Chinae Rhizoma is dried crude roots and rhizomes obtained from Smilax china Linne which belongs to Smilax Genus of Liliaceae Familia, and it has been in wife use as a folk remedy for a long time. But very little systemic studies on its pharmacological actions have been made until now. Furthermore, the effective components except saponin of Chinae Rhizoma have been not known yet. It came to the attention of the authors that Chinae Rhizoma are used as antidiarrhetics in Chinese herb medicine, therefore they attempted to observe the effects of Chinae Rhizoma Alcoholic Extracts (Chinae Rhizoma Ethanol Extract=CREE, Chinae Rhizoma Methanol Extract=CRME) from Smilax china Linne on motility of the isolated rabbit duodenum in this experiment. The results of the study were as follow: 1. The movement of the isolated rabbit duodenum was slowed down and its tonus was relaxed in proportion to the concentration of Chinae Rhizoma Alcoholic Extracts (CREE & CRME). 2. With a concentration of CREE $10^{-4}g/ml$, the contractile effects of acetylcholine $10^{-6}g/ml$, pilocarpine $10^{-5}g/ml$ and physostigmine $10^{-5}g/ml$ were remarkably blocked in the movement of isolated rabbit duodenum. 3. The concentration of the isolated rabbit duodenum induced by barium chloride $10^{-4}g/ml$ was significantly inhibited by CREE $10^{-4}g/ml$. 4. With the administrations of histamine $10^{-5}g/ml$, serotonin $10^{-6}g/ml$ and prostaglandin $A_2\;10^{-6}g/ml$ respectively, contractions were observed in the rabbit duodenum. But, treatment of CREE $10^{-4}g/ml$ inhibited markedly the tonus and motility of the duodenum pretreated with the above drugs. 5. The effects of CREE and CRME were very much alike in this experiment. From the above results, it is concluded that the mechanism of action of Chinae Rhizoma on motility of isolated rabbit duodenum was partially due to cholinergic blocking effect.
In an attempt to better understand the effect of whole body irradiation on the spontaneous motility and oxygen consumption rate of the isolated mouse duodenum, a whole body X-irradiation of 1,000r. was given to albino mouse, and 1) the total length of contraction of isolated duodenum was recorded on kymograph every five minutes for 60 minutes, 2) glucose and 5-hydroxytryptamine(5-HT) were added to the reaction medium of Kreb's-Ringer-bicarbonate buffer(KRB) and response of the isolated duodenum to the drugs was observed, and 3) the oxygen consumption rate $(QO_2)$ of the isolated duodenum as well as the effect of glucose and 5-HT on $QO_2$ were measured by Warburg's standard manometric method and the comparison was made with the control(i.e. normal) group. The results thus obtained are summarized as follows. 1. The spontaneous motility of the isolated duodenum in the irradiated groups showed a significantly elevated pattern for the first 15 minutes comparing with the control. The motility, however, decreased after 15 minutes and remained so in the irradiated groups to the level of the nonirradiated control, but 24 hours post-irradiation group showed a tendency of an increased motility while one hour post-irradiation group showed no difference comparing with the control. 2. Addition of glucose produced generally elevated motility of the isolated duodenum in both irradiated and non-irradiated groups comparing with the control throughout the experiment, but no difference was observed in contractile amplitude between the irradiated and non·irradiated groups. 3. When 5-HT was added to the irradiated group, the contractile amplitude of isolated duodenum was similar to that of the control, and 5-HT alone caused a slight increase of the motility comparing with the control. 4. The oxygen consumption rate $(QO_2)$ of the isolated duodenum was found to be ,slightly increased in one hour post·irradiated group, but similar in 24 hour post·irradiated group comparing with the control. Glucose produced a significant increase of $QO_2$ in all the groups, but 5-HT produced a tendency of decrease of $QO_2$ in all the groups.

The total length of contraction of the isolated duodenum was recorded on kymograph every five minutes for 60 minutes, and the comparison was made with the control (i.e., normal). The results thus obtained are summarized as follows: 1. The spontaneous motility of the isolated duodenum in the X·irradiated groups showed a significantly elevated pattern for the first 15 minutes comparing with the control. The motility, however, decreased after 15 minutes and remained so in the X-irradiated groups to the level of the non-irradiated control, but 12 hours post-irradiation group showed a significantly increased motility throughout the experiment comparing with the control. 2. When GSH was injected intraperitoneally Prior to the whole body X·irradiation with 1,000r, the spontaneous motility of the isolated duodenum of mouse showed a significantly decreased pattern for the first 10 and 15 minutes comparing with the X·irradiated group followed by the similar motility thereafter comparing with the control and X-irradiated groups. 3. The above results suggest that GSH is effective as a radioprotector in terms of the motility of the isolated mouse duodenum.
Diospyros Kaki Thunberg is the species of persimmon tree that grows in Korea. Although its fresh or dried fruits are often served as a desert, it has little been known if persimmon tree has any specific pharmacological action. The leaves and branches of persimmon tree has long been used as folk remedies for palsy and frostbite in the southern part of Korea and it is also in use for the treatment of hiccup and nocturnal enuresis in chinese herbal medicine. Recently it was reported that an intravenous administration of Diospyros Kaki Thunberg ethanol extract (KTEE) into the animals lowered arterial blood pressure. Lee concluded from his study on the mechanism of depressor action of KTEE that at least a part of depressor response he observed was caused by acetylcholine-like action of KTEE. On the other hand little study has been made on the effect of KTEE on the motility of isolated animal intestines. Therefore the present study was undertaken to investigate effect of KTEE and the mechanism of its action on the motility of isolated rabbit duodenum. Ethanol extract of Diospyros Kaki Thunberg was prepared by boiling 1 kg of dried branches of persimmon tree in 1 liter of ethanol and the motility of isolated rabbit duodenum was recorded on physiograph by means of force transducer connected with Magnus apparatus. Doses of KTEE used were $5{\times}10^{-4}gm/ml,\;1{\times}10^{-3}gm/ml,\;and\;2{\times}10^{-3}gm/ml$. And the isolated duodenum was separately pretreated with acetylcholine $(5{\times}10^{-7}gm/ml)$, pilocarpine $(2.5{\times}10^{-6}gm/ml)$, histamine $(5{\times}10^{-6}gm/ml)$ and barium chloride $(2.5{\times}10^{-5}gm/ml)$ in order to find out interactions of these drugs with KTEE. The results obtained are as follows: 1. At doses of $5{\times}10^{-4}gm/ml,\;1{\times}10^{-3}gm/ml$ KTEE reduced contractions of isolated duodenum, while tonus as well as contaction of duodenum were depressed with $2{\times}10^{-3}gm/ml$ of KTEE. 2. Since the inhibitory effect of KTEE on the intestinal motility was not blocked by pretreatment with acetylcholine, pilocarpine, and barium chloride, it was strongly suggested that the inhibitory action of KTEE on intestinal motility is mainly Caused by its antihistamine effect. 3. It is also concluded that the principal substance of KTEE responsible for inhibition of intestinal motility may also have a vasodilating activity and would not be an acetylcholine-like substance in case it is same substance as that cause depressor responses.
In 1944 Cavallito and Bailey first extracted an essential oil, a powerful antibacterial principle, from the garlic and named it allicin. Later Stoll and Seeback elucidated that allicin was produced from alliin by the enzymatic action of arinase. Damaru observed the depressor responses following intraperitoneal administration of garlic juice in cats. And Thiersch presented evidence that garlic had a protective action against experimental arterosclerosis in cholesterol-fed animals. On the other hand it was also reported that anemias were caused by long-term ingestion of garlic as a result of reduction in hemoglobin and RBC. From the experiment in which the effect of garlic on the blood sugar level was studied, Lee insisted garlic elevated blood sugar level. However, August and Jain claimed that hypoglycemia was induced by garlic administration. Recently Bordia and Bansal suggested that essential oils extracted from onion and garlic have a strong preventive effect on hyperlipemia and prolonged coagulation time resulted from fat-feeding. Furthermore Bordia et al indicated that garlic exerted a strong fibrinolytic activity. In early 1920 s Sugihara reported that essential oil of garlic not only decreased arterial blood pressure but also had a paralytic effect on the isolated heart and intestinal strip of animals. The present study was proposed to investigate the effect of garlic juice and the mechanism of its action on the motility of the isolated rabbit duodenum. The motility of the isolated duodenum was recorded on polygraph by means of force transducer connected with Magnus apparatus. And the isolated duodenum was separtely pretreated with $acetylcholine(5{\times}10^{-7}\;gm/ml)$, $pilocarpine(2.5{\times}10^{-6}\;gm/ml)$, $histamine(5{\times}10^{-6}\;gm/ml)$ and barium $chloride(2.5{\times}10^{-5}\;gm/ml)$ in order to find out interations of these drugs with ASJ. The results obtained were as follows; At concentrations of 0.25%, 0.5% and 1.0% ASJ markedly inhibited contractions of isolated duodenum while tonus as well as contractility of the isolated intestine were decreased also with 0.5% and 1.0% ASJ. Since ASJ markedly abolished augmented motility of isolated intestine by histamine and partly reduced that by $BaCl_2$, it is strongly suggested that inhibitory action of ASJ on the intestinal motility is caused mainly by its antihistamine effect and partly by its direct action on the intestinal smooth muscle.
It has long teen known in herbal medicine that the mugwort not only exerts a strong hemostatic and parasiticidal actions but also has therapeutic effects for stomachache and diarrhea. In recent pharmaceutical botany the mugwort is known to have antipyretic and astringent actions also. Among the major principles which have been found in the leaves and stems of mugwort are inulin, alkaloids, vitamines, and esestial oil. It is well known that santonin, one of the well known parasiticides, is one of the glucosides extracted from the limited species of mugwort. The present study was undertaken to investigate effects of mugwort(Artemisia asiatica Nakai) on the motility of isolated rabbit duodenum. The results obtained are as follows: At does of 0.2%, 0.5% and 1. 0% AAJ(Artemisia asiatica juice) markedly enhanced contractility of isolated duodenum and tonus of the intestine was also augmented with doses of 0.5% and 1.0%. The augmentative effect of AAJ on intestinal motility was not affected by pretreatment with epinephrine and avil while it was completely abolished by atropine. Therefore it is strongly suggested that augmentative action of AAJ on duodenal motility was exerted solely through muscarinic cholinergic receptors.
Helicobacter species have been identified in or isolated from domestic carnivores, but their prevalence in different population of animals and their clinical significance are still unknown. This study was peformed to evaluate the prevalence of Helicobacter in clinically healthy dogs by urease test, culture, morphological examination and polymerase chain reaction (PCR) technique. Tissue samples from 70 dogs in Kangwon and Kyunggi areas from August 1998 to April, 1999, were examined. The detection rates of Helicobacter by urease activity of tissue-samples were 84.6%, 61.3% and 4.8 % in the fundus, the antrum and the duodenum, respectively. One strain of Helicobacter was isolated from the duodenum. It was identified as H canis by biochemical and morphorogical examination. The detection rates of Helicobacter by histological examination were 92.3%, 79.0% and 4.8% in the fundus, antrum and the duodenum, respectively. Helicobacter organisms were colonized more in the gastric pits than in the surface of epithelium, the gastric gland or the parietal cell. Although most of dogs were colonized with Helicobacter in tissue, gross lesions and specific histopathological lesions caused by Helicobacter in these tissues were not observed. The detection rate of Helicobacter by PCR was 78.6%. The histological examination was more sensitive than urease test, culture or PCR technique for the detection of Helicobacter.
Although the 'Insamyangwee-tang' has been widely used in clinical purposes in the oriental medicine, its clinical efficacy is only documentated for the cases of gastritis, gastric ulcer and enteritis, but the experimental study on these has not been undertaken. So, to investigate the clinical efficacy of 'Insamyangwee-tang' comparing with animal experiments, This study was carried out. The results showed that relaxing action was shown on the isolated ileum in mice and that strong antagonistic actions were seen on $BaCl_2$, acetylcholine and histamine induced contraction of the ileum in mice, rats, rabbits and guinea-pigs that the relaxing effect of the intestinal smooth muscle was recognized. Inhibitory effects on transport rate in the small intestine of mice. Strong antagonistic actions were seen on acetylcholine induced contraction of duodenum in rats and remarkably inhibiting actions were seen of duodenum in rats. Inhibitory action on the secretion of gastric juice and pepsin, anti-ulceration effect was recognized.
Duodenal injury following blunt abdominal trauma is a relatively unusual complication, and it may sometimes be difficult to distinguish a duodenal hematoma from duodenal perforation. According to recent reports, intramural hematomas typically resolve spontaneously with conservative treatment. Surgery, however, is occasionally necessary in some cases if the diagnosis is delayed, conservative therapy fails, or a high degree of suspicion of duodenal injury persists. We experienced a case of delayed manifestation of a duodenal intramural hematoma that was surgically treated.
The effects of erythrosine on motility of frog heart, rabbit duodenum and uterus isolated, and on mice intestinal motility and voluntary activity were investigated. The effect of erythrosine $2.3{\times}10^{-5}M$ on isolated frog heart showed a slight decrease of the amplitude of motility, and the heart motility stopped in $3.5{\times}10^{-4}M$. With the administration of erythrosine $3.4{\times}10^{-4}M$, the isolated rabbit duodenum showed a remarkable contraction and this effect was inhibited by atropine $1.4{\times}10^{-7}M$. The administration of erythrosine $2.3{\times}10^{-3}M$, produced a contractile effect on the isolated rabbit uterus, and the motility of $6.9{\times}10^{-3}M$ started to increase in contractions at first and finally stopped, keeping in continuous contractions. The effects of erythrosine 0.5, 1.0, 10, and 20mg/kg on mice intestinal motility were not significantly different from this of the normal control. With 20 and 40mg/kg of erythrosine, the effects on voluntary activity showed the decrease of 21 and 58% respectively, and voluntary activity of the mice pretreated with erythrosine 20 and 40mg/kg, induced by C. N. B. 30mg/kg showed the decrease of 57 and 78% respectively in contrast with the normal control group.
