Enhanced Antitumorigenicity and Antimutagenicity of Doenjang Prepared from Mushroom Mycelia-cultured Traditional Mejus (재래식 버섯균사체 된장의 항종양성과 항돌연변이성)

  • Antitumorigenic and antimutagenic activities of the doenjangs prepared from mushroom mycelia-cultured traditional mejus (designated to MTDJ) were investigated using the model of Sarcoma-180-induced mouse ascites cancer, and 2-amino-3-methylimidazo [4,5-f] quinoline (IQ) and aflatoxin B$_1$ (AFB$_1$) -mediated S. typhimurium mutagenicity, respectively. Antioxidative activity of MTDJ was also investigated using the mouse liver microsome system. Mushroom stains used for the preparation of the mushroom mycelia-cultured traditional mejus were Synryeong (Agaricus blazei), Yeonggi (Canoderma Iucidum), Sanghwang (Phellinus linteus), and Neutari (Pleurotus ostreatus). All MTDJS showed the enhanced antitumorigenicities (12% by Synryeong, 13% by Sanghwang, 16% by Yeonggi, and 19% by Neutari), antimutagenicity (6.1~20.8% for IQ and 3.1~10.2% for AFB$_1$), and antioxidative activity (6.6~46.5%), relative to the control doenjang. The $\beta$-D-glucan content (0.75~1.71 mg/g) of MTDJs was 3~8 times higher than that (0.22 mg/g) of the control doenjang. Genistein content (769~932 Ug/g) of MTDJS was also higher than that (728 Ug/g) of control doenjang The content of $\beta$-D-glucan and genistein was not exactly correlated to the antitumorigenicity and antimutagenicity of MTDJs. These results indicate that anti-tumorigenicity and antimutagenicity of MTDJS were elevated in comparison with the control doenjang, and the observed functions were, in part, derived from $\beta$-glucan and/or genistein in the MTDJS.

Soybean Ecological Response and Seed Quality According to Altitude and Seeding Dates

  • This experiment was carried out to examine ecological response and soybean quality as affected by environmental cultivation for producing high seed quality in domestic soybean variety. The results are as follows: Under equal cumulative temperature condition, soybean plants grown in Muju showed longer days to flowering, which was an effect of the long day-length on high latitudes, and longer duration of reproductive stage as a result of low temperature within that period. Considering apparent seed quality, 100 seed weight of soybeans grown in Muju was heavier than Miryang. Ratio of seed crack and disease-damaged seeds was lower in Muju, and these parameters decreases as planting was delayed. The protein contents did not show significant difference in terms of altitude and planting date, however, crude oil contents were higher in Miryang. An opposite trend was observed in C18:1 and C18:3. In the fatty acid composition, the proportion of C18:1 decreased as seeding date was delayed, and was higher in Miryang. Opposite observations were obtained from C18:3. The anthocyanin contents were highest on June 10 planting and higher in Muju than in Miryang. Isoflavone content was higher as seeding date was delayed and is similar accross seeding dates in Muju. As a summary, for high seed quality production the optimum planting date was June 10, and Muju was more suitable region than Miryang.

  • To accept basic data of utilizing of cheongtae as a raw material of new food and industrial products, major chemical components were investigated. As a results of mineral analysis, the content of P (310.52mg/100 g) was much higher. than those of K (48.65 mg/100 g), Ca (40.93 mg/100 g), Mg (27.87 mg/100 g), Na(14.36 mg/100 g), Fe (6.31 mg/100 g), Zn (4.40 mg/100 g) and Cu(1.27 mg/100 g). The main components of free sugar in cheongtae were sucrose and glucose. The main fatty acids of total lipid were linoleic acid and oleic acid, which comprised over 75$\%$ of total fatty acids. The daidzein and genistein levels in cheongae were 40.95 mg/100g, 38.02 mg/100 g, respectively.

Fermented soybeans by Rhizopus oligosporus reduce femoral bone loss in ovariectomized rats

  • BACKGROUND/OBJECTIVES: Soy isoflavones are structurally similar to estrogen and bind to estrogen receptors, suggesting that they exhibit estrogenic activities; therefore, they are referred to as phytoestrogens. Fermentation may affect the bioavailability of isoflavones altering soy isoflavone glycosides in the form of aglycones. Thus, this study investigated the effects of fermented soybeans by Rhizopus oligosporus on bone metabolism in both young rats as a pilot test and in ovariectomized (ovx) old rats as a model of menopause. MATERIALS/METHODS: In the pilot test, a total of 24 seven-week-old female Sprague-Dawley (SD) rats were fed one of three diets for a period of four weeks: casein, unfermented soybean product, or fermented soybean product by R. oligosporus. In the ovx rat model, 20-week-old SD rats weighing 260-290 g underwent either sham-operation (n = 10) or bilateral ovariectomy (n = 30) and were then fed the AIN-93M diet for one week. Thereafter, rats were fed sham-casein, ovx-casein, ovx-soybean, or ovx-fermented soybean diet for five weeks. After decapitation, femoral bones were isolated and preserved in 9% formalin for assessment of bone mineral density (BMD), bone mineral content (BMC), and bone-breaking strength (BBS). RESULTS: Ovx rats showed significantly increased weight gain and decreased uterine wet weight. Of particular interest, ovx rats fed fermented soybeans showed increased uterine wet weights compared to control rats. Fermented soybean diet caused a significant increase in plasma 17-${\beta}$ estradiol concentrations in young rats, and 17-${\beta}$ estradiol levels were enhanced in ovx rats to match those of sham-operated ones. Significantly lower femoral BMD and BMC were observed in ovx rats compared to sham-operated controls, whereas bone areas did not differ statistically among the groups. In addition, BBS tended to be increased in ovx rats fed soybeans and fermented soybeans. CONCLUSIONS: Supplementation of fermented soybeans could have preventive and therapeutic effects against osteoporosis in postmenopausal women.

Characterization of a Novel Necrotic Response of Glycine max Line 'PI96188' to Xanthomonas axonopodis pv. glycines

  • Typical susceptible symptoms of the bacterial pustule disease caused by Xanthomonas axonopodis pv. glycines are pustule and chlorotic haloes that usually occur in leaves of Glycine max plants. The soybean genotype 'PI96188' showed an atypical response to all tested races X. axonopodis pv. glycines, accompanied with necrosis without chlorotic haloes on the underside of the necrotic symptoms. X. axonopodis pv. glycines 8ra grew to levels from 10 to 100 fold lower on PI96188 than on susceptible cultivar Jinjoo1, but 10-fold higher than on the resistant cultivar CNS. The chlorophyll content in PI96188 leaves remained unchanged until 12 days after bacterial infection. Ultrastructural observation showed that the infected leaf cells of PI96188 had intact normal chloroplasts compared to those of the susceptible cultivar Jinjoo1. Chloroplast degradation or the absence of chloroplasts in cells of the infected tissues was observed in Jinjoo1. Senescence-related ACS7 gene was significantly induced in PI96188 compared to those in Jinjoo1 at 2 days after inoculation. While photosynthesis-related rbcS gene showed the dramatic change in Jinjoo1, this gene was constitutively expressed in PI96188. However, expression of the defense-related genes, such as peroxidase and isoflavone synthase in the infected PI96188 leaves was similar to that in Jinjoo1. Together, these results suggest that the novel necrotic symptom in PI96188 is a kind of resistant response different from a typical hypersensitive response in the resistant genotypes.

Metabolite Profiling and Microbial Community of Traditional Meju Show Primary and Secondary Metabolite Differences Correlated with Antioxidant Activities

  • Meju, a type of fermented soybean paste, is used as a starter in the preparation of various Korean traditional soybean-based foods. In this study, we performed Illumina-MiSeq paired-end sequencing for microbial communities and mass spectrometry analysis for metabolite profiling to investigate the differences between 11 traditional meju products from different regions across Korea. Even though the bacterial and fungal communities showed remarkable variety, major genera including Bacillus, Enterococcus, Variovorax, Pediococcus, Weissella, and Aspergillus were detected in every sample of meju. The metabolite profile patterns of the 11 samples were clustered into two main groups: group I (M1-5) and group II (M6-11). The metabolite analysis indicated a relatively higher amino acid content in group I, while group II exhibited higher isoflavone, soyasaponin, and lysophospholipid contents. The bioactivity analysis proved that the ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)) radical-scavenging activity was higher in group II and the FRAP (ferric reducing antioxidant power) activity was higher in group I. The correlation analysis revealed that the ABTS activity was isoflavonoid, lipid, and soyasaponin related, whereas the FRAP activity was amino acid and flavonoid related. These results suggest that the antioxidant activities of meju are critically influenced by the microbiome and metabolite dynamics.

In vivo and In vitro hair growth promotion effects of extract from Glycine soja Siebold et Zucc

  • Hair is a dermal adjunctive organ that protects the body from external physical and chemical stimuli; hair undergoes anagen, catagen, and telogen phases, with hair-loss occurring during the telogen phase. Alopecia is a condition wherein a person undergoes hair-loss far exceeding the normal amount, owing to diverse external factors. Wild beans are rich in isoflavone and amino acids known to prevent hair-loss; compared to cultivated beans, many wild bean species have higher protein content. This study aimed to develop a hair growth promoting solution, with superior hair growth promoting effects and fewer side effects, using naturally obtained Glycine soja Siebold et Zucc (GSSZ) extracts. Seven-week-old C57BL/6N male mice were classified into different experimental groups. Hair growth was observed in GSSZ-treated mice, and compared against that seen in 3 % minoxidil (MXD, positive control)-treated mice. Visual observations revealed a greater reduction in hair-loss in MXD and GSSZ application groups, compared to that in TXN group (hair loss induction using 1 % testosterone). Evaluation using an image analysis software revealed that compared to the positive control, TXN + GSSZ group showed the highest hair growth. TXN + MXD and control groups exhibited similar follicular cell growth, while the hair growth promotion patterns were similar in the negative control (normal), TXN + GSSZ, and TXN groups, as observed via histological analysis. GSSZ did not induce cytotoxicity (even at 2 mg/mL) in keratinocytes and dermal papilla cells; alternately, dermal papilla cell proliferation was activated in a (GSSZ) concentration-dependent manner. Therefore, the GSSZ extract promoted hair growth and increased hair growth-related cell activity, and could therefore be utilized in alopecia treatment.

Preparation and Characterization of Wet Noodle Containing Germinated Small Black Bean Flour (발아약콩가루를 첨가한 생면의 제조 및 특성연구)

  • Small black beans (SBB) were germinated up to 72 h at 23$^{\circ}C$, and 48 h germinated SBBs having the highest isoflavone content were chosen for noodle preparation. Some of the germinated SBB were treated by 20 min boiling (B) but some were not boiled (NB). For noodles, the germinated SBB flour replaced 15% or 20% of wheat flour, so five kinds of noodles were prepared and tested: control, 15B, 15NB, 20B, and 20NB. Higher amount of bean flour resulted in significant decrease of $L^*$ and increase of $b^*$ values in noodle. After 20 min cooking, the noodles containing germinated SBB flour showed less soluble sugar content in cooked water than control, and the lowest value was observed in 15B. For textural properties, the addition of SBB flour lowered hardness and adhesiveness of noodle, but increased chewiness than control. In sensory tests, the most acceptable sample was 15B because of developed texture and better appearance which seem to be good for health.

Effects of Oral Intake of Kimchi-Derived Lactobacillus plantarum K8 Lysates on Skin Moisturizing

  • Skin is the soft outer covering of vertebrates that provides protection from pathogenic infection, physical damage, or UV irradiation, and controls body temperature and water content. In this study, we examined the effects of oral intake of kimchi-derived Lactobacillus plantarum K8 lysates on skin moisturizing. In an in vitro study, we observed that the hyaluronic acid content increased in HaCaT cells treated with L. plantarum K8 lysates. Oral administration of L. plantarum K8 lysates effectively attenuated the horny layer formation and decreased epidermal thickening in DNCB-treated SKH-1 hairless mice skin. The damage to barrier function was reduced after 8 weeks of oral administration of L. plantarum K8 lysates as compared with that in the atopic dermatitis mice. For the test with volunteers, we manufactured experimental candy containing 2.1% L. plantarum K8 lysates, while control candy did not contain bacterial lysate. A significant increase in hydration in the experimental candy-administered group as compared with the control candy-administered group was observed on the face after 4 and 8 weeks, and on the forearm after 4 weeks. Decreases in horny layer thickness and TEWL value were observed on the face and forearm of the experimental group. Together, the in vitro cell line and in vivo mouse studies revealed that L. plantarum K8 lysates have a moisturizing effect. A clinical research study with healthy volunteers also showed an improvement in barrier repair and function when volunteers took L. plantarum K8 lysates-containing candy. Thus, our results suggest that L. plantarum K8 lysates may help to improve skin barrier function.

Comparison of Biological Activity Between Soybean Pastes Adding Sword Bean and General Soybean Pastes (작두콩 첨가 된장과 일반 된장의 생리활성 특성 비교)

  • This study was carried out to analyse the biological activity in soybean pastes with the sword beans and general soybean pastes. The electron donating activity of soybean pastes with the sword beans(J2, J4) was higher than other soybean pastes with value of $81.5{\pm}2.2%$ and $88.3{\pm}0.7%$, respectively. And the content of total phenol compounds was higher in J2 and J4 compared to the others with value of $1773.8{\pm}2.9\;mg/kg$ and $1785.7{\pm}3.9\;mg/kg$ respectively. ACE inhibitory effect was higher in J2 and J4 than other soybean pastes with value of $63.1{\pm}1.0%$, $62.1{\pm}1.4%$, respectively. And aglycone type(daidzein, genistein) isoflavone content was higher than glucoside type(daidzin, genistin). But tyrosinase inhibitory effect was not different among test samples. From the results, soybean pastes with the sword beans(J2, J4) have higher anti-oxidant activity and ACE inhibitory effect than other soybean pastes, therefore, it may have potential to prevent hypertension.
