Fingerprinting of Rice Genomes Using PCR with Arbitrary Primers

  • The arbitrary primed polymerase chain reaction (AP-PCR) has been used to detect the genetic alternations in the related species. Simple and reproducible fingerprints of complex genomes can be generated using single arbitrary chosen primers and the PCR. The technique was applied to the Oryza species and characterized the relationship among three cultivars of rice species based on theresult of genomic DNA fingerprints. The results indicated that the polymorphism revealed in rice strains and the differences in the PCR product pattern could be represented for each strainis. There was many variationsin the PCR product pattern between cv. Dongin(japonica type)and cv.Hyangdo (indica type), and our chosen AP-primers can ge as markers for strain identification and verfication.

Characteristics and Genetic Segregation of a Rolled Leaf Mutant in Rice

  • Leaf structure is one of the important agronomic traits. A rolled leaf mutant was induced from an ethyl methane sulfonate (EMS)-treated japonica rice, 'Koshihikari'. The rolled leaf mutant showed phenotypes of reduced leaf width and leaf rolling. In addition, several abnormal morphological characteristics were observed, including dwarfism, defected panicle, delayed germination, and lower seed-setting. Microscopic analysis revealed that the number of small veins was decreased and the sizes of adaxial bulliform cells were reduced in the mutant leaves. The genetic study with two $F_2$ populations from the crosses of the rolled leaf mutant with 'Koshihikari' and Milyang23 suggested that the mutant phenotype might be controlled by a single dominant gene.

저온스트레스와 회복기간중 저온 내성 및 감수성 벼 품종의 생리적 반응 (Physiological Responses of Chilling - Tolerant and Susceptible Rice Cultivars during Chilling Stress and Subsequent Recovery)

  • 저온에 내성인 자포니카의 일품벼와 감수성인 인디카의 태백벼 품종간에 저온처리기간 동안 및 저온처리 후 회복동안에 나타날 수 있는 생리적 반응을 비교하였다. $5^{\circ}C$의 저온처리 3일 동안에 잎의 생체중 및 엽록소 함량은 두 품종간에 차이가 적거나 없었으나 저온처리 후 $25^{\circ}C$에 회복시킨 동안에는 태백벼에 비해 일품 벼가 더 빨리 회복되었다. 상대적 수분함량, 지질과산화 작용의 지표인 malondialdehyde 생성량 및 엽록소 형광은 저온처리기간 동안 및 저온처리 후 회복기간동안에도 두 품종간에 유의적인 차이를 보였고, 이들 생리적 지표들은 생체중 및 엽록소 함량보다는 더 민감한 생리적 지표이었다. 두 품종간에 상대적 수분함량, malondialdehyde 생성량 및 엽록소 형광의 차이는 저온처리 후 회복기간동안보다는 저온처리동안에서 더 적었다. 따라서 이들 결과를 통해서 볼 때 상대적 수분함량, malondialdehyde 생성량과 엽록소 형광은 저온에 내성을 선발하는데 중요한 생리적 지표로 이용할 수 있으리라 생각한다.

택지개발지구내 양서류 서식처로서 묵논습지의 특성과 기능평가 (Assessment of Characteristics and Functions of Abandoned Rice Paddy Wetlands as Habitats for the Amphibia within Land Development Districts)

  • The current study analyzed appropriateness of abandoned rice paddy wetlands as habitats for the Amphibia by assessing functions and value of abandoned rice paddy wetlands within land development districts as a habitat for living creatures and researching into the Amphibia living in the subject districts. For this purpose, the study designated abandoned rice paddy wetlands within Hwaseong A District and Pangyo B District under land development as subject lands. Those wetlands went through succession after abandonment of farming. Detailed study results are as follows. From classification of wetland types and an analysis of their characteristics, it was found that herbaceous plants such as smartweed communities, horsetail communities and reed communities, and woody plants such as pussy willow communities appeared in both districts. These abandoned rice paddy wetlands shows typical characteristics of palustrine wetlands. As a result of a vegetation structure research, it was observed that succession has been progressed for at least over 2~3 years. In assessment of wetland functions by item, it was found that they are developing into fine wetlands in terms of functions because they are ranked over Moderate Class except separate distance from streams. Also from a research into animals living in the abandoned rice paddy wetlands, it was found that main inhabitants are Rana nigromaculata,Rana dybowskii, Hyla japonica, Bufo bufo gargarizans, etc. It can be confirmed that the abandoned rice paddy wetlands are providing appropriate conditions as habitats to the Amphibia in a view that those species have life cycles of inhabitation (shores and wetlands) - egg-laying (rice paddy, puddles and swampy places) - hibernation (rice field banks and swampy places). From this point of view, it was found that abandoned rice paddy wetlands that developed because of abandonment of farming can be used as means to improve structure and functions of habitats.

현미의 저장조건에 따른 aflatoxin $B_1$ 생성의 변화 (Variation of Aflatoxin $B_1$ Production in Brown Rice Inoculated with Aspergillus parasiticus under Different Storage Conditions)

  • 쌀에대하여 afltoxin 생성을 위한 기질로서의 능력을 알아보기 위하여 현미 (청청벼) 시료에 Aspergillus parasiticus를 접종하고 조건을 달리하여 저장하면서 Aflatoxin $B_1$의 생성을 관찰하였다. 시료중의 Aflatoxin $B_1$의 분석은 ELISA를 이용하여 수행하였다. 현미 시료에서 Aflatoxin $B_1$ 생성에 가장 좋은 온도는 $28^{\circ}C$였으며, 시료의 수분함량을 15.8%로 증가시킨 경우 Aflatoxin $B_1$ 의 생성이 유의하게 증가하였고 (P<0.05), 고압증기멸균시킨 시료는 Aflatoxin $B_1$ 생성에 보다 효과적인 기질이 되었다. 실온에서 3개월 동안 저장한 현미에서는 15일 동안 저장 한 경우에 비해 Aflatoxin $B_1$ 생성이 유의한 증가를 보였다(P<0.05). 따라서 저장 온도 및 수분함량이 쌀에서도 Aflatoxin $B_1$ 생성에 영향을 미치는 바를 나타내었으며, 또 시료의 상태 및 저장기간도 쌀에서 Aflatoxin 생성 위험 요인으로 작용할 수 있음이 제시되었다. 이러한 결과로부터 쌀이 aflatoxin 생성에 좋은 기질이 될 수 있는 것으로 평가된다.

Current Insights into Research on Rice stripe virus

  • Rice stripe virus (RSV) is one of the most destructive viruses of rice, and greatly reduces rice production in China, Japan, and Korea, where mostly japonica cultivars of rice are grown. RSV is transmitted by the small brown plant-hopper (SBPH) in a persistent and circulative-propagative manner. Several methods have been developed for detection of RSV, which is composed of four single-stranded RNAs that encode seven proteins. Genome sequence data and comparative phylogenetic analysis have been used to identify the origin and diversity of RSV isolates. Several rice varieties resistant to RSV have been selected and QTL analysis and fine mapping have been intensively performed to map RSV resistance loci or genes. RSV genes have been used to generate several genetically modified transgenic rice plants with RSV resistance. Recently, genome-wide transcriptome analyses and deep sequencing have been used to identify mRNAs and small RNAs involved in RSV infection; several rice host factors that interact with RSV proteins have also been identified. In this article, we review the current statues of RSV research and propose integrated approaches for the study of interactions among RSV, rice, and the SBPH.

KRDD: Korean Rice Ds-tagging Lines Database for Rice (Oryza sativa L. Dongjin)

  • The Korean Rice Ds-tagging lines Database (KRDD) is designed to provide information about Ac/Ds insertion lines and activation tagging lines using japonica rice. This database has provided information on 18,158 Ds lines, which includes the ID, description, photo image, sequence information, and gene characteristics. The KRDD is visualized using a web-based graphical view, and anonymous users can query and browse the data using the search function. It has four major menus of web pages: (i) a Blast Search menu of a mutant line; Blast from rice Ds-tagging mutant lines; (ii) a primer design tool to identify genotypes of Ds insertion lines; (iii) a Phenotype menu for Ds lines, searching by identification name and phenotype characteristics; and (iv) a Management menu for Ds lines.

Identification and Characterization of New Copia-like Retrotransposon Osr1 in Rice

  • An insertion sequence identified as a solo long terminal repeat (LTR) of a new rice copia-like retrotransposon was detected in the ORE of the Pi-b gene from the rice cv. Nipponbare, and was designated as Osr1. Osr1 consists of a 6386 bp nucleotide sequence including 965 bp LTRs on both ends with an 82% nucleotide sequence identity to the wheat Tarl retrotransposon on reverse transcriptase. Nucleotide divergence was noted among the individual LTRs, as well as the coding region of Osr1. Various restriction fragment length polymorphism (RFLP) of LTR were detected in indica cultivars, whereas, only a few could be detected in the japonica cultivars. The population of Osr1 is lower in the wild-type rice compared with that in the domesticated cultivars. The insertion of LTR sequence in the h-b gene in the susceptible cultivar suggested that retro-tyansposon-mediated insertional mutation might play an important role in the resistance breakdown, as well as in the evolution of resistance genes in rice.

Karyotype Analyses of a Rice Cultivar 'Nakdong' and its Four Genetically Modified Events by Conventional Staining and Fluorescence in situ Hybridization

  • Conventional staining and fluorescence in situ hybridization (FISH) karyotypes of the non-genetically modified (GM) parental rice line, 'Nakdong' (Oryza sativa L. japonica), and its four GM rice lines, LS28 (event LS30-32-20-1), Cry1Ac1 (event C7-1-9-1), and LS28 ${\times}$ Cry1Ac1 (events L/C1-1-3-1 and L/C1-3-1-1) were analyzed using 5S and 45S rDNAs as probes. Both parental and transgenic lines were diploids (2n=24) with one satellite chromosome pair. The lengths of the prometaphase chromosomes ranged from 1.50 to $6.30{\mu}m$. Four submetacentric and eight metacentric pairs comprised the karyotype of 'Nakdong' and its four GM lines. One pair of 5S rDNA signals was detected near the centromeric region of chromosome g in both the parental and transgenic lines. The 45S rDNA signals were detected on the secondary constrictions of the satellite chromosome pair in both the parental and transgenic lines. There was no significant difference in chromosome size, length, and composition between 'Nakdong' and its four GM lines. This research was conducted as a preliminary study for chromosomal detection of transgenes in GM rice lines and would be useful for their breeding programs.

최근(最近) 개발(開發)된 수도용(水稻用) 제초제(除草劑)의 작용성(作用性)과 제초효과(除草效果) (Herbicidal Efficacy of Newly Developed Several Herbicides in Rice)

  • 최근(最近)에 수도용(水稻用) 제초제(除草劑)로 개발(開發)된 수종(數種)의 신제초제(新除草劑)들의 못자리 및 본답(本畓)의 잡초방제효과(雜草防除效果)와 이들의 작용성(作用性)을 구명(究明)하기 위해 1986년(年) 영남작물시험장(嶺南作物試驗場)에서 일련(一連)의 실험(實驗)을 실시(實施)하였던 결과(結果)는 다음과 같다. 1. 최근(最近) 개발(開發)된 수도용(水稻用) 제초제(除草劑) 중(中)에서 상면진압(床面鎭壓)못자리에서도 벼에 대(對)한 약해(藥害)가 거의 없으면서 제초효과(除草效果)가 우수(優秀)하였던 제초제(除草劑)는 pyrazolate 또는 pyrazoxyfen의 파종전(播種前) 처리(處理)와 quinclorac의 파종후(播種後)10일(日) 처리(盧理)하였다. 2. 이앙답(移秧畓)에서 일년생( 一年生) 및 다년생(多年生) 잡초(雜草)에 대(對)한 살초효과(殺草效果)가 높으면서 벼에 대(對)한 약해(藥害)가 겨의 없었던 처리(處理)는 NC-311와 butachlor 또는 quinclorac와의 혼합처리(混合處理)였는데 이들 혼합처리(混合處理)는 피의 엽기(葉期)가 2.5 엽기(葉期)까지도 90% 이상(以上)의 살초효과(殺草效果)를 보였으며 NC-311 + quinclorac의 혼합방법(混合方法)이 NC-311 + butachlor의 혼합방법(混合方法)보다 높은 살초력(殺草力)을 유지(維持)하였고 살초력(殺草力)을 유대(維待)하였고 살초력(殺草力) 지속기간(持續期間)도 길었다. 3. pyrazolate와 sulfonyl urea계(系) 제초제(除草劑)인 DPX-5384, CGA 142464, NC-311은 벼에 대(對)한 안전성(安全性)이 대단(大端)히 높았는데 지상부(地上部) 생육(生育)보다는 지하부(地下部) 생육억제영향(生育抑制影響)이 높은 경향(傾向)이었고, 이들 제초제(除草劑)에 대(對)한 품종간(品種間) 반응(反應)에 있어서도 일본형(日本型) 품종(品種)인 낙동(洛東)벼가 통일형(統一型) 품종(品種)보다 민감(敏感)한 반응(反應)을 보였다. 4. 이들 제초제(除草劑)의 흡수부위(吸收部位) 영향에 있어서는 벼의 경우 흡수부위(吸收部位)에 관계(關係)없이 大端(대단)히 안전(安全)하지만 피에 대(對)해서는 sulfonyl urea계(系)의 경우는 유아부(幼芽部) 흡수(吸收)가, pyrazolate와 Quinclorac의 경우는 유근부(幼根部) 흡수(吸收)가 주흡수(主吸收) 경로(經路)가 되었으며 피에 대(對)한 살초효과(殺草效果)는 Quinclorac이가장 탁월(卓越)하였다.
