Knockdown of cytosolic NADP+-dependent isocitrate dehydrogenase enhances MPP+-induced oxidative injury in PC12 cells

  • 1-Methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and its toxic metabolite 1-methyl-4-phenylpyridium ion (MPP$^+$) have been shown to induce Parkinson's disease-like symptoms as well as neurotoxicity in humans and animal species. Recently, we reported that maintenance of redox balance and cellular defense against oxidative damage are primary functions of the novel antioxidant enzyme cytosolic NADP$^+$-dependent isocitrate dehydrogenase (IDPc). In this study, we examined the role of IDPc in cellular defense against MPP$^+$-induced oxidative injury using PC12 cells transfected with IDPc small interfering RNA (siRNA). Our results demonstrate that MPP$^+$-mediated disruption of cellular redox status, oxidative damage to cells, and apoptotic cell death were significantly enhanced by knockdown of IDPc.

호염성 세균의 생리적 특성

  • 국내 염장식품 및 염전으로부터 세균을 분리하여, 호염성 세균의 NaCl 농도에 따른 성장범위, 생리적 및 효소학적 특성을 조사하고자 했다. 염전으로부터 NaCl 20%배지에서 14주와 총 16종류의 젓갈류에서 NaCl 10% 배지로 56균주의 호염성 세균을 분리하여 0, 5, 10, 15, 20, 25% NaCl농도에서 성장률을 조사하고 최적온도 및 배지조성과 함께 동정에 필요한 생리실험을 하였다. 또한 세포의 효소로서 Lactate dehydrogenase, Glucokinase, Glucose-6-phosphate dehydrogenase, Alanine dehydrogenase, Isocitrate dehydrogenase 등의 특성도 조사하였다. 선별한 균주중 Acinetobacter sp, 등이 관찰 조사되었으며 최적 성장 NaCl농도는 10%이고, 최적온도는 3$0^{\circ}C$이며, 25% NaCl, 45$^{\circ}C$에서 자란 Halobacterium sp. 등이 분리되었다. 그중 Acinetobacter strain H6는 단백분해효소와 탄수화물 분해효소의 생성능이 15>10>20% NaCl순이며, 특히 Lactate dehydrogenase 활성은 2>3>1>OM NaCl 순으로 나타났고, NaCl 대신 KCl을 사용했을 때는 3>2>1> OM순으로 활성이 나타났다.

Dunaliella tertiolecta에 의한 acetate의 이용 -TCA cycle과 glyoxylate pathway의 활성 조사- (The utilization of acetate for the growth and the respiration in Dunaliella tertiolecta.―Enzymes of the tricarboxylic acid cycle and glyoxylate pathway)

  • The utilization of acetate by Dunaliella tertiolecta was examined, and the detections and assays of the enzymes of the tricarboxylic acid cycle and the glyoxylate pathway were described. Acetate could not be utilized as a sole carbon source for the growth. The carboxyl carbon of acetate was incorporated more rapidly into CO2 than the methyl carbon. It was identified that malate, succinate, citrate and etc., were accumulated whne [U-14C] acetate was supplied to the cell free homogenate. The following enzyme activities were measured; acetothiokinase, isocitrate dehydrogenase, fumarase, malate dehydrogenase and aconitase. Though isocitratase, malate synthetase, succinate dehydrogenase and oxoglutarate dehydrogenase could not be detected, 14C from succinate was easily contributed to CO2 and cell component. The evidence suggested that the glyoxylate pathway was not operative and showed that the TCA cycle was the all important pathway in the oxidation of acetate to CO2 in Dunaliella.

Soluble isocitrate dehydrogenase plays a key role in obesity and hyperlipidemia

  • NADPH is an essential co-factor for fat and cholesterol biosynthesis. However, the role of cytosolic NADP$\^$+/-dependent isocitrate dehydrogenase (IDPc), a putative NADPH producer, in the control of the fat and cholesterol metabolism has not been assessed. Here we report that increased or decreased IDPc expression in 3T3-Ll fat cells promoted or retarded adipogenesis, respectively. Furthermore, overexpression of IDPc in transgenic mice exhibited fatty liver, hypertriglyceridemia, hypercholesterolemia and obesity by increasing NADPH production leading to subsequent stimulation of acetyl-coenzyme A and malonyl-coenzyme A consumption. In contrast, administrations of a synthetic IDPc inhibitor, DAl1004, to ob/ob mice effectively reduced body weight with lowering cholesterol and triglyceride levels. In addition, a positive relationship (${\gamma}$ = 0.69, $\rho$<0.0l) between plasma IDPc activity and body mass indexes was observed in 98 randomly-selected human volunteers. Our findings strongly indicate that NADPH produced by IDPc plays an important role in controlling body fat and lipid biosynthesis.

Halobacterium sp. EH10 NADH dehydrogenase의 $Na^+$ 요구성 ($Na^+$ Requirement of NADH dehydrogenase from an Extreme halophile, Halobacterium sp. EH10 Isolated from a Saltern in Korea)

  • Intracellular enzymes of an extreme halophilic bacterium, Halobacterium sp. HE10, isolated from a saltern in Korea was investigated. The membrane-bound enzyme, NADH dehydrogenase, involved in electron transport system was stimulated by the addition of 2.0 M NaCl. The respiratory enzyme activities such as NADH oxidase and NADH dehydrogenase was decreased on removal of $Na^+$ ion and restored when replaced with cations like $K^+$, $Li^+$and $NH_{4}^{+}$ ions. Furthermore, their activities were affected by the anions such like carbonate, acetate, sulfate, chloride and nitrate at the presence of $Na^+$ion. Lactate dehydrogenase activity was highest at the asturated solution of NaCl and isocitrate dehydrogenase activity was a maximum level at 1.0 M NaCl. These results suggested that the enzyme activites of the respiratory chain in Halobacterium sp. EH10 was stimulated by the presence of $Na^+$ ion.

  • The presence of considerable amount of enzymes of TCA cycle isocitrate dehydrogenase (ICDH-NADP+, EC1.1.1.42), $\alpha$-ketogluterate dehydrogenase ($\alpha$-KGD, EC1.2.4.2) and malate dehydrogenase (MDH, EC1.1.1.37) in fresh control and in vitro starved adult Isoparorchis hypselobagri establish the functional TCA cycle in this fluke. The major metabolic end products are pyruvate, lactate, oxaloacetate and malate. The ratio of oxaloacetate/malate assess that oxaloacetate is reduced to malate and in this fluke the reverse TCA cycle is active. The pyruvate/lactate ratio shows pyruvate is reduced to lactate and the fluke is homolactate farmenters.

왕대속 3종의 지역개체군간 동위효소변이 (Isozymic Variation of Local Populations in the 3 Species of Phyllostachys)

  • Intraspecific and interspecific isozyme variations and their relationship of 16 local populations in 3 species of Phyllostachys, that is, P. bambusoides, P. nigra var. henonis and P. pubescens were investigated by multi-variate analysis. Leaf isozymes of Phyllostachys such as 6-PGD (6-phosphogluconate dehydrogenase), MDH (malate dehydrogenase), PGI (phosphoglucoisomerase), PRX (peroxidase), PGM (phosphoglutamase), IDH (isocitrate dehydrogenase) showed electrophoretic variations in the number of zymotypes (7, 6, 6, 9, 3 and 5, respectively). In the cluster analysis on the isozymic characteristics, 16 populations were classified into 3 species at the euclid genetic distance of 2.041. P. nigra var. henonis and P. bambusoides were clustered first at 2.813 and then P. pubescens at 3.001. So far, 3 local types of intraspecific ariation were found in P. nigra var. henonis and P. bambusoides.

Isolation and Physiological Characterization of Bacillus clausii SKAL-16 Isolated from Wastewater

  • An alkaliphilic bacterium, Bacillus clausii SKAL-16, was isolated from soil that had been contaminated with vegetable oil. The optimal pH and general pH range for bacterial growth was 8, and 7 to 10, respectively. The bacterium could grow on tributyrin and glycerol, but could not grow on acetate and butyrate. The SKAL-16 strain excreted butyric acid during growth on tributyrin, and selectively ingested glycerol during growth on a mixture of butyric acid and glycerol. The SKAL-16 generated intracellular lipase, but did not produce esterase and extracellular lipase. The DNA fragment amplified with the chromosomal DNA of SKAL-16 and primers designed on the basis of the esterase-coding gene of Bacillus clausii KSM-KI6 was not identical with the esterase-coding gene contained in the GenBank database. Pyruvate dehydrogenase, isocitrate dehydrogenase, and malate dehydrogenase activities were detected in the cell-free extract (crude enzyme).

Aedes aegypti L. 난성숙과정중 생성되는 Mitochondrial Malate Dehydrogenase의 정제 및 특성 (Purification and Characterization of Mitochondrial Malate Dehydrogenase during Ovarian Development in Aedes aegypti L.)

  • 암컷 Aedes aegypti의 난성숙과장에서 새로 나타나는 malate dehydrogenase(L-malate, $NAD^+$ oxidoreductase, EC 1.11.37, MDH)를 DEAE-Sepharose, Sulphonyl-Sepharose, Cibacron 3FGA affinity chromatography를 이용하여 분리정제하여 그 특성을 조사하였다. 분자량은 70,000 dalton정도의 dimer 형태로 되어 있으며 최적 pH는 malate-oxaloacetate반응에서는 pH 9.0~9.2, oxaloacetate-malate 반응에서는 pH 9.8~10.2이었다. 정재된 MDH는 mitochondria에 위치하고 있으며 기질로서 malate에 대한 Km값의 경우 $1.29 \times 10^{-4}$ M, oxaloacetate에 대한 Km 값은 $6.58\times 10^{-4}$M, NAD에 대한 Km값은 $0.76\times 10^{-3}$ M이며 NADH에 대한 Km 값은 $3.8\times 10^{-3}$ M 을 보이고 있으며 각각의 기질에 의한 저해현상을 보이고 있었다. 기질에 대한 Km값을 부분적으로 분리한 DEAE-sepharose에 흡착된 원형질 MDH와 비교한 결과 malate에 대한 Km 이 $8.92\times 10^{-3}$으로 상당한 차이를 보이고 있었다. 또한 정제된 MDH는 cltrate, $\alpha$-ketoglutarate, ATP 등의 대사산물에 의하여 저해작용을 받았다. ATP 및 citrate에 의한 MDH 활성도 저해는 oxaloacetate-malate반응에서 보다는 malate-oxaloacetate 반응에서 덜 일어났다. Oxaloacetate-malate 반응의 경우 ATP에 의하여저해작용이 완전히 일어났으며 malate-oxaloacetate반응에서는 cltrate에 의하여 저해작용이 일어나지 않았다. 흡혈 후 생성되는 MDH는 난소에서 합성되며 흡혈 수 난소에서 18시간 때부터 활성도가 나타나 48시간 이후 최고 활성도가 유지되는데 TCA회로의 isocitrate dehydrogenase 의 경우 난소내에서의 활성도 변화가 MDH의 변화 양상과 같았다.

한국산 논우렁이과 ( Family Viviparidae ) 2종에서의 동위효소 변이 (Isozyme Variability in Two Species of Freshwater Viviparid Snails in Korea : Cipangopaludina chinensis malleata and C. Japonica)

  • A horizontal starch gel electrophoresis for enzyme proteins extracted from 2 species of Korean viviparid snails; Cipangopaludina chinensis malleata and C. japonica was carried out in order to elucidate their genetic relationships. A total of 10 enzymes were employed in three different kinds of buffer systems. Two loci from each enzyme of alcohol dehydrogenase, esterase, glucose phosphate isomerase, isocitrate dehydrogenase, iditol dehydrogenase, malate dehydrogenase and peptidase(VL); and only one locus dach from two enzymes, glycerlo-3-phosphate dehydrogenase and phosphoglucomutase were detected; but, four loci from peptidase(LGG) were observed. Most of loci in two viviparid species showed homozygous monomorphic banding patterns and some of them were specific as genetic markers between two different species. However, EST-1, MDH-1, PEP(VL)-1loci showed polymorphic banding patterns. Foru populations of C. chinensis malleata were more closely clustered in a dendrogram within the range of genetic identity values of 0.928-1.00, and these clusters were lineated with C. japonica at the value of 0.355. In summarizing the above results, two viviparid snail species dmployed in this study mostly showed monomorphic enzyme protein banding patterns, and genetic differences specific between two species.
