Fish Fauna and Disturbance in Odaesan National Park, Korea (오대산국립공원의 어류상과 어류교란)

  • Fish fauna of mountain streams in the Odaesan National Park area was investigated from May to October 2004. A total of 2,580 individuals were collected and classified into 26 species belonging to 9 families at the 25 sites. There were 8 Korea endemic species $(30.77\%)$, including Pseudopungtungia tenuicorpa, Silurus microdorsalis, Coreoperca herzi. Exotic species, Oncorhynchus mykiss was collected in this survey. Dominant species were Rhynchocypris kumgangensis $(37.17\%)$, and subdominant species was Rhynchocypris steindachneri$(23.02\%)$. Also Zacco platypus, Zacco temmincki, Oncorhynchus masou masou, Brachymystax lenok tsinlingensis, and Ladislavia tazanowskii were numerous. In spite of the geographical isolation by Taebaek-sanmaek, 8 species were introduced from the Han River systems to eastern water systems in Odaesan National Park.

Analysis of Toxicity in Escherichia coli from the Expression of Human Purinergic Receptor $P2X_4$ (인간 퓨린수용체 $P2X_4$를 발현시킬 때 나타나는 대장균 독성의 원인분석)

  • In general, expression of membrane protein in Escherichia coli is very toxic to the host organism, but the mechanism for the toxicity is not clear yet. Expression of human purinergic receptor $P2X_4$ was found to be extremely toxic to the host E. coli. We examined this toxicity by isolation and analysis of less toxic mutant proteins. We could isolate 30 less toxic mutants of $P2X_4$ after hydroxylamine mutagenesis. Western blot showed that all of them produced proteins smaller than the wild type $P2X_4$. DNA sequencing of two largest mutant proteins showed that they were lost its second transmembrane domain. Localization analysis of these mutant proteins showed that they are not in cytoplasmic membrane, but in inclusion bodies. These data showed that inactive truncated $P2X_4$ is not toxic to E. coli and membrane integration and functionality of $P2X_4$ may be needed to show host toxicity.

Report on 30 unrecorded bacterial species of the phylum Firmicutes isolated from Korea in 2016

  • During the course of investigation of indigenous prokaryotic species in Korea, a total of 30 bacterial strains belonging to the phylum Firmicutes were isolated from diverse environmental sites such as soil, avian feces, wastewater treatment plants, fermented vegetables, seawater, algae, sea cucumber, octopus and tidal flat sediment. Phylogenetic analysis based on 16S rRNA gene sequences revealed that each strain showed high sequence similarity (${\geq}98.7%$) to the closest type strain and formed a robust phylogenetic clade with the most closely related species in the phylum Firmicutes. To date, there is no official record of these 30 species in Korea. Therefore, we report 26 species of 12 genera in the order Bacillales and 4 species of 4 genera in the order Lactobacillales which have not been reported in Korea. Morphological and biochemical characteristics, isolation sources and NIBR deposit numbers are described in the species descriptions.

  • File integrity checking is the most reliable method to examine integrity and stability of system resources. It is required to examine the whole data whenever auditing system's integrity, and its process and result depends on administrator's experience and ability. Therefore the existing method is not appropriate to intrusion response and recovery systems, which require a fast response time. Moreover file integrity checking is able to collect information about the damaged resources, without information about the person who generated the action, which would be very useful for intrusion isolation. In this paper, we propose rtIntegrit, which combines system call auditing functions, it is called Syswatcher, with file integrity checking. The rtlntegrit can detect many activities on files or file system in real-time by combining with Syswatcher. The Syswatcher audit file I/O relative system call that is specified on configuration. And it can be easily cooperated with intrusion response and recovery systems since it generates assessment data in the standard IDMEF format.

  • In order to improve the thermal efficiency of an internal combustion engine, various ignition timing control systems were examined and the best one was chosen. The parts used for the systems were a microcomputer system with DAS, 8 bit output port (D-FLIP FLOP), three types of isolation circuit, two types of ignition timing pulse generator, three types of switching circuit and two types of high voltage ignition circuit. Most systems did not operate well due to the effects of electromagnetic waves and surge currents occurring when the ignition began or ended with resulting high voltage. The best ignition timing control system was found to be the combination of (microcomputer system)-(ignition timing pulse generator using step motor position control pick-up)-(switching circuit using TR logic)-(high voltage ignition circuit using CDI).

Mating Systems and Inbreeding Pressure in Populations of Wild Lentil Tare, Vicia tetrasperm (Leguminosae) (얼치기완두(콩과) 집단의 교배계와 내교잡 압력)

  • The mating systems of natural populations of Vicia tetrasperm in Korea were determined using allozyme analysis. The result suggests that V. tetrasperm is low rates of outcrossing or mix-mating outcrossing (self-fertilization, s < 0.5). At the population levels, the values of inbreeding coefficient of ten populations in Korea varied from 0.131 to 0.176, giving an average 0.154. For ten natural populations, multi-locus estimates of outcrossing (tm) was 0.333 across fifteen polymorphic loci, with individual population values ranging from 0.269 to 0.423. The differences between the tm and ts values were not close to zero (tm - ts > 0.154), indicating that biparental inbreeding was significant in the loci. The reason for relatively low outcrossing rates of some populations could be attributed to extensive consanguineous mating and isolation of flowering mature plants. Although heterozygote excess was observed in one natural population, most populations exhibited varying degrees of inbreeding and heterozygotes deficit. Thus, selection against homozygotes operated in the progeny populations throughout the life cycle.

  • Polytetrafluoroethylene (PTFE) is a commercialized friction material in friction pendulum systems used for earthquake hazard mitigation in structures, and it has excellent chemical resistance and frictional performance. However, PTFE has a relatively low wear resistance for the friction pendulum systems in service. As an alternative to PTFE, a cost-effective frictional material, polyvinylidene fluoride (PVDF) strengthened by magnesium oxide (MgO), with enhanced wear resistance performance is proposed in this study. The frictional performance of the developed PVDF/MgO was evaluated through experiments and compared with that of PTFE. Accordingly, a friction pendulum system was designed using the measured friction coefficient. The performance of this friction pendulum system was evaluated via nonlinear time history analyses of bridges. Subsequently, the plausibility of using PVDF/MgO as an alternative to PTFE as a friction material for friction pendulum systems was discussed.

Isolation and Characterization of Endophytic Bacteria from Rice Root Cultivated in Korea (한반도 중부지방의 벼 뿌리로부터 내생 세균의 분리와 특성 분석)

  • The 44 endophytic bacterial strains were isolated from surface-sterilized root of rice cultivated in seven different locations of Chungcheong province, Korea. Each isolate was introduced into rice seedlings grown gnotobiotically by inoculating scissor-cut first true leaf with cell suspensions, and the colonization capacity of each isolate in root tissue was analyzed at 7 days after inoculation. Sixteen out of 44 isolates were re-isolated from root successfully with the frequency of $10^{3-5}$ CFU/g tissue. Interestingly, seven out of 16 isolates were identified as Burkholderia species. The identity between inoculated strains and re-isolates was confirmed by genomic finger-printing and 16S rDNA sequence analysis. By a confocal laser scanning microscopic observation it was revealed that KJ001 strain, one of the sixteen isolates tagged with gfp colonized in root tissue especially around xylem. Six out of seven Burkholderia strains obtained in this study showed antagonizing activities against seven different fungal pathogens, contain nifH gene, and five of them enhanced growth of cucumber over 30%. The isolates showed no hypersensitive response on tobacco leaves and no pathogenecity in rice. From these results it was found that the endophytic Burkholderia strains will be useful in agriculture to develop a biocontrol agent or a bio-fertilizer.

  • Chlorophytum borivilianum Baker (Antharicaceae) commonly referred as 'Safed Musli' has been widely used in the Indian traditional systems of medicine to treat various diseases like rheumatism apart from having immunomodulating property and is used as general tonic. It is also known as 'Ayurvedic viagra' for its aphordisiac properties. C. borivilianum was screened for the first time to determine its antioxidant activity, isolation of the sapogenins and standardization of the isolated sapogenin fraction using HPTLC. Potent antioxidant activity of ethanolic extract was found by their ability to scavenge DPPH (84.51%), hydroxyl radical (48.95 %), ferryl bi-pyridyl complex (84.53%) along with the inhibition of lipid peroxidation (67.17%) at $100\;{\mu}g/ml$ concentration. The ethanolic extract also exhibited significant inhibition of superoxide anion radical generated by photochemiluminescence. Thus, the potent antioxidant activity validates the innumerable therapeutic claims of the plant in the traditional system especially its use as a Rasayana drug.

  • Obtaining mutant animals is important for studying the function of a particular gene. A chemical mutagenesis was first carried out to generate mutations in C. elegans. In this study, we used ultraviolet-activated 4,5',8-trimethylpsoralen to induce small deletion mutations. A library of mutagenized worms was prepared for recovery of candidate animals and stored at $15^{\circ}C$ during screening instead of being made into a frozen stock library. In order to isolate deletion mutations in target genes, a polymerase chain reaction (PCR)-based screening method was used. As a result, two independent mutants with deletions of approximately 1.0 kb and 1.3 kb were isolated. This modified and improved reverse genetic approach was proven to be effective and practical for isolating mutant animals to study gene function at the organismal level.
