Biological Function of Lactoferrin in Milk

  • Lactoferrin is an iron-binding glycoprotein and its bacteriostatic and bactericidal effects on Gram-positive and Gram-negative bacteria have been well-known. However, certain kind of lactic acid bacteria are resistant against its antibacterial effects. Moreover, it is reported that lactoferrin promotes the growth of bifidobacteria by in vitro and in vivo experiments. In this experiment, lactoferrin-binding protein was found both in the membrane and cytosolic franctions of Bifidobacterium. Bifidobacterium was grown in anaerobic conditions in MRS broth containing cysteine, gathered by centrifugation and processed by sonication. The lactoferrin-binding proteins on the PVDF-membrane transferred after SDS-PAGE were detected by far-western method using biotinylated lactoferrin and streptavidin-labeled horse radish peroxidase. Observation in growth effects of lactoferrin on Bifidobacterium suggested that there is a relation between the presence of lactoferrin-binding proteins on the cells and their growth.

The Nutritional Status of Iron, Magnesium and the Effects of Iron Supplementation on Serum Iron and Magnesium Concentrations of Pregnant Korean Women (임신부의 일상 식이 중 철, 마그네슘 영양상태와 철 보충제의 복용이 혈청 철, 마그네슘의 함량에 미치는 영향)

  • The purpose of this study was to assess the nutritional status of iron and magnesium and the effect of von supplementation during 8 weeks(from 20 to 28 weeks of gestation) on serum iron and magnesium status of 31 pregnant women in Kyunsin area. The age, weight, and height of the subjects before pregnancy were investigated by questionnaires. At 20 and 28 weeks of gestation, data for food and nutrient intake were obtained by questionnaires and serum sample was obtained. According to the frequency of iron supplementation, subjects were divided into two groups(4$\geq$/wk) The mean iron supplementation of 5 $\geq$/wk group(63.mg/day) was significantly higher than 4$_2$ intakes were much less than the Korean RDA. Especially, calcium, iron and magnesium intakes showed half the levels compared with Korean RDA. At 28 weeks of gestation the serum total protein(p<0.001), albumin(p<0.01) and globulin(p<0.001) were significantly decreased. Total iron binding capacity(TIBC) was significantly increased(P<0.001), but serum ferritin(p<0.01) and magnesium(p<0.01) concentrations were significantly decreased. However, even 28 weeks of gestation 5$\geq$/wk group showed higher serum iron and ferritin concentration and lower TIBC than 4$\geq$/wk group. Therefore, iron supplementation should be conducted with concerning the mineral balance like magnesium.

Effect of Pine Needle Extract on Fe ion and Active Oxygen Related Lipid Oxidation in Oil Emulsion (Fe ion과 활성산소 관련 지방산화반응에 미치는 솔잎 추출물의 영향)

  • This study was carried out to investigate the effect of Pine needle extract on lipid oxidation and free radical reaction in iron sources reacted with active oxygen species. The results were summarized as follow; The pine needle extracts didn`t show a distinct effect on reduction of lipid oxidation if the iron ion didn`t exist in oil emulsion. The pine needle extracts played role as a strong chelating agents to bind iron ion if Ferrous iron(Fe\ulcorner) exist in oil emulsion. Ferric iron(Fe) was lower effect than Ferrous iron(Fe) on free radical reaction in oil emulsion. And also, the Fe\ulcorner reacted with pine needle extract did not show distinct effect on free radical reaction, compared to Fe\ulcorner reacted with pine needle extract. And also, Pine needle extracts reacted with H\ulcornerO\ulcorner were tended to show a low oxygen scavenging ability in case of H\ulcornerO\ulcorner only was existed, compared to those of H\ulcornerO\ulcorner + Fe\ulcorner complex. Pine needle extracts were the most powerful Fe\ulcorner binding agents, compared to other strong synthetic antioxidants such as EDTA and DTPA.

Nutritional Iron Status in Pre - and Postmenopause Middle - Aged Women in Kangnung Area (일부 폐경 전 , 후 중년 여성의 철분 영양상태에 관한 연구 - 강릉지역을 중심으로)

  • The purpose of this study were to assess iron status and obesity in 82 middle aged women living in Kangnung area. Anthropometric measurements were taken for body weight, height, percentage of body fat and circumferences of waist and hip. Venous blood samples were drawn from subjects for measurement of hemoglobin(Hb), hematocrit(Hct), serum iron(Fe), total iron binding capacity(TIBC), transferrin saturation(TS) and serum ferritin. Dietary intakes of iron(heme iron and nonheme iron), the amounts of MPF(meat, poultry and fish) and ascorbic acid were assessed by modified 24-hr recall method. The results obtained are summarized as follows : Postmenopausal women had more body fat than premenopausal women. That is, postmenopausal women tend to be obeser than premenopausal women. There was no difference in Hb, Hct, Fe, TIBC and TS between pre- and postmenopausal women. But the serum ferritin concentration of postmenopausal women(83.7$\pm$42.1ng/ml) was significantly (p<0.05) higher than premenopausal women(56.4$\pm$41.0ng/ml). Prevalences of iron deficiency (20%, 20.0% and 17.1% respectively) of postmenopausal women. The mean daily intakes of total iron in pre- and postmenopausal women were 17.5$\pm$9.3mg and 15.6$\pm$6.9mg, respectively. Bioavailabilities of dietary iron were 6.5% and 4.5% in pre- and post-groups. These results indicate that individual dietary guidelines should be used to educate middle-aged women different in status of menopause. For example, premenopausal women should increase nutritional iron status and postmenopausal women should try to prevent obesity.

  • Porphyromonas gingivalis, a Gram negative, anaerobic, asaccharolytic rod, is one of the most frequently implicated pathogens in human periodontal disease and has a requirement for hemin for growth. A 30 kDa (heated 24 kDa) hemin-binding protein whose expression is both hemin and iron regulated has recently been purified and characterized in this oral pathogen. This study has identified a hemin-binding P. gingivalis protein by expression of a P. gingivalis genomic library in Escherichia coli, a bacterium which does not require or transport exogenous hemin. A library of genomic DNA fragments from P. gingivalis was constructed in plasmid pUC18, transformed into Escherichia coli strain $DH5{\alpha}$ , and screened for recombinant clones with hemin-binding activity by plating onto hemin-containing agar. Of approximately 10,000 recombinant E. coli colonies screened on LB-amp-hemin agar, 10 exhibited a clearly pigmented phenotype. Each clone contained various insert DNA. The Hind III fragment transferred to the T7 RNA polymerase/promoter expression vector system produced a sligltly smaller (21 kDa) protein, a precursor form, immunoreactive to the antibody against the 24 kDa protein, suggesting that the cloned DNA fragment probably carried an entire gene for the 24 kDa hemin-binding protein.

Elucidation of Antioxidant Activity of Phosvitin Extracted from Egg Yolk using Ground Meat

  • Phosvitin was extracted from a chicken egg yolk and the iron-binding, along with antioxidative activity of the extracted phosvitin, was determined after mixing with ground beef at the concentrations of 100 and 500 mg/kg of meat. The electrophoretic pattern of the extracted phosvitin on SDS-PAGE was found to be identical to that of the standard phosvitin. The extracted phosvitin at $1,000{\mu}g$/mL showed an ability to bind approximately 65% of the iron in a 3 mM iron solution. Lipid oxidation was inhibited in the ground beef mixed with 500 mg/kg of the extracted phosvitin, during storage at $4^{\circ}C$ compared to that of the control (p<0.05). Additionally, color stability of ground beef containing the extracted phosvitin was enhanced (p<0.05). The pH, cooking loss, texture, and sensory properties of the ground beef were not affected, by adding up to 500 mg/kg of the extracted phosvitin. This result suggests that the phosvitin extracted from egg yolk could be used as an antioxidant reagent. In particular, phosvitin would be more amenable for use in meat products because it is a natural protein derived from animal products.

Binding of Calcium and Iron by Job′s tears (Yulmoo: Coix lachryma-jobi L. var. Ma-yuen Stapf.) Total Dietary Fiber and Acid Detergent Fiber (율무 식이섬유의 칼슘, 철분과의 결합특성)

  • This study was performed to examine the mineral binding properties of total dietary fiber (TDF) and acid detergent fiber (ADF) from Job's tears (Yulmoo: Coix lachryma-jobi L. var. Ma-yuen Stapf.) bran. The endogenous Ca and Fe contents of dietary fiber extracted from Yulmoo bran were measured by atomic absorption spectrophotometer (AAS). Ca contents were 40, 118.6$\mu\textrm{g}$ Ca/g TDF and 131.6 $\mu\textrm{g}$ Ca/g ADF. Fe contents were 1,219.7 $\mu\textrm{g}$ Fe/g TDF and 277.5 $\mu\textrm{g}$ Fe/g ADF. The calcium binding capacities of dietary fiber at pH 5.0, 6.0, 7.0 were 50.77, 77.57, 86.64% for TDF, 40.27, 51.42, 40.90% for ADF, respectively. The iron binding capacity of dietary fiber at pH 5.0~7.0 was 95~97% for TDF and 85~95% for ADF.

Effects of Custom Nutrition Education on Dietary Intakes and Clinical Parameters in Patients Diagnosed with Iron Deficiency Anemia (맞춤형 영양교육이 철결핍성 빈혈환자의 임상 지표 및 영양섭취 상태에 미치는 효과)

  • This study examined the effects of custom nutrition education on dietary intakes and clinical parameters in patients diagnosed with iron deficiency anemia. A total of 34 patients visited the anemia clinic of Yeouido St. Mary's Hospital. Among these, only 16 patients were available for follow-ups. A follow-up was conducted by a clinical dietitian 2 months from the first nutrition education session. Patients were all women. For custom nutrition education, we investigated anthropometric data, dietary assessment (24 hr-recall, FFQ), and self-recognized anemic symptoms. Weight did not show a significant difference but hemoglobin, hematocrit (P<0.01), serum iron, and serum ferritin (P<0.05) were significantly increased after the nutrition education. Serum total iron binding capacity was significantly decreased (P<0.01). Self-recognized symptoms such as dizziness, fatigue (P<0.001), shortness of breath, headache (P<0.01), brittle nails, and sore tongue (P<0.05) were significantly improved. Daily intakes of protein (P<0.05), total iron (P<0.01), and animal iron (P<0.001) were significantly increased. A significantly negative correlation was observed between current serum iron and the intake of carbohydrates, fat, or phosphorus (P<0.05). But current serum ferritin showed a significantly positive correlation with the frequency of intake of meat, poultry, and fish. It could be concluded that the custom nutrition education might be effective on quality of diet as well as iron status and it might also improve the clinical parameters in patients diagnosed with the iron deficiency anemia.

The usefulness of soluble transferrin receptor in the diagnosis and treatment of iron deficiency anemia in children

  • Purpose: Soluble transferrin receptor (sTfR) is a truncated extracellular form of the membrane transferrin receptor produced by proteolysis. Concentrations of serum sTfR are related to iron status and erythropoiesis in the body. We investigated whether serum sTfR levels can aid in diagnosis and treatment of iron deficiency anemia (IDA) in children. Methods: Ninety-eight patients with IDA were enrolled and were classified according to age at diagnosis. Group 1 comprised 78 children, aged 6-59 months, and group 2 comprised 20 adolescents, aged 12-16 years. Results: In group 1, patients' serum sTfR levels correlated negatively with mean corpuscular volume; hemoglobin (Hb), ferritin, and serum iron levels; and transferrin saturation and positively with total iron binding capacity (TIBC) and red cell distribution width. In group 2, patients' serum sTfR levels did not correlate with ferritin levels and TIBC, but had a significant relationship with other iron indices. Hb and serum sTfR levels had a significant inverse relationship in both groups; however, in group 1, there was no correlation between Hb and serum ferritin levels. In 30 patients of group 1, serum sTfR levels were significantly decreased with an increase in Hb levels after iron supplementation for 1 month. Conclusion: Serum sTfR levels significantly correlated with other diagnostic iron parameters of IDA and inversely correlated with an increase in Hb levels following iron supplementation. Therefore, serum sTfR levels can be a useful marker for the diagnosis and treatment of IDA in children.

Helicobacter pylori Infection is a Risk Factor for Iron-Deficiency Anemia in Korean High School Girls

  • This study investigated the relationship of Helicobacter pylori (H. pylori) infection to iron-deficiency anemia in high school girls. Four hundred and fourty-five subjects resided in Ulsan City in Korea were evaluated by questionnaire or blood analysis for serum indicators of iron status, daily nutrient intakes, symptoms of anemia, and H. pylori IgG antibody status. In H. pylori infected subjects, total energy intake of was 1534.14±350.81 kcal (73.0% of the Korean RDA), the iron intake of subjects was 11.38±3.90 mg (71.1% of Korean RDA) and calcium intake was 467.63±175.96 mg (58.3% of the Korean RDA). Carbohydrate (p < 0.05), vitamin A (p < 0.05), vitamin C (p < 0.01) intakes were significantly lower in infected subjects than in non-infected subjects. The prevalence of anemia (hemoglobin (Hb) < 12 g/dL.) in H. pylori infected subjects was 22.7% as compared to 14.7% in non-infected subjects. The mean serum ferritin of infected subjects (21.71±21.50 ng/mL) was significantly lower than in non-infected subjects (p < 0.05) (as was the mean Hb concentration (12.54±1.51 g/dL) and hematocrit (37.99±3.64%)). Total iron binding capacity (TIBC) of H. pylori infected subjects (449.09±78.23 ㎍/dL) was significantly higher than that of non-infected subjects (432.99+76.15 ㎍/dL) (P < 0.05). The levels of iron, red blood cell (RBC) and transferrin saturation were similar in infected and non-infected subjects. Two symptoms of anemia, 'pale face (p < 0.01)' and 'decreased ability to concentrate (p < 0.005)', were significantly higher in H. pylori infected subjects than in non-infected subjects. High school girls are known to be more vulnerable to iron deficiency than other age groups, and this research demonstrates that infection with H. pylori increases their risk of iron-deficiency anemia.
