the enzyme repressed the activation of NF-κB in the cells compared to intact PA FLA. The activation of TLR5 by PA-FLA was highest in TLR-overexpressing HEK293 cells at a substrate concentration of 5,000 ng/mL, whereas PA FLA treated with the enzyme strongly repressed the activation of TLR5. Conclusion: Sweet potato PAP has the potential to be a new alternative agent against the increased antibiotic resistance of P. aeruginosa and may be a new conceptual feed additive to control unwanted inflammatory responses caused by bacterial infections in animal husbandry.
Fed-batch fermentation was used to produce the high concentrations of poly-$\beta $-hydroxybutyrate (PHB) and poly-$\beta $-(hydroxybutyrate-co-hydroxyvalerate) (PHB/V). Specific growth rate ($\mu $), yield of cell from glucose (Y$_{x/s}$) were calculated from the two samples in 3 to 5 hours of interval and they were reflected on the determination of glucose feeding rate to maintain the glucose concentration at around 10 g/l in the culture broth. PHB was accumulated after the nitrogen became limited at 60 g/l of dry cell weight by changing ammonia water to 4N-NaOH solution. As results, the final dry cell weight (DCW) of 170 g/l, PHB of 115 g/l were obtained in 50 hours and the overall productivity was 2.4 g/l$\cdot $h. After PHB accumulation, cosubstrate of glucose and propionic acid (PA) was fed to accumulate PHB/V. But, PA feeding rate was decreased from 3 g/l$\cdot $h to 1 g/l$\cdot $h to prevent PA from accumulating to high level in the broth, which is very inhibitory to the cells. As results, DCW, PHB and PHV were 147.5 g/l, 90 g/l and 8 mole % of hydroxyvalerate, respectively.
To improve utilization of boxthorn (Lycii Fructus), the boxthron and mixed boxthron (Lycii Fructus, Schizandrae Fructus, Corni Fructus, Zizyphi Fructus, Zingiberis Rhizoma, Cinnamami Cortex) hot-water extractable concentrates were prepared by vaccum evaporation and its rheological properties were investigated. The rheological properties of concentrates $(20{\sim}50^{\circ}Bx)$ followed power low model and showed a pseudoplastic behavior at the temperature range of $20{\sim}60^{\circ}C$. The apparent viscosity of $20^{\circ}Bx$, $30^{\circ}Bx$, $40^{\circ}Bx$ and $50^{\circ}Bx$ boxthron hot-water extractable concentrate was 0.0074 Pa s, 0.0175 Pa s, 0.0431 Pa s and 0.0988 Pa s, that of mixed boxthorn hot-water extractable concentrate was 0.0099 Pa s, 0.0328 Pa s, 0.0720 Pa s and 0.1940 Pa s at $20^{\circ}C$ and 1500 l/s, respectively. The yield stress of boxthron and mixed boxthron hot-water extractable concentrates ranged from 0.045 to 6.253 Pa and from 0.022 to 8.891 Pa, respectively. The activation energy for the flow of boxthron and mixed boxthorn hot-water extractable concentrates increased from 1.6182 to $2.0543{\times}10^7\;J/kg{\cdot}mol$ and from 1.7057 to $2.1462{\times}10^7\;J/kg{\cdot}mol$ with the concentrations of concentrates, respectively.

Blueberries and strawberries are highly perishable and easily contaminated with microorganisms. To maintain the quality of these commodities during refrigerated storage, the effects of two packaging materials as well as passive modified atmosphere packaging on the quality of blueberries and strawberries were investigated. The harvested blueberries and strawberries were first treated with combined non-thermal treatment of aqueous chlorine dioxide and fumaric acid, followed by packaging with polyolefin film (6,000 mL $O_2/m^2{\cdot}24hr{\cdot}atm$ at $24^{\circ}C$) and polyamide/polyamide/polyethylene film (PA/PA/PE, 60 mL $O_2/m^2{\cdot}24hr{\cdot}atm$ at $24^{\circ}C$), respectively. After combined sanitizer treatment, the populations of total aerobic bacteria in blueberries and strawberries were reduced by 2.50 and 1.97 log CFU/g while those of yeast and molds were reduced by 1.95 and 2.18 log CFU/g, respectively, compared with the control. In particular, microbial growth in these samples packed with PA/PA/PE film was reduced during refrigerated storage. In addition, the blueberries and strawberries packed with PA/PA/PE film underwent lower weight loss than those packed with polyolefin film during storage. These results suggest that appropriate packaging with proper gas permeability is necessary to maintain the quality of blueberries and strawberries during refrigerated storage.
Use of laparoscopic appendectomy (LA) for perforated appendicitis (PA) in children remains controversial because of the development of postoperative intra-abdominal abscess formation. We developed the irrigation method for the prevention of abscess formation after LA performed for PA in children with severe panperitonitis. We called it 'the shaking irrigation'. The object of this study was to analyze the efficacy of this irrigation method. All cases of PA with severe panperitonitis in children that underwent LA with massive shaking irrigation and drainage between June 2003 and December 2007 were studied retrospectively. We included only PA with panperitonitis and large amounts of purulent ascites throughout the abdomen as well as an inflamed small bowel with ileus. Thirty-four children were involved in this study. The mean patient age was eight years. The mean amount of irrigation fluid was 8.2 L (range: 4-15 L), The mean operative time was 89.5 min. The mean length of the hospital stay was 5.1 days. There were no postoperative intra-abdominal abscesses. There was no conversion to open surgery. In conclusion, Use of LA in PA with severe panperitonitis in children is safe and effective. Massive shaking irrigation and abdominal drainage appears to prevent intra-abdominal abscesses after LA for PA with panperitonitis.
Amoving aeration-membrane (MAM) bioreactor was employed for the production of 2$\mu$g/mL of tissue type Plasminogen Activator (tPA)in serum free medium from normal human fibroblast cells. This system could maintain high cell density for long periods of steady state conditions in perfusion cultivation. Under normal operating condition, shear stress was as low as 0.65 dynes/$\textrm{cm}^2$ at the agitation speed of 80 rpm. Even though cell density gradually decreased with increasing agitation speed, tPA production increased linearly with increasing shear stress within a moderate range. This culture system allowed production of 2$\mu$g tPA/mL while maintaining a high cell denisty of 1.0$\times$107 viable cell/mL.
By sequential degradation using partial acid hydrolysis of a weakly acidic polysaccharide (GL-4IIb2'), two acidic oligosaccharide fragments, PA-2' and PA-1-III were isolated and their structures were characterized. PA-2' consisted of almost equal proportion of a rhamnose (Rha) and an unusual sugar, 3-deoxy-D-manno-2-octurosonic acid (Kdo). When permethylated oligosaccharide-alditol derived from PA-2' was analyzed by GC-MS, the peak gave the fragment ions at m/z 189 $(bA_1,\;6-deoxyhexose)$ and at m/z 308 $(aJ_2,\;alditol\;from\;Kdo)$. The peak also gave the characteristic ion at m/z 162 but it did not give the fragment ion at m/z 177, suggesting that Kdo is substituted at C5 but not at C4. Methylation analysis also indicated that PA-2' was composed mainly of terminal Rhap and 5-substituted Kdo. When the reduced product from PA-2' was analyzed by $^1H-NMR$, it gave a signal at 5.09 ppm due to an anomeric proton of ${\alpha}-L-Rha$. These results indicated that PA-2' mainly contained ${\alpha}-L-Rhap-(1{\rightarrow}5)-Kdo$. On the other hand, PA-1-III mainly comprised Rha and Kdo in addition to small proportions of arabinose (Ara) and 3-deoxy-D-lyxo-2-heptulosaric acid (Dha). MS analysis of permethylated oligosaccharide-alditols from PA-1-III suggested that the major peak 1P was $Rhap-(1{\rightarrow}5)-Kdo$ whereas the minor peaks 2P and 3P possessed $Araf-(1{\rightarrow}5)-Dha$ unit and these peaks were produced as epimers during reduction of carbonyl groups in Dha.
Objective: This work was carried out to evaluate the effect of pasture (PA) feeding on buffalo meat quality compared with buffaloes reared intensively with the use of corn silage as a forage base or alternatively with polyphite meadow hay (PH). Methods: Thirty Mediterranean bull buffaloes were distributed into three experimental diet groups: maize silage (MS), PH, and PA. The animals were slaughtered at a live weight of 250 kg, and carcass and meat quality were evaluated. After 7 days of ageing, physical and chemical parameters of longissimus thoracis muscle were determined. To evaluate lipid oxidation the thiobarbituric acid reactive substances was tested at 7 and 14 days, and also the fatty acid profile was recorded by gas chromatography. Results: The PA group, even if it showed carcass parameters lower than those of the silage maize group, reported a good meat percentage (60.59% vs 58.46%, respectively) and lower fat percentage (p<0.001). PA-fed animals showed meat redness, and even if only on raw meat, shear force was higher than the others. Low values of conjugate linoleic acid, polyunsaturated fatty acids, and n-3 were reported in the silage maize group. Principal component analysis (PCA) clearly showed the influence of different diets on meat quality, and PCA1 and PCA2 explained 82% of the variability. Conclusion: Buffaloes reared on PA had meat with high nutritional value even if they showed poor carcass performance compared to the animals fed on MS. Buffaloes fed on polyphite hay were in an intermediate position, similar to grazing animals, according to the same nutritional determinations.

Objectives The purpose of this study was to examine the effects of positive and negative emotion on the quality of life(QoL) in each Sasang contitution, using Positive Affect and Negative Affect Schedule(PANAS) and Short Form-12 Health Survey Questionnaire(SF-12) questionnaire. Methods A total of 1,123 participants completed the Korean version of the PANAS and SF-12 questionnaire. The participants were classified into one Sasang constitution by QSCC II. The effects of emotion on the QoL were compared between the each Sasang contitution using analysis of variance(ANOVA). Results We found significant differences between constitutions in Positive Affect(PA), PA-Joy, PA-Interest and PA-Activation of PANAS. And we found significant differences between constitutions in Mental Component Summary(MCS) of SF-12. Negative correlation of less -0.4 was shown in Negative Affect(NA), NA-Afraid, NA-Upset of PANAS and MCS of SF-12. Conclusions The Soyangin has significantly higher score than Soeumin in PA, PA-Joy, PA-Interest and PA-Activation of PANAS as previous studies. This study also shows the correlation between negative emotion and decline in the quality of mental status.
The present study was conducted to evaluate fertigation effects of balanced manure nutrients from combined fertilizers on the growth of eggplant (Solanum melongena L.) and soil chemical properties in greenhouse cultivation. There are 4 treatments for fertigation; animal liquid manure (LM), LM supplemented with chemical ferilizer (LM+CF) to make nutrient-balanced ferilizer, LM supplemented with chemical ferilizer and phosphorus acid (LM+CF+PA), and conventional chemical fertilzer (CCF). Fruit lenth, width, and weight for LM treatment were significantly lower than LM+CF, LM+CF+PA, and CCF treatments. There was no significant differences in fruit length, width, and weight among LM+CF, LM+CF+PA, and CCF treatments. The yield of eggplants per plant for LM treatment (26.2 kg) was decreased by 6% compared to CCF treatment (27.9 kg). The growth and yield of eggplants were not significant different among CCF, LM+CF, and LM+CF+PA treatments. The marketable yield for LM treatment was lowest among 4 treatments and that for LM+CF+PA treatment (26.6 kg) was increased by 8% compared to CCF treatment (24.1 kg). The chemical properties of the soil for CCF treatment, in general, tended to be higher compared to the other treatments. In conclusion, the present study demonstrates that the application of balanced liquid manure combined with mineral fertilizer to soil is considered as a good management practices because it improves, eggplant quality and soil properties.
