Effect of Weed Managements after False Seedbed on Weed Occurrence and Growth of Organic Chinese Cabbage Growth (가묘상을 이용한 잡초관리가 잡초발생과 유기농 배추의 생장에 미치는 영향)

  • The study was conducted to evaluate effects of weed managements after false seedbed on the weed control and growth of spring and fall cabbages (Brassica oleracea L.) in an organic upland in 2013. Weed managements included rake, flame, PE mulch, and none treatment. The weed occurrence was not effectively controlled by rake treatment but controlled by flame treatment in spring and fall cultivation. PE mulch plots had 30% of tip-burn in spring cultivation. Weed managements after false seedbed were similar fresh weight, leaf number, and cabbage yield to the conventional PE mulch. Head of cabbage was the lowest on the none treatment. Fall cultivation reduced the weed density and increased yield of cabbage compared to those of spring in all treatment plots. In conclusion, rake or flame treatment after false seedbed markedly decreased weed occurrence, which was expected to use for management of environmentally-friendly vegetation as a substitute of PE mulch.

Effect of a Refrigerator with LED on Functional Composition Changes and Freshness Prolongation of Cabbage (냉장고의 LED부착이 양배추의 선도 연장에 미치는 영향)

  • A LED (light emitting diode) was attached inside the cold $(4^{\circ}C)$ compartment of a home refrigerator to investigate a possible effect on freshness prolongation of cabbage over 10 d of storage. The LED increased chlorophyll synthesis, inhibited vitamin C degradation and increased the content of phenolics, compared to values measured in cabbages stored in an identical refrigerator without the LED. In the refrigerator with the LED, the color of cabbage leaves remained green for a period fourfold longer than that seen in a conventional refrigerator. Vitamin C content was twice that of a conventionally stored cabbage, and phenolic consent was 14% higher in the LED-illuminated vegetables.

  • The cultivation area for domestic cabbage increased by 26.3% from 10,968 ha in 2019 to 13,854 ha in 2020, and among leafy vegetables, the cabbage cultivation area was 62%, and production was 78.9%. Demand for field crop production of cabbage, which has a relatively high-income level compared to rice farming, is increasing, and mechanization of the field operation is urgently needed due to the insufficient development of related farming machinery. In this study, a prototype fabrication and performance test of a tractor-attached cabbage collector was carried out. The transport section was divided into two parts, one for the feeding and transportation and the other for the screening and packaging to selectively collect cabbages in bulk bags or boxes. The length of the primary collecting conveyor was designed to meet the field conditions of the Korean cabbage cultivation standards so that six cultivation rows could be worked simultaneously. Power was controlled by a hydraulic transmission line of the tractor and was easily mounted onto the 3-point hitch links behind the tractor. When the performance was evaluated, the transfer rate, loss rate, damage rate, and work performance were 100, 0, 1.2%, and 1.9 h·10 a-1. Final improvement and commercialization of the prototype would considerably contribute to the mechanization of harvesting cabbage, the main ingredient of Kimchi.

Development of Minimal Processing Technology for Korean Fruit and Vegetables (과실 및 채소의 신선편의 식품화 개발기술에 관한 연구)

  • The purpose of this study was to investigate the effectiveness of various quality preservative treatments for extending shelf life and maintaining good quality of minimally processed fruit and vegetables produced in Korea. To determine the suitable treatments for delaying quality deterioration, fresh Asian pears and Chinese cabbages were sliced and treated with various quality preservatives (1% CaCl$_2$, 1% NaCl, 3% sucrose, 1% Ca-lactate, 1% vitamin C, 0.05% chitosan +1% vitamin C, 0.1% Sporix+1% vitamin C, hot water (60$\^{C}$), 0.2% L-cysteine), packed with polyethylene film (60㎛-thick), and stored at 4$\^{C}$/0$\^{C}$ or 20$\^{C}$. Various biological and sensory tests were performed to evaluate the quality changes in minimally processed products. Results indicated that Chinese cabbages treated with 1% CaCl$_2$ at 4, and 1% CaCl$_2$ and 1% NaCl at 20$\^{C}$ were most effective in maintaining the quality and minimizing the biochemical changes during storage. For sliced Asian pears, 0.2% L-cysteine and 1% NaCl treatments were effective to reduce browning, and 1% CaCl$_2$ treatment was the most effective to prevent softening during storage at 20$\^{C}$ and 0$\^{C}$. Modified atmosphere packaging of Pleurotus ostreatus and Lentinus edodes had a significantly different shelf life depending on packaging material, packaging thickness and storage temperature. Sealed packaging with polyethylene film (60㎛-thick) for two kinds of mushrooms maintained a good quality with an extended shelf life by 30-50% at 20$\^{C}$ and by 30-130% at 0$\^{C}$. To minimize the quality deterioration which appeared in the condition of polyethylene film packaging, quality preservatives such as KMnO$_4$ and KHSO$_2$+K$_2$S$_2$O$\_$5/ for SO$_2$ generation were added inside of mushroom packaging. The best condition for maintaining good quality longer was packaging with polyethylene film+SO$_2$ which showed 5080% extended shelf life for both Pleurotus ostreatus and Lentinus edodes at 20$\^{C}$ and 0$\^{C}$.

Development of DNA Markers for Trehalose Synthesis Genes in Brassica rapa L. (배추 trehalose 합성 유전자와 연관된 DNA 마커 개발)

  • High temperature stress might affect the yield and quality of Chinese cabbage. In order to develop cultivars resistant to high temperature stress, we developed polymorphic DNA markers for trehalose synthesis genes related to abiotic stress resistance. A total of 28 Brassica rapa ESTs homologous to trehalose synthesis genes of Arabidopsis were found from the NCBI database. The polymorphic DNA sequences were searched between Chinese cabbages - Chiifu, which is relatively susceptible to high temperature stress, and Kenshin, which is tolerant to high temperature stress. Among the 28 ESTs, we found 10 ESTs that have either insertion/deletion and/or single nucleotide polymorphism between the two cultivars. Those polymorphic sites were then targeted for the development of 10 PCR based markers. These molecular markers related to trehalose genes could be used not only to test their relationship with abiotic stress resistance in Chinese cabbage, but also the development of abiotic stress resistant cultivars using MAS.

Clubroot Affects Both Agriculture and Tourism in Kagoshima Prefecture, Japan

  • Both agriculture and local tourism of Kagoshima prefecture where is located on the south-western region of the Japanese mainland, are the important industries. Although cabbage (Brassica oleracea) has been cultivated in recent decades in Kagoshima, clubroot disease caused by Plasmodiophora brassicae had never been observed. However, the disease in cabbage was reported in four regions last couple years. Our survey showed that one region is infested severely whereas others are slightly. In the most widely infested region, the disease was also observed in turnip rape (Brassica rapa) which is grown as ornamental plants for landscape design in early spring and important tourist attraction. Consequently, both agriculture and local tourism are damaged by clubroot. The increase of clubroot incidence in this region might be caused by significant increase of cabbage production, the expansion of cropping season throughout the year and continuous turnip rape cultivation in the same fields of cabbage for almost three decades. Therefore we are trying to estimate the risk of clubroot damage cultivation throughout the year in this region. We collected five isolates of resting spores and identified them as race 3, 4 and 9 by Williams' method, and as pathotype group 3 and 4 by classification system using clubroot resistant (CR) $F_1$ cultivars of Chinese cabbage as differential hosts as described in Hatakeyama et al.(2004). Furthermore, we found that these populations were avirulent to commercial CR cabbages. These results indicate that introduction of CR cabbage and breeding of turnip rape are the effective measures to solve our problem.

Development of a Cabbage Loader

  • Cabbage is one of the most important vegetable in Korea. The cabbage production was mainly based on human labor A comprehensive research fur substituting the human labor by machines has been performed until now. In general, cabbage is cultivated on hillside in Korea. Picking up the harvested cabbages in field and carrying to a vehicle fur transportation are very laborious work. Manual transportation of cabbage is likely to damage the quality and is also a cause to increase the cost of cabbage production. This study was to develop and evaluate a prototype cabbage loader fur efficient and safe transportation of cabbage. The developed loader was a semi-tracked vehicle operated by a hydraulic system, allowing the safe transportation and loading of cabbage in a steep field. The maximum loading weight of the loader was 1.0 ton. By using two sets of safety devices attached to the loader to avoid the roll-over in a steep field, the static rollover slopes were increased up to 34.0% and 37.4% fur the left and the rear direction, respectively The maximum field speed was about 6km/hr with two cabbage pallets of 750kg at a 25% inclined field. The loading capacity of the loader was about 35 pallets/hr when picking up, carrying, and unloading two cabbage pallets for one loading operation. The loading capacity was more than 8 times in comparison with the conventional human labor. The developed loader could be used fur loading and carrying the other vegetables. The study suggested a possible approach for designing the field machines operated on hillside.

Development of a Rapid Detection Method for Pectobacterium carotovorum subsp. carotovorum Using the Loop-Mediated Isothermal Amplification (LAMP) (Loop-Mediated Isothermal Amplification (LAMP)법을 이용한 Pectobacterium carotovorum subsp. carotovorum의 신속 진단법 개발)

  • Pectobacterium carotovorum subsp. carotovorum is the causative agent of soft rot in crops such as potato and cabbages. Loop-mediated isothermal amplification (LAMP) is a simple DNA amplification method, as well as isothermal PCR technique. In this study, a new method for the rapid detection of Pectobacterium carotovorum subsp. carotovorum was developed using LAMP that named PCC-LAMP. Based on lytic murein transglycolase gene of Pectobacterium carotovorum subsp. carotovorum, a set of four primers for LAMP was designed. The optimal PCC-LAMP reaction temperature was established at $61^{\circ}C$. Under standard conditions, PCC-LAMP amplified $1{\times}10^3$ copies of clone PCC-pBX437 per reaction. Further, this method can also assay directly by SYBR Green I without electrophoresis. Amplification was not detected for five other bacterial species. In conclusion, PCC-LAMP may be a useful method for the detection Pectobacterium carotovorum subsp. carotovorum in the field.

Influence of Commercial Antibiotics on Biocontrol of Soft Rot and Plant Growth Promotion in Chinese Cabbages by Bacillus vallismortis EXTN-1 and BS07M

  • We investigated influence of three commercial antibiotics viz., oxolinic acid, streptomycin, and validamycin A, on biocontrol and plant growth promoting activities of Bacillus vallismortis EXTN-1 and BS07M in Chinese cabbage. Plants were pre-drenched with these strains followed by antibiotics application at recommended and ten-fold diluted concentration to test the effect on biocontrol ability against soft rot caused by Pectobacterium carotovorum SCC1. The viability of the two biocontrol strains and bacterial pathogen SCC1 was significantly reduced by oxolinic acid and streptomycin in vitro assay, but not by validamycin A. In plant trials, strains EXTN-1 and BS07M controlled soft rot in Chinese cabbage, and there was a significant difference in disease severity when the antibiotics were applied to the plants drenched with the two biocontrol agents. Additional foliar applications of oxolinic acid and streptomycin reduced the disease irrespective of pre-drench treatment of the PGPRs. However, when the plants were pre-drenched with EXTN-1 followed by spray of validamycin A at recommended concentration, soft rot significantly reduced compared to untreated control. Similarly, strains EXTN-1 and BS07M significantly enhanced plant growth, but it did not show synergistic effect with additional spray of antibiotics. Populations of the EXTN-1 or BS07M in the rhizosphere of plants sprayed with antibiotics were significantly affected as compared to control. Taken together, our results suggest that the three antibiotics used for soft rot control in Chinese cabbage could affect bacterial mediated biocontrol and plant growth promoting activities. Therefore, combined treatment of the PGPRs and the commercial antibiotics should be carefully applied to sustain environmental friendly disease management.

Isolation and Expression Analysis of Brassica rapa WRKY 7

  • The cDNA clone of Brassica rapa WRKY7 (BrWRKY7) was obtained from EST collection in Brassica genomics team and its DNA sequence was determined. The cDNA clone is 1,037 bp long in nucleotides and encodes an open reading frame of 307 amino acids. Based on a phylogenetic tree, BrWRKY7 belongs to group IId. BrWRKY7 was induced by wound and SA. It was also induced by pathogen attack such as Xanthomonas campestris pv. campestris (Xcc), suggesting that this BrWRKY may play an essential role in defense response of chinese cabbages.
