Effects of Vitamin E Supplementation on Renal Glycosylation Products in Diabetic KK Mice (당뇨 KK마우스에서 비타민 E 보강식이가 신장 당화단백질 생성에 미치는 영향)

  • We investigated the effects of vitamin E supplementation on protein glycosylation in early and end stage product, and light microscopic studies were done on the renal glomeruli of KK-mice of various ages and various duration of diabetes. Weaned KK-mice were fed high fat diets containing 20% corn oil(wt/wt), and sacrificed at 4,6, and 9 months of age. The high vitamin E diet was a high fit diet supplemented with an excess amount of d1-$\alpha$-tocopheryl acetate (2080IU/kg diet). We measured Hemoglobin $A_{IC}$ (Hb $A_{IC}$) as a glycosylation early product, and renal collagen-linked fluorescence as a glycosylation end product. In the diabetic group, levels of Hb $A_{IC}$ were increased within 2 months after onset of diabetes and remained at a constant level for the duration of experiment. 5 months after onset of diabetes, renal collagen linked fluorescence(CLF) was markedly increased. A quantative, morphologically demonstratable, progressive thickening of the basement membrane and calcification occured in the diabetic KK-mice. There is a statiscally positive correlation between CLF and histologic grade of diabetic nephropathy. Hepatic vitamin E levels correlated with those of Hb $A_{IC}$, renal CLF, and renal calcification. Treatment with vitamin I did not modify the level of blood glucose. However, we observered a significant lowering of CLF and Hb $A_{IC}$ in diabetic mice. Supplementation of vitamin E was found to delay the progression of diabetic nephropathy.   • An experimental study was undertaken to observe the difference in susceptibility of mouse strains to Metagonimus yokogawai infection by estimating it from worm recovery rate and dimension of worms. It was also studied the effects of prednisolone injection on the chronological pattern of worm recovery in ICR mice. The metacercariae were obtained from sweetfish and 300 in each number were given to 5 strains (CBH, A, DBA, $C^{57}BL$ and KK) of mice, and after 7 days period, the worms were collected from their intestine. Prednisolone at the dose of 10 mg/kg was injected to ICR mice every other day from 7 days prior to infection until sacrificed at 6 hours to 35th post-infection day. ICR mice infected with M. yokogawai but untreated were used for controls. The success rate in infection of mice ranged 25.0-83. 3% by strains, the worm recovery rate 1. 2-18. 9%, and the average size of worms O. 554-0. 683 mm long and 0.214-0.244 mm wide. The higher rates and larger size of worms were observed in KK and $C^{57}BL$ strains than others and the difference was statistically significant. In ICR mice for control, the worm recovery rate until 1 day after infection was relatively high (38-66%) but it became much lower (less than 0.7%) during 1-35 days. However, prednisolone injection brought about persistently high recovery rates (16-80%) until 21 days. It was concluded that the susceptibility to M. yokogawai infection is different by strains of mice but it can be elevated by prednisolone injection probably due to suppression of Immune respon3e3 in ICR mice.

  • We investigated the effects of vitamin E supplementation on lipid peroxidation and on the activities of antioxidative enzymes in kidney of KK mice of various age and duration of diabetes. Weaned KK mice were fed high fat a diet containing 20% corn oil(wt/wt), and were sacrificed at 4, 6, and 9 months of age. The high vitamin E diet consisted of the high fat diet supplemented with an excessive amount of dl-$\alpha$-tocopheryl acetate (2080 IU/kg diet) . In the diabetic groups, we found an increase in lipofuscin and decrease in antioxidative enzyme activities with aging. Treatment with vitamin E did not modify the level of fasting blood glucose. However, a significant decrease in lipofuscin and increase in antioxidative enzyme acitvities were observed in diabetic mice.

In Vitro and In Vivo Effects of Piceatannol and Resveratrol on Glucose Control and TLR4-NF-κB Pathway (피세아테놀과 레스베라트롤의 혈당조절 및 TLR4-NF-κB 경로 조절 작용)

  • Piceatannol (PIC) is a natural hydroxylated analog of resveratrol (RSV), which is a polyphenol known to extend lifespan by stimulating sirtuins. The aim of this study was to investigate the effects of PIC and RSV on the toll-like receptor 4 (TLR4)-nuclear factor kappa B ($NF-{\kappa}B$) pathway in mouse hepatocytes and an obese/diabetic KK/HlJ mouse model. AML12 mouse hepatocytes in the absence or presence of palmitic acids (PA) were treated with PIC ($50{\mu}M$) or RSV ($50{\mu}M$). Male KK/HlJ mice at 20 weeks of age were divided into three subgroups as follows: 1) obese and diabetic control (KK), 2) KK_PIC, and 3) KK_RSV. PIC and RSV were administered orally at a dose of 10 mg/kg/d for 4 weeks. Four weeks of PIC and RSV treatment did not affect body weight or food intake in KK mice. Serum fasting blood glucose was significantly reduced in KK_PIC, and 2 h oral glucose tolerance test area under the curve was significantly reduced by PIC and RSV treatment in KK mice. PIC tended to improve homeostasis model assessment of the insulin resistance index (HOMA-IR) and HOMA beta-cells in diabetic KK mice. TLR4 and $NF-{\kappa}B$ were down-regulated by PIC and RSV treatments in hepatocytes in the absence or presence of PA. Insulin receptor, AMP-activated protein kinase, peroxisome proliferator-activated receptor gamma, nucleotide oligomerization domain-like receptor family pyrin domain-containing 3, interleukin-1, and $NF-{\kappa}B$ were altered in PIC-treated livers. Collectively, PIC and RSV inhibited the $TLR4-NF-{\kappa}B$ pathway, and PIC seems to be more effective than RSV in the regulation of analyzed targets, which are involved in insulin signaling and inflammation in vivo.

  • We investigated the effects of vitamin E supplementation on the protein glycosylation in vivo. Weaned KK-mice were fed high fat diet containing 20% corn oil(wt/wt), and sacrificed at 4, 6, and 0 months of age. High vitamin E diet was the high fat diet supplemented with an excess amount of 이-$\alpha$-tocopheryl acetate(2080IU/kg diet). We measured $HbA_{1C}$ as a glycosylation early product, and collagen-linked fluorescence (CLF) of skin as a glycosylation and product. We found that diabetic group had increased levels of $HbA_{1C}$ within 2 months after onset of diabetes and during the experiments. The skin CLF increased dramatically 5 months after onset of diabetics. Treatment with vitamin E did not modify the level of blood glucose. However, we observed a significant lowering in CLF and $HbA_{1C}$ in diabetic mice.

  • The purpose of this study was to investigate the effect of vitamin E supplementation on the lipid peroxidation and activities of antioxidative enzymes in the pancreas of diabetic KK mice. KK mice were fed high ft diet containing 20% corn oil(wt/wt), and sacrificed at 2 months of diabetes. A hish vitamin E diet consisted of the high fat diet supplemented with an excessive amount of 이-$\alpha$-tocopheryl acetate (2080IU/kg diet). The incidence of diabetes mellitus was 61% when mice were fed the high fat diet, but was 44% when mice were fed the high vitamin E diet, Vitamin E supplementation fhus seems to have the effect of decreasing of decreasing the onset of diaetes. In the diabetic group, we found increases of MDA (malondialdehyde) and antioxidative enzyme activities. Treatment with vitamin E did not modify the level of fasting blood glucose. However, MDA and antiosicative enzyme activities in diabetic mice were decreased by the high vitamin E diet. Increased levels of lipid peroxidation products suggests the occurrence of oxidative damage in the pancreas of diabetic mice. The increased level of antiosicative enzyme activities could be due to an adaptive response to conditions of increased peroxidative stress. Significant normalization on catalase activity was noted in vitamin E supplemented animals.

  • The purpose of this study was to investigate the effects of vitamin E supplementation on the activities of antioxidative enzymes in liver of KK mice of various ages and various duration of diabetes. Diabetes was induced by feeding high fat diet containing 20% corn oil(wt/wt). Weaned KK mice were fed high fat diet containing 51 IU or 2080 IU vitamin E per kg diet. Animals were sacrificed at 4, 6, and 9 months of age. In nondiabetic group, we found the decrease of antionxidative enzyme activities with aging. In diabetic group, antioxidative enzyme activities were decreased, and the change of hepatic vitamin E was related to glutathione peroxidase activity (r=0.71, p<0.001). Treatment with vitamin E did not modify the level of fasting blood glucose. However, it was observered that glutathione reductase and glutathione peroxidase activities as well as hepatic glutathione levels were increased by vitamie E supplementation, whereas catalase activity did not changed. The present result suggest that high vitamin E supplementation protects against lipid peroxidative damage in diabetic KK mice.

  • This study was carried out to investigate the supplementary effects of ${\gamma}$-oryzanol on the blood glucose level in diabetic mice. We supplied 2 experimental diets (CO without and GO with 0.2% ${\gamma}$-oryzanol) to diabetic mice for 8 weeks. Diet intake, body weight and levels of blood glucose, hemoglobin $A_{lc}$ and insulin were measured. Though there was no significant difference in diet intake between experimental groups, the concentration of fasting blood glucose and blood glucose area from glucose tolerance test in diabetic mice was lower in GO group than CO group during the supplementary period of experimental diets. Hemoglobin Ai, was lower and serum insulin level was higher in GO group than CO group without significance. These results suggest that r-oryzanol decrease the blood glucose level, and ${\gamma}$ -oryzanol produced from residual product of rice may be developed with high value.

Supplementary Effect of$\gamma$-Oryzanol on Lipid Metabolism in Diabetic KK Mice ($\gamma$-Oryzanol의 급여가 KK 당뇨 마우스의 지질대사에 미치는 영향)

  • This study was carried out to investigate the supplementary effects of ${\gamma}$-oryzanol extracted from rice bran on lipid metabolism in diabetic mice. We supplied 2 kinds of experimental diets (CO without and GO with 0.2% ${\gamma}$-oryzanol) to diabetic mice for 8 weeks. Diet intake, body weight, organ weight, contents of serum and hepatic lipid profiles, and fecal lipid levels were measured. Though there was no significant difference in diet intake, body weight change and organ weight between experimental groups, the concentration of serum total cholesterol and hepatic total lipid, total cholesterol and HMG-CoA reductase activity was significantly lower in GO group treated with 0.2% ${\gamma}$-oryzanol of diet than CO group after supplementary period of experimental diets. And total lipid, triglyceride, total cholesterol, and bile acid levels excreted to feces were significantly higher in GO group than CO group. These results suggest that ${\gamma}$-oryzanol decrease the serum and hepatic lipid levels by lowing HMG-CoA reductase activity or increasing the contents of fecal lipid in diabetic KK mice.
