Ferroptosis and its role in gastric and colorectal cancers

  • Ferroptosis is a novel mechanism of programmed cell death, characterized by intracellular iron overload, intensified lipid peroxidation, and abnormal accumulation of reactive oxygen species, which ultimately resulting in cell membrane impairment and demise. Research has revealed that cancer cells exhibit a greater demand for iron compared to normal cells, indicating a potential susceptibility of cancer cells to ferroptosis. Stomach and colorectal cancers are common gastrointestinal malignancies, and their elevated occurrence and mortality rates render them a global health concern. Despite significant advancements in medical treatments, certain unfavorable consequences and drug resistance persist. Consequently, directing attention towards the phenomenon of ferroptosis in gastric and colorectal cancers holds promise for enhancing therapeutic efficacy. This review aims to elucidate the intricate cellular metabolism associated with ferroptosis, encompassing lipid and amino acid metabolism, as well as iron metabolic processes. Furthermore, the significance of ferroptosis in the context of gastric and colorectal cancer is thoroughly examined and discussed.

재즈댄스와 철분섭취가 성인여성의 신체 조성, 혈액 성분 및 골 대사에 미치는 영향 (Effect of Jazz Dance and Iron Intake on Body Composition, Blood Components and Bone Metabolism in Adult Women)

  • 본 연구는 성인여성을 대상으로, 재즈댄스+철분섭취군(7명), 재즈댄스군(7명), 대조군(7명) 총 21명으로 하였다. 재즈댄스와 철분섭취가 성인여성의 신체조성, 혈액성분 및 골 대사에 미치는 영향을 구명하기 위하여 운동 강도 RPE (9$\sim$15), 운동 빈도 주3회, 1회 60분의 지속시간으로 12주간 실시하였다. 철분은 1일 1회 식후 80 mg을 섭취시켜, 비교 분석한 결과 다음과 같은 결론을 얻었다.신체조성은 집단내 변화에서 재즈댄스+철분섭취군은 체중, 체지방량은 유의하게 감소하였고, 제지방은 유의하게 증가 하였으며, 재즈댄스군에서 체지방량이 유의하게 감소한 것으로 나타났다. 집단 간 변화에서 대조군보다 재즈댄스+철분섭취군과 재즈댄스군이 유의한 차이로 긍정적인 변화가 있는 것으로 나타났다.혈액성분은 집단 내 변화에서 재즈댄스+철분섭취군은 RBC, WBC, Hb는 유의하게 증가 하였고, MCV는 유의하게 감소하였으며, 재즈댄스군은 WBC, MCHC가 유의하게 증가하였다. 대조군은 MCHC 유의하게 감소를 하였다. 집단 간 변화에서 RBC는 재즈댄스+철분섭취군이 대조군보다 유의한 차이가 있었고, WBC, MCV, MCHC, Hb는 재즈댄스+철분섭취군, 재즈댄스군이 대조군보다 유의한 차이가 있었다. 골대사는 집단 내 변화에서 재즈댄스+철분섭취군, 재즈댄스군은 DPD, OC는 유의하게 증가하였고 대조군은 유의하게 감소하였다. 집단간 변화에서 대조군보다 재즈댄스+철분섭취군과 재즈댄스군이 유의하게 증가하였다. 이상의 연구 결과를 통해 재즈댄스가 성인여성들에게 여가시간과 건강에 도움이 되는 댄스프로그램으로서 바람직하고, 다양한 연령층에서 신체기능을 향상시키는 과학적인 연구가 필요하며, 보다 효율적이고 규칙적인 운동프로그램 개발과 영양섭취에 대한 지속적인 연구가 요구된다.

E3 ligase BRUTUS Is a Negative Regulator for the Cellular Energy Level and the Expression of Energy Metabolism-Related Genes Encoded by Two Organellar Genomes in Leaf Tissues

  • E3 ligase BRUTUS (BTS), a putative iron sensor, is expressed in both root and shoot tissues in seedlings of Arabidopsis thaliana. The role of BTS in root tissues has been well established. However, its role in shoot tissues has been scarcely studied. Comparative transcriptome analysis with shoot and root tissues revealed that BTS is involved in regulating energy metabolism by modulating expression of mitochondrial and chloroplast genes in shoot tissues. Moreover, in shoot tissues of bts-1 plants, levels of ADP and ATP and the ratio of ADP/ATP were greatly increased with a concomitant decrease in levels of soluble sugar and starch. The decreased starch level in bts-1 shoot tissues was restored to the level of shoot tissues of wild-type plants upon vanadate treatment. Through this study, we expand the role of BTS to regulation of energy metabolism in the shoot in addition to its role of iron deficiency response in roots.

장기간의 철분 부족 식이가 카드뮴중독된 흰쥐의 철분대사에 미치는 영향 (The Effect of Dietary Low Iron Levels on the Metabolism of Iron in Long Termm Cadmium Poisoned Rats)

  • This study was undertaken to investigate the effects of dietary low (Fe) levels on Fe metabolism of cadmium(Cd) poisoned rats. 40 male Sprague weaning Dawley rats weighing 80-90g were divided into 4 groups(LFe:low Fe, LFeCd:low Fe and Cd, AFe: adequate Fe, AFeCd: adequate Fe and Cd) according to Cd administration(0, 50ppm in drinking water) and Fe levels(Fe:6ppm, 40ppm in diet)for 12 weeks. The food intake and body weight gain of Cd group with low Fe(LFeCd) were significantly lower than those of without Cd group with adequate Fe(AFe)(p<0.01, P<0.05). But there was no significantly difference between Cd groups and without Cd groups in water intake. The blood levels of hemoglobin, hematocrit, and serum levels of Fe of LFeCd were significantly lower than those of AFe(p<0.01, p<0.05, p<0.001). The urinary and fecal excretion of Fe of LFeCd was significantly lower than that of AFe(p<0.05, p0.05). The levels of Fe of liver, spleen in LFeCd were lower than those of AFeCd(p<0.05, p<0.05). These results indicates that adequate iron supplementation to Cd pretreated rats induce protective effects on the reduction of Fe status by Cd poisoning.

일부 여대생의 식이섬유 섭취와 철분대사에 관한 연구 (A Study on the Dietary Fiber Intake and Iron Metabolism in Korean Female College Students)

  • This study was conducted to evaluate dietary fiber intake, iron status, and their correlations in 50 female college students living in Seoul. The nutritional status was analyzed using 3-day dietary record, duplicated diet collection, and venous blood sampling. The mean values for age, height, weight, BMI, and blood pressure of the subjects were 23.2 years, 160.2cm, 53.9kg, 21.0kg/$m^2$, and 110.1/68.4mmHg, respectively. Daily intakes of enery, protein, fiber, crude fiber, iron, and heme iron were 1635.5㎉, 54.3g, 22.5g, 6.8g, 16.2mg, and 0.2mg, respectively. Fiver intake was positively correlate with energy, protein, carbohydrate, vitamin C, iron, and crude fiber intakes. Also, iron intake was positively correlated with energy, protein, lipid, carbohydrate, and vitamin C intakes. There was a significant correlation between heme iron and MFP(meat, fish, poultry) intakes. To exame the iron balance, iron intake and excretion were measured. Iron intake and excretions through urine and feces were 19.5mg, 8.5mg, and 1.6mg, respectively. Based on these iron retention and iron apparent absorbability were calculated as 9.4mg and 52.4%, respectively. There was no significant relationship between dietary fiber intake and iron status. However, there were significant positive correlation between fiber intake and urinary iron excretion.

Mass spectrometry-based approaches to explore metabolism regulating ferroptosis

  • Ferroptosis is a type of programmed cell death distinct from apoptosis or necroptosis. Ferroptosis is well characterized by an iron-dependent accumulation of lipid peroxides and disruption of cellular membrane integrity. Many metabolic alterations can prevent or accelerate ferroptosis induction. Recent advances in analytical techniques of mass spectrometry have allowed high-throughput analysis of metabolites known to be critical for understanding ferroptosis regulatory metabolism. In this review, we introduce mass spectrometry-based analytical methods contributing to recent discovery of various metabolic pathways regulating ferroptosis, focusing on cysteine metabolism, antioxidant metabolism, and poly-unsaturated fatty acid metabolism.

Dietary Iron Intake and Body Iron Status of Myocardial Infarction Patients in Chunan Area

  • It has been known for some time that elevated body iron could be a risk factor for coronary heart disease. The present study was conducted to determine body iron status and dietary iron intake of patients with myocardial infarction(MI). Seventy five patients from the Chunam area with their first MI history within he past 2 months were recruited. The serum iron concentration, total iron binding capacity(TIBC) and percent transferrin saturation(TS) were selected as indicators of body iron status. Twenty four hour recall was conducted by trained interviewers to asses the dietary intake. Most women (91.3%) showed waist to hip ratio(W/H) greater than 0.85 while 17.3% of men were assessed to have a tendency of abdominal obesity(W/H>0.95). The average BMI of women was 25.80 and that of men was 23.98. The average diet intake of participants was below the recommended dietary allowances (RDA) for most nutrients. He average dietary iron intake was 10.03 mg/day for all subjects while women's iron intake was significantly lower than men's. However, a great proportion of participants (77%) showed a tendency to have normal iron status. About 9% of the participants were assessed as iron deficient and 14% had an iron overload. The mean serum iron concentration was 125 g/dl ranging from 13.3 to 280.6 g/dl. Iron intake from animal sources were significantly associated with body iron status (r=0.257, p=0.026) when TIBC was used as an iron status indicator. When iron status was assessed with TS, it was directly associated with iron intake from animal sources(r=0.278, p=0.05) for he subjects in the normal iron status group. He results of the present study showed that the nutrient intake of Mi patients in Chunan was not quite adequate while iron status was mostly in the normal range. Further studies are needed to investigated whether there is a possible difference in iron metabolism of the MI patients.
