Quantitative analysis of lipid hydroperoxides levels in peripheral organs of Juvenile Visceral Steatosis (JVS) Mice at 1 month of age

  • Juvenile visceral steatosis (JVS) mouse is an animal model of the systemic camitine deficiency. JVS mice first develop fatty liver following cardiac hypertrophy. hyperammonemia, etc. To clarify the relationship between fatty liver and other symptoms. lipid hydroperoxides levels of peripheral oragans in JVS mice at 1 month were determined by the use of phosphine derivatives. We also report here a new method to quantitate the lipid components level in fatty liver of JVS mice.

Organ Culture of Ovary Isolated from Juvenile Mice (약령 마우스에서 분리한 난소의 기관배양에 관한 연구)

  • This study was to assess the developmental capacity of oocytes matured in vitro after 20, 15, 10, 5 and 0 days of organ culture when ovaries were isolated from juvenile mice at 0-, 5-, 10-,15- and 20-day old, respectively, and to develop in vitro culture system that observed a view to morphology of ovaries and nucleus maturation of oocytes. The size of ovaries decreased 35.9%, 8.7%, 1.2% and 14.4% after 20, 15, 10, 5 days of organ culture when the ovaries were isolated from 0-, 5-, 10 and 15-day old mice, respectively. After organ culture, the recovery rates, diameters of oocytes and the number of oocytes progressed from GV to MII were increased as increasing age of mice.

  • To determine the infectivlty and maturity of nletacercanae of Parqgonimur westermani after keeping at low temperature for a long period, 45 mice and 45 rats were each infected with 20 metacercariae which were kept at $4^{\circ}C$ for 8 to 234 days. The worm recovery in mice increased with age of worm and reached a peak of 32% at 41-50 days and then decreased with age. The rate In rats first decreased to a lowest point of 6% at 71-100 days and then Increased with age. In 42 infected mice and 41 infected rats, 187 immature worms (183 tiny and 4 Juvenile ones) and 190 worms (164 tiny, 19 Juvenile and 7 mature ones) were recovered respectively. Two wormcysts with eggs only and 8 empty wormcysts were also found In the rats. In addition, the frozen metacercanae can still develop to mature worms in SD rats.

In Vitro Development of Preantral Follicles Isolated from Juvenile Mice (약령 마우스에서 분리한 난포난자의 체외발생)

  • The aim of this study was to assess the developmental capacity of oocytes maturated in vitro after 10 days of culture when the preantral follicles were isolated from juvenile mice 10- and 20-day old, respectively, and to develop in vitro culture system that observed a view to morphology of follicles and nucleus maturation of oocytes. The antral-like cavities became formation after 6 days of culture in follicle isolated from 10- and 20-day old mice. The number of follicles were 21.5 and 33.3 in ovary isolated from 10- and 20-day old mice, respectively. The diameters of oocytes were 51.85 and 57.50 ${\mu}{\textrm}{m}$ before culture and were grew 55.95 and 63.11 ${\mu}{\textrm}{m}$ after culture for 10 days, in follicles isolated from 10- and 20-day old mice, respectively. The observation rates up to the M II and from GV to M II were 4.3 and 22.1%, and 14.5 and 61.1% after culture for 10 days in follicles isolated from 10- and 20-day old mice, respectively.

  • To discover germ cell-specific transcripts, we prepared a cDNA library from adult testes of 35-day old mice and subtracted it with mRNA from the testes of juvenile mice. Real-time RT-PCR analysis indicated that 42 cDNA clones in the subtracted library were expressed more intensely in the adult testes than in the juvenile testes. One clone identified by subtraction is expressed preferentially in the late spermatid and is located on chromosome 17E3 in mouse and 2p22 in human. (omitted)

  • The present study was designed to investigate the appearance of the congenital aggregates of the ectodermal glial cells in the brain of the normal rodents. The brain samples were taken from mice fetus, juvenile mice, rats and rabbits. The appearance regions of the glial cell aggregates (GCA) were investigated and the cells in the GCA were identified with electron microscope. 1. GCA in the mouse fetus tended to be higher in cell density, larger in size and lower frequency in appearance than juvenile mouse. The regions of higher appearance frequency of GCA in the juveniles of mice, rats and rabbits were ordered as subependymal layer in the collateral trigone of lateral ventricles, molecular layer of the neocortex, inner layer except the molecular layer in the neocortex, cerebral medulla, corpus callosum and hippocampus. Appearance frequency of GCA in the neonatal mice tended to be higher until 5 day after birth, and were markedly decreased on 10 and 15 day after birth. 2. GCA tended to be closed on one side of the blood vessels or neurons but not perivascular or perineuronal appearance. 3. In electron microscophy, GCA were composed of immature oligodendrocytes and astrocytes in the subependymal, and tended to be more mature and loose in the neocortex and to be appended some microglia cells with age. The cells in the GCA of older mice tended to be more mature than in young mice.

Effects of Intraperitoneal N-methyl-D-aspartate (NMDA) Administration on Nociceptive/Repetitive Behaviors in Juvenile Mice

  • Dysregulation of excitatory neurotransmission has been implicated in the pathogenesis of neuropsychiatric disorders. Pharmacological inhibition of N-methyl-D-aspartate (NMDA) receptors is widely used to model neurobehavioral pathologies and underlying mechanisms. There is ample evidence that overstimulation of NMDA-dependent neurotransmission may induce neurobehavioral abnormalities, such as repetitive behaviors and hypersensitization to nociception and cognitive disruption, pharmacological modeling using NMDA has been limited due to the induction of neurotoxicity and blood brain barrier breakdown, especially in young animals. In this study, we examined the effects of intraperitoneal NMDA-administration on nociceptive and repetitive behaviors in ICR mice. Intraperitoneal injection of NMDA induced repetitive grooming and tail biting/licking behaviors in a dose- and age-dependent manner. Nociceptive and repetitive behaviors were more prominent in juvenile mice than adult mice. We did not observe extensive blood brain barrier breakdown or neuronal cell death after peritoneal injection of NMDA, indicating limited neurotoxic effects despite a significant increase in NMDA concentration in the cerebrospinal fluid. These findings suggest that the observed behavioral changes were not mediated by general NMDA toxicity. In the hot plate test, we found that the latency of paw licking and jumping decreased in the NMDA-exposed mice especially in the 75 mg/kg group, suggesting increased nociceptive sensitivity in NMDA-treated animals. Repetitive behaviors and increased pain sensitivity are often comorbid in psychiatric disorders (e.g., autism spectrum disorder). Therefore, the behavioral characteristics of intraperitoneal NMDA-administered mice described herein may be valuable for studying the mechanisms underlying relevant disorders and screening candidate therapeutic molecules.

Experimental Life History and Biological Characteristics of Fasciola gigantica (Digenea: Fasciolidae)

  • This study was conducted to investigate the life history, morphology, and maturation of larval stages and adult worms of Fasciola gigantica in experimental mice. Lymnaea auricularia rubiginosa was used as the intermediate host, and Oryza sativa was used for encystment of the metacercariae, while Mus musculus was used as the definitive host for maturation study. Fresh eggs from the gall bladder of water buffaloes fully developed into embryonated ones and hatched out at days 11-12 after incubation at about $29^{\circ}C$. Free-swimming miracidia rapidly penetrated into the snail host, and gradually developed into the next larval stages; sporocyst, redia, and daughter redia with cercariae. Fully-developed cercariae were separated from the redia and shed from the snails on day 39 post-infection (PI). Free-swimming cercariae were immediately allowed to adhere to rice plants, and capsules were constructed to protect metacercariae on rice plants. Juvenile worms were detected in intestines of mice at days 3 and 6 PI, but they were found in the bile duct from day 9 PI. Juvenile and adult flukes were recovered from 16 mice experimentally infected with metacercariae, with the average recovery rate of 35.8%. Sexually mature adult flukes were recovered from day 42 PI. It could be confirmed that experimentally encysted metacercariae could infect and develop to maturity in the experimental host. The present study reports for the first time the complete life history of F. gigantica by an experimental study in Thailand. The obtained information can be used as a guide for prevention, elimination, and treatment of F. gigantica at environment and in other hosts.

Abnormal Astrocytosis in the Basal Ganglia Pathway of Git1-/- Mice

  • Attention deficit/hyperactivity disorder (ADHD) is one of the most common neurodevelopmental disorders, affecting approximately 5% of children. However, the neural mechanisms underlying its development and treatment are yet to be elucidated. In this study, we report that an ADHD mouse model, which harbors a deletion in the Git1 locus, exhibits severe astrocytosis in the globus pallidus (GP) and thalamic reticular nucleus (TRN), which send modulatory GABAergic inputs to the thalamus. A moderate level of astrocytosis was displayed in other regions of the basal ganglia pathway, including the ventrobasal thalamus and cortex, but not in other brain regions, such as the caudate putamen, basolateral amygdala, and hippocampal CA1. This basal ganglia circuit-selective astrocytosis was detected in both in adult (2-3 months old) and juvenile (4 weeks old) $Git1^{\check{s}/\check{s}}$ mice, suggesting a developmental origin. Astrocytes play an active role in the developing synaptic circuit; therefore, we performed an immunohistochemical analysis of synaptic markers. We detected increased and decreased levels of GABA and parvalbumin (PV), respectively, in the GP. This suggests that astrocytosis may alter synaptic transmission in the basal ganglia. Intriguingly, increased GABA expression colocalized with the astrocyte marker, GFAP, indicative of an astrocytic origin. Collectively, these results suggest that defects in basal ganglia circuitry, leading to impaired inhibitory modulation of the thalamus, are neural correlates for the ADHD-associated behavioral manifestations in $Git1^{\check{s}/\check{s}}$ mice.

Effects of different algae in diet on growth and interleukin (IL)-10 production of juvenile sea cucumber Apostichopus japonicus

  • The experiment was conducted to investigate the effects of different algae in diet on growth, survival, and interleukin-10 productions of sea cucumber. At first, a 9-week feeding trail was conducted to evaluate the growth performance and survival of the sea cucumber fed one of the six experimental diets containing ST (Sargassum thunbergii), UL (Ulva lactuca), UP (Undaria pinnatifida), LJ (Laminaria japonica), SS (Schizochytrium sp.), and NO (Nannochloropsis oculata) in a recirculating aquaculture system. The result showed that survival was not significantly different among the dietary treatments, and the specific growth rate (SGR) of sea cucumber fed the UL diet ($1.58%d^{-1}$) was significantly higher than that of sea cucumber fed the other diets (P < 0.05), except for the LJ and NO diets. Secondly, interleukin (IL)-10 gene expression was determined where mice splenocytes were stimulated with $10{\mu}g\;ml^{-1}$ of sea cucumber extracts for 2 h. The result showed that IL-10 gene expression levels were significantly increased in UL, LJ, and NO diets fed sea cucumber extracts compared to other experimental diets. The results suggest that dietary inclusion with Ulva lactuca, Laminaria japonica, and Nannochloropsis oculata algae may improve the growth of juvenile sea cucumber and could upregulate IL-10 gene expression in mice splenocytes. Such detailed information could be helpful in further development of more appropriate diets for sea cucumber culture.
