Detection of Microbial Growth in an Automated Culture System (자동배양기를 이용한 미생물 검출)

  • Modern automated culture systems have increased the isolation rate of microorganisms and shortened the time to detection, reducing experimental errors in diagnosis of infecting agents. BacT/ALERT 3D system is based on the colorimetric detection of $CO_2$ produced by the growing microorganisms. In order to evaluate the efficiency of the detection system, sterility test were performed using 6 bacteria. With standard aerobic and anaerobic bottles containing the liquid media, both three aerobic bacteria (P. aeruginosa, M. luteus, B. subtilis) and a facultative bacterium S. aureus were detected up to 1 CFU in 31.44 hr. In addition, growth of anaerobic C. sporogenes was recognized up to 1 CFU in 15.96 hr. The slowly growing bacteria P. acnes was detected up to 10,000 CFU in 129.36 hr. In comparison with conventional culture method, BacT/ALERT 3D automated culture system was more sensitive and saved detection time up to$2\sim10$ hr. Therefore, this automated culture system enables to efficiently detect bacteria in clinical samples and biological medicines.

Isolation and Identification of Pathogenic Bacteria from Spinach (시금치로부터 병원성세균의 분리 및 동정)

  • Raw and washed spinaches were tested to evaluate the incidences of Aeromonas hydrophila, Escherichia coli O157:H7, Plesiomonas shigelloides, Pseudomonas aeruginosa, Salmonella spp., Shigella spp., Yersinia enterocolitica, Bacillus cereus, Campylobacter jejuni, Clostridium perfringens, Listeria monocytogenes, and Staphylococcus aureus. Four pathogenic bacteria were isolated from spinach samples, and identified by morphological and biochemical methods, including API and ATB identification systems. Isolates from MacConkey, Cereus Selective, Clostridium Perfringens, and Baird-Parker agar media were in 99.9, 99.8, 99.9, and 97.8% agreements with A. hydrophila, B. cereus, C. perfringens, and S. aureus at the species level, respectively. SET-RPLA revealed, among the five strains of S. aureus isolates, two produced type A enterotoxin. All five strains of B. cereus isolates produced enterotoxin as revealed with CRET-RPLA.

Behavior of Geotextile Tube Composite Structure by 2-D Limit Equilibrium and Plane Strain Analysis (2차원 한계평형 및 평면변형해석을 통한 지오텍스타일 튜브 복합구조물의 거동분석)

  • The geotextile have been used in filtration and drainage for over 30 years in many applications of civil and environmental projects. Geotextile tube is compound technology of filtration and drainage property of geotextile. Geotextile have been used for various types of containers, such as small hand-filled sandbags, 3-dimensional fabric forms for concrete paste, large soil and aggregate filled geotextile gabion, prefabricated hydraulically filled containers, and other innovative systems involving containment of soils using geotextile. They are hydraulically filled with dredged materials. It have been applied in coastal protection and scour protection, dewatering method of slurry, and isolation of contaminated material. Recently, geotextile tube technology is no longer alternative construction technique but suitable desired solution. This paper presents the behavior of geotextile tube composite structure by 2-D limit equilibrium and plane strain analysis. 2-D limit equilibrium analysis was performed to evaluate the stability of geotextile tube composite structure for the lateral load and also the plane strain analysis was conducted to determine the design and construction factors. Based on the results of this paper, the three types of geotextile tube composite structure is stable. And the optimum tensile strength of geotextile is 151kN/m and maximum pumping pressure is 22.7kN/m.

Studies on Soybean Protein [Part ll]-Isolation and Subunit Composition of Multiple 7S Globulins- (대두(大豆) 단백질(蛋白質)에 관(關)한 연구(硏究) 제2보[第二報]-7S Globulin중의 복합단백질(複合蛋白質)의 분리(分離) 및 그 구성(構成) Subunit에 대하여)

  • The multiple 7S globulins composed of two fractions (A and B) in the electrophoresis with Davis' method were isolated at different stages of the soybean seed development. Electrophoresis of their subunits liberated in PAWU solvent [phenol-acetic acid-water (2 : 1 : 1) solution plus 5M urea] yielded 4 major bands. Observation of both the electrophoretic bands of the multiple 7S fractions(7S-A and 7S-B) and those of their subunits was suggestive of a similarity of the subunit pattern between two 7S fractions. The two fractions in multiple 7S globulins were isolated with DEAE-Sephadex A-50 column$(2.0{\sim}100cm)$ chromatography. They were separated into 2 fractions in a linear gradient concentration of 0.28 to 0.40M NaCl with phosphate buffer (pH 7.8) containing 10mM ${\beta}-mercaptoethanol$(ME). The isolated protein was dissociated into subunits with two different solvent systems; in PAWU solvent and in Tris-HCl buffer(pH 8.0) containing 1% sodium dodecyl sulfate (SDS) and 40mM ME. The dissociated subunits were subjected to electrophoresis in PAWU-treated 7.5% acrylamide gel and in 1% SDS-treated 5.6% acrylamide gel. In PAWU gel electrophoresis, total 7S globulin was separated into 5 major bands, two of which were occupied in common by two 7S fractions(7S-A and 7S-B). In SDS gel electrophoresis, total 7S globulin was separated into 7 major bands, three of which were overlapped with the subunit of the two 7S fractions. The above results alluded us to the presence of a common and/or similar subunit between the multiple 7S globulins.

MANAGING A PROLONGED STATION BLACKOUT CONDITION IN AHWR BY PASSIVE MEANS

  • Removal of decay heat from an operating reactor during a prolonged station blackout condition is a big concern for reactor designers, especially after the recent Fukushima accident. In the case of a prolonged station blackout condition, heat removal is possible only by passive means since no pumps or active systems are available. Keeping this in mind, the AHWR has been designed with many passive safety features. One of them is a passive means of removing decay heat with the help of Isolation Condensers (ICs) which are submerged in a big water pool called the Gravity Driven Water Pool (GDWP). The ICs have many tubes in which the steam, generated by the reactor core due to the decay heat, flows and condenses by rejecting the heat into the water pool. After condensation, the condensate falls back into the steam drum of the reactor. The GDWP tank holds a large amount of water, about 8000 $m^3$, which is located at a higher elevation than the steam drum of the reactor in order to promote natural circulation. Due to the recent Fukushima type accidents, it has been a concern to understand and evaluate the capability of the ICs to remove decay heat for a prolonged period without escalating fuel sheath temperature. In view of this, an analysis has been performed for decay heat removal characteristics over several days of an AHWR by ICs. The computer code RELAP5/MOD3.2 was used for this purpose. Results indicate that the ICs can remove the decay heat for more than 10 days without causing any bulk boiling in the GDWP. After that, decay heat can be removed for more than 40 days by boiling off the pool inventory. The pressure inside the containment does not exceed the design pressure even after 10 days by condensation of steam generated from the GDWP on the walls of containment and on the Passive Containment Cooling System (PCCS) tubes. If venting is carried out after this period, the decay heat can be removed for more than 50 days without exceeding the design limits.

Population Genetic Structure of the Korean Endemic Species, Iksookimia pacifica (Pisces: Cobitidae) Distributed in Northeast Korea (한국고유종 북방종개(어류강, 미꾸리과)의 집단유전학적 구조)

  • Population genetic studies of 10 groups of Iksookimia pacifica were conducted to investigate the genetic diversity and population genetic structure across its known range in South Korea. Population DNA sequences of one mitochondrial gene (mtCOI) and three nuclear genes (IRBP, EGR2B, RAG1) were examined in samples collected from ten streams that flow into the East Sea. Both mitochondrial and nuclear sequences exhibited significant differentiation among populations except a few cases. The Bayesian analysis of the multi-locus genotypes inferred from the DNA sequences of nuclear genes clustered the individual fish largely into two geographical groups: a northern group (from Baebong stream to Cheonjin stream) and a southern group (Yangyangnamdae stream to Gangneungnamdae stream). Given that the streams flowing into the East Sea are geographically isolated water systems, such separation of genotypes can be interpreted by the geographical separation of common ancestors into north and south that had colonized South Korea. Since the initial geographical separation of the ancestral population by north and south, the ancestral groups seem to have experienced further differentiation into the current genetic clusters through the physical isolation of streams by the East Sea in each region. It is notable that many individuals in the Jasan stream formed a genetic cluster with those of Yangyangnamdae and Gangneungnamdae streams which are distant from each other. In addition, mitochondrial gene showed low genetic differentiation between some neighboring populations and very low level of genetic diversity in several populations. The present population genetic study will provide valuable information for the conservation and management of the Korean endemic fish species, I. paicifica.

Comparative Experiments to Assess the Effects of Accumulator Nitrogen Injection on Passive Core Cooling During Small Break LOCA

  • The accumulator is a passive safety injection device for emergency core cooling systems. As an important safety feature for providing a high-speed injection flow to the core by compressed nitrogen gas pressure during a loss-of-coolant accident (LOCA), the accumulator injects its precharged nitrogen into the system after its coolant has been emptied. Attention has been drawn to the possible negative effects caused by such a nitrogen injection in passive safety nuclear power plants. Although some experimental work on the nitrogen injection has been done, there have been no comparative tests in which the effects on the system responses and the core safety have been clearly assessed. In this study, a new thermal hydraulic integral test facility-the advanced core-cooling mechanism experiment (ACME)-was designed and constructed to support the CAP1400 safety review. The ACME test facility was used to study the nitrogen injection effects on the system responses to the small break loss-of-coolant accident LOCA (SBLOCA) transient. Two comparison test groups-a 2-inch cold leg break and a double-ended direct-vessel-injection (DEDVI) line break-were conducted. Each group consists of a nitrogen injection test and a nitrogen isolation comparison test with the same break conditions. To assess the nitrogen injection effects, the experimental data that are representative of the system responses and the core safety were compared and analyzed. The results of the comparison show that the effects of nitrogen injection on system responses and core safety are significantly different between the 2-inch and DEDVI breaks. The mechanisms of the different effects on the transient were also investigated. The amount of nitrogen injected, along with its heat absorption, was likewise evaluated in order to assess its effect on the system depressurization process. The results of the comparison and analyses in this study are important for recognizing and understanding the potential negative effects on the passive core cooling performance caused by nitrogen injection during the SBLOCA transient.

Current Status and Future Research Directions of Separator Membranes for Lithium-Ion Rechargeable Batteries (리튬이차전지용 분리막 이해 및 최신 연구 동향)

  • Lithium-ion rechargeable batteries (LIBs) have garnered increasing attention with the rapid advancements in portable electronics, electric vehicles, and grid-scale energy storage systems which are expected to drastically change our future lives. This review describes a separator membrane, one of the key components in LIBs, in terms of porous structure and physicochemical properties, and its recent development trends are followed. The separator membrane is a kind of porous membrane that is positioned between a cathode and an anode. Its major functions involve electrical isolation between the electrodes while serving as an ionic transport channel that is filled with liquid electrolyte. The separator membranes are not directly involved in redox reactions of LIBs, however, their aforementioned roles significantly affect performance and safety of LIBs. A variety of research approaches have been recently conducted in separator membranes in order to further reinforce battery safeties and also widen chemical functionalities. This review starts with introduction to commercial polyolefin separators that are currently most widely used in LIBs. Based on this understanding, modified polyolefin separators, nonwoven separators, ceramic composite separators, and chemically active separators will be described, with special attention to their relationship with future research directions of advanced LIBs.

A Distributed Real-Time Concurrency Control Scheme using Transaction the Rise of Priority (트랜잭션 우선 순위 상승을 이용한 분산 실시간 병행수행제어 기법)

  • As real-time database systems are extended to the distributed computing environment, the need to apply the existing real-time concurrency control schemes to the distributed computing environment has been made. In this paper we propose an efficient concurrency control scheme for distributed real-time database system. Our proposed scheme guarantees a transaction to commit at its maximum, reduces the restart of a transaction that is on the prepared commit phase, and minimizes the time of the lock holding. This is because it raises the priority of the transaction that is on the prepared commit phase in the distributed real-time computing environment. In addition, it reduces the waiting time of a transaction that owns borrowed data and improves the performance of the system, as a result of lending the data that the transaction with the raised priority holds. We compare the proposed scheme with DO2PL_PA(Distributed Optimistic Two-Phase Locking) and MIRROR(Managing Isolation in Replicated Real-time Object Repositories) protocol in terms of the arrival rate of transactions, the size of transactions, the write probability of transactions, and the replication degree of data in a firm-deadline real-time database system based on two-phase commit protocol. It is shown through the performance evaluation that our scheme outperforms the existing schemes.

A survey of respiratory pathogens in dogs for adoption in Gwangju metropolitan city animal shelter, South Korea (광주광역시동물보호소 입양 대상 유기견의 호흡기 질병 실태 조사)

  • Canine infectious respiratory disease (CIRD), also known as infectious tracheobronchitis or kennel cough occurs in a multiple-dog environment such as a shelter. In this study, we were collected 300 of nasal swab samples from dogs and 145 of environmental samples from a shelter to investigate respiratory pathogens of dogs in the Gwangju metropolitan city animal shelter from February to October, 2019. Bacteria cultures for isolation of Bordetella (B.) bronchiseptica and polymerase chain reaction (PCR) tests were performed for detection of eleven canine respiratory pathogens, namely Mycoplasma (M.) cynos, canine distemper virus (CDV), canine influenza virus (CIV), canine parainfluenza virus (CPIV), canine respiratory coronavirus (CRCoV), alpha-coronavirus (CCoV), canine pneumovirus (CnPnV), canine hepacivirus (CHeV), canine adenovirus type 2 (CAdV-2), canine herpesvirus-1 (CHV-1) and canine bocavirus (CBoV). Among 300 nasal swab samples, 148 samples (49.3%) were positive for at least one pathogens. CHV-1 was the most common pathogen, found in 95/300 (31.7%) samples. Subsequently, M. cynos (22.0%), B. bronchiseptica (2.3%), CPIV (2.0%), CBoV (1.7%), CCoV (0.7%) were detected. The detection rates of M. cynos and CHV-1 according to the duration of stay in the shelter were statistically significant. Among environmental samples, M. cynos, CCoV, CBoV and CHV-1 were detected in 45/145 (31.0%). These results indicated the need for disease control and prevention systems in the shelter.
