가축 사료에서 유당비분해 그람음성균의 분리 및 분리균의 항생제 감수성 (Isolation of lactose non-fermenting Gram negative bacilli from animal feedstuffs and antibiotics susceptibility of isolates)

  • Total viable cells and lactose non-fermenting cells were counted from animal feedstuffs (n=65). And isolation of Gram negative lactose nonfermenting enterobacteria and antibiotics susceptibility of isolates were performed. 1. The ranges of total viable cells / lactose non-fermenters in animal feedstuffs from Korean cattle were counted as 9$\times$$10^4$-1$\times$$10^7$ / 1$\times$$10^2$-6$\times$$10^3$, milking cow as 1$\times$$10^4$-2$\times$$10^8$ / 2$\times$$10^2$-8$\times$$10^3$, pig as 1$\times$$10^4$-1$\times$$10^6$ / 2$\times$$10^2$-6$\times$$10^3$, and chicken as 7$\times$$10^4$-1$\times$$10^9$ / 4$\times$$10^2$-1$\times$$10^5$ cfu/g, respectively. 2. Among the 214 isolates from feedstufs, 87 from Chinan(n=23), 66 from Changsu (n=23) and 61 from Mooju(n=19) were isolated. Of these isolates, 60 from pigs (n: 19), 51 from milking cows(n=15), 45 from chikens(n=11) and 58 from Korean cattle(20) were isolated. 3. Among the 6 genuses of Gram negative lactose nonfermenting enterobacili, Salmonella sp, Y pseudotuberculosis, Ent agglomerans and Sal choleraesuis were frequently encountered. 4. A majority of isolates were sensitive to 19 antibiotics, singly or in combination. These isolates were completely susceptible to Cp, Gm, Imp and Pi, 93% to Ak and To, 73% to Cax and Ts, 66% to Cft and Tim, 46-53% to Caz, Cf and Cz, 33-40% to Am, Azt, Cfz and Ti, and 6% to Cfx, in order, but not susceptible to Crm. 5. Among the antibiotic resistant strains, a total of 23 resistant patterns was noted, and of these Crm 40(18.7%), Am Cf Cfx Cfz Crm Ti 27(12.6%), each of Azt Ctx Crm and Azt Cax Caz Cft Cfx Crm 22(10.3% ) were frequently encountered.

사료 첨가용 생균제를 위한 Probiotics 유산간균의 분리 및 동정 (Isolation and Identification of Probiotic Lactobacillus Isolates for Calf Meal Supplements)

  • 한우와 홀스타인의 분변으로부터 MRS 배지와 LAPT 배지를 이용하여 무작위 선발법으로 54균주의 유산균을 1차로 분리하였다. 1차로 분리된 54균주에 대해 내담즙성이 우수한 10균주를 분리한 다음 내산성을 조사한 결과 인공위액 pH 2.5에서 LS1, LS15 및 LL6 균주가 각각 66.5%, 82.6% 및 80.7%의 생존율을 나타내었다. Sal. typhimurium, Sta. aureus 및 Cl. perfringens의 병원균에 대해 가장 큰 항균력을 보인 균주는 LL6와 LL7이었다. API CHL kit로 동정한 결과 LS1, LS2 및 LMI 균주는 모두 L. fermentum, LL6와 LL7 균주는 L. acidophilus, LS3 균주는 L. plantarum으로 각각 동정되었고, 나머지 4균주는 Lactobacillus spp. 로 동정되어 분리된 10균주 모두 안전성 있는 유산간균임을 확인하였다. 10종류의 항생제에 대한 내성을 조사한 결과 ampicillin, amoxicillin과 erythromycin에 대해서는 감수성이 있으나 colistin과 ciprofloxacin에 대해 모두 내성을 나타내었다. LB1, LL6 및 LL7 균주는 gentamicin과 neomycin에 대해 내성을 보여 주었다. 분리 동정된 균주 중에 내산성, 내담즙성 및 병원성 균에 대한 특성을 기준으로 판단할 경우 probiotic 유산균으로 사용 가능성이 높은 것은 LL6인 L. acidophilus로 나타났다.

Prevention of Scours in Neonatal Kids after Oral Administration of an Organic Acid Solution or Antibiotics

  • The efficacy of an organic acid solution in the prevention of neonatal kid diarrhoea, was examined in this study and was compared with that of different antibiotics. In a goat farm, kids and their mothers were divided in 4 experimental groups. The kids of Group I were not given any treatment serving as negative controls. The kids of Group II were treated orally for 3 consecutive days with a solution of organic acids starting approximately 12 h after their birth. The kids of Groups III and IV were treated once orally with ampicillin and oxytetracycline respectively, approximately 12 h after birth. All groups were compared as regards the occurrence of diarrhoea, its duration, and the rate of mortality. The results have shown that the number of cases and the duration of diarrhoea were significantly reduced in all treated groups, when compared to the control group (p<0.05). However, no or little difference was noted with respect to morbidity, mortality and diarrhoea characteristics when treated groups were compared with each other (p>0.05). Samples of diarrhoeic faeces from kids in the control group resulted in the isolation of K88 and K99 strains of Escherichia coli. It was concluded that, early oral administration of organic acids can be effective in the prevention of scours in neonatal kids, possibly caused by enterotoxigenic E. coli strains, and to a degree of protection similar to that seen with the used antibiotic schemes.

High prevalence of Enterococcus spp. from dogs with otitis externa

  • Otitis externa (OE) is a frequent disease in the ear canals of dogs. To identify the pathogens causing OE in dogs and to determine their antimicrobial resistances, specimens were collected from animal hospitals in Daejeon. The isolates were examined by morphological and biochemical tests, 16S rRNA analysis and antimicrobial susceptibility tests. We analyzed correlation between the isolated pathogens and external factors of dogs such as breed, age, gender, ear mite, hair in ears and experience with antibiotic therapy. Thirty three strains of bacteria were isolated from 26 of the 68 heads of dogs with OE. The most isolated bacteria were Enterococcus faecalis (E. faecalis) followed by Staphylococcus aureus (Sta. aureus), Sta. pseudointermedius, E. faecium, E. avium and Streptococcus canis (Strep. canis) in order of frequency of occurrence. Isolation frequency of Enterococcus spp. and Staphylococcus spp. were 51.5% and 45.5%, respectively. E. faecalis and E. faecium isolates showed VanB phenotype, which is resistant to vancomycin but sensitive to teicoplanin were 58% and 25%, respectively. Nine isolates among total twelve isolates of E. faecalis were isolated from the dogs treated with antibiotics. There was no methicillin-resistant Sta. aureus (MRSA), but were MR-Sta. pseudointermedius (MRSP) (57.1%) and vancomycin-resistant (VR)-Sta. pseudointermedius (14.3%) (VRSP) showing VanB phenotype. However, vanA, vanB and vanC genes were not detected in VR isolates from the dogs. Taken together, VR-Enterococcus spp. (VRE) is one of the major pathogens in domestic animals, as well as community-and hospital-acquired infection.

Isolation, Characterization, and Comparative Genomics of the Novel Potential Probiotics from Canine Feces

  • Lactic acid bacteria (LAB) are commonly used as probiotics; however, not all LAB strains have the same beneficial effects. To successfully use LAB as probiotics in canines, LAB species should originate from the canine intestinal tract as they display host specificity. The objective of this study was to investigate the phenotypic and genomic traits of potential probiotic LAB isolated from canine fecal samples. Twenty LAB samples were evaluated for their potential probiotic characteristics including resistance to low pH, bile salts, hydrophobicity, auto-aggregation, co-aggregation, adhesion to epithelia or mucosa, and production of inhibitory compounds. Additionally, we evaluated their safety and other beneficial effects on canine health, such as DPPH free radical scavenging, and β-galactosidase. Four strains demonstrated potential probiotic characteristics and were selected: Enterococcus hirae Pom4, Limosilactobacillus fermentum Pom5, Pediococcus pentosaceus Chi8, and Ligilactobacillus animalis FB2. Safety evaluations showed that all strains lacked hemolytic activity, could not produce biogenic amines, and did not carry any pathogenic genes. In addition, L. fermentum Pom5 and P. pentosaceus Chi8 displayed susceptibility to all antibiotics and concordant with the absence of antibiotic resistance genes. Based on their phenotypic and genomic characteristics, L. fermentum Pom5 and P. pentosaceus Chi8 were identified as potential probiotic candidates for canines.

경남 북부지역 오리 분변에서 분리된 Campylobacter spp.의 항생제 내성 (Antimicrobial resistance of Campylobater spp. from duck feces in northern area of the Gyeongnam province, Korea)

  • The purpose of this study was to investigate prevalence and antimicrobial resistance patterns of Campylobacter spp. from duck feces in northern area of the Gyeongnam province, Korea. Samples of 121 duck feces were taken from April to December 2014 for this survey. Samples were examined by bacteria isolation and reverse transcriptase-polymerase chain reaction assay for detection of Campylobacter spp. Campylobacter were isolated in 37 samples (30.6%). Among these samples, C. jejuni and C. coli were isolated in 35 samples and 2 samples, respectively. Minimum inhibitory concentration (MIC) test is performed to investigate antimicrobial resistance patterns of Campylobacter spp. C. jejuni were resistant to ciprofloxacin (85.7%), nalidixic acid(82.9%), tetracycline (77.1%), gentamicin (57.1%), azithromycin (40.0%), clindamycin (34.3%), erythromycin (22.9%), and florfenicol (8.6%). These data support a database of pollution and antimicrobial resistance of Campylobacter spp. from duck feces and provide a basic information of reducing the secondary damage of antibiotic misuse.

Epidemiological Investigation and Antibiotic Sensitivity of Salmonellosis in Goats at the Selected Areas of Bangladesh

  • Salmonellosis is one of the life-threating diseases of goat in Bangladesh. Therefore, the present study was designed to study the prevalence of Salmonellosis, and isolation and characterizations of the Salmonella spp. from apparently healthy and diarrheic goat. A total of 47 faces samples were collected from selected place and cultured onto different prescribed medium to isolate it. In this study, 12.76% (6/47) samples were found to be positive for Salmonella spp. During culture on SS agar medium, all of the Salmonella isolates produced round, smooth, opaque, translucent and black color colonies on SS agar media. All of the isolated Salmonella spp. fermented dextrose, maltose and mannitol with production of acid and gas but did not ferment sucrose and lactose. However, these isolates had showed Indole and Voges-Proskauer test negative, Methyl-Red test positive. All of these isolates were subjected to rapid plate agglutination test with polyvalent "O" (Poly 'O') and polyvalent "H" (poly 'H') antisera where positive agglutination were observed. They were highly sensitive to ciprofloxacin, spiramycin and gentamycin; moderately sensitive to oxytetracyline, streptomycin and amoxicillin; less sensitive to sulphamethoxazole and resistant to penicillin-G. Based on the present findings, it may be concluded that the investigated Salmonella spp. from goats might be S. typhimurium, S. enteritidis, S. brandenburg, S. salford, S. newbrunswick, S. newport or S. dublin. Further study will be needed, therefore it requires further characterization using other serological and molecular techniques.

동물원(動物園) 야생동물(野生動物)에서 분리(分離)한 Yersinia enterocolitica 의 생물형(生物型), 혈청형(血淸型) 및 항생제(抗生劑) 감수성(感受性) (Biotype, serotype and antibiotics susceptibility of Yersinia enterocolitica isolated from zoo animals)

  • A study on the isolation of Yersiniae from the feces of wild animals(mammals 376, birds 19 and reptiles 13) in zoo and the biotype and serotype and susceptibility of 12 antibiotics was carried. Out of 408 animals, Yersiniae were isolated from 28 animals(6.9%). Of 28 isolates, 27 isolates(96.4%) were Y. enterocolitica and 1(3.6%) was Y. kristensenii. According to the species, 25(6.6%) of Y. enterocolitica and 1(0.3%) of Y. kristensenii were isolated from 376 mammals, 2(15.4%) of Y. enterocolitica from 13 reptiles but not isolated from 19 birds. According to the eating pattern, 8(5.2%) of Y. enterocolitica were isolated from 155 carnivora, 13(10%) of Y. enterocolitica from 123 herbivora, and 6(4.9%) of Y. enterocolitica and 1(0.8%) of Y. enterocolitica from 123 omnivora. Out of 27 isolates of Y. enterocolitica, all were biotype 1. And predominant serotype was 0:21(40.7%), and 0:5(37.0%), 0:6(11.1%), 0:1(3.7%), 0:9(3.7%) and untypable(3.7%). Yersiniae isolated from zoo animals were resistant to cephalothin(100%), ampicillin(96.4%), carbenicillin(96.4%) and tetracycline(14.3%) and streptomycin(3.6%) and susceptible to chloramphenicol(100%), colistin(100%), gentamicin(100%), kanamycin(100%), nalidixic acid(100%), polymyxin B(100%) and tobramycin(100%). The predominant multiple resistance pattern was Am-Cf-Cb(82.1%), and Am-Cf-Cb-Te(10.7%) and Am-Cf-Cb-Te-Sm(3.7%).

Isolation, In vitro Antibacterial Activity, Bacterial Sensitivity and Plasmid Profile of Lactobacilli

  • The present research work was conducted to evaluate the beneficial effects as well as the safety aspects of lactobacilli as probiotic. Lactobacilli were isolated from poultry faecal samples, feed samples and from some known preparations procured from poultry feed manufacturers. L. acidophilus and L. sporogenes were tested for the antibacterial activity against four poultry pathogens viz. Escherichia coli, Salmonella spp., Proteus spp. and Pseudomonas aeruginosa. Cell free supernatant (CFS) of L. acidophilus exhibited significantly higher antibacterial activity against Salmonella spp. at original pH (4.50${\pm}$0.02). At the adjusted pH (6.50${\pm}$0.02) significantly higher antibacterial activity was recorded against indicator organism except for P. aeruginosa. Likewise, L. sporogenes exhibited similar antibacterial activity at original as well as adjusted pH except for E. coli. Antibacterial activity against E. coli was significantly higher at adjusted pH than at original pH of CFS. The competitive exclusion of E. coli by lactobacilli over the intestinal epithelial cells (IEC) was checked. L. acidophilus strain I, which was of poultry origin, exhibited maximum attachment over IEC as compared to other three strains of non-poultry origin viz. L. acidophilus strain II, L. sporogenes strain I and II. Overall, L. acidophilus exhibited higher competitive exclusion as compared to L. sporogenes. All the lactobacilli of poultry origin were most sensitive to penicillin G, amoxycillin, ampicillin and chloramphenicol, least sensitive to sulphamethizole, ciprofloxacin, neomycin, norfloxacin and pefloxacin and resistant to metronidazole and nalidixic acid. The isolates from probiotic preparations were most sensitive to ampicillin, amoxycillin and tetracycline, least sensitive to sulphamethizole, norfloxacin, neomycin and ceftriazone and resistant to nalidixic acid and metronidazole. Eight of the multiple drug resistant lactobacilli isolates were studied for the presence of plasmids. Plasmids could be extracted from six isolates of lactobacilli. These plasmids could be responsible for bacteriocin production or for antibiotic resistance of the strains. The lactobacilli need further studies regarding their safety for use in the probiotic preparations.

Isolation and Characterization of Bacteriophages Against Pseudomonas syringae pv. actinidiae Causing Bacterial Canker Disease in Kiwifruit

  • Pseudomonas syringae pv. actinidiae causes bacterial canker disease in kiwifruit. Owing to the prohibition of agricultural antibiotic use in major kiwifruit-cultivating countries, alternative methods need to be developed to manage this disease. Bacteriophages are viruses that specifically infect target bacteria and have recently been reconsidered as potential biological control agents for bacterial pathogens owing to their specificity in terms of host range. In this study, we isolated bacteriophages against P. syringae pv. actinidiae from soils collected from kiwifruit orchards in Korea and selected seven bacteriophages for further characterization based on restriction enzyme digestion patterns of genomic DNA. Among the studied bacteriophages, two belong to the Myoviridae family and three belong to the Podoviridae family, based on morphology observed by transmission electron microscopy. The host range of the selected bacteriophages was confirmed using 18 strains of P. syringae pv. actinidiae, including the Psa2 and Psa3 groups, and some were also effective against other P. syringae pathovars. Lytic activity of the selected bacteriophages was sustained in vitro until 80 h, and their activity remained stable up to 50℃, at pH 11, and under UV-B light. These results indicate that the isolated bacteriophages are specific to P. syringae species and are resistant to various environmental factors, implying their potential use in control of bacterial canker disease in kiwifruits.
