Lantibiotics, Class I Bacteriocins from the Genus Bacillus

  • Antimicrobial peptides exhibit high levels of antimicrobial activity against a broad range of spoilage and pathogenic microorganisms. Compared with bacteriocins produced by lactic acid bacteria, antimicrobial peptides from the genus Bacillus have been relatively less recognized despite their broad antimicrobial spectra. These peptides can be classified into two different groups based on whether they are ribosomally (bacteriocins) or nonribosomally (polymyxins and iturins) synthesized. Because of their broad spectra and high activity, antimicrobial peptides from Bacillus spp. may have great potential for applications in the food, agricultural, and pharmaceutical industries to prevent or control spoilage and pathogenic microorganisms. In this review, we introduce ribosomally synthesized antimicrobial peptides, the lantibiotic bacteriocins produced by members of Bacillus. In addition, the biosynthesis, genetic organization, mode of action, and regulation of subtilin, a well-investigated lantibiotic from Bacillus subtilis, are discussed.

Antifungal Effects on Plant Pathogenic Fungi and Characteristics of Antifungal Substances Produced by Bacillus subtillis SJ-2 Isolated from Sclerotia of Rhizoctonia solani (벼 잎집무늬마름병균(Rhizoctoniz solani)의 균핵에서 분리한 Bacillus subtilis SJ-2의 식물 병원 곰팡이에 대한 항균 활성 및 항균 물질의 특성)

  • 벼 잎집무늬마름병균(Rhizoctonia solani)의 균핵에서 분리한 길항세균 SJ-2를 대치 배양 방법에 의해 벼도열병균(Pyricularia oryzae)외 6종의 식물 병원균에 대한 억제 효과를 조사하였다. 형태 및 생리적 특성을 조사한 결과, 이 균은 Bacillus subtilis로 동정되었다. 이 균은 AM 1(antibiotic medium 1)액체 배지에서 항균 물질을 분비하였으며, 배양 2일째 가장 항균 활성이 높았다. Butyl alcohol로 배양액에서 항균 물질을 조추출하여 100$\mu\textrm{g}$/ml의 농도로 조제한 감자 한천 배지에서 P. oryzae의 15개균에 대한 생육을 조사한 결과 P. oryzae, R. solani, Cochliobolus sativus에 대해서는 100% 생장을 억제하였으며, C. miyabeanus, Alternaria alternata, Botrytis cinerea, Cladosporium fulvum, Colletotrichum gloeosporioides, Fusarium moniliforme, F. oxysporum에는 80% 이상의 억제 효과를 보인 반면 난균강에 속하는 병원균에 대한 효과는 낮게 나타났다. 활성물질을 분리 정제하여 동정한 결과 B. subtilis가 분비하는 항균 물질로 알려진 polypeptides계의 iturins로 밝혀졌다. 이러한 항균 물질은 벼 도열병균(P. oryzae)의 포자 발아 및 발아관의 팽대(swelling)를 야기했으며, 벼 잎집무늬마름병균(R. solani)에는 균사의 용균(lysis)현상을 일으켰다.

Antagonistic and Plant Growth Promoting Activity of Bacillus species Isolated from Brackish Environment

  • Bacteria of the Bacillus sp. are well known to possess antagonistic activity against numerous plant pathogens. In the present study, 11 Bacillus sp. were isolated from the brackish environment and assayed for antagonistic activity under in vitro and in vivo conditions. Among the 11 isolates tested, 9 isolates effectively inhibited the growth of various plant pathogens, namely Phytophthora capsici, Phytophthora citropthora, Phytophthora citricola, Phytophthora sojae, Colletotricum coccodes, Colletotricum gloeosporioides, Colletotricum acutatum, Rhizoctonia solani, Fusarium solani, Fusarium graminearum, Pyricularia sp. and Monilina sp. The effective isolates were further screened for Phytophthora blight suppression in Capsicum annuum L. under green house conditions. The isolate SB10 exhibited the maximum (72.2%) reduction in disease severity. The antifungal compounds from the isolate were isolated and characterized. The isolated compounds exhibited high thermo stability ($100^{\circ}C$ for 30 min). Matrix-Assisted Laser Desorption Ionization-Time of Flight investigation of the antifungal compounds revealed three lipopeptide complexes, the surfactins, the iturins, and the fengycins.

Genomics and LC-MS Reveal Diverse Active Secondary Metabolites in Bacillus amyloliquefaciens WS-8

  • Bacillus amyloliquefaciens is an important plant disease-preventing and growth-promoting microorganism. B. amyloliquefaciens WS-8 can stimulate plant growth and has strong antifungal properties. In this study, we sequenced the complete genome of B. amyloliquefaciens WS-8 by Pacific Biosciences RSII (PacBio) Single Molecule Real-Time (SMRT) sequencing. The genome consists of one chromosome (3,929,787 bp) and no additional plasmids. The main bacteriostatic substances were determined by genome, transcriptome, and mass spectrometry data. We thereby laid a theoretical foundation for the utilization of the strain. By genomic analysis, we identified 19 putative biosynthetic gene clusters for secondary metabolites, most of which are potentially involved in the biosynthesis of numerous bioactive metabolites, including difficidin, fengycin, and surfactin. Furthermore, a potential class II lanthipeptide biosynthetic gene cluster and genes that are involved in auxin biosynthesis were found. Through the analysis of transcriptome data, we found that the key bacteriostatic genes, as predicted in the genome, exhibited different levels of mRNA expression. Through metabolite isolation, purification, and exposure experiments, we found that a variety of metabolites of WS-8 exert an inhibitory effect on the necrotrophic fungus Botrytis cinerea, which causes gray mold; by mass spectrometry, we found that the main substances are mainly iturins and fengycins. Therefore, this strain has the potential to be utilized as an antifungal agent in agriculture.

Diversity and Chemical Defense Role of Culturable Non-Actinobacterial Bacteria Isolated from the South China Sea Gorgonians

  • The diversity of culturable non-actinobacterial (NA) bacteria associated with four species of South China Sea gorgonians was investigated using culture-dependent methods followed by analysis of the bacterial 16S rDNA sequence. A total of 76 bacterial isolates were recovered and identified, which belonged to 21 species of 7 genera, and Bacillus was the most diverse genus. Fifty-one percent of the 76 isolates displayed antibacterial activities, and most of them belonged to the Bacillus genus. From the culture broth of gorgonian-associated Bacillus methylotrophicus SCSGAB0092 isolated from gorgonian Melitodes squamata, 11 antimicrobial lipopeptides including seven surfactins and four iturins were obtained. These results imply that Bacillus strains associated with gorgonians play roles in coral defense mechanisms through producing antimicrobial substances. This study, for the first time, compares the diversity of culturable NA bacterial communities among four species of South China Sea gorgonians and investigates the secondary metabolites of gorgonian-associated B. methylotrophicus SCSGAB0092.
