개똥쑥의 천연화학물질에 의한 항균효과와 성분확인 (The Antifungal Activity of Chemical Substances from Artemisia annua)

  • 개똥쑥(Artemisia annua)에 함유되어 있는 천연화학물질이 곰팡이에 미치는 항균효과를 조사하고 천연화학물질의 성분을 분석하였다. 개똥쑥의 수용성물질과 휘발성물질로 인한 곰팡이의 생장실험은 곰팡이의 종류에 따라 생장률이 다르게 나타났고 수용성물질과 휘발성물질의 농도가 증가 됨에 따라 곰팡이의 생장률이 억제되었다. 특히 Fusarium oxysporum의 경우 휘발성물질보다 수용성물질에서의 생장 억제가 높았고 반면, Aspergillus nidulans는 수용성물질보다 휘발성물질에서 생장이 더 억제되었다. 따라서 이러한 모든 억제작용을 일으키는 화학물질을 찾기 위해 GC/MS를 사용하여 개똥쑥의 정유를 분석한 결과 2,4-hexadienal, β-pinene, cineole, thujone, camphor, citronellal, (-)-menthone, terpineol, (1R)-(-)myrtenol, mytenol, (S)-(-)-perillaaldehyde, perillalcohol, 4-tert-buthylaniline, eugenol, isosafrole, isoeugenol, α-humulene 등이 동정되었다. 이와 같은 결과로 볼때, 개똥쑥에 함유된 수용성 물질과 휘발성 물질이 실험에 쓰인 곰팡이에 대한 항균효과에 관계된다는 사실을 밝혀내었다.

The Effect of Roasting Temperature on the Formation of Volatile Compounds in Chinese-Style Pork Jerky

  • The purpose of this work was to study the effect of roasting temperature on the production of volatile compounds in Chinese-style pork jerky. The pork jerky was roasted by far-infrared grill at $150^{\circ}C$ or $200^{\circ}C$ for 5 min. The analysis of volatile compounds using a Likens-Nickerson apparatus coupled to a gas chromatograph and a mass spectrometer enabled us to identify 21 volatile compounds. The results showed that the volatile compounds coming from pork jerky can be divided into two groups in accordance with their possible origins. The first group of volatile compounds derived from oxidation of lipid included hexanal, ethylbenzene, nonanal, benzaldehyde, 2,4-decadienal, 1-octen-3-ol, octadecanal, and 9-octadecenal. The second group of volatile compounds generated from degradation of natural spices included 1,8-cinene, 4-terpineol, ${\alpha}$-terpineol, e-anethole, methyl-eugenol, panisaldehyde, elemol, eugenol, methyl-isoeugenol and myristicin. Significant differences (p<0.05) were found between 2 different roasted temperatures at levels for all volatile compounds.

Acaricidal Components of Medicinal Plant Oils Against Dermatophagoides farinae and Dermatophagoides pteronyssinus

  • The oils of Acorus gramineus, Cinnamomum sieboldii, Eugenia aromatica, and Inula helenium were tested for their acaricidal activity against Dermatophagoides farinae and D. pteronyssinus. Responses varied according to dose and mite species. As compared to the oils, the oil most toxic to D. farinae and D. pteronyssinus was E. aromatica, followed by C. sieboldii, A. gramineus, and I. helenium. On the basis of $LD_{50}$ values of the oils in A. gramineus, C. sieboldii, and E. aromatica, the compound most toxic against D. farinae and D. pteronyssinus was eugenol congeners (isoeugenol>eugenol>acetyleugenol) followed by benzyl benzoate, salicylaldehyde, safro1, DEET, cinnamyl alcohol, and 3-carene. As a naturally occurring acaricide, these oils and eugenol congeners could be useful as new acaricidal agents against Dermatophagoides spp.

식물정유 10 종의 라디칼 소거 활성과 주요 활성 성분의 탐색 (Radical Scavenging Effects of 10 Plant Essential Oils and Active Compound Screening Analysis)

  • Background: A growing interest in health has increased the need for the development of potent antioxidant materials known to play a role in various physiological activities. Currently research and development of non-toxic natural antioxidants with high activity is ongoing. Methods and Results: In this study, we measured 2,2'-azinobis- (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging ability of 10 plant essential oils, selecting samples of Dendranthema indicum, Dendranthema zawadskii, and Citrus sunki essential oils. The samples were analyzed using liquid chromatography (LC) and the radical scavenging activity on LC-based systems with the same conditions. In the LC-mass spectroscopy (MS)/MS analysis of the active compound peak, 2-methoxy-4-vinylphenol with a molecular weight of 150.1 g/mol was identified in C. sunki essential oils. Eugenol or isoeugenol with a molecular weight of 164.1 g/mol as identified in D. indicum and D. zawadskii essential oils as radical scavenging active compounds. Conclusions: In the LC-based measurement system, the active ingredient can be identified by simultaneously conducting profile analysis and the radical scavenging activity of essential oil samples. In addition, LC-MS/MS analysis of the active compound peaks can be performed under the same separation conditions to obtain data that can identify the active compounds in the sample.

Three Predictive Tests Using Mice for the Identification of Contact Sensitizer

  • Predictive tests for the identification of contact sensitizing chemicals have been developed. We measured the sensitization potential with three predictive tests, the in vitro and the in vivo Local Lymph Node Assay(LLNA), ELISA to detect interferon-gamma(IFN-${\gamma}$) from supernatant and flow cytometry to detect change of cell surface proteins, using draining lymph nodes of mice. BALB/c mice were exposed to various chemicals or vehicles on the ears daily for 3 consecutive days in all experiments. With some exceptions of propyl paraben, neomycin sulfate, the in vivo LLNA was able to detect the sensitizing capacity of test chemicals and was more sensitive than the in vitro LLNA for chemicals used in the present study. In another experiment, contact sensitivity was assessed by the ELISA to detect IFN-Υ from the supernatants of the cultured LNCs after sensitization with chemicals. There was a good correlation between the LLNA and the IFN-Υ production for test chemicals. We also examined the change of cell surface proteins on LNCs after sensitization by flow cytometry for some cell adhesion molecules(ICAM-1, E-cadherine, B7 molecule), T cell markers(CD3, CD4, CD8, T$\alpha$$\beta$,T${\gamma}$$\delta$) and B cell markers(LR1, CD45R, I-Ad). The number of ICAM-1 positive cells and B cells in LNCs were increased after sensitization with DNCB, TNCB, isoeugenol and 25%, 50% cinnamic aldehyde compared with that of vehicle as a control. In conclusion, the in vivo LLNA could provide more sensitive screening test for moderate to strong sensitizers and some weak sensitizers including cosmetic raw materials than the in vitro LLNA. The production of IFN-Υ by allergen-activated LNCs might be a values indicators without radioisotopes for the identification of contact allergens. Detection of allergens by testing the increase of ICAM-1 positive cells and B cells in LNCs by flow cytometry might be used as a test method to detect allergens.

꾸지 뽕나무 휘발성 향기성분의 항산화활성 (Antioxidant Activities of Volatile Aroma Components from Cudrania tricuspidata (Carr.) Bureau Extracts)

  • 꾸지 뽕나무(C. tricuspidata)의 휘발성 향기추출물의 항산화활성을 2 가지의 시험방법을 이용하여 측정하였다. 그리고 꾸지 뽕나무의 향기추출물에서 동정된 10종의 휘발성 향기성분에 대한 항산화활성도 측정하였다. 꾸지 뽕나무의 줄기 및 뿌리 향기추출물들은 2가지 실험법에서 모두 뚜렷하게 처리농도에 비례하여 항산화활성을 나타내었다. 즉 aldehyde/carboxylic acid assay에서 $500{\mu}g/mL$의 농도로 처리하였을 때, 줄기와 뿌리의 향기추출물은 각각 $77.02{\pm}8.12%$$74.19{\pm}6.82%$의 항산화활성을 나타내었으며, lipid/malonaldehyde assay에서 $160{\mu}g/mL$의 농도로 처리하였을 때, 줄기 및 뿌리의 향기추출물은 각각 $61.43{\pm}2.11$$76.17{\pm}4.25%$의 항산화활성을 나타내었다. 꾸지 뽕나무의 향기추출물에서 동정된 향기성분들은 7종의 terpenes and terpenoides, 14종의 alkyl compounds, 11종의 nitrogen containing heterocyclic compounds, 3종의 oxygen containing heterocyclic compounds, 12종의 aromatic compounds, 9종의 lactones와 7종의 miscellaneous compounds였다. 꾸지 뽕나무의 향기추출물에서 동정된 향기성분들 중에서 eugenol, isoeugenol 및 2,4-bis(1,1-dimethylethyl)phenol은 lipid/malonaldehyde assay에서 $160{\mu}g/mL$의 농도로 처리하였을 때, 각각 $91.74{\pm}1.33$, $94.00{\pm}0.59$$91.22{\pm}4.74%$의 항산화활성을 나타내었다. 이는 동일한 방법에서 BHT와 ${\alpha}$-tocopherol의 항산화활성이 각각 $91.90{\pm}0.42%$$89.47{\pm}3.04%$인 것과 비슷한 수준이었다. 한편 동일한 처리농도에서 vanillin과 2-acetylpyrrole은 각각 $63.36{\pm}5.83$$58.62{\pm}6.83%$로서 중간 정도의 항산화활성을 나타내었다. 따라서 꾸지 뽕나무는 산화적 손상으로 인한 질병의 예방과 사람들의 건강에 도움을 줄 수 있을 것으로 생각되었다.

$AQUI-S^{\circledR}$의 조피볼락(Sebastes schlegeli)에 대한 진정 및 마취 효과 (Investigation on Sedation and Anesthetic of $AQUI-S^{\circledR}$ for Black Rockfish(Sebastes schlegeli))

  • Along with olive flounder(Paralichthys olivaceus), black rockfish(Sebastes schlegeli) is another very popular maricultured species in Korea. As there is many difficulties in handlling live fish for aquaculturist, use of suitable anesthesia for proper handling of fish is very important in the field. In this view, the effect of $AQUI-S^{\circledR}$ has analysed for its use in the field. $AQUI-S^{\circledR}$, contains 50% isoeugenol, is a new anesthics for fish and zero-withdraw time required since it was approved as a safe additives of food. In the present study, black rockfish adult exhibted sedation effect from 5 ppm at $10^{\circ}C\;and\;15^{\circ}C$, and 7.5 ppm at $20^{\circ}C$, on the other hand, anesthesia was at least required 7.5 ppm at $10^{\circ}C\;and\;15^{\circ}C$, and 10 ppm at $20^{\circ}C$. The fish was recovered from sedation and anesthesia after approximately 5 and 10 minutes, respectively. In case of black rockfish fry, sedation was recorded from 2.5 ppm at $20^{\circ}C$, and 5 ppm at $15^{\circ}C\;and\;20^{\circ}C$. The least concentraion of anesthesia was 2.5 ppm at $10^{\circ}C$, 7.5 ppm at $15^{\circ}C$, and 5 ppm at $20^{\circ}C$. The acute toxic test showed that black rockfish adult and nv showed mortality above 12.5 and 15 ppm concentration of $AQUI-S^{\circledR}$, respectivly. In sum, $AQUI-S^{\circledR}$ is considered as useful anesthetic drug for sedation and anesthesia of black rockfish, it is highly availbale to be prevented from a varity of stress.

생물공학에 기반한 천연 바닐린 생산에 관한 최근 연구 (Recent Advances in the Biotechnological Production of Natural Vanillin)

  • 바닐린은 천연 바닐라의 주요 향미 화합물이며 식품, 음료, 향수, 제약 산업 및 기타 응용 분야에서 광범위하게 사용된다. 바닐린은 화학합성법, 바닐라 꼬투리를 이용한 식물추출법, 천연 전구체를 이용한 생물전환법, 그리고 포도당을 사용한 직접 발효법에 의해 생산될 수 있다. 현재 상업적으로 이용 가능한 대부분의 바닐린은 큐어링 공정을 거쳐 얻어진 바닐라 꼬투리에서 추출하는 방법과 구아이아콜과 글리옥실산을 원료로 사용하여 화학적 합성법에 의해 생산된다. 환경 문제, 건강 준수, 천연 원료에 대한 선호, 천연 바닐라의 제한된 공급 및 치솟는 가격으로 인해 생명 공학 기반 바닐린 생산은 유망한 대안으로 간주된다. 페룰산, 유제놀, 이소유제놀, 리그닌을 포함한 여러 천연 전구체를 대사하고 바닐린을 축적할 수 있는 많은 미생물이 선별되고 평가되면서, 상업적으로 실행 가능한 생산 기술 개발을 위해 많은 노력을 기울였다. 본 총설은 이러한 천연 전구체를 사용하여 천연 바닐린의 생물공학적 생산에 대한 최근의 발전을 간략하게 설명한다. 또한, 포도당에서 바닐린의 새로운 생합성 경로를 기반으로 재생 가능한 탄소원에서 천연 바닐린을 생산하기 위한 최신의 개발 전략과 생산 농도를 높이는 데 발생하는 문제를 극복하기 위한 적절한 해결방안을 소개한다.
