Production of Itaconic Acid by Various Immobilization Methods (다양한 고정화 방법에 의한 이타콘산 생산)

  • Aspergillus terreus NRRL 1960 was immobilized on various alginate gel beads, Celites, and polyurethane foam cubes, and the comparisons were made for the production of itaconic acid according to the types and sizes of each carrier. The levels of itaconic acid produced from Ca- alginate and Sr-alginate were similar, and the addition of bentonite to Ca- and Sr-alginate resulted in an increase of itaconic acid. The addition of 1.67% bentonite and 0.33% starch to Sr-alginate (SABS bead) produced higher level of itaconic acid (11.59 g/1) than other gel beads. A decrease in the size of Celite increased the itaconic acid production, and the smallest size of Celites, R- 634, produced 6.37 g/l of itaconic acid. Among various types of polyurethane foam cubes, HR 08 (2X2X2 cm) produced about 19 g/l of itaconic acid, which was more efficient than other carriers. In a repeated batch culture using immobilized cells on polyurethane foam cubes (HR 08, 2X2X2 cm), the stability of itaconic acid production was maintained up to 4 batches. Also, the possibility of itaconic acid production by continuous culture was shown in a packed-bed reactor.

Production of Itaconic Acid at Various Bioreactors (다양한 생물반응기에서 이타콘산의 생산)

  • A suitable culture method and bioreactor type for itaconic acid production were chosen by comparing the maximal concentration of itaconic acid produced in various systems. In batch culture, the maximal concentration of itaconic acid produced in a bubble column reactor was about 5% greater than that produced in stirred-tank or external-loop airlift reactor. These results were thought to be due to lower shear force and higher mass transfer efficiency in a bubble column reactor in comparison with other reactors. Moreover, the fed-batch mode in a bubble column was found to be a suitable one, producing about 25% higher concentration of itaconic acid compared to batch mode.

  • Ultrathin networks of itaconic acid copolymers and poly(allylamine) were produced by a Langmuir-Blodgett (LB) technique employing a double-chain amine as a monolayer template which was subsequently removed by extraction after thermal crosslinking. Itaconic acid copolymers used were copoly (itaconic acid-ethyl vinyl ether) and copoly (itaconic acid-n-butyl vinyl ether). The polyion-complexed monolayers of three components consisting of template amine, itaconic acid copolymer and poly (allylamine) were formed at the air-water interface. The Langmuir film properties have been studied by the surface pressure-area isotherm and fluorescence microscopy. The monolayers were transferred on solid substrates and were characterized by FT-IR spectroscopy, X-ray photoelectron spectroscopy, and scanning electron microscopy (SEM). Two-dimensional polymer networks were formed through imide or amide linkages by heat treatment under vacuum. The heat-treated films were extracted with chloroform after immersion in aq. sodium chloride to remove the template amines. SEM observation of a LB film on a porous fluorocarbon membrane filter with pore diameter of 0.1 μm showed covering of the pores by six layers in the polyion complex state.

  • Itaconic acid has been produced during the cutivation of Aspergillus terreus DSMZ 5770 by using several starchs as carbon sources. The starchs were pretreated by partial hydrolysis with some acids at various pH conditions. The highest yield for the production of itaconic acid has been found when rice starch was pretreated by sulfonic acid at pH 2.5 and utilized for the cultivation. Using the results from shaker fermentation A. terreus has been cultivated in 2.5 L bioreactor for the production of itaconic acid and its on-line monitoring.

  • Chitosan was prepared from chitin which was abstracted from wasted crab shells. Then itaconic acid was graft-copolymerized onto chitosan using ceric ammonium nitrate as a reaction initiator. To investigate the optimal grafting conditions, the influences of several factors on the grafting were studied, i. e., the concentrations of CAN and itaconic acid, the reaction temperature and time. And to find out its flocculation ability. the flocculation test was carried out with a metal plating factory waste water. The state of graft-copolymer was identified through IR spectra analysis. The optimal grafting conditions and flocculation results were shown to be : concentration of ceric ammonium nitrate is $3.5{\times}10^3M$, reaction temperature is $40^{\circ}C$ and reaction time is 4hrs with 0.25M of the monomer(itaconic acid). Though flocculation tests using chitosan, grafted chitosan and cation, CODcr. metal ions removal rates were measured. The order of superiority is Itaconic acid grafted chitosan>Chitosan>Cationic polymer.

  • In this study, we confirmed water content and oxygen permeability of new polymeric materials synthesized from itaconic acid used for soft contact lenses. In this study, we polymerized materials for soft contact lenses using HEMA (2-hydroxyethyl methacrylate), the based-monomer of soft contact lenses, EGDMA (ethylene glycol dimethacrylate) as a cross linkage agent, and the new additives mono-ester or di-ester derived from itaconic acid commercially produced by the fermentation of A. itaconicus. New polymer materials for contact lenses were synthesized with the mixture of HEMA and mono- or di-ester at different ratios and water content and oxygen permeability (Dk) was analyzed. In polymerizing HEMA and mono-ester (15%), the water content and oxygen permeability of contact lenses were found to be of good value at 57.7% and 28.6 Dk respectively. The mixture of HEMA and mono-ester is more excellent than HEMA/di-ester in regards to water content and oxygen permeability. The water content and oxygen permeability of soft contact lenses made by new polymeric materials were highly represented.

  • Synthesis of methyl 3-methyloctanoate, a perfume component isolated from African orchid Aerangis confusa (or Aerangis kirkii) was achieved starting from itaconic acid in 9 steps. Itaconic acid is one of the cheapest organic compounds which is the fermentation product of microorganism Asp. terreus. As the key intermediate, 2-methyl-1,4-butanediol 4-acetate was obtained through the enzymatic regioselective hydrolysis of 2-methyl-1,4-butanediol diacetate with lipase. After Grignard reaction and oxidation, 3-methyloctanoic acid was obtained and converted to the various corresponding scented esters with a variety of alkyl alcohols, and the resulting fragrancy esters are expected to be utilized as the aroma additive materials in cosmetics, drinks and foods.

Studies on the Optimal Culture Conditions for Itaconic Acid Production by Aspergilus Terreus (Aspergillus Terreus에 의한 이타콘산 생산을 위한 최적배양조건에 관한 연구)

  • The production of itaconic acid by Aspergillus terreus NRRL 1960 was studied. The optimal culture conditional such as pH, inoculum size and medium composition were established. Maximum production of itaconic acid, $19.18g/\ell$, was obtained when the cultivation was carried out at $37^{\circ}C$ and pH 2.5 for 7days, with medium containing 5%(w/v) glucose, 0.5%(w/v) NH4Cl, 0.2%(w/v) yeast extract 0.1%(w/v) CaC12, 0.1%(w/v) MgSO4 and 0.2%(w/v) NaCl. A proper medium for inoculum culture was found to be 2%(w/v) malt extract. The batch production of itaconic acid with free cells in a stirredtank reactor was not efficient compared to the shake-flask culture.

Statistical Optimization of Production Medium for Enhanced Production of Itaconic Acid Biosynthesized by Fungal Cells of Aspergillus terreus (Aspergillus terreus에 의해 생합성되는 이타콘산의 생산성 증가를 위한 통계적 생산배지 최적화)

  • Statistical optimization of the production medium was carried out in order to find an optimal medium composition in itaconic acid fermentation process. Itaconic acid utilized in the manufacture of various synthetic resins is a dicarboxylic acid biosynthesized by fungal cells of Aspergillus terreus in a branch of the TCA cycle via decarboxylation of cis-aconitate. Through OFAT (one factor at a time) experiments, six components (glucose, fructose, sucrose, soluble starch, soybean meal and cottonseed flour) were found to have significant effects on itaconic production among various carbon- and nitrogen-sources. Hence, using these six factors, interactive effects were investigated via fractional factorial design, showing that the initial concentrations of sucrose and cottonseed flour should be high for enhanced production of itaconic acid. Furthermore, through full factorial design (FFD) experiments, negative effects of $KH_2PO_4$ and $MgSO_4$ on itaconic acid biosynthesis were demonstrated, when excess amounts of the each component were initially added. Based on the FFD analysis, further statistical experiments were conducted along the steepest ascent path, followed by response surface method (RSM) in order to obtain optimal concentrations of the constituent nutrients. As a result, optimized concentrations of sucrose and cottonseed flour were found to be 90.4g/L and 53.8g/L respectively, with the corresponding production level of itaconic acid to be 4.36 g/L (about 7 fold higher productivity as compared to the previous production medium). From these experimental results, it was assumed that optimum ratio of the constituent carbon (sucrose) and nitrogen (cottonseed flour) sources was one of the most important factors for the enhanced production of itaconic acid.

Acylation of Wool Keratin with Dibasic Acid Anhydrides - on reaction mechanism - (2염기산 무수물에 의한 양모섬유의 아실화(I) - 반응메카니즘을 중심으로 -)

  • Wool yarns were treated in dimethylformamide solutions containing various concentrations of three dibasic acid anhydrides : succinic, itaconic, and phthalic anhydrides in various conditions. The structural aspects of these dibasic acid anhydrides are different : the succinic, itaconic, and phthalic acid anhydrides have saturated aliphatic etylene, unsaturated aliphatic vinyl and aromatic phenyl groups, respectively. The reaction mechanism of the acylation of wool keratin and some resction conditions were invastigated. And the results are as follows. 1. The N-acylation and formation of free carboxyl group were dominant rather than the O-acylation cross-linked on the side chain of polypeptide. The acylation of wool keratin is easier than that of silk fibroin. 2. The higher molecular weight, steric hinderance and resonance caused lower acylating reactivity. By the determination of acyl contents for acylated keratin, it was rerealed that the degree of acylation was succinic acid anhydride > itaconic acid anhhydride > phthalic acid anhydride.
