척추동물의 Isozyme에 관한 비교연구: III. 한국산 뱀목의 Lactate 및 Malate Dehydrogenase Isozyme

  • MDH and LDH isozymes of various tissues of serveral snakes which belong to Colubridae, Crotalidae and Viperidae were studied. MDH isozymes were appeared as two bands in all species. Four LDH isozymes were found inall species of Colubridae, Crotalidae and Viperidae. LDH isozymes of muscle and liver had fundamentally the same pattern. On the other hand, there were two LDH isozymes in the heart of all species and the pattern of heart LDH isozyme was reverse of the pattern in muscle. The pattern of LDH isozymes of stomach tissue can be thought as the same pattern with that of heart in all species, but in the stomach tissue of Crotalidae and Viperidae there were four LDH isozymes.

  • A cellulosse acetate electrophoretic survey of Korean Anura has revealed the presence of diverse lactate and malate dehydrogenase (LDH and MDH) isozymes. The pattern of LDH and MDH isozymes in the tissues of the central nervous system of the six species of Anura examined are species specific and differ from those of mammals and birds. Both Rana nigromaculata nigromaculata and Rana nigromaculata coreana have two molecular forms of LDH and MDH, respectively, with almost the same pattern. Whole brain homogenate of Rana temporaria shows also a maximum of only two LDH isozymes. Both Bufo bufo asiaticus and Bombina orientalis have five molecular forms of LDH with an entirely different spacing on the zymograms, whereas Rana rugosa has three. Two molecular forms of MDH are present in all animals examined and one band is shown in olfactory lobe and mixture of cerebellum and medulla oblongata of Rana nigromaculata nigromaculata.

  • Electrophoretic isozyme patterns from mycelia, primordia, cap and stem of Pleurotus spp. collected in Korea were compared. Primordia, cap and stem of fruitbody showed very similar isozyme patterns but mycelial isozyme patterns were different from those of fruitbody. Isozyme patterns of malate dehydrogenase, acid phosphatase in Pleurotus ostreatus collected from different regions in Korea were similar but those of esterase, peroxidase, leucine amino peptidase and superoxide dismutase were different. Interspecific comparison of esterase isozyme patterns among Pleurotus ostreatus P. cornucopiae and, P. florida was very different and may be valuable subsidiary tool to conventional taxonomic techniques for identifying species of Pleurotus. A dendrogram by similarities of isozyme band pattern was presented.

The LDH and MDH isozymes in the tissues of the brain, heart, stomach, skeletal muscle and liver of the six species of Anura examined show the species specific patterns which differ from those of mammals and birds. Two isozymic forms of LDH and MDH exist in both Rana nigromaculata and Rana nigromaculata coreana, respectively, with almost the same pattern. LDH of Bombina orientalis has five isozymic forms, and that of Rana temporaria ornativentris contains four isozymes. Bufo sp. has 3 to 5, and Rana rugosa has 3 to 4 isozymic bands according to the tissues. MDH's of all animals have two isozymic forms with different spacing on the zymograms.

Purification and Characterization of Peroxidase Isozyme C from Mung Bean (녹두의 Peroxidase Isozyme C의 생화학적 성장)

  • Peroxidase isozyme C was isolated from mung bean cotyledon and purified to homogeneity as ascertained by chromatography and polyacrylamide gel electrophoresis, and then crystallized. Purification procedures included ammonium sulfate precipitation and column chromatography on Sephadex G-75, DEAE-cellulose and DEAE-Sephadex A-50. Peroxidase isozyme C was purified about 63 fold with 5% recovery. Isozyme C showed optimal activity at pH 5.0 with o-dianisidine and at pH 6.0 with guaiacol as substrate, and the optimal temperature was $70^{\circ}C$. Molecular weight of 50,000 was estimated for the isozyme C by SDS-polyacrylamide gel electrophoresis. At $70^{\circ}C$, it took 30 min to inactivate the isozyme to 50%, and at $80^{\circ}C$, this isozyme was almost completely inactivated in 20 min. The Km value of isozyme C for o-dianisidine was 0.11mM and that for guaiacol was 60.98mM using hydrogen peroxide as cosubstrate, and the kinetic pattern showed a competitive cyanide inhibition with respect to substrate. The crystalline structure of isozyme C was rectangular in shape.

  • This study was planned to identify the variety of Italian ryegrass using electrophoresis. Thirty seven varieties of Italian ryegrass were tested by starch gel electrophoresis. The specific electrophoretic zymograms of each variety were observed by Alcohol dehydrogenase(ADH) and Esterase. The results were surnrnerized as follows; 1. AU varieties displayed two band zones by ADH and Rf values were 0.63 and 0.6 (Table 2, Fig. 2). 2. There were five band type for ADH isozyme of 37 varieties classified with isozyme banding pattern. According to the isozyme band type 7, 2, 6, 18 and 4 varieties belong to group, I, II, III, IV, and V, respectively (Table 2). 3. The varieties displayed single band zone for Esterase isozyme and Rf value was 1.00 (Table 2, Fig. 4). 4. According to banding type, Esterase isozyme of 37 varieties classified into 3 groups, 22, 8 and 7 varieties belong to group, I , II, and III, respectively (Table 2).

Changes of the Malate Dehydrogenase Isozymes in Oyster (Crassostrea gigas) Exposed to Different Temperature, pH and Salinity (온도, pH 및 염도가 굴(Crassostrea gigas)의 MDH isozyme에 미치는 영향)

  • Changes of malate dehydrogenase isozyme in oyster exposed to different temperature, pH and salinity were investigated by polyacrylamide gel electrophoresis. MDH isozyme in control group was separated into two bands on the positive side. In case of temperature and pH stress, MDH isozyme was separated into only one band after 12 hours exposure but two bands after 24, 48 hours exposure on the positive side. In case of salinity stress, after 12 hours exposure, MDH isozyme bands were separated into two bands in 5 ppt, 30 ppt and three bands in 10 ppt, 40 ppt concentration on the positive side. After 24 hours and 48 hours exposure case in salinity stress, MDH isozyme bands was separated into two bands on the positive side in all concentration. Activities of isozyme bands show their characteristics according to the condition of experiment. In conclusion, changes of MDH isozyme was a biochemical defense mechanism in oyster and result from effect of environmental stress to oyster.

  • A electrophoretic study on seveal important enzymes was conducted with seed, leaf and stem of Benincasa hipida (Thunb) Cogn and Commelina communis L. Isozyme patterns of esterase were different from the parts and fruit of wax gourd appeared only one band. In the peroxidase isozyme, fruit of wax gourd appeared four bands, but stem of common dayflower no appeared. In the amylase isozyme, seed and fruit of 'gourd were appeared clearly one band, respectively.

  • The changes in peroxidase activity and its isozyme pattern in the different parts of mung bean sprout were investigated; The enzyme activity in cotyledon and root showed a tendency to increase at an early stage and then decreased gradually as germination continued. However, the crude homogenate of epicotyl and hypocotyl showed a continuous decline in the enzyme activity. In particular, the enzyme activity of the root was $1.5{\sim}3.5$ times higher than that of other parts. Gel electrophoresis of the crude homogenate revealed that the number of isozyme in every part of the mung bean sprout increase during germination up to 6th days. Two isozymes from root were partially purified by ammonium sulfate fractionation, gel filtration by Sephadex G-75 and DEAE cellulose column chromatography. One of the isozymes (A) was purified 16-fold by the present procedure, but the purity of the other isosyme (B) was not increased , significantly. Isozyme A was the most active at $65^{\circ}C$ and isozyme B at $70^{\circ}C$, while both isozyme (A, B) have a optimal pH of 5.6. The Km values of isozyme A and B for 0-dianisidine as a hydrogen donor determined to be 0.071 mM and 0.052mM, respectively, and those for isozyme A and B using $H_2O_2$ as a hydrogen acceptor were 0.28mM and 0.23mM, respectively.
