The objective of this study was to evaluate the quality characteristics of lotus leaf Dasik prepared with various sweeteners, including honey (HON), starch syrup (SS), isomaltooligosaccharide(IMOS), galactooligosaccharide (GOS), and fructooligosaccharide (FOS). The moisture contents of samples were highest in GOS. L-values were significantly higher in the HON group. Whereas a- and b-values were not significantly different among the groups. According to the results for mechanical characteristics, hardness was significantly lower in HON while cohesiveness, gumminess, and chewiness were highest in SS; however, adhesiveness and springiness were not significantly different among the groups. The sensory evaluation results showed that the lotus leaf Dasik containing FOS received the highest preference scores.

The digestibility of xylooligosaccharide(XO) by juices of the digestive tract and retardation effect of XO on the adsorption of bile acids were compared with fructooligosaccharide(FO) and isomaltooligosaccharide(IO). In vitro digestion experiments showed that any hydrolyzed products of FO, IO and XO were not detected by HPLC after reaction with saliva, pancreatic, artifical intesteinal, and large intestinal luices, and artifical sera for 4 hours at 37$^{\circ}C$. However, IO were mostly digested by the small intestinal juice, and some quantity of FO were digested. XO were not digested at all by any enzyme of digestive tract. In order to investigate retardation effect of XO on the bile acid absorption. In vitro, permeability of bile acid against dialysis membrane was determined in the mixture which contained guar gum instead of XO was set 100%. The premeability of bile acid showed about 50% in the FO and IO mixture and 43% in the XO mixture. The activity of lactase in FO group and activity of sucrase and maltase in XO group in rat small intestinal mucosa were significantly decreased. Consequently, the present results indicate that XO is indigestible in digestive tract and has retarding effect of adsorption of bile acid compared with the other oligosaccharides. The disaccharidase activity of the XO dietary group was lower than that of the other oligosaccharides dietary group. Furthermore, it was suggested that hydrolysis of sugar may be retarded in digestive tract and glucose level in blood may be controlled effectively by the XO.

Prebiotic isomaltooligosaccharide preparations contain $\alpha$-D-glucooligosaccharides comprising isomaltooligosaccharides (IMOs) and non-prebiotic maltooligosaccharides (MOs). They are both glucose oligosaccharides characterized by their degree of polymerization (DP) value (from 2 to $\sim$10), linkages types and positions (IMOs: $\alpha$-(1$\rightarrow$2, 3, 6 and in a lower proportion internal 1$\rightarrow$4) linkages, MOs: α-(1$\rightarrow$4) linkages). Their structure is the key factor for their prebiotic potential. In order to determine and elucidate the exact structure of unknown IMOs and MOs, unambiguous assignments of $^{13}C$ and $^1H$ chemical shifts of commercial standards, representative of IMOs and MOs diversity, have been determined using optimized standard one and two-dimensional experiments such as $^1H$ NMR, $^{13}C$ NMR, APT and ${^1}H-{^1}H$ COSY, TOCSY, NOESY and <$^1H-{^{13}}C$ heteronuclear HSQC, HSQC-TOCSY, and HMBC. Here we point out the differential effect of substitution by a glucose residue at different positions on chemical shifts of anomeric as well as ring carbons together with the effect of the reducing end configuration for low DP oligosaccharides and diasteroisotopic effect for H-6 protons. From this study, structural $^{13}C$ specific spectral features can be identified as tools for structural analysis of isomaltooligosaccharides.

A marine bacterium Bacillus cereus ASK202 showing a high agar degrading activity, was incubated in the culture medium containing agar. After incubation for 30 hr, the productivity of agarase in the culture broth reached to maximum value (160.8 units/L). As the results of TLC and HPLC analysis, agarooligosaccharides (degrees of polymerization 2, 4 and 6) were produced from the hydrolysis of agar by using the crude agarase. Physical and chemical properties of agarooligosaccharides were compared with the manufactured products of other oligosaccharides (fructooligosaccharide; isomaltooligosaccharide; maltotetraoligosaccharide) and agarooligosaccharides showed higher viscosity, higher contents of oligosaccharides, higher stability at low pH's and higher temperatures, and lower sweetness than other oligosaccharides.

This study was conducted to examine the effects of dietary oligosaccharide on the blood glucose and serum lipid composition in streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male rats weighing 150$\pm$10g were randomly assigned to one normal and four STZ-induced diabetic groups. Diabetic groups were classified to basal diet(DM group) 10% xylooligosaccharide diet(DM-XO group) 10% isomaltooligosaccharide(DM-IMO group) and 10% fructooligosaccharide (DM-FO). Diabetes was experimentally induced by intravenous injection of 50 mg/kg of body weight of STZ in citrate buffer (pH 4.3) after feeding of experimental diets for 4 weeks. The rats were fed with experimental diet for further 4 weeks in diabetic state. The oligosaccharide diets were not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the body weight food intakes and food efficiency ratio. The oligosaccharide diets were also not effected on the weights of liver kidney and small intestine but the weight of cecum was significantly increased on the groups of xylooligosaccharide and isomaltooligosaccharide diet. The levels of oral glucose tolerance test was more effectively improved by DM-XO group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. The levels of blood glucose were markedly lower in oligosaccharide supplemented groups than that of DM group. Activities of two intestinal enzymes such as lactase and sucrase in DM-XO and DM-FO groups were lower than that of DM group while activity of maltase was lower only in DM-XO in DM-FO groups than that of DM-group respectively. The levels of serum triglyceride in DM-XO group were lower than that of DM-group respectively. The levels of serum triglyceride in DM-XO groups were lower than that of DM group however was no significant differences among the oligosaccharide groups. These results suggest that dietary oligosaccharide may act as functional food to be capable of improving carbohydrate and lipid metabolism in diabetic rats.
In this study, transglucosidase (TG), an enzyme produced by Aspergillus niger, synthesized isomaltooligosaccharide from ${\alpha}-(1{\rightarrow}4)$ linked substrates. The highest TG-producing A. niger KCTC6913 was selected from six kinds of species, and optimized TG producing conditions were established. Five different carbon sources (potato starch, sweet potato starch, corn starch, wheat starch, and dextrin) and three different nitrogen sources (yeast extract, malt extract, and beef extract) were tested to establish the carbon and nitrogen sources favorable for TG production. Measurements of TG activity after an initial culture at pH 5.0 for 15 days revealed that potato starch and yeast extract, which are basic culture media, resulted in the highest TG activity. In addition, A. niger KCTC6913 increased TG production under aerobic conditions and a controlled carbon/nitrogen ratio. In conclusion, to evaluate TG activity in the established optimal medium, it is confirmed that the basal and potato dextrose broth medium were used as a control, and the highest TG production was measured, which was highlighted in the established optimal medium.
A simple, fast and reproducible quantitative analysis method for sugar concentration composed in oligosaccharide mixture was developed. Two glass TLC plates were prepared per sample. After dipping one plate into the copper bicinchoninate reagent and the other plate into 5% sulfuric acid solution, both plates were baked in microwave oven until sugar spots were developed or the surface temperature of TLC plate becomes 60 to 70 $^{\circ}C$. The corrective factor values [F value =(the value of total sugar concentration converted as glucose unit/the value of reducing sugar concentration converted as glucose unit)/(polymerization degree of sugar)] of different molecular weight sugars were determined. Within the concentration of 0.25∼1.0 $\mu\textrm{g}$ in each sample loaded, the fructose-F (corrective factor value of fructose) was 0.45, yet for the higher concentration (2.5∼7.5 $\mu\textrm{g}$) fructose-F was 1.0. In case of glucose, in the range of 0.5∼7.5 $\mu\textrm{g}$, glucose-F was same as fructose-F, 1.0. However, as the molecular weight of sugar was increased, the F values were decreased in both maltodextrin and isomaltodextrin oligosaccharides in 0.5∼7.5 $\mu\textrm{g}$ of each sample loaded. Interestingly, F values were equal for the same molecular weight sugars, although the structures were different from each other. Using F value of each sugar, we could determine and compare the exact total sugar concentration of different molecular weight maltooligosaccharide and isomaltooligosaccharide. We also could determine if the unknown sugar was a reducing or non-reducing compound by using optimized TLC with microwave oven method.
The characteristics of sponge cakes prepared with replacement of 30, 50, 75, or 100% of sucrose with isomaltooligosaccharide (IOS), fructooligosanharide (FOS), maltitol symp (MS), or maltitol powder (MP), were examined through physical measurement and sensory evaluation. The specific gravities of foams and cake batters were not significantly different among samples (p .0.05). The use of IOS increased the viscosity of cake batter. The cakes containing IOS, FOS, MS, or MP were softer than control cakes (sucrose 100%). Especially cakes prepared with 30, 50% IOS, or 75, 100% MP, appeared to be fragile. When increasing levels of IOS or FOS were incorporated in the cake formula, cake crust color was getting darker than that of control cake, whereas cake containing maltitol was getting lighter as the levels of replacement increased. Generally, the volume of cake containing FOS were superior to that of control cake, whereas those of cakes containing above 50% MP were inferior. Sensory analysis of experimental cakes did not show significant differences from the control cake for softness, springiness and gumminess. Sweetness increased replacing the sucrose with FOS, MS or MP (30, 50%) and moistness increased using IOS, FOS, MS, or MP without agreement with moisture contents of cakes. Replacement of 30, 50% sucrose with MS or 30% with MP did not affect greatly the physical measurements or sensory characteristics studied.

This study mechanically measured physical properties of jelly made by adding kappa carrageenan and agar-agar to the sipjeondaebo extract, varying the addition ratio. The sipjeondaebo extract used in the study showed 4.95 pH, 7.0 Brix$^{\circ}$, and high turbidity of more than 3.0 NTU. The sugar content was fixed at 25.2 Brix$^{\circ}$ when isomaltooligosaccharide was added in a 25-percent concentration. When the jelly was mechanically measured after it became hard in the refrigerator, the measurement results did not show any significant difference in cohesiveness between different addition ratios of agar-agar to kappa carrageenan. In the measurement of chewiness and gumminess, the jelly show a significantly decreasing tendency in both the two properties as the increase in the addition rate of agar-agar was made. Hardness was also measured lower as the more addition of agar-agar was made, and its the highest level was shown when there was the addition of kappa carrageenan only. Springiness was the highest when the ratio of kappa carrageenan to agar-agar was 0.7%:0.3%, while it was measured the lowest when agar-agar was added in a 1.0% concentration. In the measurement of the jelly's physical properties, the results showed the highest level of appearance when the ratio of kappa carrageenan to agar-agar was 0.7% but indicated that there was no significant difference in flavor. The overall Quality was evaluated to maintain the highest level at the kappa carrageenan to agar-agar ratio of 0.7%:0.3%, followed by a 0.5%:0.5% addition ratio.
The effects of storage temperature, moisture content and the concentration of additives, such as sucrose fatty acid ester(SE), isomaltooligosaccharide(IO) and glycerin(GL), on texture properties, hardness(HA), cohesiveness(CO) and chewiness(CH) of Baikseolgi after 7 days storage were analyzed by response surface methodology(RSM). The contour values of HA of SE added Baikseolgi at 20, 50 and $80^{\circ}C$ of storage temperate were $1500{\sim}3200,\;500{\sim}1300$ and $100{\sim}400\;g_f$, respectively. The HA of IO or GL added Baikseolgi decreased with increased storage temperature, moisture content and additive concentration. The storage temperature was the most significant factor affecting the HA of Baikseolgi. However, the second and third significant factors were moisture content and additive concentration, respectively. These results imply that the control of storage temperature is the most effective method to increase the storage stability of Baikseolgi. The CO of IO or GL added Baikseolgi was increased by the change of strage temperature from $20{\circ}\;to\;50{\circ}$. While, there was no significant difference between $50{\circ}\;and\;80{\circ}$ of storage temperature. The CO of IO or GL added Baikseolgi was maximized around 40% of moisture content and that of GL added Baikseolgi was minimized around 0.5% of GL concentration. The storage temperature, additive concentration and moisture content were the first, second and third affacing factors on the CO of Baikseolgi, respectively. The CH of Baikeolgi was decreased by increasing storage temperature, moisture content and additive concentration.
