Comparison of electrophoretic karyotypes in fusarium

  • The electrophoretic karyotypes of 6 species in different Fusarium sections were examined by using contour-clamped homogeneous electric field (CHEF) gel electrophoresis. Intact chromosomal DNA was prepared from protoplasts and up to 9 distinct bands were separated on 0.7% or 0.8% agarose gel under several different conditions. Putative chromosome numbers varied from 6 to 9 amd polymorphic karyotypes were observed in different Fusarium sections. Using Schizosaccharomyces pombe and Saccharomyces cerevisiae chromosomes as standards, the sizes of the Fusarium spp. chromosomes were estimated. The electrophoretic karyotypes of F. moniliforme and F. subglutinans (section Liseola) were similar. Unidentified filamentour fungi, F. beomiforme was much closer to F. axysporum (section Elecgans) in karyotype and the karyotypes of F. napiforme were more similar to those of section Liseola than any other sections. F. graminearum (section Discolor) had a distinctive electrophoretic karyotype.

Karyotypes of Korean Endemic Land Snail, Koreanohadra koreana (Gastropoda: Bradybaenidae)

  • The karyotypes of Korean endemic land snail, Koreanohadra koreana, using air-drying method wereinvestigated. Somatic cells of this species had 2n = 58. Karyotypes were also analysed with 16 metacentric, 12 submetacentric and one subtelocentric chromosome pairs. Observed chromosomes ranged from 2.6 to $8.9{\mu}m$ and the total length was $122.3{\mu}m$. This is the second report on the chromosome numbers and the karyotype of K. koreana.

Variation of Univariate Flow Karyotypes and Chromosomal DNA Contents in Maize (Zea mays L.)

  • Analyses of now karyotypes using different maize (Zea mays L.) inbred lines have been performed. The accumulation and isolation of high quality and quantity metaphase chromosomes from root tips can be achieved from many kinds of maize lines. The chromosome suspensions were prepared by a simple slicing method from synchronized maize root tips and analyzed with a now cytometry. The variations of experimental now karyotypes were detected among inbred lines in terms of the positions and/or the numbers of chromosome peaks. The 2C DNA amount among 8 inbred lines ranged from 5.09 to 5.52 pg. The variability of DNA content in maize chromosome 1 was 9.1 % ranging from 0.685 to 0.747 pg. The selection of appropriate maize lines is critical for sorting specific single chromosome types. At least five different chromosome types can be discriminated and sorted from five maize lines.

  • None of the numerous published canis idiogram and karyotypes has yet been generally accepted as a standard one because the dog has 76 acrocentric autosomes of similar size and shape. The karyotypes of DongGyeongi dog were analysed by conventional trypsin/Giemsa staining (GTG-banding techniques), and were compared with one another. There were no variations in karyotypes which were analysed by conventional GTG-banding techniques, but differences were observed in G-banding patterns with sapsaree (or canis familiaris strains). It is not clear that these disagreements in G-banding patterns between strains of dog were caused by chromosome polymorphism or a difference in interpretation.

  • Giemsa C-banded karyotypes of A. thunbergii, A. deltoide-fistulosum, A. cyaneum and A. cyaneum var. deltoides were analyzed, and the interspecific relationships were investigated on the basis of the C-banding patterns. The banding pattern of each species was unique and made possible an easy and clear separation among them. A. thunbergii and A. deltoide-fistulosum revealed very close banding relationship but a. cyaneum and A. cyaneum var. deltoides showed significant difference in banding pattern in spite of their close plant systematic position.

Allium속 이배체 2종과 사배체 2종의 Giemsa C-분염핵형 (Giemsa C-banded Karyotypes in Two Diploid and Two Tetraploid Allium Species)

  • C-banded karyotypes in A. anisopodium, A. sacculiferum, A. deltoide-fistulosum and A. splendens were investigated. The chromosome compositions were diploid of 2n=16 in A anisopodium and A. splendens, and teiraploid of 2n=32 in a. sacculiferum and A. deltoide-fistulosum. The bands were distributed in telomeric parts of the chromosomes dominantly in addition to interstitial regions sporadically, resulting in the specific C-banded karyo types according to the species. No centromeric band was observed in these species except only one chromosome in A. deltoidefistulosum. The interspecific relationship based on the C-band distribution will be discussed.

  • The karyotypes of 4 Korean Myotis species (M. mystucinus gracilis, M. jormous tsuensis, M. duubentonii ussuriensis and M. macrodoctvlusp and 2 Pipistrelfus species (p. coreensis and p. abramusl belonging to the Vespertilionidae were examined. The 4 Mvotis species had the karyotypes of 2n : 44 with FN : 50 (M. m. gracilis and M. f. tsuensisl or 52 (M. d. ussuriensis and M. mncrodactylus). Furthermore the karyotype of R obrumus (2n : 26, FN : 441 seemed advanced compared with that of U coreensis (2n : 44, FN : 501 which is similar to the original karyotype of Mvotis species (2n : 44, FN : 50).

국내 토종개의 염색체 조사에 관한 연구 (Study on chromosomes survey of Korea native dogs)

  • The karyotype of the domestic dog is widely accepted as one of the difficult mammalian karyotypes to work. In contrast to many other animals, knowledge about the canine karyotype is quite sparse. The dog has a total of 78 chromosomes; all 76 autosomes are acrocentric in morphology and show only a gradual decrease in length. But appear to be quite small and difficult to identify unambiguously. To purchased standardization of chromosome in Korea native dog, there were analyzed by conventional trypsin/Giemsa staining (GTG-banding techniques), and were compared with 4, 6, 8, 11, 13, 17 chromosome. There were no variations in karyotypes which were analyzed by conventional GTG-banding techniques, but differences were observed in G-banding patterns with Sapsaree, Jindo, Gyeongju DongGyeong dogs, Welshi-Corgi. It is not clear that these disagreements in G-banding patterns between strains of dog were caused by chromosome polymorphism or a difference in interpretation. Comparative analysis of the distribution patterns of conserved segments defined by dog paints in the genomes of the Korea native dogs demonstrates that their differences in the karyotypes of these three species could have resulted from acrocentric banding patterns.

두꺼비 종류의 아이소자임과 염색체 (Isozymes and Karyotypes of Bufo Species)

  • 두꺼비와 물두꺼비 종류의 아이소자임과 염색체를 조사 비교 하였다. 전기영동법에 의하여 23가지 종류의 효소성 및 비효소성 단백질을 분석하였으며, 두꺼비 종류는 23가지중 6종류의 단백질이 다형이었고 물두꺼비 종류에서는 23가지중 4종류의 단백질만이 다형이었다. 23가지 종류의 단백질중 11가지는 두꺼비와 물두꺼비 사이의 종간차이가 없었으며 12가지는 뚜렷한 종간차이를 보였다. 두꺼비의 이형율은 4.8%이었고 물두꺼비의 이형율은 3.7%이었다. 두꺼비와 물두꺼비의 유전적 유사성은 0.5 이하였다. 두꺼비와 물두꺼비는 모두 22개의 염색체를 가지고 있었으며 서로 유사한 핵형이었으나 두꺼비에 있어서는 한쌍의 염색체에 부수 염색체가 있었다. 본 실험결과는 두꺼비와 물두꺼비는 그들의 핵형은 서로 유사하나 유전자 구성에는 큰 차이가 있으며 진화학상 비교적 오래전에 2종이 서로 분화 하였음을 알 수 있었다.

한국산 강준치아과(Cultrinae) 2종의 핵형분석 (Karyotypes of Two Species of Cultrinae (Cyprinidae) from Korea)

  • 한국산 강준치아과 (Cultrinae)의 백조어와 강준치 2종의 염색체 수 및 핵형 분석을 실시하였다. 백조어와 강준치의 염색체 수 및 핵형은 2n=48 (7M+10SM+7ST), FN=96 그리고 2n=48 (6M+10SM+8ST), FN=96으로 밝혀졌다. 염색체의 크기는 백조어에서 가장 큰 염색체 쌍과 가장 작은 염색체 쌍은 각각 $5.4{\mu}m$$2.2{\mu}m$였으며, 강준치의 경우 $6.1{\mu}m$$2.3{\mu}m$를 보였다.
